자원식물의 기능성 정유성분 이용 고찰 (Review of Functional Volatile Component in Essential Oil of Medicinal and Aromatic Plants)

  • The number of natural products obtained from plants has now reached over 100,000 and new chemical compounds are being discovered ever year. Medicinal and Aromatic plants and their extracts have been used for centuries to relieve pain, aid healing, kill bacteria and insects are important as the antifungal and anti-herbivore agents with further compounds being involved in the symbiotic associations. Although their functions in plants have not been fully established, it is Known that some substances have growth regulatory properties while others are involved in pollination and seed dispersal. The complex nature of these chemicals are usually produced in various types of secretory structures which is an important character of a plant family and also influenced and controlled by genetic and ecological factors. Detailed anatomical description of these structures ave relevant to the market value of the plants, the verification of authenticity of a given species and for the detection of substitution or adulteration. Volatile oils are used for their therapeutic action for flavoring of lemon, in perfumery of rose or as starting materials for the synthesis of other compounds of turpentine. For therapeutic purposes they are administered as inhalations of eucalyptus oil, peppermint oil, as gargles and mouthwashes of thymol and transdermally many essential oils including those of lavender, etc. With these current trend for using volatile components in essential oil will be increasing in the future in Korea and in the world as well.

훈연공정이 훈연액 중의 다환방향족 탄화수소의 함량에 미치는 영향 (Effect of Smoking Process on the Contents of Polycyclic Aromatic Hydrocarbons in Smoke Flavouring)

  • The contents of holocellulose, one of the main components of the wood, were 83.9% in oak wood and 76.9% in apple wood, respectively. Those of hemicellulose were 16.41 and 20.33%, and in lignin 23.0 and 19.7%, respectively. Six species of domestic oak wood and apple wood were considered to be suitable for smoking materials due to the low content of lignin. Benzo(a)pyrene contents in smoke flavoring prepared with oak wood at 150, 400 and 500$^{\circ}C$ were 0. 4, 3. 7 and 5.6$\mu\textrm{g}$,/kg, respectively. The amounts of phenanthrene were 112.7, 131.4 and 255.9$\mu\textrm{g}$/kg, respectively, in each temperature. The amounts of polycyclic aromatic hydrocarbons(PAH) in smoke flavory were in the order of phenanthrene>anthracene>pyrene>benzo(a)anthracene>chrysene>benzo(b)fluoranthens>benzo(a)pyrene Benzo(a)pyrene contents in smoking extracts prepared with apple wood at 150, 400 and 500$^{\circ}C$ were 0.4, 3.3 and 5.5$\mu\textrm{g}$/kg, respectively. Phenanthrene contents in those samples were 72.7, 100.2 and 220.5$\mu\textrm{g}$/kg, respectively. Contents of each PAH showed the same order as in oak wood.

Chemical Constituents of Helichrysum conglobatum Growing in Egypt

  • Five aromatic compounds, of which two are new glucosides, and six flavonols were isolated and identified for the first time from the flower heads and aerial shoots of Helichrysum conglobatum (Asteraceae). Their structures were established on the basis of chemical and spectroscopic methods including UV, MS, 1D- and 2D-NMR. Some fractions and isolates were screened for anti-microbial activities. This is the first report of the isolation of the chemical constituents of this species.

  • A new aromatic amide, methyl-N-(3-carbamoylpropionyl) anthranilate was isolated for the first time as a natural compound and one known $C_{19}-diterpene\;alkaloid$, avadharidine was also obtained from the root of Aconitum pseudolaeve var. erectum. The\;LD_{50}$ values of water extract and MeOH extract of the root of Aconitum pseudolaeve var. erectum in mice were 1. 23 g (13. 6 g crude drug) and 0. 77 g(5. 13 g crude drug)/kg, p.o., respectively.

Metabolic Activation of Ester- and Amide-Type Drugs by Carboxylesterases

  • Carboxylesterase is widely distributed in the tissues of vertebrates, insects, plants and mycobacteria. Among various tissues of animals and humans, the highest esterase activity with various substrates is found in the liver. Kidney has moderate carboxylesterase activity in the proximal tubules. Considerable esterase activity is also found in the small intestine epithet elial cells and serum of mammals. Besides these tissues, carboxylesterase has been found in the lung, testis, adipose tissue, nasal mucosa and even in the central nervous system. Hepatic microsomal carboxylesterase catalyzes the hydrolysis of a wide variety of endogenous and exogenous compounds such as carboxylester, thioester and aromatic amide. Since carboxylesterases are important for metabolic activation of prodrugs and detoxification of xenobiotics, differences in substrate specificity and immunological properties of this enzyme are important in connection with choosing a suitable laboratory animal for the evaluation of biotransformation and toxicity of drugs. On the other hand, liver, kidney, intestine and serum were found to contain multiple forms of carboxylesterases in animal species and humans. In fact, we have purified more than fifteen isoforms of carboxylesterases from microsomes of liver, kidney and intestinal mucosa of nine animal species and humans. and characteristics of these isoforms were compared each other in terms of their physical and immunochemical properties. On the other hand, we have reported that hepatic microsomal carboxylesterases are induced by many exogenous compounds such as phenobarbital, polycyclic aromatic hydrocarbons, Aroclor 1254, aminopyrine and clofibrate. Later, we showed that some isoforms of hepatic carboxylesterase were induced by glucocorticoids such as dexamethasone and 16 ${\alpha}$-carbonitrile, but other isoforms were rather inhibited by these compounds. These findings indicate that involvement of carboxylesterases in the metabolism and toxicity of drugs should be explained by the isoforms involved. Since 1991, we have carried out detailed research investigating the types of carboxylesterases involved in the metabolic activation of CPT-11, a derivative of camptothecin, to the active metabolite, SN-38. The results obtained strongly suggest that some isoforms of carboxylesterase of liver microsomes and intestinal mucosal membrane are exclusively involved in CPT-11 metabolism. In this symposium, the properties of carboxylesterase isoforms purified from liver, kidney and intestine of animal species and humans are outlined. In addition, metabolism of CPT-11, a novel antitumor agent, by carboxylesterases in relation to the effectiveness will also be discussed.

Characterization of $\beta$-Ketoadipate Pathway from Multi-Drug Resistance Bacterium, Acinetobacter baumannii DU202 by Proteomic Approach

  • In this study, the biodegradative activities of monocyclic aromatic compounds were determined from the multi-drug resistant (MDR) Acinetobacter baumannii, which were studied in the form of clinical isolates from a hospital in Korea. These bacteria were capable of biodegrading monocyclic aromatic compounds, such as benzoate and p-hydroxybenzoate. In order to determine which pathways are available for biodegradation in these stains, we conducted proteome analyses of benzoate, and p-hydroxybenzoate-cultured A. baumannii DU202, using 2-DE/MS analysis. As genome DB of A. baumannii was not yet available, MS/MS analysis or de novo sequencing methods were employed in the identification of induced proteins. Benzoate branch enzymes [catechol 1,2-dioxygenase (CatA) and benzoate dioxygenase $\alpha$ subunit (BenA)] of the $\beta$-ketoadipate pathway were identified under benzoate culture condition and p-hydroxybenzoate branch enzymes [protocatechuate 3,4-dioxygenas $\alpha$ subunit (PcaG) and 3-carboxy-cis,cis-muconate cycloisomerase (PcaR)] of the $\beta$-ketoadipate pathway were identified under p-hydroxybenzoate culture condition, respectively, thereby suggesting that strain DU202 utilized the $\beta$-ketoadipate pathway for the biodegradation of monocyclic aromatic compounds. The sequence analysis of two purified dioxygenases (CatA and PcaGH) indicated that CatA is closely associated with the CatA of Acinetobacter radiresistance, but PcaGH is only moderately associated with the PcaGH of Acinetobacter sp. ADPI. Interestingly, the fused form of PcaD and PcaC, carboxymuconolactone decarboxylase (PcaCD), was detected on benzoate-cultured A. baumannii DU202. These results indicate that A. baumannii DU202 exploits a different $\beta$-ketoadipate pathway from other Acinetobacter species.

촉매담체내에서 방향족 화합물의 역확산 (Counter-diffusion of Aromatic Compounds in Catalyst Supports)

  • 촉매 담체로 사용되는 알루미나와 실리카-알루미나에 흡착된 tetra-phenylprophine과 coronene이 아세톤의 주입에 의해 탈착되는 역확산 과정과, coronene이 tetra-phenylporphine 용액에 의한 역확산 과정을 조사하였다. 이상흡착용액 이론을 적용한 역확산 모델을 이용하여 역확산 과정을 해석하고 모사한 결과로부터 역확산 계수를 결정하였다. 과량의 아세톤 주입에 의한 탈착에서 tetra-phenylporphine과 coronene의 역확산 계수는 ${\sim}10^{-15}m^2/sec$ 범위로 얻어졌다. 흡착용액과 비슷한 농도를 갖는 tetra-phenylprophine 용액에 의한 역확산의 경우 탈착되는 coronene의 역확산 계수의 값도 ${\sim}10^{-15}m^2/sec$ 범위에 있으며, 세공내로 들어가는 tetra-phenylporphine의 확산 계수는 ${\sim}10^{-11}m^2/sec$ 이었다. 흡착제 밖으로 탈착되어 나오는 coronene의 역확산 계수는 아세톤이나 tetra-phenylporphine 용액의 주입에 의한 흡착-탈착의 경우 탈착제는 종류와 양에 관계없이 모두 비슷한 범위의 값을 보여 주었다. 역확산계수는 비어 있는 세공내로의 확산에 대한 유효확산계수에 비해 현저히 감소하였다. 이러한 현상은 역확산 과정에 나타나는 확산 흐름의 교차에 기인하는 것으로 설명되었다.

Unrecorded liverwort species from Korean flora III. New data on the distribution of Mannia Opiz (Marchantiophyta)

  • While conducting a floristic study of Korean hepatics, we discovered two unrecorded species, which were collected from wind holes near the Donggang River, Korea. Mannia fragrans (Balb.) Frye & L. Clark and Mannia androgyna (L.) A. Evans are hereby reported for the first time in Korea. M. androgyna is characterized by pale grayish oil bodies in both the aerenchyma and basal tissue, and saccate spores with a conspicuous proximal disc. M. fragrans is characterized by a gynoecial segment with a whitish apical brush of scales, a commonly aromatic smell, and areolate spores with a conspicuous proximal disc. Two unrecorded species are described and illustrated based on Korean material.

Penicillium Diversity from Intertidal Zone in Korea

  • Penicillium species are commonly isolated from various outdoor and indoor environments, including marine environments such as sponges, algae and sand. Penicillium is especially important because numerous bioactive compounds have been isolated. Penicillium was the most common species in intertidal zone in Korea, however the diversity and ecological roles of Penicillium in intertidal zone are not clarified. We explored diversity and ecological roles of marine-derived Penicillium from tidal flat and sea sand in Korea. The diversity of marine-derived Penicillium from Korea was investigated using both culture-dependent and culture-independent approach by ${\beta}$-tubulin sequence. In addition, we evaluated optimal temperature, halo-tolerance, and enzyme activity of Penicillium strains, such as extracellular alginase, endoglucanase, ${\beta}$-glucosidase, and protease. For culture-dependent approach, a total of 182 strains of 62 Penicillium species were isolated, with 53 species being identified. The most common species was Penicillium oxalicum, followed by P. crustosum, P. brasilianum, P. koreense, and P. griseofulvum. Species richness and composition were not significantly different by season, substrates, and seaside. For culture-independent approach using Illumina sequencing, 73 OTUSs were detected. The most frequently observed species was P. antarcticum, followed by P. koreense, P. crustosum, and P. brevicompactum. Diversity of Penicillium was higher during winter season than during summer season and in western sea than in southern sea, respectively. Community structure was significantly different by season and sea side. 52 species were detected by both methods. Unique species were isolated from each of methods - 10 from culture methods and 21 from Illumina sequencing. Furthermore, salinity adaption of the Penicillium varied depending on species. Many Penicillium species showed endoglucanase, ${\beta}$-glucosidase, and protease activity. Some species including P. paneum and P. javanicum degraded the polycyclic aromatic hydrocarbons. Thus, our results demonstrate that intertidal zone in Korea harbors diverse Penicillium community and marine-derived Penicillium play important ecological roles as decomposers of organic material in marine environments.

세신의 생약학적 연구 (Pharmacognostical Studies on the "Se Sin")

  • Chinese crude drug 'Se Sin'(細辛)has been used to cure headache, cough and expectorant. To the botanical origin of 'Se Sin', Zhong-yao-zhi(中葯志) reported Asarum heterotropoides var. mandshuricum. A. sieboldii and A. sieboldii var. seoulensis, and Zhong-yao-da-ci-dian(中葯大辭典) reported Asarum heterotropoides var. mandshuricum and A. sieboldii of Aristolochiaceae. In Korea, this crude drug has been used as a Korean folk remedy for tothache and aromatic. The botanical origin of 'Se Sin' is considered to be Asarum species of Aristolochoaceae. But there has no pharmacognostical confirmation on it. To clarify the botanical origin of 'Se Sin', we studied on the anatomical characteristics of Asarum species growing wildly in Korea i.e. A. maculatum, A. sieboldii, A. sieboldii var. seoulensis, and of 'Se Sin' from Korea on Korean market. Through our studies, the botanical origin of 'Se Sin' from Korea was proved to be whole plant of Asarum sieboldii and A. sieboldii var. seoulensis.
