Isolation and Characterization of Elastase Inhibitor from Areca catechu (빈랑으로부터 Elastase 저해물질의 분리 및 특성조사)

  • We have previously screened 150 medicinal plants on the inhibition of elastase and found a significant inhibitory effects of the extracts of Areca catechu L. on the aging and inflammation against the skin tissues. To isolate and identify the compounds having biological activity, we was further purified by each of the solvent fractions, silica gel column chromatography, preparative TLC and reversed-Phase HPLC. Peak in HPLC, which coincided with the inhibitory activity against elastase, was identified as Phenolic substance using various colorimetric methods, UV, and IR. $IC_{50}$/ values of phenolic substance purified from Areca catechu were 26.9 $\mu\textrm{g}$/$m\ell$ for porcine pancreatic elastase (PPE) and 60.8 $\mu\textrm{g}$/$m\ell$ for human neutrophil elastase (HNE). This Phenolic substance showed more potent activity than those of reference compounds, oleanolic acid (76.5 $\mu\textrm{g}$/$m\ell$ for PPE, 219.2 $\mu\textrm{g}$/$m\ell$ for HNE) and ursolic acid (31.0 $\mu\textrm{g}$/$m\ell$ for PPE, 118.6 $\mu\textrm{g}$/$m\ell$ for HNE). According to the Lineweaver-Burk Plots, the inhibition against both PPE and HNE by this phenolic substance was competitive with substrate. Phenolic substance from Areca catechu exhibited high free radical scavenging effect ($SC_{50}$/ : 6 $\mu\textrm{g}$/$m\ell$) and inhibited effectively hyaluronidase activity ($IC_{50}$/: 210 $\mu\textrm{g}$/$m\ell$). These results suggest that the Phenolic substance Purified from Areca catechu showed anti-aging effect by protecting connective tissue proteins.

알카로이드독을 이용한 약물

  • 毒에는 비소, 수은처럼 無機毒과 식물, 동물성의 有機毒이 있으며, 有機毒에는 알카로이드, 배당체, 단백毒 등이 있다. 식물의 알카로이드毒을 중심으로 이것이 어떻게 藥物로서 이용되어 왔는지에 관해 다루었다. Morphine의 발견을 계기로 여러 종류의 식물 알카로이드가 계속 분리되었으며, 藥의 탐구의 새로운 길을 열었다. Morphine은 우수한 진통의 효과를 가졌지만, 흡수억제 및 습관성의 결점을 가지고 있다. 또, Areca catechu L. 의 종자(빈낭자)에는arecoline, arecaidine 등의 piperidine系의 알카로이드를 함유하고 있다. Arecoline은 副交感神經 흥분작용이 있고, 침분비 촉진, 比瀉 등으로 사용되어 왔지만, 지금은 그 용도가 적어졌다. 그리고 화살독이 되는 毒을 약물로 사용한 것은 Acontium屬 식물이 유일한 것이다. 중국에서는 이를 身體四肢關節의 마비, 疼痛의 회복, 대사기능실조의 회복 등의 목적으로 사용해 왔다.

Antimutagenic Effect of Tansen (Salvia miltriorrhiza Bunge) (단삼의 항산화적 항돌연변이 효과)

  • To confirm the effects of binlang(Areca catechu L.) and tansen(Salvia miltriorrhiza bung) on the mutagenicity induced by hydrogen peroxide, SOS Chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA104 were perlormed. Methanol-soluble parts of their water extracts showed high inhibitory effect against the mutagenicity of hydrogen peroxide in two bacterial mutation assays. Step-wise fractionation of methanol-soluble part from tansen was done using ethyl acetate, butanol and water. Among these fraction was further partitioned by Sephadex LH-20 column chromtography, and 6 subfractions were obtained. The fraction III showed the strongest inhibitory effects against the mutagenic activities induced by hydrogen peroxide. The inhibition rates of fraction III at concentration of 500$\mu\textrm{g}$/assay were 28%, 30% and 15% against 4-NQO, MNNG and B(a)P, respectively. But the mutagenic potency of AFB1 was increased.

  • In order to evaluate the effect of the extracts of eidble and medicinalplant son the activation of immune cells, measurements were made by ELISA and radioimmunoassay on the degree of release for the tumor necrosis factor (TNF) and neopterin by the edible and medicinal plants in peripheral blood cells. The results of measurements of TNF in the supernatant cultured liquid showed nothing in t도 control which does not have any edible and medicinal plants. However, measurements of TNF 9pg/ml) in the samples are given as follows : 716.7 in lipopolysaccharide (LPS 1 g/ml), 465.2 Rheum plamatum L.m302.7 Sanguisorba offciinalis L. 818.2 Rubus coreanus M, 328.3 Terminalia chebula R., 426.6 AReca catechu L. 227.0 Eugenia caryophiliata T., 272.9 Ephedra sinica S., 30.1 Caesalpinia sappan L., 474.0 Chaenomeles japonica L., 396.0 Cornus officinalis S.in edible and medicinalplants. ENopterin (n mole/L) value showed below the check point in the control group, however, the values are 11.0 in LPS, and edible and medicinal plants, 5.3 Rheum palmatum L., 11.6 Eugenia caryophiliata T., 5.5 Ephedra sinica S., 4.5 Caesalpinia sappan L., 4.3 Chaenomelees japonica L.3.7 Cornus officinalis S. In order to find m RNA levels of Cytokines increased by edible and medicinal plants, total RNA was separated from mononuclear cells treated 5 hrs with Rubus coreanus M. and then administrated for RT-PCR. The considerable increases of the m RNA of TNF, IL-1 $\alpha$ and IL-6 were observed.

Isolation and Quantitative Determination of Arecoline from Arecae Semen (빈랑자(Arecae Semen)로부터 arecoline의 분리 및 함량분석)

  • The seeds of Areca catechu L. has been used for the treatment of the diseases caused by parasites in East Asia. As a part of a research for standardization of crude drugs, we have determined the content of arecoline in the seeds of Arecae Semen purchased from various regions of Korea. The HPLC method for quantitative analysis of arecoline in Arecae Semen was established and reproducible results and chromatographic isolation of arecoline was accomplished successively. It suggested that the content of arecoline in Arecae Semen was $0.2726\;{\pm}\;0.05532%$.
