입체장애아민 AMP (2-amino 2-methyl 1-propanol) 수용액의 CO2 흡수평형 (Absorption Equilibrium of CO2 in the Sterical Hindered Amine, AMP Aqueous Solution)

  • 본 연구는 그 동안 주로 석유화학의 개질공정 등에서 적용되었던 이산화탄소 흡수분리기술을 연소배가스에 적용하기 위한 기본적인 연구로서, 이산화탄소의 흡수분리공정에 일반적으로 많이 사용되는 흡수제인 MEA(mono-ethanol amine)를 연소배가스에 적용하였을 경우 예상되는 열화, 재생에너지 및 흡수능력 등의 문제점을 개선하기 위하여 거론되는 입체장애아민인 AMP(2-amino 2-methyl 1-propanol) 수용액과 이산화탄소와의 흡수평형 특성을 비교 평가하였다. 실험결과 AMP는 MEA에 비하여 이산화탄소의 흡수능력이 반응 메카니즘에 의하여 약 2배 정도 증가함을 확인하였고, 연소배가스와 같은 저농도 이산화탄소 흡수분리공정 설계에 필요한 흡수평형자료를 부분적으로 제시하였다.

Scrophulariae Radix Aqueous Extracts Ameliorate the Pressure Overloaded Heart Failure by Transverse Aortic Constriction in Mice

  • Objectives: The purpose of this study was to investigate the cardioprotective effect of the potent antioxidant properties of Scrophulariae Radix (SR) aqueous extracts by examining pressure overload (PO) heart failure (HF) induced by transverse aortic constriction (TAC) of C57BL/6 mice. Methods: SR (500, 250, 125 mg/kg) and resveratrol (10 mg/kg) were administered orally once a day for 14 days, after the TAC operation. Changes in mortality, the body and heart weights, histopathology of the heart, and antioxidant defense system of the heart were analyzed. Results: After the TAC operation, increases were observed in mortality, heart weights, left ventricular hypertrophy, and lytic and focal fibrotic histological change, and destruction of the heart antioxidant defense system. However, the HF signs showed dose-dependent inhibition following 14 days of continuous oral treatment with SR. A SR dose of 125 mg/kg gave a similar inhibition to that obtained with resveratrol at 10 mg/kg. Conclusions: Oral administration of SR beneficially improves PO-induced HF following TAC surgery by increasing the activity of the heart antioxidant defense system. The overall effect of SR at 125 mg/kg is similar to the effect of resveratrol at 10 mg/kg. However, more detailed mechanistic studies should be performed by screening of the biologically active compounds in SR.

효소공정에 의한 트립토판 생산 (Production of L-Tryptophan by Enzymatic Processes)

  • Tryptophanase를 이용하여 트립토판 합성시 인돌의 효소활성 저해를 억제하기 위하여 유가식 조업, 유기용매 이상계의 사용, 그리고 cyclodextrin의 첨가등에 대하여 연구하였다. 효소 농도가 0.5mg/ml일때 인돌 농도 0.4mM 부근에서 트립토판 생성이 가장 빨랐으며 그 이상에서는 효소활성이 심한 저해를 받았다. 초기 인돌 농도가 20mM일 때는 27시간 반응후 인돌의 전환율이 20인데 비하여 반응기내 인돌 농도를 5mM 이하로 유가식 조업을 하였을 때 전환율이 80%로 향상되었다.

Optimization of Maillard Reaction in Model System of Glucosamine and Cysteine Using Response Surface Methodology

  • Sulfur-containing amino acids play important roles in good flavor generation in Maillard reaction of non-enzymatic browning, so aqueous model systems of glucosamine and cysteine were studied to investigate the effects of reaction temperature, initial pH, reaction time, and concentration ratio of glucosamine and cysteine. Response surface methodology was applied to optimize the independent reaction parameters of cysteine and glucosamine in Maillard reaction. Box-Behnken factorial design was used with 30 runs of 16 factorial levels, 8 axial levels and 6 central levels. The degree of Maillard reaction was determined by reading absorption at 425 nm in a spectrophotometer and Hunter's L, a, and b values. ${\Delta}E$ was consequently set as the fifth response factor. In the statistical analyses, determination coefficients ($R^2$) for their absorbance, Hunter's L, a, b values, and ${\Delta}E$ were 0.94, 0.79, 0.73, 0.96, and 0.79, respectively, showing that the absorbance and Hunter's b value were good dependent variables for this model system. The optimum processing parameters were determined to yield glucosamine-cysteine Maillard reaction product with higher absorbance and higher colour change. The optimum estimated absorbance was achieved at the condition of initial pH 8.0, $111^{\circ}C$ reaction temperature, 2.47 h reaction time, and 1.30 concentration ratio. The optimum condition for colour change measured by Hunter's b value was 2.41 h reaction time, $114^{\circ}C$ reaction temperature, initial pH 8.3, and 1.26 concentration ratio. These results can provide the basic information for Maillard reaction of aqueous model system between glucosamine and cysteine.

시호가 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상샘 기능저하증에 미치는 영향 (Effects of Bupleuri Radix on the Rat Hypothyroidism Induced by PTU (6-n-propyl-2-thiouracil))

  • The object of this study was to evaluate the effect of Bupleuri Radix, aqueous extracts of the root part of Bupleurum falcatum on the 6-n-propyl-2-thiouracil (PTU)-induced rat hypothyroidism. Aqueous extracts of Bupleuri Radix (BR; yield = 11.73%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 300 and 150 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), triiodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride, liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT), serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were observed with histopathology of thyroid glands. Results were compared with $LevoT_4$ 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, triglyceride contents, liver CAT activities and changes of serum thyroid hormone levels were observed with increases of serum AST, HDL contents, liver $H_2O_2$ and SOD activities and thyroid gland weight. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contents and sizes were demonstrated at histopathological inspections. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of BR extracts, and BR extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that BR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

  • The object of this study was to evaluate the effect of Lonicerae Flos, aqueous extracts of the dried flower bud part of Lonicera japonica on the 6-n-propyl-2-thiouracil (PTU)-induced rat hypothyroidism. Aqueous extracts of Lonicerae Flos (LF yield = 23.80%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500 and 250 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), tri-iodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride were observed with liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT) and serum asparte aminotransferase (AST) and alanine aminotransferase (ALT) analysis. Results were compared with Levothyroxine ($LT_4$) 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, serum thyroid hormone levels and triglyceride contents, liver $H_2O_2_3$ and SOD activities were observed with increases of serum AST and HDL contents, liver CAT activities, thyroid gland weight. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of LF extracts, and LF extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. $\alpha$-Chymotrypsin 을 이용한 Kyotorphin 유도체의 합성 (The Synthesis of Kyotorphin Derivative by $\alpha$-Chymotrypsin)

  • 효소를 이용한 생리활성 펩티드의 합성 연구에 관한 기초자료를 얻기 위하여, Kyotorphin(진통작용을 가진 펩티드) 유도체가 $\alpha-chymotrysin$를 이용한 이상계(two phase system;유기상과 수용액상)조건하에서 Ac-Tyr-OH 와 $Arg-NH_2$로부터 합성되었다. Ac-Tyr-OH(10 mM)과 $Arg-NH_2$ (20 mM)와의 Kyotorphin 유도체 합성에 대한 유기용매의 효과에서 ethyl acetate계에서의 합성 수율이 다른 유기용매(dichloromethane, n-butanol, n-hexane, chloroform)에서 보다도 더 높았다. Kyotorphin 유도체의 합성에 미치는 최적조건을 보면, 효소 농도는 10 ${\mu}M$, 온도 및 pH는 각각 $35^{\circ}C$ 및 7.0이었으며, 유기상/수용액상의 비$(\alpha)$는 15였다. Kyotorphin 유도체 합성의 최적 반응조건하에서 수율은 70.2%였으며, 이때의 반응은 24시간 후에 평형에 도달하였다.

콜로이드 계면화학을 이용한 저온형 고체전해질용 $CeO_2$계 복합 산화물의 소결체 제조 (The Preparation for Sintered Body of $CeO_2$ Based Complex Oxide in Low Temperature Solid Oxide Fuel Cells Using Colloidal Surface Chemistry)

  • In this study, the dispersion stability of CeO2 based complex oxide was studied, and density, porosity, and microstructure of green body were investigated using colloid surface chemistry to manufacture the Gd2O3 doped CeO2 solid electrolyte in an aqueous system. To prepare the stable slurry for slip casting, the dispersion stability was examined as a function of pH using ESA(electrokinetic sonic anplitude) analysis. The dynamic mobility of particles was enhanced with anionic and cationic dispersant were added the amount of 0.5wt% respectively, but pH value in slurries didn't move to below 6.0 because of the influence of dopants. This phenomenon also appeared in the CeO2-Y2O3 and CeO2-Sm2O3 systems, so it could be inferred that rare earth dopants such as Gd2O3, Sm2O3 and Y2O3 not only have the similar motion with changing pH in an aqueous system but also can be dissolved in the range of pH 6.0∼6.5. In CeO2-Gd2O3 system, when the anionic dispersant was added the amount of 0.5wt% and pH value in slurries was fixed at 9.5, the green body density was 4.07g/㎤, and the relative density of sintered body was 95.2%. It could be inferred from XRD analysis that Gd3+ substituted into Ce4+ site because there was no free Gd2O3 peak.

Poly(ethyleneglycol)과 인산염용액이 형성하는 2상계에서의 단백질 분획에 관한 연구 (Protein Partition in an Aqueous Poly (ethyleneglycol)-salt Two-phase System)

  • Poly(ethylene glycol)과 인산염 용액(PPB)이 형성 하는 액상 이상계에서 단백질의 분획 거동을 관찰하였다. PEG(12%)/PPB(12%) 액상 이상계에서 단백질의 표면 소수성이 다른 BSA, ${\beta}-lactoglobulin$, ovalbumin 모두 PPB-rich phase인 하층부로 몰려 그 분획계수는 대단히 낮았다. PPB의 농도를 15%로 증가 시키면 하층부의 전위는 +50mV 에서 0으로 낮아져 단백질의 분획계수가 다소 증가하는 경향을 나타내었다. PPB의 제2인산염과 제일인산염의 몰 비율을 1.43에서 9.55로 증가시키면 이상계의 상 하층부 부피비(Vt/Vb)가 다소 감소되면서 분획계수는 증가하였다. PPB농도를 $14{\sim}26%$로 증가시키면 이상계의 부피비는 1.5에서 0.39로 현저히 감소하면서 단백질의 분획계수는 ${\beta}-lactoglobalin$의 경우 74배, BSA 32배, ovalbumin 12배, lysozyme 5배로 크게 증가하였다.

AOT/이소옥탄/역미셀계에서의 Tyrosinase 반응 (Tyrosinase Reaction in AOT/Isooctane/Reverse Micelles)

  • 지용성 물질은 물에 용해되지 않아 수용액 효소반응계에서의 특정효과를 평가하기가 어렵다. 생체내에서 멜라닌을 합성하는 효소인 tyrosinase의 지용성 저해제를 탐색하고자 유기용매, 계면활성제 및 물로 구성된 광학적으로 투명한 RM을 도입하였다. Tyrosinase의 겉보기 반응이 물에서와 유사한 RM의 형성조건 중 유기용매는 isooctane, 계면활성제는 100 mM의 dioctyl sulfosuccinate(AOT)가 적절하였다. RM에서 물과 AOT의 몰 농도 비율이 15일 때 tyrosinase(103.5 units)는 기질인 3,4-dihydroxy-L-phenylalanine(0.18 mM)에 대하여 수용액에서와 유사한 거동을 나타내었다. 또한 tyrosinase 저해제인 cinnamic acid 존재 하에서도 tyrosinase 촉매 반응의 반응 생성물이 반응 시간에 비례하여 형성됨이 확인되어 지용성 물질도 RM에서는 tyrosinase 저해활성이 가능함을 시사하였다. 따라서 RM은 수용성인 효소와 기질 그리고 지용성인 저해제를 모두 하나의 상(phase)에 용해시킬 수 있어 지용성 물질의 tyrosinase 저해 효과를 분석할 수 있는 중요한 수단으로 활용할 수 있을 것으로 판단되었다.
