We investigated the effects of aqueous extracts from twelve medical herbs on ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis in B16F10 mouse melanoma cell. The cells were incubated with ${\alpha}$-MSH and aqueous extracts 5 days and 2 days and then analysed melanin amount and tyrosinase activities, respectively. Nine aqueous extract of herbs examined at 1 mg/m${\ell}$ level decreased melanin synthesis in B16F10 cell, especially in Agastache rugosa, Leonurus siviricus and Murus bombycis. The significant decrease of released extracellular melanin were also observed in treated cells with aqueous extract from Leonurus siviricus, Murus bombycis and Ledebouriella seseioides. The ${\alpha}$-MSH-induced activation of tyrosinase was inhibited in cells treated with aqueous extract from Cuscutae semen, Angelica tenuissima and Agastache rugosa. These results suggest that herbs inhibiting melanogenesis through tyrosinase activity may apply to develop whitening drugs and cosmetics.
The protective effects on hepatic fibrosis of an aqueous extract from the roots of Platycodon grandiflorum A. DC (Campanulaceae), Changkil (CK), in hepatic stellate cell line, CFSC-2G. The increased deposition of extracellular matrix by hepatic stellate cells following liver injury in a process known as activation is considered a key mechanism for increased collagen content of liver during the development of liver fibrosis. Objectives: This study evaluated the effect of aqueous extract from roots of Siberian ginseng on mTORC1 pathway. Methods: mTORC1 activity was measured by the phosphorylation status of p70 S6 kinase (S6K) in HeLa cells as well as the brain, liver and muscle tissues in diabetic db/db mice. Autophagy induction after the treatment of Siberian ginseng extract was evaluated by monitoring the conversion of cytoplasmic LC3I into lipidated LC3II in cultured human HeLa GFP-LC3 cells. Cell cycle analysis was performed in HeLa cells treated with Siberian ginseng using flow cytometry. Results: Among >2,800 plant products used for oriental medicine, Siberian ginseng was found to inhibit mTORC1 to phosphorylate S6 kinsase (S6K) in HeLa cells as well as the brain, liver and muscle tissues in diabetic db/db mice. Siberian ginseng-mediated mTORC1 activity was reversible unlike the prolonged suppression of mTORC1 by rapamycin when HeLa cells were grown in fresh media after the removal of the inhibitors. Siberian ginseng extract at concentrations to inhibit mTORC1 was not overly cytotoxic in cultured HeLa cells whereas rapamycin was obviously cytotoxic. The conversion of cytoplasmic LCI into lipidated LCII was increased by fivefold in HeLa GFP-LC3 cells treated with Siberian ginseng extract. Progression of cell cycle was attenuated at G2/M phase by the treatment of Siberian ginseng extract. Conclusions: These results suggest that the aqueous extract of Siberian ginseng possibly plays a good therapeutic role in various diseases involving mTORC1 signaling.
We studied antioxidant activity and inhibitory effect of ${\alpha}$-glucosidase from aqueous, ethanolic and methanolic fractions of Galla rhois. In FRAP and ORAC assay for measuring antioxidant activity, we confirmed that Galla rhois extracts had strong antioxidant activity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. We used trolox as a positive control. In order to measure the inhibitory effect of ${\alpha}$-glucosidase, we compared acarbose and Galla rhois extracts. As a result of ${\alpha}$-glucosidase inhibitory assay, aqueous, ethanolic and methanolic extracts of Galla rhois showed high inhibitory activitity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. The 50% inhibitory concentrations (IC50s) of acarbose, aqueous, ethanolic and methanolic fractions were 0.45 mM, $0.53{\mu}g/ml$, $0.415{\mu}g/ml$ and $0.37{\mu}g/ml$, respectively. These results suggest that Galla rhois extracts can be a clinically useful anti-diabetic ingredient, indicating that it needs to be fractionated and isolated and should be further investigated.

This study was conducted to find out the effects of aqueous extract from the leaves of Artemisia capillaris (AA) on the reduction of hepatotoxicity induced by ethanol in rats. In this experiment, Sprague Dawley rats were used in the experimental groups, which were divided into 5 groups; normal group, ethanol+UDCA (ursodeoxycholic acid)-treated group (positive control), ethanol-treated group (control), ethanol+Artemisia capillaris aqueous extract-treated group (200 mg/kg of BW) and ethanol+Artemisia capillaris aqueous extract-treated group (400 mg/kg of BW). AST (aspartate aminotransferase), ALT (alanine aminotransferase), GGT (gamma(${\gamma}$)-glutamyl transferase) and LDH (lactate dehydrogenase) activities of the ethanol+Artemisia capillaris aqueous extract-treated group (400 mg/kg of BW) were significantly decreased compared to that of the ethanol-treated group (P<0.05). The triglyceride level of the ethanol-treated group was significantly increased and the HDL-cholesterol level was significantly decreased compared to the normal group (P<0.05). On the other hand, the triglyceride level was significantly decreased (P<0.05) and the HDL-cholesterol level was significantly increased (P<0.05) in the ethanol+Artemisia capillaris aqueous extract-treated groups. Superoxide dismutase (SOD) activity was enhanced significantly (P<0.05) in the ethanol+Artemisia capillaris aqueous extract-treated groups. Also, malondialdehyde contents were decreased in this group (P<0.05). Histologically, in the control group, there was a mild degenerative change around central venule. The AA treated group showed well preserved lobular architectures with no evidence of steatosis or liver damage in aqueous extract from the leaves of Artemisia capillaris treated group (H&E, ${\times}20$). As the results of this study, it is thought that Artemisia capillaris aqueous extract may have effects on the improvement of hepatic damage by ethanol.
In this study, analysis of active compounds that are believed to be highly relevant to antioxidant activity was carried out on the methanol extract and its solvent fractions of Corni fructus. The DPPH radical scavenging activity for the comparison of antioxidant activity was higher in order of aqueous fraction > methanol extract > ethyl acetate fraction > n-hexane fraction. It is similar to the order of total polyphenol contents in the samples. As a result of LC-MS analysis, phenolic acid compounds such as caffeic acid, gallic acid and chlorogenic acid and lognin, which is known as a representative active ingredient of Corni fructus, were identified as active compounds. And the antioxidative activity and the total polyphenol content of the extracts and solvent fractions were found to be related to the contents of the compounds. Particularly, it was confirmed that phenolic acid such as caffeic acid contributes to the antioxidative activity of the aqueous fraction of Corni fructus methanol extract.
Anti-diabetic effect was observed with Thespesia lampas Dalz & Gibs (Family: Malvaceae) when given as a root extract in normal as well as alloxan induced diabetic rats. The effects, however, were more pronounced in diabetic animals in which administration of plant extract for 15 days after alloxan induced diabetes, significantly reduced blood glucose levels. After alloxan induced diabetes it was observed that both standard drug (glibenclamide) and aqueous extract of Thespesia lampas were significantly superior to control in reducing blood sugar on long term treatment (15 days). The aqueous extract of T. lampas (300 and 600 mg/kg) reduced the blood glucose levels from $349.2{\pm}7.2$ to $120.7{\pm}4.6$ and $346.3{\pm}3.4$ to $101.8{\pm}6.3$, respectively. The data suggested that T. lampas could be of beneficial in diabetes mellitus in controlling blood sugar. The present investigation established pharmacological evidence to support the folklore claim as an anti-diabetic.
When iron oxide magnetic nanoparticles were synthesized by co-precipitation method using aqueous ammonia as reducing agent, the synthesized particles were aggregated and thus precipitation occurred. Using Magnolia kobus leaf extract as reducing agent, spherical nanoparticles of 50~200 nm were synthesized with low yield. By using both Magnolia kobus leaf extract and aqueous ammonia as reducing and stabilizing agents, smaller nanoparticles of 40~120 nm could be synthesized with various shapes. The synthesized magnetic nanoparticles were characterized with field emission transmission electron microscopy (FE-TEM) and scanning electron microscopy (SEM). TEM and SEM images showed that the magnetic nanoparticles are a mixture of triangles, tetragons, rods and spherical structures.
In this research, the genotoxic effect of Picrorrhiza Rhizoma (PR) aqueous extract was evaluated using the mouse micronucleus test. PR extract was administered once a day for 2 continuous days by oral gavage to male ICR mice at doses of 2000, 1000 and 500 mg/kg. Cyclophosphamide was used as a known genotoxic agent in a positive control. The appearance of a micronucleus (MN) in polychromatic erythrocyte (PCE) is used as an index for genotoxic potential, and PCE ratio is used as an index of cytotoxicity. Although significant (p < 0.01) increase of the number of PCE with one or more nuclei (MNPCE) was detected in cyclophosphamide treated groups, no significant increases of MNPCE numbers were observed in all three different dosages of PR extracts treated mice with over 0.39 of the individual polychromatic erythrocyte ratio in all mice used in this study. The results obtained indicated that PR extract shows no genotoxicity effects up to 2000 mg/kg dosing levels.
Objectives : The present study purposed to compare the anti-oxidative effect of Fructificatio Inonoti Obliqui aqueous extract according to extraction temperature. Methods : We measured the total phenol content and anti-oxidative activity of Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low-temperature leachate. Results : Anti-oxidative activity of Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low-temperature leachate was different according to extraction temperature, concentration, and antioxidant effect measuring method. In all experiments, the total extract showed higher antioxidant than n-BuOH fraction. The total phenol content was higher in Fructificatio Inonoti Obliqui total extract than $50^{\circ}C$ low-temperature leachate. Conclusions : Both Fructificatio Inonoti Obliqui total extract and Fructificatio Inonoti Obliqui $50^{\circ}C$ low-temperature leachate have significant anti-oxidative effect.
