Lactobacillus brevis를 활용한 흰점박이꽃무지 유충과 산양삼의 발효물에 대한 특성 연구 (Properties of Aqueous Extract of Protaetia Brevitarsis Larva and Mountain Ginseng Fermented by Lactobacillus Brevis)

  • 본 연구에서는 김치로부터 분리된 L. brevis SM61을 생화학적 특성과 16s rRNA 염기서열 분석을 통해 분리 및 동정하였으며, 흰점박이꽃무지 유충 열수추출물과 산양삼 분말을 L. brevis SM61을 이용하여 48시간동안 발효를 수행하여 발효물을 제조하였다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 농도에 따른 세포 독성은 없는 것으로 판단되었다. 그리고, 흰점박이꽃무지 유충 열수추출물과 L. brevis SM61을 사용한 발효물에 대해 발효 시간에 따른 폴리페놀 함량을 분석한 결과 발효 초기에는 약 $1.49{\mu}g/mL$ 비해 48시간 이후 약 $5.09{\mu}g/mL$ 수준으로 증가하였다. 또한, 항산화능은 흰점박이꽃무지 유충 열수추출물이 약 60%였으나, L. brevis SM61을 이용한 발효물이 약 98%로 높아졌다. 항균 활성의 경우 L. brevis SM61을 이용한 발효물에서만 항균 효과가 확인되었다. 따라서, 본 연구를 통해 분리된 신규 L. brevis SM61을 이용한 흰점박이꽃무지 유충의 열수추출물에 대한 발효물은 식용 곤충의 기능성을 강화시킬 수 있을 것으로 판단되며, 해당 유산균을 활용하여 다양한 식용 곤충에도 적용이 가능할 것으로 사료된다.

Allelopathic Effects of Chrysanthemum boreale on Seed Germination and Seedling Growth of the Selected Plants

  • To examine the allelopathic potentials of Chrysanthemum boreale, aqueous extracts and essential oil of the plant were used in these experiments. Seed germination of the receptor species was inhibited by the aqueous extracts and the inhibitory effect was increased in proportion to the concentration of extracts. In contrast, seedling elongation showed varied results. Achyranthes japonica, Bidens bipinnata, Raphanus sativus var. hortensis for. acanthiformis, Plantago asiatica, Pimpinella brachycarpa and Lactuca sativa were inhibited by increasing concentration of the aqueous extract, while Brassica campestris subsp. napus var. pekinensis and Echinochloa crus-galli were stimulated by the extract. Dry weight was also inhibited proportionally by increasing concentration of the aqueous extract, while some species were stimulated by a lower concentration of the extract. The volatile substances of C. boreale did not affect the seed germination of receptor plants, but seedling elongation and dry weight of some species were inhibited dose-dependently. Root hair development of selected plants was inhibited along with the concentration of essential oil. The above mentioned results, therefore, confirmed that the natural substances from C. boreale had allelopathic potentials to other plants.

Effect of Dichrostachys cinerea (Linn.) Root Extract on Ethylene Glycol Induced Urolithiasis in Rats

  • Dichrostachys cinerea (Linn.) is commonly known as Vadatalla and used as phytotherapeutic agent. Tender shoots of the plant bruised and applied to the eyes in case of ophthalmia. The root is astringent and used in rheumatism, urinary calculi and renal troubles. The effect of the Ethanolic and aqueous extract of the root of D. cinerea were studied for its anti-urolithiatic and diuretic activity at 200 mg/kg dose level in male Wistar albino rats. Ethylene glycol feeding resulted in hyperoxaluria as well as increased renal excretion of calcium and phosphate. Supplementation with aqueous and ethanolic extract of the plant significantly reduced the elevated urinary oxalate, showing a regulatory action on endogenous oxalate synthesis. Compared to ethanolic extract, aqueous extract exhibited significant anti-urolithiatic activity. Both the extracts showed significant diuretic activity. The results of our present study supports folklore claim of D. cinerea.

침향의 항알레르기 효과 (Antiallergic Effect of Aquilariae Lignum)

  • Effects of the aqueous extract of Aquilariae Lignum (Thymelaeaceae) on the allergic reactions were investigated. Oral administration of this extract (50, 250, and 500mg/kg) exhi bited a dose-dependent inhibition on passive cutaneous anaphylactic reactions in rats. Administrations of this extract (500mg/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) decreased the mortality rates to 0, 0, and 14.2%, respectively. The aqueous extract of Aquilariae Lignum (0.05 ~ 1.6mg/ml) showed a dose-related inhibition on histamine release from rat peritoneal mast cells. The morphological examination also clearly showed that the aqueous extract of Aquilariae Lignum prevented the degranulation of mast cells in rats.

  • Corrosion inhibition of 304 stainless steel (SS) in 1 M HCl by aqueous extract of coriander seeds was studied using weight loss, potentiodynamic polarization, electrochemical impedance spectroscopy (EIS) and electrochemical frequency modulation (EFM) techniques. Values of inhibition efficiency obtained are dependent upon the concentration of extract and temperature. Generally, inhibition was found to increase with inhibitor concentration, but decrease with temperature. Physical adsorption mechanism has been proposed for the inhibition with Langmuir adsorption isotherm obeyed. Values of activation energy of the inhibited corrosion reaction of 304 SS are greater than the value obtained for the blank. Thermodynamic consideration reveals that adsorption of aqueous extract of coriander seeds 304 SS surface is spontaneous.

Tenderization of Bovine Longissimus Dorsi Muscle using Aqueous Extract from Sarcodon aspratus

  • The aim of this study was to investigate the effects of aqueous extract from Sarcodon aspratus on tenderization of the bovine longissimus dorsi muscles in comparison with commercial proteolytic enzymes. Furthermore, meat quality and muscle protein degradation were examined. We marinated meat with 2% Sarcodon aspratus extract, 2% kiwi extract, and 0.2% papain. Beef chunks (3×3×3 cm3) were marinated with distilled water (control), Sarcodon aspratus extract (T1), kiwi extract (T2) or papain (T3) for 48 h at 4℃. There were no significant differences in muscle pH and lightness between control and treated samples. T1 had the lowest redness (p<0.01), and higher cooking loss and water holding capacity than control and T2 (p<0.05). T1 and T3 exhibited lower shear force values than control (p<0.05). Total protein solubility did not differ significantly between T1 and control, but T1 had less myofibrillar protein solubility than control and T2 (p<0.001). The degradation of myosin heavy chain in T1 and T3 was observed. This degradation of myofibrillar protein suggests that Sarcodon aspratus extract could influence tenderization. These results show that aqueous extract of Sarcodon aspratus extract actively affect the tenderness of the bovine longissimus dorsi muscle.

Effects of Drynariae Rhizoma on the Proliferation of Human Bone Cells

  • Drynariae Rhizorna (DR), an herbal medicine known for its effect to purify blood quality and improve circulation, frequently appears as the main ingredient in prescriptions for bone injuries. Currently, how pharmacologically it contributes to the reformation of bone is unclear. In the present study, the effect of the aqueous extract of DR on bone cells was investigated in vitro for the first time. The human osteoprecursor cells (OPC-I) were incubated in the medium with different concentrations of the aqueous extract of DR and the cell proliferation was studied. When the concentration of DR aqueous extract was $<120{\;}\mu\textrm{g}/ml$, the proliferation of OPC-I was enhanced. However, the proliferation of OPC-I was inhibited by DR extract with the concentrations $>250{\;}\mu\textrm{g}/ml$. Under most treatments, the cells presented very pale expression for cyclooxygenase-2 (Cox 2) protein; a slightly intensified band showed at the highest DR concentration, 1.0 mg/ml during the course of culture. From the results, it was concluded that the aqueous extract of DR was found to directly stimulate the proliferation, alkaline phosphatase (ALP) activity, protein secretion and particularly type I collagen synthesis of OPC-I at dose-dependent manner.

Cytotoxic Activities of Red Algae Collected from Jeju Island Against Four Tumor Cell Lines

  • Methanolic and aqueous extracts of 26 red algae species collected from Jeju Island coast were prepared at a high $(70^{\circ}C)$ and a room temperature $(20^{\circ}C)$ and were examined for their cytotoxic activity against 4 tumor cell lines: U-937 (human monoblastoid leukemia cell line), HL-60 (human promyelocytic leukemia cell line), B-16 (murine melanoma cell line) and HeLa (woman cervical carcinoma cell line). $20^{\circ}C$ methanolic extract of Polysiphonia japonica showed cytotoxic activity of over 50% against U-937, HL-60 and B-16 cells. On the other hand, the $20^{\circ}C$ aqueous extract of Scinaia okamurae and $70^{\circ}C$ aqueous extract of Chondrus crispus showed cell growth inhibition activity of more than 50% against HL-60 and B-16 cells. The highest cytotoxic activity was observed in the $20^{\circ}C$ aqueous extract of Scinaia okamurae against B-16 cells (80.55%).

셀룰로오스와 나일론 직물의 쑥 추출물에 대한 염색성과 항균성 (Dyeing and Antimicrobial Properties of Cellulose and Nylon Fabrics Treated with Artemisia Extracts)

  • We examined dyeing properties using cotton, Tencel, general nylon 66 and hollow nylon 66 treated with aqueous and ethanol extracts without mordant. The antimicrobial properties of fabrics treated with Artemisia extracts against gram positive Staphylococcus aureus (S. aureus) and gram negative Klebsiella pneumonia (K. pneumonia) were also examined. The dying solution concentrations were determined from a calibration curve of the concentration and absorbance of Artemisia extracts. FTIR spectra confirmed that antimicrobial components and colorants (such as 1,8-cineol, thujone, caffeoylquinic acid and chlorophyll) were more present in ethanol extract than in aqueous extract. Nylons had higher $a^*$ and $b^*$, and lower $L^*$ values than cellulose fabrics dyed with aqueous solutions of Artemisia extracts; however, the dyed nylon fabrics were brown. Fabrics dyed with ethanol-extract added solutions were greener and had higher antimicrobial properties than those dyed with aqueous solutions; however, they faded and lost their antimicrobial properties after laundering. Fabrics regained their antimicrobial properties (especially against S. aureus) by the spraying of Artemisia ethanol extract; therefore, the application of Artemisia ethanol extract onto underwear is expected to relieve atopic dermatitis.

  • This study was conducted to evaluate the allelopathic effect of aqueous extract of Ganghwa domestic mugwort (Artemisia spp.) on vegetables and its related allelo-chemicals. When the receptor vegetables, such as Chinese cabbage, lettuce, and red radish, were treated with aqueous extract obtained from Sajabalssuk (A. $sp^*I$), Ssajuarissuk (A. $sp^*II$) or Ssajarissuk (A. $sp^*III$), their germination rate, leaf number, plant height, and root length were restricted with increasing concentration of aqueous extract. Allelopathic effect was the highest in radish, than lettuce and Chinese cabbage in order. The growth of topplant were more inhibited then root growth observing in restriction of plant height, root length, and chlorophyll contents. The plant height, the root length of red radish were 53.3 and 61.2% and their fresh weights were 19.8 and 26.4% compared to those of controls, respectively. A. $sp^*III$ showed the highest allelopathic effect among the donor plants. In HPLC analysis, 7 phenol compounds were identified in A. $sp^*I$ and A. $sp^*II$, and, in A. $sp^*III$, and hydroxybenzoic acid and phenylacetic acid were further identified as allelochemicals. It is considered that their plant growths were variously inhibited by the amounts and types of allelochemicals in aqueous extracts. To increase the productivity of farm land after cultivation of mugwort, these results can be useful to select the following field crops.
