To develop the continuous water quality monitoring system using the daphnid Daphnia sp., the growth of test animal, sensitivity, and behaviour response of toxicants were observed. Growth of test animal significantly increased with increasing the food density under the 90~105 mg/L ($CaCO_3$) hardness, except the concentration of food (Chrollela sp.) was exceeded than optimal food supply. Behaviour responses of test animals were continuously analyzed by changes of fractal dimension value (FDV). The FDV sharply decreased after exposure to the concentrations of 0.13 mg/L copper, 0.06 mg/L lead, and 0.38 mg/L cadmium. In these concentrations, mortality and abnormal behaviour of daphnids exhibited within ca. 1.0-h after exposure. Comparison of 24-h $LC_{50}$ values with other zooplankton species indicated that sensitivity of the Daphnia sp. was higher than most zooplankton for lead, and brain shrimp, rotifer, and water flea (Ceriodaphnia dubia, D. magna) for copper, and brain shrimp, water flea (D. lumholzi), and amphipod for cadmium. Based on the above experimental results, significant relationship between toxicity and behaviour response of Daphnia sp. was supported the high potential of water quality monitoring system. Consequently, behavioural monitoring method in this study suggests a good estimation tool for detection of the discharged toxicants in water body and for ecotoxicological assessment aquatic organisms.
Leachate from municipal solid waste (MSW) landfill, effluent from leachate treatment plant, and ground water sample from a monitoring well near landfill site were tested for an acute toxicity. Microtox toxicity test was used for testing the acute toxicity of leachate and other samples. EC$_{50}$ values which a concentration of pollutant for reducing 50% light output from luminescent bacteria, Photobacterium phosphoreum were determined to assess the toxicity of pollutants as well as the relative toxicity. In addition, characteristics of leachate were studied and compared to those of phenol and pentachlorophenol (PCP) which are typical aquatic toxic pollutants. For leachate, EC$_{50}$ for 30 min incubation was 10.8%, while for phenol and PCP, 46 ppm and 1.2 ppm, respectively. the relative toxicity of treated leachate by in situ aeration with activated sludge was reduced to more than 75% of toxicity of the untreated leachate. Microtox toxicity test was failed to figure out EC$_{50}$ values for groundwater from a monitoring well since the relative toxicity of the unconcentrated sample was too low to estimate EC$_{50}$. Addition of activated carbon to leachate was reduced the relative toxicity. The reduction Pattern of the relative toxicity of leachate by mechanical aeration was similar to that of PCP, but different from that of phenol. These findings suggest that the toxicity of leachate may come from PCP-like toxic compounds rather than phenol-like one. In conclusion, the process of aeration with activated sludge might be very important to reduce the environmental toxicity of leachate. And Microtox test could be a reasonable bioassay for screening and monitoring the environmental toxicity of leachate from municipal solid waste landfill as well as for determining the reduction efficiency of the leachate toxicity by various treatment processes in leachate treatment plant.

Objects: Chironomus riparius, a non-biting midge (Chironomidae, Diptera), is extensively used as a model organism in aquatic ecotoxicological studies, and considering the potential of C. riparius larvae as a bio-monitoring species, little is known about its genome sequences. This study reports the results of an Expressed Sequence Tags (ESTs) sequencing project conducted on C. riparius larvae using 454 pyrosequencing. Method: To gain a better understanding of C. riparius transcriptome, we generated ESTs database of C.ripairus using pyrosequencing method. Results: Sequencing runs, using normalized cDNA collections from fourth instar larvae, yielded 20,020 expressed sequence tags, which were assembled into 8,565 contigs and 11,455 singletons. Sequence analysis was performed by BlastX search against the National Center for Biotechnology Information (NCBI) nucleotide (nr) and uniprot protein database. Based on the gene ontology classifications, 24% (E-value${\leq}1^{-5}$) of the sequences had known gene functions, 24% had unknown functions and 52% of sequences did not match any known sequences in the existing database. Sequence comparison revealed 81% of the genes have homologous genes among other insects belonging to the order Diptera providing tools for comparative genome analyses. Targeted searches using these annotations identified genes associated with essential metabolic pathways, signaling pathways, detoxification of toxic metabolites and stress response genes of ecotoxicological interest. Conclusions: The results obtained from this study would eventually make ecotoxicogenomics possible in a truly environmentally relevant species, such as, C. riparius.
Crucian carp (Carassius auratus) has been used as the sentinel species for POPs (Persistent Organic Pollutants) monitoring in aquatic environment. However, there is little information for dioxin toxicity and especially, early life stage toxicity in crucian carp have been never carried out. In this study, we investigated several toxic effects for 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) in fertilized egg obtained by natural fertilization from crucian carp. The embryos at 3 h post-fertilization (hpf) were treated with 0.039, 0.156, 0.625, and 2.5 (${\mu}g/L$) TCDD by waterborne exposure for 60 minutes and changed with fresh water 2 times per day. Fertilized eggs started hatching at 51 hpf and TCDD exposed embryo showed decrease of hatching rate in a dose-dependent manner at 75 hpf. Pericardial edema was continuously observed in larvae exposed to TCDD from hatching start time (51 hpf), followed by the onset of mortality. In addition, AhR-related gene, CYP1A was clearly increased by TCDD in a dose dependent manner. These results indicated that fertilized eggs obtained from crucian carp have the TCDD related gene regulation and a distinct TCDD developmental toxicity syndrome by TCDD exposure. Therefore, we suggested that early life stage test in crucian carp could be used as test methods on dioxins toxicity.
During the last decade, perfluorinated compounds (PFCs) have gained more attention due to their toxicity and global distribution. The aim of this study was to examine the distribution and bioaccumulation of perfluorinated compounds (PFCs) in aquatic wildlife effected from a sewage treatment plant. The concentrations of 12 PFCs were determined in water, sediment and fish samples. PFOS were predominantly detected in both ambient environment and fish. In fish, the concentration of PFCs in blood was the highest (i.e., 112.47 ng/mL wet-wt. PFOS) in comparison to other tissues. However, PFOA and PFHpS were highly detected in gonad as 3.87 and 4.58 ng/g wet-wt., respectively. The bioconcentration factor (BCF) of PFCs was greatest in the blood > liver${\cong}$gonad > kidney > gill, and lowest in the muscle tissue. The BCFs of PFUnDA (39,000), PFDA (2,700) and PFOS (1,100) were rated as high values based on wet weight concentration. BCFs increased with increasing the length of the perfluoralkyl chain.
Acute and chronic toxicities of sediment elutriate and surface water samples collected at Lake Shihwa were evaluated using standard toxicity testing organisms including Vibrio fischeri, Daphnia magna and Moina macrocopa. Acute exposure resulted in toxic effects in all surface water or sediment elutriate samples, except for those collected from the reed swamp and Okgu stream. The rainy season influenced the toxicity of the water samples, presumably either by dilution of point discharge or through introduction of non-point source contaminants through runoff. In the sediment, elutriate and surface water samples, copper was detected above potentially lethal concentration, which may in part explain the observed toxicity. Considering acute toxicities of the surface water streams that direct to the Lake Shihwa, efforts should be warranted to control and reduce discharge of point and non-point sources along Lake Shihwa.
Objectives: For suitable risk management of the domestic aquatic environment, it is necessary to conduct toxicity tests using species native to Korea. In the present study, we performed toxicity ring tests using endemic freshwater arthropoda Neocaridina denticulata and evaluated its validity and reproducibility as an international standard test species. Methods: To evaluate the sensitivity levels of N. denticulata to hazardous chemicals, toxicity values for several chemicals were compared with other standard test species. Intra- and inter-laboratory acute toxicity tests were performed both within a single laboratory and among four laboratories respectively using 3,4-Dichloroaniline, which is generally used as a reference test substance in fish toxicity tests. In addition, intra- and inter-laboratory coefficient of variations (CVs) were calculated to evaluate reproducibility based on the estimated toxicity values. Results: The sensitivity of N. denticulata to several chemicals was found to be similar with D. manga, indicating that the species is valid as a test species. The CVs of the intra- and inter-laboratory tests were 22.946% with four qualified runs and 8.828% among the four laboratories, respectively. Conclusions: N. denticulata serves in an important role in the food chain of Korean aquatic ecosystems and also inhabits several other Asian countries. Since the validity and reproducibility of the species were confirmed as a toxicity test species in this study, further efforts are needed to establish N. denticulata as the international standard test species for the appropriate risk assessment of aquatic ecosystems at home and abroad.
Benzoyl peroxide is a high production volume chemical, which was produced about 1,375 tons/year in Korea as of 2001 survey. Most of them are used as initiators in polymerization, catalysts in the plastics industry, bleaching agents for flour and medication for acne vulgaris. The substance is one of the sever chemicals of which human and environmental risks are being assessed by National Institute of Environmental Research under the frame of OECD SIDS Program. It has a melting point of 104-106 $^{\circ}C$ and has solubility of 9.1 mg/1 in water at 25 $^{\circ}C$. The substance was readily biodegradable (83 % after 21days) and had toxic effects to aquatic organisms. The range of 72 hr-EbC50 (biomass) for algae was 0.07-0.44 mg/1 and 48 hr-EC50 for daphnia was 0.07-2.91 mg/1. The LC50 of acute toxicity to fish was 0.24-2.0 mg/1. Although the toxic effects of benzoyl peroxide to aquatic organisms were investigated, environmental monitoring data were not studied. In this study, distribution of the chemical among multimedia environment was estimated using EQC model based on the physical-chemical properties to evaluate the risk of benzoyl peroxide in environment. In level I, II calculation the chemical was distributed to soil (68.3 %) and water (28.7 %). In level III calculation it was primarily distributed to soil (99.9 %) and overall residence time of 3.4 years was estimated. Benzoyl peroxide could be persistent in environment.
Microalgae are functional foods because they contain special anti-aging inhibitors and other functional components, such as ecosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and omega-3 polyunsaturated fatty acids. Many of these functional dietary components are absent in animals and terrestrial plants. Thus, microalgae are widely utilized in human functional foods and in the feed provided to farmed fish and terrestrial livestock. Many marine organisms consume microalgae, often because they are in an appropriate portion of the cell size spectrum, but also because of their nutritional content. The nutritional requirements of marine organisms differ from those of terrestrial animals. After hatching, marine animals need small live forage species that have high omega-3 polyunsaturated fatty acid contents, including EPA and DHA. Euglena cells have both plant and animal characteristics; they are motile, elliptical in shape, 15-500 μm in diameter, and have a valuable nutritional content. Mixotrophic cell cultivation provided the best growth rates and nutritional content. Diverse carbon (fructose, lactose, glucose, maltose and sucrose) and nitrogen (tryptone, peptone, yeast extract, urea and sodium glutamate) supported the growth of microalgae with high lipid contents. We found that the best carbon and nitrogen sources for the production of high quality Euglena cells were glucose (10 g L–1) and sodium glutamate (1.0 g L–1), respectively.
The quality and quantity of live food sources strongly influence the success of fish production in farming operations. Thus, critical studies of live forage species are a crucial element for progress in fish aquaculture. The fat content of food is an especially important determinant of growth in marine fish. Omega-3 highly unsaturated fatty acids (HUFA) are essential components of diet that determine the nutritional value of larval fish. Euglena is a protist that has potential as a forage species. These single-celled organisms have plant and animal characteristics they are motile, elliptical in shape and 15–500 μm in diameter. Their nutritional content is excellent, but most studies have focused on cells raised in autotrophic culture. We therefore examined differences in the lipid and fatty acid contents, and the growth of Euglena cells grown under autotrophic, heterotrophic, and mixotrophic conditions. Biomass production reached 15.03 g/L, 12.28 g/L, and 3.66 g/L under mixotrophy, heterotrophy, and autotrophy, respectively. The proportional n-3 HUFA content differed among culture methods: 10.04%, 5.80% and 10.01% in mixotrophic, heterotrophic and autotrophic cultures, respectively. Mixotrophy was to be the best form of cultivation for improving the growth and nutritional content of Euglena.
