Dietary Intake and Potential Health Risk of Polycyclic Aromatic Hydrocarbons (PAHs) via Various Marine Organisms in Korea

  • Sixteen polycyclic aromatic hydrocarbons (PAHs) were analyzed in seventy marine organisms (40 species) from the Korean coast. PAHs were present in all the organisms. The level of total PAHs in the organisms varied from 0.45 to 224 ng/g dry weight and the carcinogenic PAHs varied from 0.05 to 49.8 ng/g dry weight. The PAHs residues according to the marine organisms showed a highest content in bivalve species, and followed by crustaceans, cephalopods, fish and gastropods. Human dietary intake of total PAHs through marine organism in Korea was estimated to be 4.12 ng/kg body weight/day and 0.67 ng/kg body weight/day for carcinogenic PAHs. The relative contributions of individual species to the total dietary intake of PAHs were in the order of bivalves $(53.4{\%})$, fish $(21.9{\%})$, crustaceans $(15.3{\%})$, cephalopods $(8.8{\%})$ and gastropods $(0.6{\%})$. Daily dietary intake of $PAH_{TEQ}$ expressed as a TEQ value was estimated to be 0.13 pg TEQ/kg body weight/day, which did not exceed a tolerable daily intake (TDI) proposed by the KFDA and the WHO as well as the UK toxicity committee. Lifetime cancer risk due to ingestion of marine species by the Korean adult was evaluated using the equation estimating exposure of food ingestion. Although approximately $23{\%}$ of cumulative frequency of the sampled marine species exceeded the cancer risk guideline, lifetime cancer risk associated with marine organism consumption was negligible. Results indicate that dietary intake of PAHs through the consumption of the Korean marine organisms seems to be safe for human ingestion with negligible cancer risk.

  • The short-term acute toxicities of mercury, cadmium and copper for the clam, Meretrix lusoria were determined from 28 June to 15 July, 1978. In the test with mercury and copper, the rate ot mucus excretion increased gradually at a higher concentration. But the clams did not excrete mucus in the solutions of cadmium and natural sea water. The rate of mucus excretion in mercury was $42.9\%$, and that in copper was $14.3\%$ in a test solution of 1mg/l. mercury was more toxic than copper. The median lethal concentration after 96 hours (96 hr-Lc 50) was 0.67mg/l in mercury, 7.04mg/l in copper and 7.10mg/l in cadmium. Consequently it was found that mercury was the most toxic substance and cadmium was the least. meanwhile, it was considered that exposure time by stimulation in a fixed concentration caused the test animals to respond.

Cryopreservation of Common Korean Bitterling Acheilognathus signifer Sperm (묵납자루(Acheilognathus signifer) 정자의 냉동보존)

  • This study aimed to find an optimal diluent and cryoprotective agent (CPA) during cryopreservation of common Korean bitterling Acheilognathus signifier sperm. The bitterling is an endangered species in Korea, and this study will enable conservation and further technical development of artificial seed production. We tested the effects of cryopreservation and toxicity by the type of diluent and CPA. The optimal combination of diluent and CPA for cryopreservation was 300 mM glucose+10% methanol, resulting in a survival rate and sperm activity index (SAI) of $96.3{\pm}1.5%$, and $3.0{\pm}0.0$ respectively, and no significant difference compared to fresh sperm. The survival rate and SAI of post-thawed sperm was $9.7{\pm}1.5%$, $0.8{\pm}0.3$ respectively, which was significantly higher than had been achieved with other diluents and CPAs.

Toxic Algal Bloom Caused by Dinoflagellate Alexandrium tamarense in Chindong Bay, Korea

  • Monospecific red tide by a toxic dinoflagellate belonging to the genus Alexandrium occurred at Chindong Bay in the southern coast of Korea and continued from April 6th to 15th in 1997. The ratio of its cell number to total phytoplankton cell number was much higher than $95\%$. This organism was identified as Alexandrium tamarense, although slight morphological differences were found comparing to the original and successive descriptions of the species. We found neither anterior nor posterior attachment pores in these cells of the bloom population. The occurrence of red tide caused by A. tamarense was first reported in Korea. Its plate formula is Po, Pc, 4', 6"c, 8s, 5"' and 2"". Thecal plates are thin with pore-like ornamentation. In those plates, the anterior part of the first apical plate (1') is narrower and its posterior end has sometimes a block-like accessory, but this variation was considered within the range of the morphological variability of this taxon. The cell density during the red tide exhibited a wide range of variation by the depth of water column, ranging from $2\times10^6$ cells$l^{-1}$ to $5\times10^6$ cells·$l^{-1}$. Water temperature varied from 11.8 to $12.3^{\circ}C$. Toxicity of A. tamarense during red tide was measured as $8.8\times10^5$. $MU\;\cdot\;cell^{-1}$ by mouse bioassay.

Bioactive Components from Red Tide Plankton, Cochlodinium polykrikoides (Cochlodinium polykrikoides 적조 조체의 생리활성 성분)

  • Large amounts (300 grams) of natural red tide plankton, Cochlodinium polykrikoides, was collected at the Yokji island coastal waters, Kyeongnam, Korea, on October, 1993. Using the bioassay systems, bioactive materials were screened from methanol extracts of C. polykrikoides. Live C. polykrykoides was toxic to fishes, however, the water soluble and chloroform soluble fraction of their methanol extracts did not shown ichthyotoxicity (5 mg/ml), and toxicity to mice (50 mg, i.p,). These fractions did not show any peaks corresponding to paralytic shellfish toxins or diarryhetic shellfish toxins on the fluorometric HPIC chromatograms. Neither fractions did not show antibiotic activities by paper disk (10 mg/disc) test and chloroform soluble fraction showed only $20\%$ growth inhibition activity on the Lymphoid P-388 at the concentration of $180\;{\mu}g/ml$. Hemolytic activity was detected by both fractions. Fatty acid analysis by GC, GC/MS and proton NMR showed that the chloroform soluble fraction composed of $25.3\%$ of DHA (dorosahexaenoic acid) and $15.3\%$ of IPA (eicosapentaenoic acid) as the hemolytic components.

Development of Physiological and Biochemical Bioindicators of Barnacle, Megabalanus rosa for Marine Pollution Assessment (해양오염 평가를 위한 빨강따개비(Megabalanus rosa)의 생리생화학적 오염지표의 개발)

  • Using Megabalanus rosa a sessile filterfeeder, its scope for growth (SFG) by analysis of energy budget were examined and free amino acids composition of whole body tissue were analyzed under the exposure to different concentrations of Hg Cu and Cd. The $96\;hr-LC_{50}$ of the barnacle after 96 hr exposure to Hg, Cu, and Cd were 0.220, 0.269 and $1.380\;mgL^{-1}$, respectively. Hg and Cu showed stronger toxicity than Cd, while Hg and Cu had similar influence on the survival of the barnacle. SFG of the barnacles exposed to sublethal concentrations of mercury was 18.936 $Jgdrywt.^{-1}hr^{-1}$ in control group and as increase of mercury concentration the SFG remarkably reduced to 0.041 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}$ concentration of Hg. In the case of Cu, the SFG was 29.841 $Jgdrywt.^{-1}hr^{-1}$ in control group and as increase of concentration, the SFG remarkably reduced to -8.304 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}$ concentration. In Cd the SFG was 15.852 $Jgdrywt.^{-1}hr^{-1}$ in control group, and as increasing concentration, the SFG remarkably reduced to -19.490 $Jgdrywt.^{-1}hr^{-1}\;at\;0.1\;mgL^{-1}.$ Content of free amino acid (FAA) of whole body tissue of the barnacle was 45084 $mgkg^{-1}$ in control group, but it was reduced remarkably to 28,130, 37,500 and 37,106 $mgkg^{-1}$ at 0.1 $mgL^{-1}$ concentration of Hg and Cu, and 0.4 $mgL^{-1}$of Cd, respectively. Sum of threonine + serine was 1,334 $mgkg^{-1}$ if control but reduced remarkably to 1,223, 849 and 888 $mgkg^{-1}$ at 0.1 $mgL^{-1}$ of Hg and Cu, and 0.4 $mgL^{-1}$ of Cd, respectively.

Effects of Sediment Elutriates on the Early Reproductive Outputs in the Pacific oyster, Crassostrea gigas

  • This is a subsequent study to our previous finding that Pacific oysters, Crassostrea gigas, gained a so-called upper plateau concentration, around 30,000 ng/g dry weight digestive gland for benzo(a)pyrene, showed reproductive behavior but produced their ensuing reproductive outputs damaged. A serial dilution of sediment elutriates from Jinhae Bay, Korea, where pollution was progressive, were exposed to gametes of the Pacific oyster for 0, 5, 10, 20, 30, and 60 min to detail the pollutant effects on very young specimens. There was an apparent critical dilution over which adverse effects are evident. This was $10\%$ of the present sediment elutriate, corresponding to 0.3 ng/g on the basis of total polycyclic aromatic hydrocarbons (PAHs) for the oyster. Within the dilution the embryonic development was not influenced by the duration of exposure to its gamete stage. At higher dilutions over the critical dilution, occurrence of abnormality increased dependent on the pollutant dilution and the duration of exposure. Similar trends were also found in larval mortality. However, overall, the chemical toxicity was more significant to morphogenesis than to survival, suggesting a potential recruitment of the pollutants-induced abnormal larvae in the wild population to threaten the population integrity.

GC-MS Analysis of Organophosphorus Pesticide Residue in Seawater From the Kwangyang Bay, Korea (광양만 해수 중 유기인 잔류 농약성분의 GC-MS 분석)

  • Sea water samples collected in August, 1994 from 20 stations in the Kwangyang Bay were analyzed by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM) to investigate persistence and distribution pattern of four organophosphorus pesticides (DDVP, Diazinon, IBP, EDDP). Except for IBP, the contamination by DDVP, Diazinon, and EDDP in marine aquatic environment in Korea has not been reported previously. In this study, however, all these four pesticides were detected in all stations (except DDVP) and their concentrations were in ng/L level. The concentrations ranged from detection limit to 15.3ng/L for DDVP, 1.8-27.7ng/L for Diazinon, 7.3-63.5ng/L for IBP, and 22.2-1100.1ng/L for EDDP. It is noteworthy that the measured concentrations of IBP and EDDP in this study would be much lower than usual, since the use of IBP and EDDP was less than 50% of average annual consumption due to unusually dry and hot weather condition in the summer of 1994. It was very surprising to find that the highest concentrations of organophosphorus pesticides were observed at stations near Daesa Streamlet instead of Seomjin River, which has more point source of the pesticides. This result suggests that the small river discharge during heavy ram period in summer can give harmful effect on marine biota (both wild and aqua-cultured) with its organophosphorus pesticide residue, despite of their short residence time in aquatic environment. In order to protect the marine life properly from acute toxicity of the organophosphorus pesticides, it needs to be emphasized that monitoring the level of agricultural pesticides in river run-off should be done during active consumption period rather at regular intervals.

Study on Anti-estrogenic Activity of DEHP as an Endocrine Disruption Chemical (내분비 교란성 DEHP의 항-에스트로젠 활성에 관한 연구)

  • Di-2-ethylhexyl phthalate (DEHP), is a widely used plasticizer known to be a suspected endocrine disrupter, but its exact effects on aquatic organisms are not yet known. When Japanese medaka (Oryzias latipes) were exposed from the time of hatching to 3 months of age to an aqueous DEHP solution at nominal concentrations of 1, 10, and 50 $\mu\textrm{g}$/l, DEHP treated female fish showed distinct reproductive effect. And the midge (Chironomus riparius.). an aquatic invertebrate, was exposed to DEHP to evaluate the effects on reproductive processes via sediment toxicity. The test endpoints included emergence, sex ratio, fecundity, and the viability of F1 offspring egg ropes. The result implied that the normal developmental and/or reproductive processes in C. riparius had been disrupted when exposed to DEHP, the effect also being displayed in the next generation. In summary, DEHP hinders the development of reproductive organs in the female Japanese medaka and C. riparius.

Polychlorinated Dibenzo-p-dioxins and Polychlorinated Dibenzofurans in Marine Sediments from Mokpo Coastal Water of Korea

  • Concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) were determined in marine sediments from Mokpo coastal water of Korea. Total and toxic equivalent (TEQ) concentrations of PCDD/Fs ranged from 19.4 to 175pg/g dry weight and ranged from 0.195 to 2.87 pg WHO-TEQ/g dry weight, respectively. These PCDD/F concentrations were below the safety sediment value (20 pg TEQ/g dry weight) of chronic toxicity. A highly significant correlation between the concentrations of PCDD/Fs and total organic carbon (TOC) in marine sediments was observed. Overall PCDD/F concentrations in the sediments were elevated in rivers and at inner locations close to harbors, indicating that these contaminants derived from local discharges of human activities. PCDD/F concentrations measured in our study were lower than those in several industrialized areas in Korea. Nonparametric multidimensional scaling (MDS) ordination showed that combustion processes from industrial complexes are the major source of PCDD/Fs in Mokpo coastal water.
