A protease inhibitor from fish eggs was fractionated using chromatographic methods. The fractionation efficiency was evaluated in terms of specific inhibitory activity (SIA, U/mg), purity (fold), total inhibitory activity (TIA, U), and recovery (%). The protease inhibitor (PI) from egg extracts of skipjack tuna (ST Katsuwonus pelamis), yellowfin tuna (YT Thunnus albacares) and Alaska pollock (AP Theragra chalcogramma) was fractionated using Sephadex G-50 gel filtration and DEAE-Sepharose CL-6B anion exchange chromatography based on protein size exclusion and net charge, respectively. Fractions exhibiting strong inhibitory activity were contained in the 30-50 kDa fraction on gel filtration and in the range of 0.4-0.7 M NaCl gradient fraction on anion exchange chromatography. The respective TIA and percent recovery of the fraction obtained with gel filtration toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 2,758.7 U and 29.6% for ST, 1,005.5 U and 25.6% for YT, and 1,267.5 U and 26.0% for AP. Gel filtration chromatography was more effective at fractionating PI than using ion exchange chromatography. These results suggest that fish eggs act as serine protease inhibitors and might be useful for protease inhibition in foodstuffs.

Background: Large rivers are ecological treasures with high human value, but most have experienced decades of degradation from industrial and municipal sewage, row-crop agricultural practices, and hydrologic alteration. We reviewed published analyses of long-term fish diversity publications from three intensively managed large river ecosystems to demonstrate the conservation potential of large river ecosystems. Results: We show how the incorporation of recent advances in river concepts will allow a better understanding of river ecosystem functioning and conservation. Lastly, we focus on the Wabash River ecosystem based on high conservation value and provide a list of actions to maintain and support the ecosystem. In the Wabash River, there were originally 66 species of freshwater mussels, but now only 30 species with reproducing populations remain. Although there were multiple stressors over the last century, the largest change in Wabash River fish biodiversity was associated with rapid increases in municipal nutrient loading and invasive bigheaded carps. Conclusions: Like similarly neglected large river systems worldwide, the Wabash River has a surprising amount of ecological resilience and recovery. For instance, of the 151 native fish species found in the 1800s, only three species have experienced local extinctions, making the modern assemblage more intact than many comparable rivers in the Mississippi River basin. However, not all the changes are positive or support the idea of recovery. Primary production underpins the productivity of these ecosystems, and the Wabash River phytoplankton assemblages shifted from high-quality green algae in the 1970s to lower less nutritional blue-green algae as nutrient and invasive species have recently increased. Our recommendations for the Wabash River and other altered rivers include the restoration of natural hydrology for the mainstem and tributaries, nutrient reductions, mechanisms to restore historical hydrologic patterns, additional sediment controls, and improved local hydraulics.
Maturity and spawning of the snailfish, Liparis ochotensis was investigated based on samples collected by gill net in the East Sea of Korea from December 2008 to December 2009. The average total length of L. ochotensis was 63.4 cm and 64.1 cm for females and males, respectively. Gonadosomatic index (GSI) began to increase in July, reaching a maximum in November, then began to decrease in December. Monthly changes in the GSI values of L. ochotensis were similar for females and males. Annual reproductive cycle of this species can be divided into five successive stages in females: early growing stage (May), late growing stage (June to July), mature stage (August to September), ripe and spent stage (September to December), and recovery and resting stage (January to April). Males show four successive stages; growing stage (May to July), mature stage (August to September), ripe and spent stage (September to December), and recovery and resting stage (January to April). Relationship between total length (TL) and fecundity (F) was F=$0.00003TL^{4.002}$ ($R^2$ = 0.703), and F increased with TL. Total length at 50% group maturity was estimated to be 55.8 cm. Our findings suggest that the spawning period takes place from September to December, with the main spawning period occurring from October to December.
Due to the shortage of the fingerling/juvenile mud loach, Misgurnus mizolepis in Korea, these fish have been imported from China. However, the mortality rate during and after their transportation is very high. In this study, we examined various physiological and histological parameters to evaluate the effect of salt treatment on the survival and recovery of mud loaches in holding farms during the quarantine process. Glucose, osmolality, $Na^+$, $Cl^-$, and histological changes were assessed for three different salinities. Non-treated fish (control 0.0%) exhibited lower levels of osmolality, and $Na^+$ and $Cl^-$ concentrations compared with those kept in solar salt solution (0.5% and 1.0%). Glucose levels in control fish were higher than those in fish exposed to 0.5% and 1.0% solar salt solution. Histologically, control fish showed thinner epidermis of skin, branchial hyperplasia and lamellar fusion with an abundance of eosinophilic granule cell-like cells. After solar salt solution treatment, damaged gill structures in the fish almost recovered within 5 days. The present study demonstrates that mud loaches transported from China suffer from skin and gill damage and physiological dysfunction which may increase the mortality and morbidity. Moreover, saline treatment might alleviate the stress responses and ionic/osmotic imbalances, and help heal gill damage.
This study investigated the survival rates and physiological responses of Korean rockfish, Sebastes schlegeli, in live fish containers ($8^{\circ}C$, 33 psu) for 18 days. The survival rate was 99% and 97% in the control and experimental groups, respectively. Hematocrit and hemoglobin did not differ significantly between the control and experimental groups. Glucose and cortisol rose immediately on the first day of containment, but both gradually normalized. Aspartate aminotransferase and alanine aminotransferase did not differ significantly between the two groups after recovery. $NH_3$ continued to rise after the first day, but decreased to a level not significantly different from that of the control group during the recovery period. Plasma ions and osmolality did not change abnormally. The hemocyte population was not significantly different from that of the control. The ratios of apoptotic and necrotic cells indicated no specific variation in hemocyte viability. The histological changes in the skin and gills were not significantly different from those seen in the control. The experimental data obtained in this study suggest that live fish containers may be used to transport Korean rockfish without significantly affecting their physiology or survival.

Chum salmon (Oncorhynchus keta) is a species of anadromous salmonid inhabiting coastal rivers in the North Pacific and the Arctic in the Bering and is the most widely distributed among Pacific salmon species. It is an important fish species in Korea as the salmon releasing project is being actively carried out. This study was conducted to investigate changes in the physiological activity of O. keta according to water temperature. Three experiments were conducted according to the water temperature and period, and the plasma concentrations of cortisol and glucose were analyzed from the blood samples of the experimental groups. Experiment I is a short-term water temperature experiment, in which water temperature stimulation was given for 4 hours at water temperatures of 12℃, 16℃ (control), 20℃, and 24℃, and a recovery period was given for 4 hours. Experiment II is an experiment in which water temperature stimulation was given for 24 hours, 48 hours, and 72 hours at a high temperature of 24℃, and a recovery period was given for 12 hours, respectively. Experiment III is a long-term water temperature experiment, in which the water temperature was 12℃, 16℃ (control), 20℃, and 24℃ for 8 weeks. As a result of the experiment, in Experiment I, there was no significant difference in the survival rate between the experimental groups, but the concentration of cortisol and glucose in the plasma according to the set water temperature showed a significant difference. In Experiment II, there was no significant trend according to the maintenance time of the high-temperature state, but as the temperature increased, the plasma cortisol and glucose levels significantly increased compared to the control group. In Experiment III, all of the experimental group C (24℃) died in the 1st week, and there was no significant difference in the plasma glucose at the 1st and 8th weeks among the remaining experimental groups.
After an outbreak of viral disease in an aquafarm, release of virus (es) from infected fish into environmental seawater has been suspected. In the present study, we utilized a negatively charged membrane (HA type) as an efficient method for concentration and detection of fish pathogenic viruses, specifically, megalocytivirus and viral hemorrhagic septicemia virus (VHSV) present in field-collected seawater samples or inoculated into seawater artificially. Positively charged viruses adsorbed onto the negatively charged membrane and were eluted with 1 mM NaOH (pH 10.5) following rinsing with 0.5 mM $H_2SO_4$ (pH 3.0). Megalocytivirus and VHSV particles isolated using anegatively charged HA membrane from seawater inoculated with each virus at a concentration of 10 viral particles/mL were of sufficient quantity to show positive results in atwo-step PCR (or RT two-step PCR); however, despite it being negatively charged, a cellulose acetate (CA) membraneshowed negative results. In quantitative PCR, the detection limits of the HA membrane for megalocytivirus and VHSV in seawater were 1.20E+00 viral particles/mL and 1.22E+01 viralparticles/mL, respectively. The calculated mean recovery yields from 1 L seawater spiked with known concentrations of megalocytivirus and VHSV particles were 28.11% and 23.00%, respectively. The concentrate of a 1-L sample of culturing seawater from the aquatank of flounder suffering from VHSV showed clear positive results in PCR when isolated with an HA, but not a CA, membrane. Thus, viral isolation using an HA membrane is a practical and reliable method for detection of fish pathogenic viruses in seawater.
나일 틸라피아에 실험적 출혈성 빈혈 (EHA)과 용혈성 빈혈 (ELA)을 제작하여 빈혈 과정과 회복 과정에서의 병리학적 차이를 알기 위해서 본 실험을 수행하였다. EHA는 어체중의 약 1%에 해당하는 혈액을 반복 채혈하여 유도하였다. ELA는 1% phenylhydrazine을 2회 복강 주사하여 유도하였다. 회복 과정은 16일째부터 49일째까지로 하였다. 빈혈 및 회복 기간 동안 아가미의 빈혈성 퇴색 외 특기할 임상 소견은 관찰되지 않았다. 두 빈혈 경우 모두, 12일째에 세포질 염색성이 없는 적혈구들이 출현하기 시작하였으며 EHA의 경우 49일째까지, ELA의 경우 20일째까지 출현하였다. 빈혈 유형은 빈혈 유도 기간에는 EHA의 경우 정구성 저혈색소성이었고 ELA의 경우 대구성 정혈색소성이었으며, 회복 기간에는 두 경우 모두 정구성 저혈색소성이었다. 간장에서 EHA의 경우에 12일째에서 38일째까지 중심정맥 중심성 지방변성이 확인되었고, ELA의 경우에 12일에서 26일째까지 중심정맥 중심성 초자 변성이 확인되었다. EHA의 경우 12일째부터 26일째까지 두신에서 조혈활성 소견이 확인되었으며, ELA의 경우에서는 2일째부터 12일째까지 관찰되었다. 두신에서 적혈구 탐식 대식세포들이 EHA의 경우 16일째부터 38일째까지, ELA의 경우에서는 2일째부터 12일째까지 관찰되었다. 비장 협조직의 비후 소견이 EHA의 경우 12일째부터 26일째까지, ELA의 경우에서는 2일째부터 20일째까지 확인되었다. ELA의 경우 2일째에서 49일째까지 두신과 비장에서 hemosiderin 축적이 현저하였다. 49일째에 두 빈혈 경우 모두 Ht는 회복되었지만 Hb는 완전한 회복을 보이지 않았다.
버들치 Rhynchocypris oxycephalus와 버들개 R. steindachneri를 대상으로 마취 수온 10도C, 15도C 및 20도C에서 염산리도카인(염산리도카인/NaHCO하(3))의 마취 효과를 조사하였다. 마취 시간과 회복 시간을 고려시 효과적인 염산리도카인 농도는 버들치에서 300ppm ($20^{\circ}C.$), 400ppm ($15^{\circ}C.$) 및 600ppm ($10^{\circ}C.$) 이었고 버들개에서 400ppm ($20^{\circ}C.$), 500ppm ($15^{\circ}C.$) 및 600ppm ($10^{\circ}C.$) 이었다. 마취시간과 회복시간에 있어 버들치 속 2종은 염산리도카인 농도 및 마취 수온 의존적이었다. 각 염산리도카인 농도에서 개체 크기가 증가할수록 마취 시간이 길어졌으며, 개체 크기가 큰 군은 마취 농도가 증가할수록 회복 시간 역시 길어졌다. 본 연구 결과 염산리도카인은 버들치 속어류 마취에 효과적인 것으로 판명되었다.
세팔로스포린계열의 반합성 항생제인 세팔렉신은 법적 허가를 받은 여러 나라의 양식장에서 사용되어 왔다. 따라서 이 항생제는 갑각류 중 흰다리새우와 같은 어류를 제외한 다른 수생 생물 종에서 광범위하게 사용할 수 있다. 본 연구는 흰다리새우에서 세팔렉신의 잔류기간 설정을 위해 잔류분석법을 개발하였고, 이를 바탕으로 잔류실험을 수행하였다. 분석법 개발을 위하여 이미 존재하고 있는 여러 식용동물 조직에서의 분석법을 바탕으로 반복 시도하여 고성능 액체 크로마토그래피와 질량분석기(high-performance liquid chromatography-tandem mass spectrometry, HPLC-MS/MS)를 이용한 분석법을 개발하였다. 그 결과, 0.1 mg/kg의 농도에서 회수율이 81.79%, C.V.값은 8.2%로 Codex guideline에서 권장하는 회수율 및 C.V.에 충족하는 결과를 나타내었다. 개발한 분석법의 적정성을 검증 후, 흰다리새우에 세팔렉신을 경구투여 한 뒤 잔류량을 분석하여 흰다리새우 조직에서의 적용 가능성을 확인하였다. 그 결과, 투여 종료 3일 후에 대부분의 근육 시료에서 정량한계(0.05 mg/kg)이하로 검출되었고, 투여 종료 14일 후에 대부분의 간췌장 시료에서 정량한계 이하로 검출되었다. 특히 본 연구에서 개발한 분석법에서 설정된 정량한계(0.05 ppm)는 어류에서 법적으로 설정된 잔류허용 기준 0.2 mg/kg에 대해 충분한 감도를 나타낸다.
