Bacteriological Study about the Death of Cultured Doctor Fish, Garra rufa in the Aquarium

  • Since April 2012, doctor fish in the breeding tank and in the quarantine tank in Hanwha Aquaplanet Yeosu Aquarium have been dying, accompanied by diffuse bleeding around the mouth, in the chin, and at the bottom of the abdomen. In this study, the cause of death would be examined through the bacteriological study of doctor fish and the rearing water quality in the aquarium. The water quality and the bacterial counts of the rearing water in the exhibit tank and in the quarantine tank were analyzed once a week, starting from August to November 2014. Water quality was measured based on the following data: temperature was in the range of 24.5~26.8℃, pH at 6.77~7.94, DO at 6.15~8.61 ppm, ammonia at 0~0.93 ppm, nitrite at 0.009~0.075 ppm, and nitrate at 1.1~40.9 ppm. Studies revealed that the differences in these water quality factors were not related to the death of doctor fish. Bacterial counts in the rearing waters of Garra rufa slightly increased to 103~104 CFU/ml, just before the death of the doctor fish. Twelve strains of bacteria were isolated from the dead fish and rearing waters. The isolates were identified as Aeromonas veronii, Citrobacter freundii, Pseudorhodoferax aquiterrae, Shewanella putrefaciens, and Vibrio anguillarum on the basis of 16S rRNA gene sequences. The most dominant species was C. freundii, which showed medium sensitivity to florfenicol and norfloxacin, and was resistant to amoxacillin, doxycycline, oxytetracycline, tetracycline, and trimethoprim. Ten isolates were confirmed to be pathogenic to the doctor fish. Doctor fish infected with C. freundii and S. putrefaciens showed high mortality in the experimental groups. These results indicate that the variation in bacterial numbers in the rearing water was related to the death of doctor fish. C. freundii and S. putrefaciens were directly implicated in causing the death of doctor fish in the aquarium.

  • 가상현실(VR) 기법을 이용한 가상 수족관 시스템은 3차원 형상 모델링 작업을 통하여 해저지형과 여러 종류의 물고기들을 가상공간에 위치시켜서 기본적인 가상 수족관을 제작하고, 제작된 가상 수족관 에서 자연스럽게 움직이는 물고기의 모습을 표현하는 연구를 통하여 가상 수족관 시스템을 완성하였다.

Experimental Infection of Clonorchis sinensis to Cyprinus curpio nudes (간흡충의 이스라엘잉어(향어)에 대한 감염실험)

  • As a part of observation on the suitability as a second intermediate host of Clonorchis sinensis, various infection experiments were tried to Cyprinus carpio nodus. The results obtained were summarized as follows: 1. The cercariae which attempted to contact with Cyprinus carpio nodus in the water were observed under the stereomicroscope. After contact, cercariae began to separate its tails from the bodies at 4 minutes increasingly, and then the numbers ranged to 80% at 13 minutes after the encounter. But very few cercariae could actually invade into the epidermis of the fish. 2. When Cyprinus carpio nudes were exposed to a number of cercariae in the beaker, only a few cercariae could invade through the epidermis. Most of the invaded cercariae were killed before forming the cyst. Also rare encysted cercariae were found to be dead within 48 hours. 3. Cyprinus carpio nudes were reared with Parafossarulus manchouricus shedding numerous cercariae in the indoor aquarium. When observed after 12 days, to determine the susceptibility, no metacercaria was found infected. 4. Cyprinus carpio nudum were reared with Parafossarulus manchouricus shedding numerous cercariae in Kimhae lake for 2 months in summer of 1982, in order to check the natural infection. No metacercaria of Clonorchis sinensis was found from the fish. 5. In the cases of control fish, Pseudoraibora larva, numerous cercariae were inspected on the course of invasion through epidermis of the fish under the stereomicroscope. And many metacercariae were also found from the Ssh whenever they were kept in beaker, indoor aquarium, or the lake for natural infection. The results suggested that Cyprinus carpio kudus might not be proper intermediate host of Clonorchis sinensis.

Attachment of the Microcotyle sebastis (Monogenea: Microcotylidae) to the Gills of Black Rockfish, Sebastes melanops (볼락류 Sebastes melanops 아가미에 단생 흡충류 Microcotyle sebastis의 부착에 관하여)

  • The gills of seven black rockfish, Sebastes melanops collected from Hatfield Marine Science Center Aquarium during September 19-October 3,2001 were examined for parasites attached to the gills. The species of Sebastes melanops were Microcotyle sebastis baled on the number of clamps and testes. The prevalence of 42.9% was recorded for host and fourteen worms of three black rockfish were recovered. Mean intensity of infection of Microcotyle sebastis individuals per infected host for black rockfish was 2.0(range 1-11). Monogenea helminth sites were mainly consist of second and third gill arch's filaments of black rockfish. The results revealed that surface area of the 2nd and 3rd pair of gills might affect the distribution of Microcotyle sebastis.

Molecular detection of Aeromonas hydrophila isolated from albino catfish, Clarias sp. reared in an indoor commercial aquarium

  • Moribund albino catfish, Clarias sp., displayed from an indoor private commercial aquarium were submitted in the laboratory for diagnostic examination. Dense culture of bacteria was recovered from the kidney and was characterized using Vitek System 2 and showed 98% probability to Aeromonas (A.) hydrophila. PCR result showed positive using A. hydrophila extracellular hemolysin gene ahh1 (130 bp) and aerolysin gene aerA (309 bp). The 16S rRNA gene was identical and exhibited 97% sequence similarity with the other known isolates of A. hydrophila available in the GenBank. In this paper, we reported the isolation and molecular detection of A. hydrophila from an albino catfish.

Infection of Clinostomum complanatum in Korean Barbel (Hemibarbus mylodon) (어름치(Hemibarbus mylodon)에서 Clinostomum complanatum 감염증)

  • The metacercariae species have often been isolated from fresh water fish in Korea. In this study, we reported Clinostomum complanatum, a species of metacercariae isolated from gill and muscle of Korean barbel (Hemibarbus mylodon), a natural monument fresh water fish in Korea, by morphological observation. The excysted metacercariae, tongue-shaped and progenetic, were 3.11-4.62 mm in length and 0.90-1.43 mm in width. Fresh water fishes are known as the second intermediate hosts of C. complanatum. This is the first isolation report from Korean barbel.

A Study on the Egg Deposition and the Development of Pettancylus nipponicus (Basommatophora: Ferrissidae) (담수삿갓조개(Pettancylus nipponicus)의 산란(産卵)과 발생(發生)에 관(關)한 연구(硏究))

  • Pettancylus nipponicus (freshwater limpet) collected from the Lake Choon Cheon were cultrued in aquarium and studied in the respects of an egg deposition and the developmental characters of cleavage. The species were collected only in the Lake Uiam and Choon Cheon and easily sampled on the debris like vinyls and cans. Those smooth surfaces were suitalble substrata for attaching. P. nipponicus, about 4 mm in adult, deposited an egg in one calcareous egg-capsule on the aquarium walls, while many eggs are surrounded in a jelly-like egg-envelope in the most gastropods. And another special features was that an egg-capsule was consisted of two valves and the suture was opended as hatching. A diameter of its egg was $55{{\mu}m}$ and the length of an egg capsule was $620{{\mu}m}$ and $510{{\mu}m}$ in long and short diameter, respectively. It took about 8 hours from beginning cleavage to 16-cell stage and about one year to become an adult.

Rescue, rehabilitation and release of finless porpoise (Neophocaena asiaeorientalis) in Korea (한국 상괭이(Neophocaena asiaeorientalis)의 구조, 재활, 방류)

  • The coastal water of Korean Peninsula is the major habitat for finless porpoises. However, hundreds of finless porpoises are currently caught incidentally by stow-net fishery every year. To prevent the mass mortality of finless porpoises and conserve marine animals, Busan Aquarium has been designated as a "Marine Life Ex-Situ Conservation Institution and Marine Animal Conservation Center," and Cetacean Research Institute, under the National Institute of Fisheries Science have started RRR (rescue, rehabilitation and release) project together since 2011. From 2012 to 2013, four individuals of finless porpoises had been rescued and two of them finally released to their original habitat after fully recovered in July 2013. As a result, according to growth curve of finless porpoise of Korea more than 140 cm of body length, 38 kg of total weight referred to adult, results of observation of the change in the hormone revealed that mating time of finless porpoises is January, Jun and July. To become the initial institution which rescues, treatment, husbandry and releases finless porpoise in Korea.

Fluid Flow Characteristics in the Aquaculture Tank for a Breeding Fish

  • The aquaculture tank is used for breeding fish in sea water which was pumped up to land. The flow characteristics in the aquaculture were investigated with varying the tank geometry and flow rate. The numerical analysis has been employed for calculating the velocity and temperature distributions in a water tank of rectangular type. The finite volume method and SIMPLE algorithm with 3-dimensional standard $\kappa$-$\varepsilon$ turbulence model were used for the numerical analysis. For comparison with experimental results, the PIV system was applied to visualize the flow patterns quantitatively. The numerical results showed good agreements with the experimental results. The mean velocity and temperature versus aquarium depth were represented for various circulating flow rates. Especially, the aquaculture environment is recommended that the aquarium depth has to be d=0.5 m, and this case is the condition of higher velocity and temperature in winter season.
