암모니아 노출에 따른 조피볼락(Sebastes schlegeli)의 생리학적 반응 (Physiological Responses in Korean Rockfish (Sebastes schlegeli) Exposed to Ammonia)

  • 본 연구에서는 암모니아 노출에 따른 조피볼락의 생리학적 반응을 조사하고자 아가미 $Na^+/K^+$-ATPase (NKA) 활성을 비롯한 혈장 parameters를 분석하였다. 실험구의 암모니아 농도는 대조구(자연해수), 1, 2, 4, $8mg\;L^{-1}$였으며, 조피볼락을 각각의 실험구에 3시간동안 노출한 다음 혈액 및 아가미 조직을 샘플링하였다. 실험구의 암모니아 농도가 높아질수록 혈장 암모니아 농도가 증가하였으며, 아가미 NKA 활성 또한 증가하는 경향을 보였다. 혈장 $Cl^-$를 제외한 외부의 암모니아 영향을 받지 않았으나, 4, $8mg\;L^{-1}$구의 $Na^+$, $K^+$ 및 삼투질농도는 대조구 및 1, $2mg\;L^{-1}$구보다 유의하게 높았다. 암모니아 노출에 따른 조피볼락의 혈장 코티졸은 암모니아 농도와 선형관계를 보였으며, 혈장 글루코스 또한 코티졸과 동반상승하는 것으로 나타났다. 1, $2mg\;L^{-1}$구의 hematocrit는 대조구와 차이를 보이지 않았으나, 4, $8mg\;L^{-1}$구는 나머지 실험구보다 유의하게 높았다. 암모니아 농도가 높을수록 아가미 조직 손상은 심하였으며, 특히 4, $8mg\;L^{-1}$구에서는 상피세포의 과증식, 분리, 괴사 및 2차새변의 곤봉화(club-shaped lamella) 현상이 관찰되었다.

굴, Crassostrea gigas 정자의 냉동보존 (Cryopreservation of Pacific Oyster, Crassostrea gigas Sperm)

  • 본 연구에서는 굴, Crassostrea gigas의 계획적인 인공종묘생산 및 우량형질의 정자를 보존하기 위하여 냉동보존 시 적정 결빙억제제 (cryprotective agent, CPA) 및 농도를 알아보고자, CPA 종류 및 농도별 독성 및 냉동보존 효과를 파악하고 해동 후 정자의 세포 손상을 조사하였다. CPA 종류 및 농도별 침지시간에 따른 굴 정자의 독성평가를 실시한 결과, 생존율 및 운동성은 DMSO가 가장 좋았으며, 다음으로 EG, glycerol, Methanol 순이었다. 희석액으로 여과해수를 사용하여 CPA 종류 및 농도별 냉동보존 결과, 15% DMSO로 냉동보존 한 정자의 생존율 및 운동성이 가장 높았다. 굴의 냉동/해동 정자를 주사전자현미경으로 관찰한 결과, DMSO, EG, methanol, glycerol 순으로 세포 손상이 적었으며, 농도는 15, 20, 10, 5% 순이었다. 이상의 결과를 종합한 결과, 굴 정자의 냉동보존 시 여과해수를 희석액으로 사용할 경우 적정 CPA는 DMSO이며, 농도는 15%였다.

  • 본 연구결과 glucose, NH3, AST, ALT는 4℃일 때 가장 높게 나타났으며, glucose와 AST는 4℃일 때 다른 염분구보다 26 psu에서 유의하게 높았다. 참전복에는 총 18가지 FAA가 존재하였으며, taurine, arginine, glutamic acid, alanine, glycine, lysine이 전체의 90% 가량을 차지하는 우점 FAA로 나타났다. Taurine은 30 psu 이상에서는 수온 간 유의한 차이가 없었으나, 26 psu에서는 6, 8℃일 때 유의하게 증가하였다. Glycine은 수온이 낮아질수록 유의하게 감소하여 6℃일 때 가장 높게 나타났다. 따라서 참전복은 30~34 psu, 8~10℃에서의 조건으로 수송 시 스트레스를 최소화할 수 있을 것으로 여겨진다.

Impact of Seepage from Land Treatment of Pulp and Paper Effluent on Water Quality and Aquaculture

  • Pulp and paper mill wastewater has been treated by biological treatment, but the secondary effluent still contains high lignin, chemical oxygen demand, color and total dissolved solids. Tertiary treatment by land application, referred to as 'Project Green,' has been implemented to treat such high quantities of undesirable matters. The impacts of seepage from Project Green diffusing into receiving streams on the water quality and fish pen aquaculture were studied via the integration of technical and social approaches. The determination of the water quality was performed for 13 sampling stations along the receiving stream, including the Chot stream, Chot lagoon and the Pong River. The water quality was generally at normal levels, with the exception of total dissolved solids. The levels of matter were higher at the Chot stream, but became more diluted at the Chot lagoon and the Pong River, respectively. The social approach was conducted through the voluntary participation of the villagers as research assistants for the fish aquaculture study. Fish could grow at three fish pens within the study sites at the location of Project Green, the Chot lagoon and the Pong River. Fish growth at the Chot lagoon was better at the site of Project Green and the Pong River. The integration of technical and social approaches was a meaningful tool not only for the technical feasibility but in helping to solve the conflict between the community and industry.

급격한 염분변화에 따른 수온별 참전복(Haliotis discus hannai) 치패의 생존율 및 체성분 변화 (Survival Rate and Body Composition Changes in Juvenile Abalone Haliotis discus hannai by Temperature/Salinity Change)

  • Physiological studies on the salinity tolerance with respect to survival rate and body composition changes of the abalone Haliotis discus hannai were conducted by rapidly changing the salinity in an indoor rearing system. The survival rate of the control (35 psu), 30 and 25 psu groups at $15^{\circ}C$ was 100%. The survival rate of the 20 psu group was $35{\pm}5.0%$. Survival rate of the 15 psu group was 0%. At $15^{\circ}C$ and 35, 30, 25, 20, 15 psu, the moisture contents of abalone muscle were $82.1{\pm}0.7$, $82.5{\pm}0.7$, $84.9{\pm}0.5$, $86.9{\pm}0.3$ and $86.6{\pm}0.4%$, respectively. Crude lipid contents were $0.47{\pm}0.03$, $0.47{\pm}0.03$, $0.47{\pm}0.09$, $0.77{\pm}0.09$ and $0.63{\pm}0.03%$, respectively and crude ash contents were $1.30{\pm}0.12$, $1.33{\pm}0.15$, $1.13{\pm}0.23$, $1.87{\pm}0.15$ and $1.40{\pm}0.31%$, respectively. At salinity below 20 psu, these values increased compared with the control. The general components of abalone muscles significantly increased below 20 psu, while amino acid composition showed no significant difference with salinity and water temperature.

Effects of land-based fish farm effluent on the morphology and growth of Ascophyllum nodosum (Fucales, Phaeophyceae) in southwestern Nova Scotia

  • Phenotypic plasticity was examined in the economically and ecologically important brown alga Ascophyllum nodosum in southwestern Nova Scotia, considering specifically how nutrient loading affected its vegetative and reproductive features. To determine this, we examined morphometric changes in A. nodosum from two sites receiving direct effluent impacts from a land-based finfish aquaculture facility and from two control sites, approximately 2 km away from the aquaculture facility in opposite directions. Fronds from test sites were significantly younger than from control sites (5 y vs. 8 y); however, fronds from farm sites were significantly larger (219 g vs. 90 g) because of their higher growth rates. Thalli from farm sites had greater reproductive potential, as shown by numbers of receptacle initials (797 initials vs. 281 initials). These results suggest limited nutrient inflows from land-based aquaculture may positively affect adjacent Ascophyllum populations by inducing higher growth rates. We conclude that the coordination of effluent management from land-based aquaculture with natural resource harvesting of A. nodosum may be beneficial. Further study is necessary to determine the limits of nutrient loading for this potentially beneficial outcome.

Design of Closed Seawater Recirculating Aquaculture System for Korean Rockfish Sebastes schlegeli Culture

  • Recirculating aquaculture system (RAS) consists of different treatment compartments that maintain water quality within the ranges commonly recommended for fish cultures. However, common RASs still exert considerable environmental impact since concentrations of organic matter and nutrients in their effluents are high. Compared with the traditional RAS, the model RAS developed here use a sedimentation basin for digestion purposes and then use the released volatile organic matter to stimulate a denitrification process. Different treatment compartments for solids, total ammonia nitrogen, and nitrate removal have been reviewed. This paper provides the basic information on designing different treatment compartments as well as the engineering criteria in closed seawater RAS, consisting of circular tanks for fish cultures; dual drain systems, sedimentation basins and foam fractionators for removal of solids; nitrification biofilters for TAN removal; denitrification biofilters for nitrate removal; and aerators for aeration. The main purpose is to outline a common procedure in designing of closed RAS for marine fish culture with an emphasis on easy management and low expense, as well as reduction of the environmental impact.

Tracing the origin of fish without hatchery information: genetic management of stock enhancement for mangrove red snapper (Lutjanus argentimaculatus) in Taiwan

  • Stock enhancement is considered to be a valuable approach for restoring fishery resources. Because no specific official institution in Taiwan is responsible for the production of fry, the released fry are purchased directly from the private sector. However, fishermen from the private industry have not established a genetic background, so the genetic composition for each batch of released fry is unclear. Mangrove red snapper (Lutjanus argentimaculatus), a prominent species released in Taiwan, was collected after its official release. One hundred and two field samples were compared with four batches of hatchery fry (n = 685) by using a microsatellite-based multiplex PCR assay. Four of the field samples (3.9%; 4/102) were revealed to be from a fish farm and most likely from a single batch. This study revealed that wild mangrove red snappers are genetically different from those originating from farms, and their origins can be traced through molecular markers, even without information on breeding stocks.

  • The effects of food, temperature, salinity, and rearing density on the survival and growth of the swimming crab Portunus trituberculatus, were studied. With regard to food, the larval survival rate to zoea IV was the highest (53.9%) in the mixed feed group (rotifers+ Artemia), followed, in order, by the rotifer, commercial feed, and Artemia groups. The growth of young crabs reared on different diets was the highest in group VI. The survival rate to zoea II at 20 to $30^{\circ}C$ ranged between 77.2% and 83.1%, and was the highest for crab I (17.3%) at $25^{\circ}C$. When young crabs were cultured individually at four different temperatures (20, 25, 30, and $35^{\circ}C$), the growth did not differ significantly at 25 to $30^{\circ}C$, but was higher than at $20^{\circ}C$. The survival rate of larvae to crab I was 12.7 and 11.4% at 25 and 28 psu, respectively, while all of the larvae died at 15 psu. For young crabs, there was no significant difference in growth and survival from 20 to 30 psu. The survival rate decreased with increasing rearing density from zoea I to crab I. Each molting cycle took 1 month from crab I to VIII. Subsequently, the interval increased with growth. The mean body weight of crab XV after 24 months was $428.05{\pm}57.63\;g$.

  • This study compared the survival rates, growth, and fatty acid content of larval Pacific cod Gadus macrocephalus fed with rotifers raised on different enrichment diets. We used four commercial rotifer enrichment products (one domestic, ER1, and three imported, ER2, ER3 and ER4). Twelve 200-L tanks were used, with three replicates per treatment. Larvae were fed rotifers 3 times daily at a rate of 5 rotifers/ml/feeding from 7 to 21 days post-hatch. At the end of the feeding trial, the survival rate and total length of larvae fed ER3 were greater than those of larvae fed ER1, ER2, or ER4. ER3 had the highest proportions of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and polyunsaturated acid (PUFA). Partially reflecting this composition, larvae fed ER3 had the highest proportions of DHA and PUFA. These results show a positive effect of rotifer DHA and PUFA proportions on the survival and growth rates of Pacific cod larvae.
