Growth and Survival of Juvenile Grey Mullet (Mugil cephalus) in Rearing System with Recirculated Seawater and Freshwater (순환여과 사육시스템에서 해수와 담수에 사육한 숭어(Mugil cephalus) 치어의 성장과 생존율)

  • Total length and body weight of grey mullet were 6.4cm and 2.1g, respectively at the beginning of the experiment. After 60 days of rearing, body weight of the mullet(363%) was significantly higher in freshwater group than that (187%) in seawater group. However, the condition factor showed no significant differences between seawater (9.0%) and freshwater (8.8%) groups. Survival of the mullet was 98.3% and 64.3% in seawater and freshwater groups, respectively.

Genetic Similarity in Crucian Carp(Carassius carassius) by PCR-RAPD Analysis (PCR-RAPD 분석에 의한 붕어(Carassius carassius)의 유전적 유사성)

  • Genomic DNA from the blood of crucian carp(Carassius carassiu) from lake and aquaculture facility in Kunsan, Korea was extracted in order to identify genetic differences by polymerase chain reaction-randomly amplified polymorphic DNAs(PCR-RAPD). Out of 12 primers, 6 generated 266 highly reproducible RAPD markers, producing approximately 2.1 polymorphic bands per primer in crucian carp from lake. The degree of similarity varied from 0.18 to 0.76 as calculated by bandsharing analysis in crucian carp from lake. The RAPD outlines obtained with DNA of two different crucian carp populations from Kunsan were different(0.47 from lake and 0.70 from aquaculture facility, respectively). The electrophoretic analysis of polymerase chain reaction-randomly amplified polymorphic DNAs(PCR-RAPD) products showed high levels of similarity between different individuals in crucian carp from aquaculture facility. This result implies the genetic similarity due to raising in the same environmental condition or inbreeding within the crucian carp from aquaculture facility in Kunsan. In other words, crucian carp may have high levels of genome DNA diversity due to the introduction of the wild population from the other sites of Knsan even if it may be the geographical diverse distribution of this species. Generally, the RAPD polymorphism generated by these primers may be useful as a genetic marker for strain or population identification of important aquacultural fish species, crucian carp. However, in future, additional populations and sampling sites will be necessary to complement weak points.

Development of Bivalve Culture Management System based on GIS for Oyster Aquaculture in GeojeHansan Bay (거제한산만 굴 양식장에 대한 GIS 기반 어장관리시스템 개발)

  • Oyster production is playing an important role in domestic aquaculture, but facing some problems such as exports decrease, a slowdown in domestic demand and marine environmental deterioration. In order to obtain the suitable and sustainable oyster production, suitable sites selection is an important step in oyster aquaculture. This study was conducted to identify the suitable sites for lunging culture of oyster using Geographic Information System(GIS)-based multi-criteria evaluation methods. Most of the parameters were extracted by Inverse Distance Weighted(IDW) methods in GIS and eight parameters were grouped into two basic sub-models for oyster aquaculture, namely oyster growth sub-model(Sea Temperature, Salinity, Hydrodynamics, Chlorophyll-a) and environment sub-model(Bottom DO, TOC, Sediment AVS, Benthic Diversity). Suitability scores were ranked on a scale from 1(leased suitable) and 8(most suitable), and about 80.1% of the total potential area had the highest scores 5 and 6. These areas were shown to have the optimum condition for oyster culture in GeojeHansan Bay. This method to identify suitable sites for oyster culture may be used to develop bivalve culture management system for supporting a decision making.

Investigation of Optimal Temperature and Salinity for Long Distance Transport of the pacific abalone (Haliotis discus hannai) (참전복(Haliotis discus hannai)의 장거리 수송을 위한 적정 수온 및 염분 조건 탐색)

  • This study was carried out to investigate the stress response of pacific abalone exposed to various water temperatures (4, 6, 8, and 10℃) and salinities (26, 30, and 34 psu) for 7 days, with the aim of finding optimum conditions for long-distance ocean transport of pacific abalone. At the end of the experiments, the survival rate was ranged from 98.7~100% at 8 and 10℃ but dropped to 25~55% at 4℃ in all salinity levels. The levels of SOD and glutathione in hemolymph were significantly higher at 4 and 6℃ than the control in all salinity groups, indicating that these temperatures induce severe stress in pacific abalone. It was found that THC was lowest at 6℃ in the 26 psu groups. The study showed that the hemocyte of pacific abalone populations mostly consisted of blast-like cells and hyalinocytes with the ratio of hyalinocytes being significantly lower at 4 and 6℃ than the other temperatures in the 26 psu groups. Percentages of apoptotic cells and necrotic cells were higher in the 26 psu group and 4 and 6℃ temperature groups. These results explicit that pacific abalone was exposed to greater stress at 26 psu and at 4 and 6℃ but experienced no significant higher stress at 30 and 34 psu and 8 and 10℃. It was therefore concluded that the optimum temperature and salinity for the long distance transport of pacific abalone range from 8~10℃ and 30~34 psu, respectively.

  • Glucose was continuously increased as time was elapsed under the temperature at 6℃ and 4℃. NH3 was significantly increased at 4℃. AST was increased as temperature and salinity were decreased which indicated that it was affected by temperature, salinity and interaction of temperature×salinity. ALT was significantly increased as temperature was decreased. Free amino acid contents, excluding aspartic acid, threonine, serine and glycine were increased as temperature was decreased. This decreasing tendency was also observed when salinity was decreased. Taurine, aspartic acid, glutamic acid and arginine were significantly increased as salinity was decreased, while glycine, alanine, methionine, tyrosine, and histidine were decreased as salinity was decreased. The result of this study suggested that transport condition to minimize the stress is considered in the range of 30~34 psu, 8~10℃.

Gonadal Development and Reproductive Cycle of the Ark shell Scapharca subcrenata (Bivalvia: Arcidae) from Yeoja Bay (여자만 새꼬막 Scapharca subcrenata의 생식소 발달과 생식주기)

  • Gonadal development and reproductive cycle of the ark shell Scarpharca subcrenata were investigated by histological observations. Samples were collected monthly from March 2007 to February 2008 in the Yeoja Bay, Yeosu, Jeollanam-do, Korea. S. subcrenata was dioecious. The gonads consist of a number of oogenic follicle and acinus. Monthly changes in the gonad index reached a maximum in June and a minimum in September. Monthly changes in the condition index reached a maximum in April and a minimum in September. The reproductive cycle of this species can be divided into five successive stages: early active stage (January to April), late active stage (March to June), ripe stage (May to August), spent stage (July to September) and recovery and resting stage (September to March). The main spawning of S. subcrenata occurred in July and August in Yeoja Bay. The sex ratio of female to male was not significantly different from 1:1.

Mixing and Oxygen Transfer Characteristics of Three Phase Fluidized Bed for Water Quality Management of Aquaculture (양어장 수질관리를 위한 삼상유동층 반응기의 혼합 및 산소전달특성)

  • This study was conducted to elucidate mixing characteristics of the media to dertermine hydraulic model parameters of the three phase fluidized bed for the water quality management of aquaculture. The volume and H/D ratio of the reactor were 1.76 Liter and 10.9, respectively. The experimental results showed that variance $(\sigma^2)$ and dispersion number $(D/{\mu}L)$ were above 0.47 and $0.35\~\infty$, respectively. Therefore, the condition of the reactor was almost ideal complete mixing. The dependence of the overall oxygen-transfer coefficient $(K_{La})$ on each experimental variables through a multple linear regression was given by the following equation ; $$K_{La}\;=\;44.9(D_i/D_c)^{-0.4611}\;A_f^{0.8622}\;C_m^{-0.0746}\;d_p^{-0.4302}$$

  • The ecological changes in the ocean due to the drastic global warming require that action be taken to sustain the productivity of fisheries. Proper ocean facilities could help prevent the loss of the expenditures made on marine aquaculture and reduce the related compensation for various ocean conditions. The aim of this study was to develop a floating ocean wave-breaker using an eco-friendly concrete and conducting a site survey, a structural analysis, and a test of towing the tank. As a result, the wave at the fish farm would be reduced. The results of the holding power of anchors and the capability of moving the floating structures were considered in the design of the wave-breaker. The analyses of the material properties of concrete and the steel structures, as well as the CAPEX and OPEX analyses of the manufacturing and operation processes confirmed the superiority of the floating concrete wave-breaker. In particular, this study demonstrated that the concrete floating breakwater can protect the fish farm against typhoons and reverse-waves, thereby reducing losses of the fish.

  • This paper introduces to a simple culture method and growth of axenic (bacteria-free) rotifer Brachionus rotundiformis for seed stock of rotifer mass culture. This rotifer axenic culture method is based on the washing and transferring with sterilized sea water and modified antibiotic mixture AM9 solution. Population growth (final density on day 16) of axenic cultured rotifer were maintained with a high density and stable growth compared with the control of non-axenic culture (general culture style) through the 3 times-rerunning experiments (trial 1, 2 and 3). But the egg carrying rates of amictic females were not different between the axenic-and non-axenic culture condition. Although, rotifer density was higher in axenic culture, the food (Nannochloropsis oculata) was still remained unutilized than that of the non-axenic culture in third trial culture. These results suggest the possible existence of harmful bacteria for the rotifer population growth in the trial 1 and 3 of non-axenic culture compared to the trial 2. This axenic rotifer culture method is valuable for seed stock of the stable rotifer mass cultures.

Effect of Temperature and Salinity on Production of Resting Egg in Korean Rotifer, Brachionus plicatilis (L and S-type)

  • Production of resting egg from the Korean rotifer, Brachiunus plicatilis (L and S-type) was investigated at different temperatures (L-type : 20, 24, $28^{\circ}C$, S-type : 28 32, $36^{\circ}C$) and salinities (10, 20,30 ppt). The rotifer was cultured in 25 ml test tube and fed on Nannochloris oculata. With regard to mixis rate, L-type rotifer showed higher rate at lower temperature, and the highest rate was observed at 20 ppt of salinity at each temperature of the experiment. However, for S-type rotifer, the optimum temperature and salinity were $28\~32^{\circ}C$ and 20 ppt, respectively. The highest number of resting egg was 173 eggs/ml in 16 days at $24^{\circ}C$, 10 ppt for L-type rotifer and 410 eggs/ml in 14 days at $28^{\circ}C$, 10 ppt for S-type rotifer. The maximum number of resting egg produced per 10,000 rotifers was 8,122 eggs at $20^{\circ}C$, 20 ppt for L-type rotifer and 8,700 eggs at $28^{\circ}C$, 20 ppt for S-type rotifer. The maximum number of resting egg produced $10^8$ cells of N. oculata was 50.7 eggs for L-type rotifer ($24^{\circ}C$, 20 ppt) and 79.6 eggs for S-type rotifer ($32^{\circ}C$, 10 ppt). The number of resting egg produced per day was $1\~11$ eggs/ml for L-type rotifers and $21\~35$ eggs/ml for S-type rotifer in 9 combination experiments. In this study, S-type rotifer is better than L-type rotifer in resting egg production, and the optimum temperature and salinity for resting egg production were $20^{\circ}C$, 20 ppt for L-type rotifer and $28^{\circ}C$, 20 ppt for S-type rotifer. This result shows the difference of Korean rotifer in the optimum condition for resting egg production from other rotifers reported earlier.
