Early gonadal maturation and vitellogenin mRNA expression in Siberian sturgeon Acipenser baerii cultured in a semi-closed water recirculating system in Korea

  • Changes of gonadal morphology and mRNA expression patterns of vitellogenin were investigated in Siberian sturgeon Acipenser baerii (Chondrostei) during its early gonadal maturation period. Early differentiations and morphological transitions of both ovaries and testes appeared to occur actively until the age of 3 years, however from then on, the maturation patterns to full maturity were largely gender-dependent, in which males showed a faster progression of maturation than did females while females experienced a steady-state progress with a lagged interval before entering the final maturation. Expression of vitellogenin mRNAs are closely correlated with transitional patterns of gonadal appearances. In both females and males, hepatic mRNA levels of vitellogenin exponentially increased in the earliest interval (up to 1-year-old). However, in subsequent periods, vitellogenin expression in females continued to increase with age, whereas in males, the expression stabilized at a younger age. Nevertheless, at the age older than or equal to 7-year-old, fully matured individuals showed a quite low level of vitellogenin expression in both females and males. Collectively, results from this study could be useful as a fundamental guideline to address the gonad maturation of this sturgeon species, which is helpful for making practical decisions about farming practices and management for caviar production on local sturgeon farms.

Changes in Sex Steroid Hormones and Ovarian Development during Artificial Maturation of Female Eel, Anguilla japonica

  • The present study demonstrates the changes in body weight (BW) and plasma sex steroid hormone profiles during artificial maturation induced by human chorionic gonadotropin (HCG) or salmon pituitary extract (SPE) injections in cultured eel, Anguilla japonica, kept in seawater for 3 months. In the weekly SPE-injected female group, BW was relatively stable during vitellogenesis. Following induction of vitellogenesis, females exhibited a rapid increase of BW, and the oocytes were observed to be in the migratory nucleus stage at the end of the experiment. Plasma testosterone (T) and $estradiol-17{\beta}$ ($E_2$) levels increased slightly during vitellogenesis and peaked at an average of 5.82 ng/mL and 4.76 ng/mL, respectively, at the end of the experiment. In the weekly control and HCG-injected female groups, BW slowly decreased during the experimental period, and the oocytes of the two groups were observed to be at the primary yolk globule stage. In the weekly HCG-injected female group, plasma T and $E_2$ levels increased slightly during vitellogenesis and decreased afterward. In the control female group, however, plasma T and $E_2$ levels were not altered during the experimental period. Furthermore, plasma $17{\alpha},20{\beta}-dihydroxy-4-pregnen-3-one$ (DHP) was not detected in all experimental groups. Fertility and hatching rates of SPE-injected females were significantly higher in those that ovulated 15 h after DHP injection than 18 h. These results indicate that long rearing in seawater increases responsiveness to SPE in ovarian maturation of the Japanese eel, resulting in shortened period from completion of vitellogenesis by sex steroid hormone production.

  • The quality and quantity of live food sources strongly influence the success of fish production in farming operations. Thus, critical studies of live forage species are a crucial element for progress in fish aquaculture. The fat content of food is an especially important determinant of growth in marine fish. Omega-3 highly unsaturated fatty acids (HUFA) are essential components of diet that determine the nutritional value of larval fish. Euglena is a protist that has potential as a forage species. These single-celled organisms have plant and animal characteristics they are motile, elliptical in shape and 15–500 μm in diameter. Their nutritional content is excellent, but most studies have focused on cells raised in autotrophic culture. We therefore examined differences in the lipid and fatty acid contents, and the growth of Euglena cells grown under autotrophic, heterotrophic, and mixotrophic conditions. Biomass production reached 15.03 g/L, 12.28 g/L, and 3.66 g/L under mixotrophy, heterotrophy, and autotrophy, respectively. The proportional n-3 HUFA content differed among culture methods: 10.04%, 5.80% and 10.01% in mixotrophic, heterotrophic and autotrophic cultures, respectively. Mixotrophy was to be the best form of cultivation for improving the growth and nutritional content of Euglena.

Effects of Feeding Rate and Water Temperature on Growth and Body Composition of Juvenile Korean Rockfish, Sebastes schlegeli (Hilgendorf 1880)

  • Three feeding trials were conducted to evaluate the effects of feeding rate and water temperature on growth and body composition of juvenile Korean rockfish, Sebastes schlegeli rearing at 3 different water temperatures. A total of 270 fish (each experiment) individual body weight (BW) averaging $16{\pm}0.3g$ ($mean{\pm}SD$) were fed a commercial diet for 4 wk at $16^{\circ}C$, $20^{\circ}C$, and $24^{\circ}C$. At each temperature, triplicate tanks were assigned to one of 6 feeding rates: 1.5%, 2.5%, 2.8%, 3.1%, 3.4%, and satiation (3.7% BW/d) at $16^{\circ}C$, 1.9%, 2.9%, 3.2%, 3.5%, 3.8% and satiation (4.1% BW/d) at $20^{\circ}C$ and 1.7%, 2.7%, 3.0%, 3.3%, 3.6%, and satiation (3.9% BW/d) at $24^{\circ}C$ water temperature. Weight gains of fish in satiation and 3.4% groups at $16^{\circ}C$, in satiation and 3.8% groups at $20^{\circ}C$ and in satiation and 3.6% groups at $24^{\circ}C$ were significantly higher than those of fish in the other treatments (p<0.05). A broken line regression analysis of weight gain indicated that optimum feeding rates of juvenile Korean rockfish were 3.41% at $16^{\circ}C$, 3.75% at $20^{\circ}C$ and 3.34% at $24^{\circ}C$ water temperature. Results of the present study indicate that the optimum feeding rate could be >3.1% but <3.41% at $16^{\circ}C$, >3.5% but <3.75% at $20^{\circ}C$ and >3.0% but <3.34% at $24^{\circ}C$. As we expected results suggest that fish performed better at $20^{\circ}C$ than $16^{\circ}C$ or $24^{\circ}C$ water temperature and the optimum feeding rate could be 3.1% BW/d to 3.7% BW/d in 16 g of juvenile Korean rockfish.

Effects of Dietary Arachidonic Acid (20:4n-6) Levels on Growth Performance and Fatty Acid Composition of Juvenile Eel, Anguilla japonica

  • This study was conducted to evaluate the effects of dietary arachidonic acid (AA, 20:4n-6) levels on growth performance and body composition in juvenile eel, Anguilla japonica. Six semi-purified experimental diets were formulated to be isonitrogenous and iso-caloric containing 55.0% crude protein and 15% crude lipid (18.3 kJ of available energy $g^{-1}$). Six different levels of AA were added to the basal diet, with 0, 0.2, 0.4, 0.6, 0.8 or 1.2% on a dry matter (DM) basis, respectively ($AA_{0.07},\;AA_{0.22},\;AA_{0.43},\;AA_{0.57},\;AA_{0.78}\;or\;AA_{1.23}$). After a conditioning period, fish initially averaging 27${\pm}$0.5 g (mean${\pm}$SD) were randomly distributed into each aquarium as triplicate groups of 20 fish each. One of six experimental diets was fed on a DM basis to fish in three randomly selected aquaria at a rate of 2-3% of total body weight twice a day. At the end of the 12-week feeding trial, weight gain (WG) and feed efficiency (FE) of fish fed $AA_{0.78}$ and $AA_{1.23}$ diets were significantly higher than of fish fed $AA_{0.07},\;AA_{0.22},\;AA_{0.43}$ diets (p<0.05). Specific growth rate (SGR) of fish fed the $AA_{0.78}$ diet was significantly higher than of fish fed $AA_{0.07},\;AA_{0.22},\;AA_{0.43}$ diets (p<0.05). However, there were no significant differences in WG, SGR and FE among fish fed $AA_{0.57},\;AA_{0.78}\;or\;AA_{1.23}$ diets (p>0.05). Whole body AA deposition of fish fed the $AA_{1.23}$ diet was significantly higher than for the other diets (p<0.05). Broken-line model analysis on the basis of WG and SGR indicated that the dietary AA requirement could be greater than 0.69% but less than 0.71% of the diet in juvenile eel. The growth-promoting activity of AA observed in the present study provides strong support for the contention that dietary AA is essential for juvenile eel.

Evaluation of the Efficacy of Fermented By-product of Mushroom, Pleurotus ostreatus, as a Fish Meal Replacer in Juvenile Amur Catfish, Silurus asotus: Effects on Growth, Serological Characteristics and Immune Responses

  • The present experiment was conducted to evaluate the efficacy of dietary fermented by-product of mushroom, Pleurotus ostreatus, (FBPM) as a fish meal (FM) replacer in juvenile Amur catfish, Silurus asotus. A total number of 225 fish averaging $5.7{\pm}0.1g$ ($mean{\pm}standard$ deviation) were fed one of the five experimental diets formulated to replace FM with FBPM at 0%, 5%, 10%, 20%, and 30% ($FBPM_0$, $FBPM_5$, $FBPM_{10}$, $FBPM_{20}$, and $FBPM_{30}$, respectively). At the end of eight weeks of the experiment, average weight gain (WG) of fish fed $FBPM_0$ or $FBPM_5$ were significantly higher than those of fish fed $FBPM_{20}$ or $FBPM_{30}$ diets (p<0.05). However, there was no significant differences in WG among the fish fed $FBPM_0$, $FBPM_5$ or $FBPM_{10}$, and between fish fed $FBPM_{10}$ or $FBPM_{20}$, and also between those fed $FBPM_{20}$ or $FBPM_{30}$ diets. Lysozyme activity of fish fed $FBPM_0$ or $FBPM_5$ were significantly higher than those of fish fed $FBPM_{10}$, $FBPM_{20}$ or $FBPM_{30}$ diets (p<0.05). The chemiluminescent response of fish fed $FBPM_5$ was significantly higher than those of fish fed $FBPM_0$, $FBPM_{20}$ or $FBPM_{30}$ diets (p<0.05). Broken line regression analysis of WG suggested that the maximal dietary inclusion level for FBPM as a FM replacer could be 6.3% without any adverse effects on whole body composition and on serological characteristics. Therefore, these results may indicate that the maximal dietary inclusion level of FBPM as a FM replacer could be 6.3% in juvenile Amur catfish.

Factors influencing farmed fish traders' intention to use improved fish post-harvest technologies in Kenya: application of technology acceptance model

  • Improved fish post-harvest technologies (IFPT) have been promoted as more efficient methods of fish processing, preservation, and value addition than the traditional methods prevalent in developing countries. The adoption rates, however, do not appear to be convincing. The purpose of this study was to determine the socio-demographic and psychological factors that influence intention of Kenyan farmed fish traders to use IFPT. The technology acceptance model (TAM) was used to properly explain the impact of TAM constructs such as perceived usefulness (PU), perceived ease of use (PEOU), and attitude (ATT), as well as socio-demographic factors such as gender, age, education level and fish trading experience on traders' intention to use the technologies. A cross-sectional survey was conducted to collect data using a semi-structured questionnaire from 146 traders in Busia, Siaya and Kakamega counties. At a significance level of p = 0.05, a linear regression model was used to examine the socio-demographic and psychological determinants of the traders' behavioral intention to use the improved technologies. The regression analysis revealed that PU (β = 0.443; p = 0.000), PEOU (β = 0.364; p = 0.000) and ATT (β = 0.615; p = 0.000) influence traders' intention to use IFPT, with ATT having the highest influence on intention. However, the traders' socio-demographic characteristics have no effect on their intention to use the technologies, as the coefficients for gender (β = 0.148; p = 0.096), age (β = 0.016; p = 0.882), level of education (β = -0.135; p = 0.141) and fish trading experience (β = 0.017; p = 0.869) are all insignificant. These findings show that the traders intend to use IFPT and will use them when it is in their best economic interests.

시판 어육포의 위생학적 특성 (Sanitary Characterization of Commercial Fish Jerky)

  • We assessed the sanitary quality of fish jerky based on domestic standards (Korean FDA, Standards on Quality of Seafood and Seafood Products, KS) and compared the characteristics of fish jerky with those of other commercial animal jerky products. The standards encompassed sensory properties (form, flavor, color, texture, and foreign matter), moisture, and microbial properties (Escherichia coli and Staphylococcus aureus). Based on the standards, some fish jerkies did not meet standards on sensory form (code 5) and color (code 11), moisture content (code 7 and 12), E. coli (code 2, 4, 5, 9, 10 and 14) and S. Aureus (code 5). These results suggest that commercial fish jerky should be monitored and controlled on safety to ensure the distribution of high-quality products.

Molecular detection of Aeromonas hydrophila isolated from albino catfish, Clarias sp. reared in an indoor commercial aquarium

  • Moribund albino catfish, Clarias sp., displayed from an indoor private commercial aquarium were submitted in the laboratory for diagnostic examination. Dense culture of bacteria was recovered from the kidney and was characterized using Vitek System 2 and showed 98% probability to Aeromonas (A.) hydrophila. PCR result showed positive using A. hydrophila extracellular hemolysin gene ahh1 (130 bp) and aerolysin gene aerA (309 bp). The 16S rRNA gene was identical and exhibited 97% sequence similarity with the other known isolates of A. hydrophila available in the GenBank. In this paper, we reported the isolation and molecular detection of A. hydrophila from an albino catfish.

Impact of glucose and pyruvate on adenosine triphosphate production and sperm motility in goats

  • Objective: This study evaluates goat sperm motility in response to metabolic substrates and various inhibitors, aiming to assess the relative contribution of glycolysis and mitochondrial oxidation for sperm movement and adenosine triphosphate (ATP) production. Methods: In the present study, two main metabolic substrates; 0 to 0.5 mM glucose and 0 to 30 mM pyruvate were used to evaluate their contribution to sperm movements of goats. Using a 3-chloro-1,2-propanediol (3-MCPD), a specific inhibitor for glycolysis, and carbonyl cyanide 3-chlorophenylhydrazone as an inhibitor for oxidative phosphorylation, cellular mechanisms into ATP-generating pathways in relation to sperm movements and ATP production were observed. Data were analysed using one-way analysis of variance for multiple comparisons. Results: Sperm motility analysis showed that either glucose or pyruvate supported sperm movement during 0 to 30 min incubation. However, the supporting effects were abolished by the addition of a glycolysis inhibitor or mitochondrial uncoupler, concomitant with a significant decrease in ATP production. Although oxidative phosphorylation produces larger ATP concentrations than those from glycolysis, sperm progressivity in relation to these two metabolic pathways is comparable. Conclusion: Based on the present study, we suggest that goat sperm use glucose and pyruvate to generate cellular energy through glycolysis and mitochondrial respiration pathways to maintain sperm movement.
