Apple is the most economically important fruit in Korea. The suspected viroid disease of dapple apple was found in apple fruits cultivated in Kyungpook province. Symptoms begin in mid-July as small circular spots, which stand out against the background color on the young fruit. Dappling of the fruit becomes more intense and easier to detect as the fruit approaches maturity; the affected spots remain yellowish as the fruit matures. no leaf or bark syndromes have been associated with this disease. The infected fruits are downgraded considerably during quality grading. The low molecular weight RNA containing viroid RNA molecules were extracted from the peels of the apples with dapple symptoms. The RNA molecules were extracted from the apples using Qiagen column chromatography. The purified RNAs were used for the synthesis of cDNA with RT-PCR. The PCR products were then ligated into a pGEM-T Easy vector, cloned and sequenced. The sequence of the viroid RNA molecule shows 331 nucleotides with one base difference ("G" insertion between the position of 133 and 134) compared with that of the Apple scar skin viroid (ASSVd) reported by Hashimoto and Koganezawa in Japan. This is the first report on the occurrence of the ASSVd in apple trees cultivated in Korea, as well as the identification of a new Korean strain of the ASSVd.the ASSVd.

This paper is the design for the welfare robot of apple artificial pollination by a survey for 25 elders' needs of Daegu-Gyeongbuk rural region. The results are summarized as follows. 1) In the apple farms of the region, most of farmers are the couples of 50 years thru 70 years to plant the apple trees only. The apple farm requires the senior farmers lots of painful works including their bone and muscle system problem to result in safety accidents. 2) For the aging progress of rural area, the senior farmers spend lots of labor hours and cost to do artificial pollination, a proper deletion of small fruits, and a speed spray, etc.. Most of them agree to the development of the welfare robot for an apple artificial pollination. 3) The survey results could be applied to the conceptual design in the users' point of view. Therefore, they are used as the important effects for the detail design and commercial design to develop the welfare robot for the rural seniors.
The Present survey was conducted to clarify species of moths infesting apple and the general biology of the dominant species as a basis for effective control. From the results of a survey in Suweon area, 24 species of leafrollers, 3 species of fruit-moths, 4 species of leaf-miners, 4 species of fruit-piercing moths and 28 other leaf-feeders were identified as pests of apple or apple trees.
Apple cankers caused by Valsa mali Miyabe, Botryosphaeria ribis Cross, Phomopsis truncicola Miura, are distributed in the main apple growing areas, Choong Chung Puk Do, Choong Chung Nam Do, and Kyung Sang Puk Do. According to the survey in 1970 that Valid mali is the dominant causal organism among the three in the most severely damaged area. Valsa mali and Botryosphaeria ribis were found in ail 3 provinces, however, Phomopsis truncicola was not found in Choong Chung Nam Do. Especially, Botryosphaeria ribis was most abundant in Chunwon of Choong Chung Nam Do. Effects of chemical control of apple canker caused by Valsa mali were tested both on potato dextrose agar with inhibition zones produced by the fungicides and relative growth of assay fungus of peridermis was treated with tile fungicides. Ifost effective was mercuric and followed by Lime sulfur, Hydroquinone sulfate, in general.
Soil physical properties in organic farming apple orchard were evaluated in relation to conventional farming to better understand the effects of organic farming system on soil quality. Two adjacent apple orchards, matched by soil type, were chosen to ensure the same pedological conditions except management system. Soil samples were collected from middle of two adjacent trees along the tree line at two depths of 5-20 and 20-35 cm in September 2006. Contents of organic matter in organic farming soil were twice as much as those found in soil of conventional farming. The higher level of organic matter in organic farming soil was reflected through a consequent trend in improved soil physical properties. Organic farming produced greater aggregation in >2 mm size and increased aggregate stability. Bulk density was lower by 13% and hence porosity was higher in soils of organic farming as compared with conventional farming. Water holding capacity was significantly greater with organic farming by >17% over conventional farming. The capacity of organic farming to improve soil physical properties can be contributed to the regular application of relatively large amount of organic materials and the sustainable ground-cover managements, mulching with compost and cover crop cultivation.
This study was carried out to establish optimum smoking conditions i.e., smoking temperature, time, and smoking material for meat products. Smoking materials employed for smoking were oak and apple trees. Roast ham and wiener produced by various smoking conditions wree subjected to polycyclic aromatic hydrocarbons (PAHs) analysis. Benzo(a)pyrene was not detected in roast ham and wiener prepared with oak wood smoked and apple wood smoked at 250$^{\circ}C$. The content of benzo(a)pyrene in roast ham and wiener with apple wood smoked at 400$^{\circ}C$ were 0.6 and 0.7 $\mu\textrm{g}$/kg, and those sample smoked at 500$^{\circ}C$ is not suitable for meat products due to the high production rate of benzo(a)pyrene. The higher smoking temperature of the products, the higher the content of PAHs. The similar tendency was observed in smoking time. PAHs contents of the products smoked with oak wood was ranged from non-dection to 0.5 $\mu\textrm{g}$/kg. Meat products smoked for 75 min. were found to be highly valuable from the viewpoints of flavour and color.
Apple stem grooving virus (ASGV) is a high-risk viral pathogen that infects many types of fruit trees, especially pear and apple, and causes serious economic losses across the globe. Thus, rapid and reliable detection assay is needed to identify ASGV infection and prevent its spread. A reliable reverse transcription loop-mediated isothermal amplification (RT-LAMP) was developed, optimize, and evaluated for the coding region of coat protein of ASGV in pear leaf. The developed RT-LAMP facilitated the simple screening of ASGV using visible fluorescence and electrophoresis. The optimized reaction conditions for the RT-LAMP were 63℃ for 50 min, and the results showed high specificity and 100-fold greater sensitivity than the reverse transcription polymerase chain reaction. In addition, the reliability of the RT-LAMP was validated using field-collected pear leaves. Furthermore, the potential application of paper-based RNA isolation, combined with RT-LAMP, was also evaluated for detecting ASGV from field-collected samples. These assays could be widely applied to ASGV detection in field conditions and to virus-free certification programs.

Three fungal strains designated as KNUF-23-MG32, KNUF-23-YC8, and KNUF-23-MJ82 were isolated from the abnormal symptomatic apple trees during screening fungal pathogens collected in Jeollanam-do and Gyeongsangbuk-do, Korea. These fungal strains were found to have similar cultural and morphological characteristics close to the genus Botryosphaeria. Morphological characteristics were matched with B. kuwatsukai CBS 135219 but different with B. dothidea KACC 45481 and B. sinensis HMAS 246714T. Pathogenicity tests of strain KNUF-23-MG32 showed that this strain causes rot in Fuji apples. Molecular phylogenetic analysis based on the sequences of the internal transcribed spacer (ITS) regions, translation elongation factor 1-alpha (TEF1α), and partial β-tubulin (TUB2) showed that these strains are B. kuwatsukai. To the best of our knowledge, this is the first report of an apple ring rot fungus Botryosphaeria kuwatsukai in Korea.

To explore fungal diversity in orchard soil where fire-blighted apple trees are buried, we collected soil samples from apple orchards in Chungju, Korea. Fungal isolates were obtained from DG18 agar and identified at the species level based on morphological features and phylogenetic analyses. The colony characteristics and microstructures were examined using a light microscope and a scanning electron microscope after culturing on potato dextrose agar (PDA), malt extract agar (MEA), Czapek yeast agar (CYA), and oatmeal agar (OA) The PCR-amplified products of the ITS1-5.8S-ITS2 region and 28S large subunit of the nuclear ribosomal RNA gene, as well as partial sequences of the β-tubulin, calmodulin, and translation elongation factor 1-α genes were sequenced and analyzed phylogenetically. Seven previously unknown fungal species were explored in Korea. All samples, including Aspergillus aureolatus, Botryotrichum atrogriseum, Dactylonectria novozelandica, Fusarium denticulatum, Paecilomyces tabacinus, Sarcopodium tibetense and Talaromyces stollii, had ascomycetes. Herein, we report their descriptions and features.

The two-spotted spider mite Tetranychus urticae Koch, overwinters in apple culture areas of apple orchards. The mite overwinters in various places on the apple trees, usually in groups of 5-15 females. Overwintering females were mostly found in bark crevices with the hibernation cocoons of the oriental fruit moth, Grapholita molesta Busck. They were also found in small crevices of the bark, and in pedicels left after fruit harvesting. Furthermore, overwintering females were found on rough bark at the bases of buds and spurs, on small limbs and twigs, and in limb crevices and forks. Empty scales of dead diaspidids and coccids, as well as cocoons of lacewings and mummies of parasitized aphids sporadically found on trees, were found to be less common overwintering sites for females of the two-spotted spider mite. In two successive years, the mortality of overwintering females was very high, reaching approximately 72 and 80%. This could be due to the low temperature of winter season in Korea and the low cold tolerance of the mite.
