Antioxidant, Anti-acetylcholinesterase and Composition of Biochemical Components of Russian Deer Velvet Antler Extracts

  • Russian deer velvet antlers were divided into three parts and subjected to a extraction process using hot water at 100, 110, and $120^{\circ}C$ or an extraction with 70% ethanol. Each extract was analyzed for its biochemical components, including uronic acid, sulfated-glycosaminoglycans (sulfated-GAGs), and sialic acid, and the antioxidant and anti-acetylcholinesterase activities were investigated. Different levels of uronic acid and sulfated-GAGs were observed in the extracts according to the water temperature used for the extraction, and contents decreased with increasing extraction temperature. The upper layer of each extract showed high amounts of uronic acid and sulfated-GAGs, followed by the middle and base layers. Ethanol extraction was more effective for recovering uronic acid than sulfated-GAGs. Sialic acid content was the highest in the $110^{\circ}C$ extracts but was not observed in the ethanol extracts. Velvet antler extracts showed strong antioxidant activities against DPPH and hydrogen peroxide as well as strong reducing power in a dose-dependent manner. However, the antioxidant activities were different in each layer and according to the extraction method. Additionally, velvet antler extracts exhibited inhibitory activity against acetylcholinesterase, which is associated with Alzheimer's disease, in a dose-dependent manner. These results suggest that velvet antler extracts are useful as a functional food ingredient and/or a pharmaceutical.

Prevention of Collagen-induced Arthritis in Mice by Deer Antler Extract(DAE)

  • Objectives : The effect of water extract of the pilose antler of Cervus korean TEMMINCK var. mantchuricus Swinhoe (Nokyong), a traditional immuno-suppressive and immuno-activating Korean oriental medicine, on collagen-induced arthritis (CIA) mice model was studied. Identification of common Nokyong capable of affording protection or modulating the onset and severity of arthritis may have important human health implications. Methods : Nokyong has shown to possess anti-inflammatory and anticarcinogenic properties in experimental animals. In this study we determined the effect of DAE on collagen-induced arthritis in mice. Results : In three independent experiments mice given DAE in water exhibited significantly reduced incidence of arthritis (33% to 50%) as compared with mice given no DAE in water (84% to 100%). The arthritis index also was significantly lower in DAE-fed animals. Western blot analysis showed a marked reduction in the expression of inflammatory mediators such as cyclooxygenase 2 (Cox-2), $Interferon-{\gamma}\;(INF-{\gamma})$, and tumor necrosis factor ${\alpha}\;(TNF-{\alpha})$ in arthritic joints of DAE-fed mice. The neutral endopeptidase (NEP) activity was approximately 6-fold higher in arthritic joints of non-DAE-fed mice in comparison to nonarthritic joints of nonimmunized mice whereas it was only 2-fold higher in the arthritic joints of DAE-fed mice. Additionally, total IgG and type II collagen-specific IgG levels were lower in the arthritic joints of DAE-fed mice. Conclusion : Taken together our studies suggest that DAE may be useful in the prevention of onset and severity of arthritis.

Red Deer Antler Extract Accelerates Hair Growth by Stimulating Expression of Insulin-like Growth Factor I in Full-thickness Wound Healing Rat Model

  • In order to investigate and evaluate the effects of red deer antlers on hair growth in the full-thickness wound healing model, Sprague-Dawley rats were given incision wounds through the full thickness of their dorsal skin and deer antler was applied for 40 days. At specified intervals thereafter (4, 8, 16, 32 and 40 days), the animals were sacrificed and the wound site skins were excised, processed, and sectioned. At post-injury days 16, 32 and 40, longer and more active new hair appeared around the healing wound of antler-treated skin. Histological studies showed that the antler extract markedly increases the depth, size, and number of hair follicles. Expression of IGF-I (insulin-like growth factor) mRNA was detected by RT-PCR and real time RT-PCR. The result showed that the expression of IGF-I (days 16, 32, and 40) was obviously up-regulated in antler-treated skins compared to control skins. Similar results were seen in the ELISA analysis to quantify the IGF-I expression. These results support the notion that wound healing can cause hair growth by enhancing the expression of IGF-I. Deer antler extract appears to have the potential to promote hair growth and could be used in hair growth products.

정천화담강기탕가녹용(定喘化痰降氣湯加鹿茸)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향 (Anti-Inflammatory Effect of Jungcheonhwadamgangki-tang ga Antler in Xylene-Application Mouse Ear Acute Inflammation Model)

  • In the present study, the anti-inflammatory effect of "Jungcheonhwadamgangki-tang ga Antler" water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The obtained results were as follows. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, thickness and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang ga Antler-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang ga Antler extracts has clear anti-inflammatory effect on the acute inflammation such as bronchial asthma.

수용성 녹용 추출물이 랫드의 지방대사 및 혈액성분에 미치는 영향 (Effect of the Water Soluble Extracts from Velvet Antler on Lipid Metabolism and Blood Components in Rats)

  • 본 시험은 elk의 수용성 부위별 녹용의 추출물이 랫드의 지방대사에 미치는 영향을 조사하고 혈액의 변화유무를 조사하고자 실시하였다. Elk 녹용을 분골, 상대, 중대 및 하대별로 35g당 60ml 되게 녹용추출액을 제조한 다음 10ml/kg 체중의 용량으로 4주령의 Sprague-Dawley 웅성 랫드 80수에 1회/1일 경구투여 하였다. 녹용투여 3주 후에 혈청의 AST는 대조군에서 보다 녹용 처리군에서 유의하게 낮았으며(P< 0.0027), 4주 후에는 대조군에서보다 하대 처리군에서 유의하게 낮았다(P<0.0136). 녹용투여 4주 후에는 복강지방(P<0.0035)과 간 조직(P< 0.0003)의 total-cholesterol은 전체 처리군에서 대조군보다 유의하게 낮았다. 랫드 복강지방의 지방산은 4주 후에는 C14:0(P<0.0037), C16:1(P< 0.0061), C18:1(P<0.0066), C18:2(P<0.0069)과 C18:3(P<0.0035) 함량이 대조군 보다 높았다. 본 시험은 elk의 수용성 부위별 녹용의 추출물이 랫드의 지방대사에 미치는 영향을 조사하고 혈액의 변화유무를 조사하고자 실시하였다. 녹각 추출액의 젖산발효를 통한 고농도 감마-아미노부티르산 생산 최적화 (Enrichment of gamma-aminobutyric acid (GABA) in old antler extract fermented by Lactobacillus plantarum)

  • 본 연구에서는 녹각 추출물을 이용하여 GABA 생산 최적화를 위해 추출액의 농도, 질소원인 YE, 탄소원 포도당, GABA 생성전구물질 MSG 농도에 따른 최적조건으로 YE 0.5%, 포도당 1.5%, MSG 3.5%를 첨가하여 $30^{\circ}C$에서 7일간 젖산발효를 하였다. 발효 7일 pH 6.56, 산도 0.77%로 나타났으며 $2.0{\times}10^8CFU/mL$로 높은 균수를 유지하였고 GABA를 1.4% 생성하는 것으로 나타났다. 결론적으로 녹각 추출액의 젖산균을 이용한 정치배양을 통해 고농도의 GABA 생산이 가능하였으며, 발효물은 GABA, 프로바이오틱(probiotic) 등을 함유하여 식품 및 기능성 소재로 이용가능성이 높다고 사료되었다.

녹용의 영양.생화학적 고찰 (A Study on the Biochemical and Nutritional Inquiry of Antler)

  • Deer horn has been used in oriental medicine and folk remedy. It has had much effects on 'health and tonic efficacy' But this word, 'health and tonic efficacy', is very obscure in modern sense This report attempts to investigate their scientific and objective effects among many papers. The biochemical components of antler are composed of pentoses, hexoses, acid mucopolysaccharides, free and ester fatty acids, glycolipids, phospholipids, prostaglandins, sterols, protein and peptides, free amino acids, minerals, hormones etc. Deer horn extract especially increases body weight gain and feed efficiency, and it stimulates the ability of spermatogenesis of animal. However, a few papers have reported concerning the toxicity and side effects of antler.

  • Contents of nucleic acid metabolites, such as uracil, uridine and hypoxanthine in the unossified pilose antler of Cervus nippon Temminck var. mantchuricus Swinhoe and the frozen and dried meat of Theragra chalcogramma (Thallas) were determined by high-performance liquid chromatography. The amount of hypoxanthine in the water extract of the meat (39.2 mg %) was higher than that of the antler (24.6 mg %).

녹각추출물이 Benzo(a)pyrene에 의한 간손상에 미치는 영향 (Effect of Old Antler Extracts on the Benzo(a)pyrene-Induced Hepatotoxicity in Rats)

  • 녹각이 동양의학에서 보혈강장제라는 사실에 근거하여 생체내 물질대사에 중요한 역할을 하는 간장의 장해시 효소활성 변동에 미치는 녹각의 효능을 구명할 목적의 일환으로 간독성 물질인 benzo(a)pyrene으로 간장해를 유도한 후 각각의 녹각추출물을 급여함으로써 간해독 과정에 관여하는 효소의 활성을 생화학적 측면에서 비교하였다. 각각의 녹각추출물 급여로 인한 체중증가량, 식이섭취량 및 식이효율은 녹각추출물 군간의 유의성은 나타나지 않았으나, Control군에 비하여 유의적인 증가를 보였고 B(a)P 투여군에서도 Cont-B군에 비하여 녹각추출물 급여군에서 유의적인 증가를 나타내었다. 체중 100g당 장기무게는 간장의 경우 benzo (a)pyrene 투여로 증가하는 경향을 보였으나 비장과 신장 및 심장에는 별다른 차이가 없었다. 녹각추출물 급여로 4주간 사육한 흰쥐에서 간해독계 효소중 cytochrome P-450의 함량은 Control 군에 비하여 ben-zo(a)pyrene 투여로 유의적인 감소를 보였으며, 녹각추출물과 benzo(a)pyrene을 병행 투여한 실험군 중 Water-B군이 정상군에 가깝게 증가되었다. 간조직 중의 glutathione함량은 Water군과 Neutral군에서 증가를 보였으나 benzo(a)pyrene 투여로 감소하였다. 또한 간조직 중의 glutathione peroxidase 활성은 녹각추출물 급여에 따른 영향은 없었으며, benzo(a)pyrene 투여로 감소된 활성이 water-B군에서 정상군에 가까운 증가를 나타내었다. 간조직 중의 과산화지질 함량은 Water-B군에서 그 감소 정도가 가장 현저하였다. 이상의 결과를 종합해 볼 때 녹각추출물 중의 water-ext.와 neutral-ext.에 함유되어 있는 유효성분이 간장의 해독기구 효소활성을 유도하고, benzo(a)pyrene의 대사를 촉진시키므로 간손상을 억제할 수 있을 것으로 생각된다.

정천화담강기탕가녹용(定喘化痰降氣湯加鹿茸)이 Formalin으로 유발된 마우스의 만성 염증에 미치는 영향 (Effects of Jungcheonhwadamgangki-tang Plus Antler in Formalin-injected Chronic Inflammation Model in Mice)

  • In the present study, the anti-inflammatory effect of Jungcheonhwadamgangki-tang plus antler water extract was tested in formalin-injected chronic inflammation model in mouse hind paw. The test articles were dosed once a day for 10 days, and changes on the body weight, paw weights were observed with histopathology of induced paw dorsum pedis. In addition, histomorphometry was also monitored at sacrifice. 15 mg/kg/10 mL of dexamethasone (DEXA) and diclofenac (DICLO) intraperitoneally dosed groups were used as reference groups. A significantly decrease of both absolute and relative paw weights were observed in all dosing groups including DEXA and DICLO groups compared to that of control, and a significantly decrease of the differences between intact and induced paw weights were also observed Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. These histological signs-hypertrophy of paw dorsum pedis tissues were detected results from edematous changes on the cutaneous and subcutaneous tissues with severe infiltration of chronic inflammatory cells-were dramatically decreased in all dosing groups including DEXA and DICLO dosing groups compared to that of control. Especially, dose dependently decreases were detected in Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. Base on these aforementioned results, it is concluded that Jungcheonhwadam- gangki-tang plus antler have clear anti-inflammatory effect on the chronic inflammation induced by formalin injection.
