Antioxidative Activity of Korean Wild Leaf Vegetables : Pleurospemum Kamtschaticum, Aderophora Remotiflor and Aster Glheni

  • The purpose of this study was to evaluate the antioxidative potential of Korean wild leaf vegetables in vivo as well as in vitro. The antloxidative activities of Pleurospemum kamtschaticum, Aderophora remotiflor and Aster gfheni were evaluated as a reference for Spinacia oleracea. Fifty rats were find either a control diet or one of several vegetable diets for 4 weeks. The level of thiobarbituric acid reactive substance (TBARS) and the activity of catalase and superoxide dismutase (SOD) in the liver were compared. The levels of total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and triglyceride (TG) in plasma were also compared. Also, the contents of $\beta$-carotene, vitamin C, vitamin E and total phenolic compound, including flavonoid and thiobarbituric acid (TBA) value using linoleic acid model system, were measured in Korean wind leaf vegetables. The TBARS values of Aderophora remotiflor and Aster giheni tended to be lower than that of Spinacia olerncea. The SOD activity of Artrr glheffi was significantly higher than that of Spinacia oleracea and reached 265% that of the control animals, whereas there was no difference between the control animals and the vegetable diet animals in terms of catalase activity. Compared to the control animals, TG and TC levels were significantly solver and showed a lower artheriosclerotic index. TBA values of Pleurospemum Kamschaticum and Aderophora remptiflor were only 18% of control value. TBA value of Spinacia olerucra was 41% of control value. These results suggest that Aderophora remotinor and Aster glheni could have antioxidative potency in vivo as well as in vitro and potential value far functional flood to improve the plasma lipid profile. flavonoid and phenolic compounds could be the major contributing factor in the antioxidative potential of Aderophora remotiflor and Aster glheni.

In Vitro and in Vivo Effects of Nitrofurantoin on Experimental Toxoplasmosis

  • Toxoplasma gondii is an important opportunistic pathogen that causes toxoplasmosis, which has very few therapeutic treatment options. The most effective therapy is a combination of pyrimethamine and sulfadiazine; however, their utility is limited because of drug toxicity and serious side effects. For these reasons, new drugs with lower toxicity are urgently needed. In this study, the compound, (Z)-1-[(5-nitrofuran-2-yl)methyleneamino]-imidazolidine-2,4-dione (nitrofurantoin), showed anti-T. gondii effects in vitro and in vivo. In HeLa cells, the selectivity of nitrofurantoin was 2.3, which was greater than that of pyrimethamine (0.9). In T. gondii-infected female ICR mice, the inhibition rate of T. gondii growth in the peritoneal cavity was 44.7% compared to the negative control group after 4-day treatment with 100 mg/kg of nitrofurantoin. In addition, hematology indicators showed that T. gondii infection-induced serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, biochemical parameters involved in liver injury, were reduced by nitrofurantoin significantly. Moreover, nitrofurantoin exerted significant effects on the index of antioxidant status, i.e., malondialdehyde (MDA) and glutathione (GSH). The nitrofurantoin-treated group inhibited the T. gondii-induced MDA levels while alleviating the decrease in GSH levels. Thus, nitrofurantoin is a potential anti-T. gondii candidate for clinical application.

Status of Lipid Peroxidation in Normal Cycling and α-Tocopherol Supplemented Anestrus Buffalo Heifers (Bubalus bubalis)

  • The present study was undertaken to analyze the status of erythrocytic malonyl dialdehyde (MDA) as an index of lipid peroxidation in normal cycling and ${\alpha}$-tocopherol supplemented anestrus buffalo heifers. Before supplementation, the blood samples were collected at weekly interval in anestrus and normal cycling buffalo heifers and data from four samples was pooled to establish pre-supplementation base line. Thereafter, the blood samples from anestrus and ${\alpha}$-tocopherol supplemented anestrus buffalo heifers were taken at fortnightly interval for 12 weeks during supplementation and for four weeks during the post-supplementation period. The pre-supplementation endogenous erythrocytic MDA, 0.3% $H_2O_2$ induced-erythrocytic MDA and 1.5% $H_2O_2$ induced-erythrocytic MDA levels (nmol/g Hb) increased significantly (p<0.05) in anestrus heifers ($372.3{\pm}5.0$, $564.4{\pm}5.7$, $862.4{\pm}8.5$) when compared to normal cycling animals ($289.6{\pm}3.0$, $508.2{\pm}6.0$, $777.7{\pm}4.9$). Oral supplementation of 3,000 mg of ${\alpha}$-tocopherol per week per animal in anestrus heifers resulted in a progressive and significant (p<0.01) decline in erythrocytic MDA levels. These observations implied that supplementation of ${\alpha}$-tocopherol to anestrus buffalo heifers ameliorated their antioxidant status by alleviating the effects of oxidative stress.

밀웜(갈색거저리) 분말 첨가 머핀의 품질 특성 (Quality Characteristics of Muffins Containing Mealworm(Tenebrio molitor))

  • 본 연구는 밀웜 분말의 첨가량을 달리하여 제조한 머핀의 품질특성을 살펴보는 것이다. 머핀을 제조할 때 밀웜 분말은 밀가루를 대체하여 0, 1, 2, 4, 6, 8 %를 첨가하였다. 중량은 밀웜 분말 첨가량이 증가함에 따라 감소하였으며, 높이는 대조군에 비해 밀웜 분말을 첨가한 머핀이 더 높게 나타났다. 머핀의 부피는 MW2(2%)가 가장 높게 나타났다. pH는 밀웜 분말의 첨가량이 증가함에 따라 높게 나타났으며, 수분함량은 밀웜 첨가군이 대조군에 비해 낮게 나타났다. 명도와 황색도는 밀웜분말 첨가량이 증가할수록 감소하였으나, 적색도는 증가하는 경향을 나타내었다. 경도는 밀웜 분말 첨가량의 증가에 따라 유의적인 차이를 보이지 않았다. 총 폴리페놀 함량과 DPPH 라디칼 소거능을 측정한 결과, 밀웜 분말의 첨가량이 증가 할수록 유의적으로 증가하였다. 1~8% 밀웜 분말을 첨가하게 제조한 머핀의 관능검사 결과, 풍미, 맛, 전반적인 기호도가 높게 나타났다. 이와 같은 결과로 밀웜 분말은 소비자 기호도를 반영한 기능성 머핀을 제조할 수 있는 좋은 식품소재라 할 수 있다.

대두 함유 방사선 조사식이를 섭취한 Mouse의 산화적 스트레스 (Oxidative Stress of Mouse Fed with ${\gamma}$-Irradiated Soybean Diet)

  • Food irradiation has been steadily increased in many countries concomitantly with increasing international trades. Harmful contaminants naturally occurred from foods which contain high levels of unsaturated fatty acids that are easily oxidized can affect the human anti-oxidation system through the generation of free radicals. Moreover, previous studies proved that ${\gamma}$-irradiation may cause production of free radicals in food. We investigated the effect of ${\gamma}$-irradiated soybeans in relation to oxidative stress in mice. Oxidative index of mice was evaluated by TBARS, DNA fragmentation in various organs such as blood lymphocytes, liver and kidney. Forty male ICR mice were equally divided into 4 groups and fed control diet or ${\gamma}$-irradiated diet containing 50% soybeans (5, 10, and 20 kGy, respectively) for 8 weeks. Peroxide values of the irradiated diets were higher than that of the non-irradiated one and increased according to the storage period. There was no significant difference in weight gain as well as in TBARS value in plasma and kidney of all groups. Liver TBARS value of the group fed with irradiated diet at 20 kGy increased significantly compared with the control group (p < 0.05). DNA oxidative damage as measured by alkaline comet assay showed that % tail DNA in the blood lymphocytes of 5 kGy and 10 kGy groups increased significantly over the control group (p < 0.05). Also, tail moments of 5 kGy and 10 kGy groups were higher than that of the control group. Ultrastructural examination shows myeline figures and swollen mitochondria in parietal and intestinal epithelial cells of the group fed with irradiated diet. Therefore, considering unsaturated fatty acid content, consumption of soybeans ${\gamma}$-irradiated with over 20 kGy or repeatedly may decrease the body's antioxidant mechanism.

The Protective Effect of Melatonin Administration against Adria-mycin-induced Cardiotoxicity in Rats

  • Adriamycin is a commonly used chemotherapeutic agent for cancer, including acute leukemia, lymphoma, and a number of solid human tumors. However, recent studies have recognized severe cardiotoxicity after an acute dose, which are likely the result of generation of free radicals and lipid peroxidation. Therefore, the clinical uses of adriamycin have been limited. Melatonin, the pineal gland hormone known for its ability to modulate circardian rhythm, has recently been studied in its several functions, including cancer growth inhibition, stimulating the immune system, and acting as an antioxidant and radical scavenging effects. In the present study, we evaluated the effect of melatonin administration on adriamycin-induced cardiotoxicity in rat. Heart slices were prepared using a Stadie-Riggs microtome for the measurement of malondialdehyde (MDA) content used as an index of lipid peroxidation and lactate dehydrogenase (LDH) release as an indicator of lethal cell injury. Serious adriamycin-induced lethality was observed in rat by a single intraperitoneal injection in a dose-dependent manner. A single injection of adriamycin (25 mg/kg, i.p.) induced a lethality rate of 86%, with melatonin (10 mg/kg s.c. for 6 days) treatment reducing the adriamycin-induced lethality rate to 20%. The severe body weight loss caused by adriamycin was also significantly attenuated by melatonin treatment. Treatment of melatonin marked reduced adriamycin-induced the levels of MDA formation and LDH release. A cell damage indicated by the loss of myofibrils, swelling of the mitochondria as well as cytoplasmic vacuolization was seen in adriamycin-treated group. Melatonin attenuated the adriamycin-induced structural alterations. These data provide evidence that melatonin prevents adriamycin-induced cardiotoxicity and might serve as a combination with adriamycin to limit free radical-mediated cardiotoxicity.

복분자(覆盆子)의 세포내 ROS, $ONOO^-$ 생성 및 $Ca^{2+}$ 증가 억제에 의한 혈관내피세포 보호작용 (Cytoprotective action of Rubi Fructus by modulation of Reactive Oxygen Species, peroxynitrite and $Ca^{2+}$)

  • Objectives : Poststroke depression is a frequent and specific entity that impaires the rehabilliation and functional recovery of patients with hemiplegia. The author evaluated the effect of Banhahubak-tang(Banxiahoupotang) in patients with poststroke depression. Methods : 38 patients suffering from poststroke depression(determined by Diagnostic and Statistical Manual of Mental Disorders, revised. 3rd edition. and Beck Depression Inventory[BDI] cutoff $point{\geqq}10$) in Kyunghee Oriental hospital were randomized into two groups; treatment group(n=19) and control group(n=19). The treatment group was prescribed with Banhahubak-tang(Banxiahoupotang) three times a day fur a week. Control troop was prescribed with other herbal medicines used for stroke Patients three times a day for a week. Patients were evaluated by use of BDI scale, Modified Barthel Index, Depression of Ki score, Yin syndrome score, and Yang syndrome score. Among 38 patients, 24 patients got BDI scores above 21, which is the cut-off score for depression in Korean. The same procedures and assessments described above were applied. Results : Treatment group did not significantly improve compared with control group. Results yielded only slight significance (P=0.086). Especially. patients with poststroke depression as yin syndrome improved more significantly on BDI than those classified as yang syndrome. When BDI cutoff point for depression was defined as being ${\geq}\;21$, treatment group did not significantly improve compared with control group(P=0.114). However, patients with poststroke depression classified as yin syndrome were also significantly improved on BDI than those classified as yang syndrome. Conclusions : This study suggests that Banhahubak-tang(Banxiahoupotang) is significantly effective in patients with poststroke depression classified as yin syndrome.

고콜레스테롤 식이로 사육한 흰쥐 혈청 지질의 성분에 미치는 대두 배아의 영향 (Effects of Soybean Germ on the Lipid Composition of Serum in Cholesterol Fed Rats)

  • 대두(Glycin max Merrill) 배아의 기능적 활성을 조사하기 위하여 대두를 발아시켜 만든 배아(soybean sprout)을 시료로 하여 0.75% 콜레스테롤 첨가식이로 유발된 고지혈증 Sprague Dawley계 수컷의 흰쥐에 대두 배아의 유기용매 추출물을 급여시킨 후 혈청의 지질 개선효과를 알아보기 위하여 4주간 동안 대두 배아를 섭취시켰다. 식이 급여가 대조군의 체중 및 장기 무게에 미치는 영향은 거의 찾아볼 수 없었으며 혈청 총 콜레스테롤 및 LDL-콜레스테롤의 농도는 대조군에 비하여 낮았고. 콜레스테롤 식이군에 비하여 콜레스테롤 식이와 대두 배아 메탄올 추출물의 혼합투여시 낮게 나타났다. 총 콜레스테롤에 대한 HDL-콜레스테롤의 비는 대두 배아 추출물 급여군이 높게 나타났으며, 동맥경화 지수는 고콜레스테롤 식이군에 비하여 대두 배아 추출물이 낮았다. 또한 혈청중의 유리 콜레스테롤 및 고콜레스테롤 에스테르의 농도는 고콜레스테롤 식이군에 비해 대두 배아의 혼합 급여군이 감소하였고, 인지질의 농도도 콜레스테롤 식이군 보다 콜레스테롤 식이와 대두 배아 급여군이 낮은 경향을 나타내었다.

Anti-Inflammatory Effect of Flavonoids from Brugmansia arborea L. Flowers

  • Brugmansia arborea L. (Solanaceae), commonly known as "angel's trumpet," is widely grown in North America, Africa, Australia, and Asia. It has been mainly used for ornamental purposes as well as analgesic, anti-rheumatic, vulnerary, decongestant, and anti-spasmodic materials. B. arborea is also reported to show anti-cholinergic activity, for which many alkaloids were reported to be principally responsible. However, to the best of our knowledge, a phytochemical study of B. arborea flowers has not yet been performed. Four flavonol glycosides (1-4) and one dihydroflavanol (5) were for the first time isolated from B. arborea flowers in this study. The flavonoids showed significant antioxidant capacities, suppressed nitric oxide production in lipopolysaccharide (LPS)-treated RAW 264.7 cells, and reduced inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX-2) protein production increased by LPS treatment. The contents of compounds 1-4 in n-BuOH fraction were determined to be 3.8 ± 0.9%, 2.2 ± 0.5%, 20.3 ± 1.1%, and 2.3 ± 0.4%, respectively, and that of compound 5 in EtOAc fraction was determined to be 12.7 ± 0.7%, by HPLC experiment. These results suggest that flavonol glycosides (1-4) and dihydroflavanol (5) can serve as index components of B. arborea flowers in standardizing anti-inflammatory materials.

Salvianolic acid B ameliorates psoriatic changes in imiquimod-induced psoriasis on BALB/c mice by inhibiting inflammatory and keratin markers via altering phosphatidylinositol-3-kinase/protein kinase B signaling pathway

  • Salvianolic acid B (SAB) is an active phytocomponent of a popular Chinese herb called Radix Salvia militiorrhiza with numerous biological properties. The anti-psoriasis activity of SAB was examined by evaluating various psoriasis inflammatory and keratin markers against imiquimod (IMQ)-induced psoriasis on BALB/c mice. Totally 50 healthy BALB/c mice were evenly divided into 5 groups including control, drug control (SAB; 40 mg/kg), IMQ-induced psoriasis (5%), IMQ exposure and treated with SAB (40 mg/kg), or standard methotrexate (MTX; 1 mg/kg). Mice supplemented with either SAB or MTX significantly lowered the values of psoriasis area severity index (PASI), erythema, scaling, skin thickness, inflammatory markers (interleukin [IL]-22/23/17A/1β/6) and lipid peroxidation product (malondialdehyde). Also, IMQ exposed BALB/c mice treated with SAB or MTX display lesser histopathological changes with enhanced antioxidant activities (catalase, superoxide dismutase). Moreover, the protein expression of keratin markers (K16 and K17) and phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt) signaling proteins (pAkt/Akt and pPI3K/PI3K) were significantly downregulated after administration with SAB and MTX as compared with IMQ induced mice. Taking together, SAB and MTX significantly ameliorate psoriatic changes by inhibiting psoriatic inflammatory and keratin markers through abolishing PI3K/Akt signaling pathway. However, further studies (clinical trials) are needed to confirm the anti-psoriatic property of SAB before recommending to psoriasis patients.
