짝잎모자반(Sargassum hemiphyllum) 추출물의 항산화 효과 (Antioxidant Effect of Sargassum hemiphyllum Extracts)

  • 본 연구는 짝잎모자반(S. hemiphyllum)을 95% 에탄올과 물로 각각 추출하여 항산화 활성을 조사하였다. 짝잎모자반 추출물의 총 페놀 화합물 함량을 측정한 결과, 에탄올 추출물과 물 추출물 각각 17.91 mg/g과 13.44 mg/g으로 에탄올 추출물이 물 추출물보다 더 높은 페놀화합물 함량을 나타내었다. DPPH radical 소거능 측정 결과 1, 0.5, 0.1 mg/ml의 농도에서 에탄올 및 물 추출물은 각각 96, 96, 62% 및 92, 90, 44%로 에탄올 추출물이 더 높은 radical 소거능을 보였으며, 에탄올 추출물은 천연 항산화제인 BHT와 유사한 소거능을 보였다. 환원력의 경우 에탄올과 물 추출물 모두 대조구인 ascorbic acid에 비하여 낮은 값을 보였으며 에탄올 및 물 추출물 간에는 유의적인 차이가 나타나지 않았다. 금속봉쇄력의 경우 1 mg/ml의 농도에서 에탄올 추출물은 9%의 활성을 나타냈으며 물 추출물은 28%의 활성을 보여 에탄올 추출물보다 더 높은 활성을 나타내었다. Rancimat에 의한 유지 산화 억제능의 경우 에탄올 추출물이 물 추출물보다 더 높은 활성을 보였지만 대조구인 BHP에 비해 낮은 항산화력을 나타내었다. 짝잎모자반 추출물은 뛰어난 DPPH radical 소거능으로 높은 항산화 활성을 가지며 특히 물 추출물보다 에탄올 추출물에서 더 높은 활성을 가지는 것을 알 수 있었다. 따라서 본 연구 결과, 짝잎모자반은 천연 항산화 소재로서의 활용 가능성이 내재되어 있다고 사료된다.

  • This study was conducted to investigate the antioxidant activity in the leaves and stems of 50 tomato accessions, in order to examine the possibility of using tomato by-products as a functional material. The extracts of the leaves (LE) and stems (SE) were analyzed for DPPH, ABTS, and total polyphenol content (TPC). Antioxidant activities and TPC differed significantly between the LE and SE of the 50 tomato accessions. TPC in LE and SE showed wide variation, ranging from 24.4 to 60.6 and 12.5 to 18.8 mg GAE/g, respectively. The DPPH and ABTS antioxidant activities of LE ranged from 10.0 to 38.2% (scavenging effect) and 20.8 to 59.0 mg ASC/g, respectively, while the DPPH and ABTS measurements of SE were 1.4 to 8.8% and 2.2 to 22.5 mg ASC/g, respectively. As assessed by the relative antioxidant capacity index (RACI), IT033117 and IT203466 had the highest antioxidant activity in LE and SE, respectively. These results will expand the knowledge of antioxidant activity and provide information on tomato accessions valuable for the development of functional foods and food additives.

Effect of Quercetin on the Activity and mRNA Expression of Antioxidant Enzymes and Physiological Responses in Olive Flounder (Paralichthys olivaceus) Exposed to Cadmium

  • We investigated the antioxidant efficacy of quercetin (0% Diet 1, 0.25% Diet 2, and 0.5% Diet 3) pretreatment for 30 and 60 days in response to cadmium (Cd) toxicity in the olive flounder, and measured the plasma lysozyme activity to understand the immune effects of quercetin. The lysozyme activity with Diets 2 and 3 was higher than with Diet 1. Based on this result, to examine the immune ability and antioxidant role of quercetin, we exposed olive flounder fed quercetin to Cd and then measured the expression and activity of antioxidant enzymes (superoxide dismutase (SOD) and catalase (CAT)) and lipid peroxidation (LPO). With Diets 2 and 3, the expression and activity of antioxidant enzymes and the $H_2O_2$ concentration were lower than with Diet 1. In addition, the LPO levels were lower than with Diet 1, which protected the cell membrane. Therefore, quercetin removed the reactive oxygen species (ROS) produced by Cd, indicating that quercetin has antioxidant ability. In addition to its antioxidant ability, quercetin has immune effects.

  • Flavonoids are class of polyphenolic compounds widely distributed in the plant kingdom, which display a variety of biological activities, including antiviral, antithrombotic, antiinflammatory, antihistaminic, antioxidant and free-radica 1 scavenging abilities. The antioxidant enzyme (AOE) system plays an important role in the defense against oxidative stress insults. To determine whether flavonoid, myricetin can exert antioxidative effects not only directly by modulating the AOE system but also scavenging free radical, we investigated the influence of the flavonoid myricetin on cell viability, different antioxidant enzyme activities, ROS level and the expression of different antioxidant emzyme in B16F10 murine melanoma cells. Myricetin in a concentration range from 6.25 to $50\;{\mu}M$ decreased superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzyme activities, but catalase (CAT) activity was increased. In the myricetin-treated group, ROS levels were decreased dose-dependently. Antioxidant enzyme expression was measured by RT-PCR. Myricetin treatment of B16F10 cells increased catalase expression. Expression levels of copper zinc superoxide dismutase (CuZn SOD) were not affected by exposure of myricetin. Manganese superoxide dismutase (Mn SOD) and GPx expression levels decreased slightly after myricetin treatment. In conclusion, the antioxidant capacity of myricetin was due to CAT and free-radical scavenging.

Evaluation of the Antioxidant Activity of Cooked Gomchwi (Ligularia fischeri) Using the Myoglobin Methods

  • This study evaluated the influence of cooking methods (blanching, pan-frying, and microwaving) on the antioxidant activity of Ligularia fischeri (LF) using myoglobin methods against five different reactive oxygen species (ROS) (i.e., 2,2-diphenyl-1-picrylhydrazyl (DPPH), hypochlorite ions, hydroxyl radicals, peroxyl radicals, and peroxynitrite ions). With respect to DPPH scavenging activity, the antioxidant activities of blanched LF and pan-fried LF were significantly higher (P<0.05) than that of fresh LF for all cooking times. Pan-fried LF and microwaved LF showed higher antioxidant activities against hydroxyl radicals and peroxyl radicals than uncooked LF, while the protective effect of blanched LF was low, except for the at the 3 min cooking time point. Microwaved LF showed high antioxidant activity against all ROS at the 2 min cooking time point. Interestingly, LF that had been microwaved for 2 min and 3 min had high antioxidant values, whereas blanched LF and pan-fried LF had low antioxidant values. The activities of cooked LF against five different ROS were characterized comprehensively by 5-axe cobweb charts.

DLPC Liposome에 미치는 도라지 추출성분의 비타민 C 첨가에 의한 항산화력 상승효과 (Enhancement of Antioxidation Effect of Platycodon grandiflorum with Vitamin C on the DLPC Liposomes)

  • 본 연구는 불포화 인지질인 DLPC liposome에 도라지(Platycodon grandiflorum A. DC, PG)의 용매별 분획물 일정 량을 가하고, 산화제 benzoyl peroxide를 가한 후 시료의 항산화 작용을 비교 분석하였다. 도라지 용매별 추출물 중 ethylether 분획물인 PGMEE와 ethylacetate 분획물인 PGMEA는 $\alpha$-tocopherol의 경우 보다 훨씬 그 산화지수가 낮아두 분획물에 의한 강한 항산화력을 측정할 수 있었으며, 잘알려진 강력한 항산화제인 BHT(butylated hydroxytoluence)의 경우와 비슷한 항산화 효과를 나타내었다. 한편 PGMEE 분획층에 비타민 C를 일정량을 첨가함으로서 상호 보강 작용에 의한 항산화 상승효과를 본 결과, Vit C를 첨가하지 않았을 때보다 산화지수가 횔씬 낮아졌다. BHT 함유의 DLPC liposome의 경우도 Vit C에 의한 비슷한 추과를 나타내었다. 이상의 결과에 의하여 도라지의 PGMEE 분획층에는 BHT와 비슷한 강한 항산화 효과를 가진 물질의 존재가 분명하다고 사료되며 비타민 C 첨가로 인한 항산화 상승 효과도 본 연구결과에 의해 뚜렷이 알 수 있었다. 이와 같은 연구에 의해 여러 질병들의 원인이 될 뿐만 아니라 세포막 손상을 일으키는 활성 산소 또는 유리기의 생성억제 등 균형된 식생활을 통한 식품 섭취의 중요성이 매우크다고 인식되며 특히 식품으로서 애용되고 있는 도라지의 인지질 세포막에 미치는 항산화 효과가 식품으로서 약이 되는 기전 규명에 기초자료가될 것을 바라는 바이다.

식용대두유에 대한 Chelating agent의 항산화 효과 (Antioxidant Effect of some Chelating Agents on Soybean Oil)

  • 식용대두유의 산화방지를 위하여 항산화제로서 Chelating agents의 항산화효과를 mild한 조건(67ml $O_{2}/min$, $50^{\circ}C$)하에서 A.O.M.법으로 측정하였으며 기기분석법 (UV,IR,MMR)에 의한 측정의 가능여부를 시험한 결과 다음과 같다. 1. Chelating agents의 항산화 효과는 diphenic acid(109.3%), naphthoquinone(111.2%), pyremellitic acid(168.6%), Quinolinic acid(183.6%), naphthalic acid(921.2%) 순으로 증가하였으며, Control에 비하여 각각 9.3, 11.2, 68.6, 83.6, 821.2% 증가하였다. 2. Chelating agents는 항산화 효과를 나타냈으며, 철과 등을 첨가했을 경우에 현저하였다. 또 이들의 항산화 효과는 화합물의 관능기와 분자구조에 따라 다르며 diphenic acid, naphthoquinone 등에서 낮았다. 3. 본 실험과 같이 mild한 조건하에서 NMR 분석법을 이용하여 항산화 효과를 측정할 수 있었으나 UV와 IR은 적당치 않았다.

니파야자(Nypa fruticans Wurmb) 싹 추출물 및 분획물의 항산화, 세포 보호 및 항균 효과에 관한 평가 (Evaluation of Antioxidant, Cytoprotective and Antimicrobial Activities of the Extract and Fractions Obtained from Young Shoots of Nypa Fruticans Wurmb)

  • Nypa fruticans Wurmb is a species of palm, which is widely distributed in the mangrove forest of Southeast Asia. Various parts of N. fruticans has been used as a traditional medicinal plant. However, the physiological activities of N. fruticans has not yet been clarified well. Therefore, in this study, the 50% ethanol extract and its aqueous and ethyl acetate fractions of young shoots of N. fruticans were investigated for their antioxidant, cytoprotective effect, and antimicrobial activities. Every sample possessed very high free radical and various ROS scavenging capacities assessed by employing different in vitro assays such as $DPPH^{\cdot}$, $O_2^{{\cdot}-}$, ${\cdot}OH$, and $^1O_2$ scavenging activities. Based on these results, the cytoprotective effect was investigated using the oxidative hemolysis of erythrocyte. We found that the extract and fractions provide a greater protective effect compared with (+)-${\alpha}$-tocopherol. Furthermore, antimicrobial activities were confirmed against skin pathogens by broth microdilution assay. The ethyl acetate fraction had much higher antimicrobial activities than methyl paraben against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans. Taken together, our results indicated that the young shoots of N. fruticans may have the potential role as a natural active ingredient through their antioxidant, cytoprotective effect, and antimicrobial activities.

배양 인체피부섬유모세포에 있어서 활성산소의 산화적 손상에 대한 산사(山査)추출물의 항산화 효과 (Antioxidant Effect of Crataegi Fructus Extract on the Oxidative Stress of Reactive Oxygen Species in Cultured Human Skin Fibroblast)

  • This study was done to evaluate the antioxidant effect of Crataegi Fructus (CF) extract on the oxidative stress induced by reactive oxygen species (ROS), The human skin fibroblasts (Detroit 551) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cytotoxicity of $H_2O_2-induced$ oxidative stress was performed by XTT assay for the cell viability according to the dose- and time-dependent treatment. For the protective effect of CF extract on $H_2O_2-mediated$ oxidative stress, cell viability, lactate dehydroganase (LDH) activity, and ferric thiocyanate (FTC) assay for the inhibitive activity of lipid peroxidation on CF extract were carried out. In this study, $H_2O_2-mediated$ oxidative stress was decreased cell viability dose-, and time-dependent manner and increased LDH activity compared with the control in these cultures. In the protective effect, CF extract increased cell viability and decreased LDH activity on $H_2O_2-mediated$ oxidative stress, especially, CF extract has antioxidant effect by the showing the inhibitive activity of lipid peroxidation by FTC assay. From these results, It is suggested that $H_2O_2-mediated$ oxidative stress was highly toxic, and also, CF extract showed the protective effect on $H_2O_2-mediated$ oxidative stress by showing the increased cell viability, decreased LDH activity and lipid peroxidation inhibition in these cultures.

히비스커스 추출물이 인간 각질 형성 세포의 밀착 연접 관련 유전자 발현에 미치는 영향 연구 (Investigation of the effect of Hibiscus sabdariffa L. extracts on tight-junction related genes in human keratinocyte HaCaT cells)

  • Objectives : Hibiscus (Hibiscus sabdariffa L.) is rich in antioxidants such as flavonoids and anthocyanins and is known to have anti-inflammatory activity and anti-aging function of the skin, but there is no study on its effect on the skin barrier. This study aim to investigate the positive effect on the skin barrier by confirming the effect of water extracts of Hibiscus sabdariffa L. (WEHS) on the tight junction-related gene expression. Methods : The antioxidant efficacy of WEHS was investigated through ABTS and DPPH assays, and 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium was performed to examine the effect on cell viability. quantitative Reverse transcription polymerase chain reaction and immunoblot analysis were performed to confirm the effect of WEHS on the expression of tight junction-related genes, and immunofluorescence microscopy was used to confirm the movement of Claudin 1 protein into tight junctions. Results : WEHS showed strong antioxidant activity and induced an increase in both mRNA and protein expression levels of Claudin 1 among tight junction-related genes. The strong localization of Claudine 1 protein increased by WEHS to the tight junction was confirmed by immunofluorescence microscopy. Conclusions : Hibiscus was confirmed through this study to show antioxidant activity and the function of promoting the expression of the tight junction Claudin 1 gene, suggesting that Hibiscus can be used as a material for the prevention and treatment of skin moisturizing and atopy, which have an important influence on tight junction.
