본 연구는 scopoletin 식이 보충이 알코올로 인해 유발되는 인슐린저항성과 항산화방어계에 미치는 영향을 구명하고자 하였다. 실험동물은 4주령의 수컷 SD계 흰쥐에게 총 열량의 36%에 해당하는 알코올을 액체식이 형태로 8주간 공급하였으며, scopoletin은 알코올 액체식이 리터당 0.01 g과 0.05 g 두 수준으로 첨가하였다. 정상군은 알코올대조군과 동량의 에너지를 섭취하도록 하였다. 8주간의 알코올 급여는 공복 시 혈당 변화를 일으키지 않았으나 혈청 인슐린 함량을 증가시켰으며, 이는 인슐린저항성과 내당능 장애를 유발하였다. 그러나 scopoletin 저농도와 고농도 급여군 모두 인슐린 함량, 인슐린저항성 지표 및 내당능을 효과적으로 개선하는 것으로 나타났다. 알코올대조군은 p-PI3K의 단백질 발현을 유의적으로 낮추어 glucokinase 유전자 발현과 활성을 억제한 반면, 당신생 효소인 glucose-6-phosphatase의 유전자 발현과 활성을 유의적으로 높였다. 그러나 scopoletin 급여에 의하여 이들 변화는 완화되었다. 다른 당신생 효소인 phosphoenolpyruvate carboxykinase의 유전자 발현과 활성에는 영향을 미치지 않았다. 또한 scopoletin 급여군 모두 간조직의 aldehyde dehydrogenase의 활성은 알코올 대조군에 비해 증가된 반면, cytochrome P450 2E1 활성은 억제되었다. 또한 알코올로 인하여 낮아진 간조직 중의 항산화 효소(superoxide dismutase, catalase와 glutathione peroxidase)의 유전자 발현과 활성을 높임으로써 과산화수소 및 지질과산화물의 함량을 낮추었다. 이와 같이 0.001%의 scopoletin 급여량에서도 당대사의 유전자 변화를 통하여 만성 알코올로 유도되는 인슐린저항성을 개선하였으며, 알코올대사계 활성 및 항산화방어계 효소의 유전자 발현을 증가함으로써 알코올로 인한 과산화수소와 지질과산화물 생성을 개선하는 것으로 나타났다.

Low soil pH causes from $H^+$ rhizotoxicity results in nutrients unavailability in the growing media, inhibits plant growth, development and reduces crop yields. The present study was carried out to reveal morpholophysiological and biochemical responses of wheat (Triticum aestivum L.) to acidity stress. Four wheat varieties viz. BARI Wheat-21, BARI Wheat-25, BARI Wheat-26 and BARI Wheat-30 were used in the study. Eight-day-old seedlings were exposed to different pH levels (3.5, 4.5, 5.5 and 6.5) of growing media. Acidity stress at any level reduced biomass, water, and chlorophyll contents in all the varieties; whereas BARI Wheat-26 showed the least damage. $H^+$ rhizotoxicity also caused oxidative stress through excess production of reactive oxygen species and methylglyoxal which increase lipid peroxidation in all the varieties but the lowest oxidative damage was observed in BARI Wheat-26 due to better performance of the antioxidant defense and glyoxalase systems. Considering the growth, physiological and biochemical attributes BARI Wheat-26 may be considered as acidity stress tolerant, among the variety examined.
The object of this study was to evaluate the effect of Bupleuri Radix, aqueous extracts of the root part of Bupleurum falcatum on the 6-n-propyl-2-thiouracil (PTU)-induced rat hypothyroidism. Aqueous extracts of Bupleuri Radix (BR; yield = 11.73%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 300 and 150 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), triiodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride, liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT), serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were observed with histopathology of thyroid glands. Results were compared with $LevoT_4$ 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, triglyceride contents, liver CAT activities and changes of serum thyroid hormone levels were observed with increases of serum AST, HDL contents, liver $H_2O_2$ and SOD activities and thyroid gland weight. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contents and sizes were demonstrated at histopathological inspections. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of BR extracts, and BR extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that BR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), tri-iodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride were observed with liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT) and serum asparte aminotransferase (AST) and alanine aminotransferase (ALT) analysis. Results were compared with Levothyroxine ($LT_4$) 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, serum thyroid hormone levels and triglyceride contents, liver $H_2O_2_3$ and SOD activities were observed with increases of serum AST and HDL contents, liver CAT activities, thyroid gland weight. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of LF extracts, and LF extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that LF extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.
Objectives : In present study, therefore, possible beneficial pharmacological activities of standard potato protein extracts (SPE) were observed on the mild diabetic obese mice. Methods : After end of 12 weeks of continuous oral administrations of three different dosages of SPE 400, 200 and 100 mg/kg, or metformin 250 mg/kg, analyzed the hepatoprotective, hypolipidemic, hypoglycemic, nephroprotective and anti-obesity effects, separately. In addition, liver antioxidant defense systems were additionally measured with lipid metabolism-related genes expressions and hepatic glucose-regulating enzyme activities for action mechanism. Results : All of diabetes and related complications including obesity were significantly inhibited by treatment of SPE 400, 200 and 100 mg/kg, dose-dependently, and they also dramatically normalized the hepatic lipid peroxidation and depletion of liver endogenous antioxidant defense system, the changes of the hepatic glucose-regulating enzyme activities, also changes of the lipid metabolism-related genes expressions including hepatic $AMPK{\alpha}1$ and $AMPK{\alpha}2$ mRNA expressions, dose-dependently. Especially, SPE 200 mg/kg constantly showed favorable inhibitory activities against type II diabetes and related complications as comparable to those of metformin 250 mg/kg in HFD mice, respectively. Conclusions : The present work demonstrated that SPE 400, 200 and 100 mg/kg showed favorable anti-diabetic and related complications including obesity refinement activities in HFD mice, through AMPK upregulation mediated hepatic glucose enzyme activity and lipid metabolism-related genes expression, antioxidant defense system and pancreatic lipid digestion enzyme modulatory activities.

Oxidative stress is a constant threat to all living organisms and an immense repertoire of cellular defense systems is being employed by most pro- and eukaryotic systems to eliminate or to attenuate oxidative stress. Ischemia and reperfusion is characterized by both a significant oxidative stress and characteristic changes in the antioxidant defense. Heme oxigenase-l (HO-l) is up-regulated by various stimuli including oxidative stress so that it is thought to participate in general cellular defense mechanisms against ischemic injury in mammalian cells. Higenamine, an active ingredient of Aconite tuber, has been shown to have antioxidant activity along with inhibitory action of inducible nitric oxide synthase (iNOS) expression in various cells. In the present study, we investigated whether higenamine and related analogs protect cells from oxidative cellular injuries by modulating antioxidant enzymes, such as HO-l, MnSOD etc. R-form of YS-51 was the most potent inducer of HO-l in bovine endothelial cells, which inhibited apoptotic cell death by H$_2$O$_2$. HO-1 induction by YS 51 was mediated by PI3 kinase activation in which PKA- as well as PKG pathway is considered as important regulators. YS-51 also induced Mn-SOD mRNA expression by activating c-jun N-terminal kinase in endothelial cells and Hela cells. In ROS 17/2.1 cells, higenamine and enetiomers of related compounds inhibited iNOS expression by cytokine mixtures. Taken together, higenamine and related compounds can be developed as possible protective agents from oxidative cell injury or death.
Smilacis Chinae Radix is root of Smilax china L.. It has been known as a korean folk medicine for anti-inflammatory, anti-microbial diuresis, detoxification and relieving enteropathy as diarrhea. Reactive oxygen species(ROS) are continuously produced at a high rate as a by-product obtained in the aerobic metabolism. A major portion of living organisms has defense system as superoxide dismutase or catalase against damage produced by ROS. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. In this study, we have investigated the antioxidant activities of Smilacis Chinae Radix in order to screen the antioxidant substances from natural products. As a result, EtOAc extract of Smilacis Chinae Radix exhibited potent antioxidant effect on various antioxidant experiment. The major components of antioxidant activity were isolated from EtOAc extract of Smilacis Chinae Radix. Their structure of compounds were identified as quercetin and (-)-epicatechin by spectroscopic evidence, respectively.

Antioxidant enzyme genes play a key role in cell defense against the lethal effects of oxidative stresses in animals and have an essential function which has allowed the evolution of aerobic respiration starting from an ancient form of oxygen-insensitive life. Piscine antioxidant enzymes are also involved in the rapid response to various toxic chemicals as well as many biological stresses, indicating that they could be used as biomarkers for health and aquatic environment. With the purpose for developing fine molecular probing tool to assess the stresses in marine fish, we identified three major antioxidant enzyme genes (superoxide dismutase, catalase and glutathione-S-transferase) from Korean rock bream using expressed sequence tag analysis and/or high density filter screening. Here we report the molecular information on these gene transcripts including complete sequence data and expression profiles.
In this study, the effects of replacing inorganic copper, zinc and manganese with different levels of organic complexes of the same trace minerals on the lipid peroxidation and antioxidant defense systems in broilers were investigated. Two-hundred Ross-308 one-day-old broiler chickens were placed on controlled diets until 42 d of age. The experimental animals were divided into four groups comprising three experimental groups and one control group, each consisting of 50 chickens. All groups were also divided into five subgroups each containing 10 broiler chicks. The mineral content of the control group diet was controlled using a standard inorganic mineral premix with supplement levels and sources of trace minerals typical of commercial broiler diets according to the National Research Council (NRC) (containing 8 mg Cu as $CuSO_4$, 40 mg Zn as $ZnSO_4$, and 60 mg Mn as MnO, per kg). In the experimental diets, mineral premix was also comprised of inorganic formulations, except for those of Cu, Zn and Mn. Organically-complexed Cu, Zn, and Mn were separately added to the basal diet at 1/3 (L1), 2/3 (L2) and 3/3 (L3) levels with respect to the NRC recommendation, as Bioplex $Cu^{TM}$, Bioplex $Zn^{TM}$, Bioplex $Mn^{TM}$. At the end of the trial, the plasma Zn level significantly increased when the plasma Cu level significantly decreased (p<0.05) in chickens fed at 2/3 and 3/3 levels of organically complexed minerals. The liver trace mineral concentrations were significantly higher in chickens fed inorganic trace minerals in comparison to those fed organically-complexed minerals. The plasma malondialdehyde (MDA) level of experimental chickens was decreased in groups receiving levels of organic Cu, Zn and Mn in comparison to those fed inorganic forms (p<0.01). The erythrocyte superoxide dismutase (SOD) activity was higher in all groups receiving the organic mineral supplements in comparison to those fed inorganic forms (p<0.01). No differences were observed on either the erythrocyte catalase (CAT) activity or the plasma ceruloplasmin (Cp) levels, and the liver MDA levels and liver CAT and SOD activities in any of the groups that received the organic supplements of Cu, Zn, and Mn. It was concluded that supplementation of lower levels of organically-complexed copper, zinc, and manganese instead of their inorganic forms in diets had no negative effects on the antioxidant defense system in broilers.
