Protective effect of soursop (Annona muricata linn.) juice on oxidative stress in heat stressed rabbits

  • Background: Preventing oxidative stress in heat stressed animals may be possible by increasing antioxidant defence via exogenous administration of antioxidant substrate and/or its precursors. The study aimed to investigate the effect of Soursop juice in mitigating oxidative stress induced by heat stress in rabbit. Methods: Sixty mixed breed rabbit bucks aged 12-18 months old with the average weight of $1826{\pm}8.35$ g/rabbit, randomly allotted to experimental treatments of four replicates each, in a completely randomized design during high-temperature humidity index in Ado Ekiti, Southwest Nigeria. Soursop juice (SSJ) was administered via oral drenched daily per kg body weight (BW), to designated treatment 1 to 5; $0.55mlkg^{-1}BW$ distilled water (control), $0.55mlkg^{-1}BW$ SSJ, $1.11mlkg^{-1}BW$ SSJ, $1.67mlkg^{-1}BW$ SSJ and $2.22mlkg^{-1}BW$ SSJ, respectively. Fastened blood samples were collected at days 28 and 56, and assay for serum protein, cholesterol, triglycerides, superoxide dismutase, catalase, reduced glutathione and lipid peroxidation using standard procedures. Result: Result revealed that SSJ demonstrated hypocholesterolemic effect in a dose-dependent manner throughout the study. Effect of chronic administration of SSJ to heat stressed rabbits proved beneficial, as SSJ reduced serum lipid peroxidation and enhanced antioxidant activity over 8 weeks. Conclusion: Administration of soursop juice to heat-stressed bucks at $2.22mlkg^{-1}BW$ offered optimum antioxidant defense against oxidative stress.

Protective Effects of the BuOH Fraction from Laminaria japonica Extract on High Glucose-induced Oxidative Stress in Human Umbilical Vein Endothelial Cells

  • This study investigated the protective effect of the butanol (BuOH) fraction from Laminaria japonica (BFLJ) extract on high glucose-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). Freeze-dried L japonica was extracted with distilled water, and the extracted solution was mixed with ethanol then centrifuged. The supernatant was subjected to sequential fractionation with various solvents. The BuOH fraction was used in this study because it possessed the strongest antioxidant activity among the various solvent fractions. To determine the protective effect of the BFLJ, oxidative stress was induced by exposing of HUVECs to the high glucose (30 mM) or normal glucose (5.5 mM) for 48 hr. Cell viability, lipid peroxidation, glutathione (GSH) concentration, and antioxidant enzyme activities such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), and glutathion reductase (GSH-re) were measured. Exposure of HUVECs to high glucose for 48 hr resulted in a significant (p<0.05) decrease in cell viability, SOD, GSH-px and GSH-re and a significant (p<0.05) increase in thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5.5 mM glucose or untreated with glucose. BFLJ treatment decreased TBARS formation and increased cell viability, GSH concentration, and activities of antioxidant enzymes including catalase, SOD, GSH-px, and GSH-re in high glucose pretreated HUVECs. These results suggest that BFLJ may be able to protect HUVECs from high glucose-induced oxidative stress, partially through the antioxidative defence systems.

  • The environmental pollutions were a serious problem in Korea recently. So many researcher have studied the effect of environmental pollution on plants and agro-ecosystem, but the basic mechanisms of environmental stresses were various. One of the important mechanisms was oxidative stress caused by active toxic oxygen. The toxic oxygen was generated by several stresses, abnormal temperature, many xenobiotics, air pollutants, water stress, fugal toxin, etc. In the species of toxic oxygen which is primary inducer of oxidative stresses, superoxide, hydrogen peroxide, hydroxyl radical and singlet oxygen were representative species. The scavenging systems were divided into two groups. One was nonenzymatic system and the other enzymatic system. Antioxidants such as glutathione, ascorbic acid, and carotenoid, have the primary function in defense mechanisms. Enzymatic system divided into two groups; First, direct interaction with toxic oxygen(eg. superoxide dismutase). Second, participation in redox reaction to maintain the active antioxidant levels(eg. glutathione reductase, ascorbate peroxidase, etc.).

백두산 자생참돌꽃의 생육과 이용 II. 유식물의 부위별 superoxide dismutase 활성 (Growth and Utility of Rhodiola sachalinensis in Baekdu Mountain II. Activities of Superoxide Dismutase in Portions of the Seedlings)

  • 참돌꽃 유식물에서 줄기와 뿌리의 부위별로 나타나는 superoxide dismutase(SOD)의 활성과 환경 스트레스 또는 항산화분자 처리에 따른 SOD활성도를 조사하여 다음과 같은 결론을 얻었다. SOD의 활성은 줄기보다는 뿌리에서 높았고 각 기부보다는 선단부에 이를수록 높았으며 이러한 현상은 유식물 생육의 초기보다 후기에 더 상승된 결과를 나타내고 있다. 또한 환경 스트레스에 대한 SOD의 활성도는 유식을 초기생육 상태에서는 NaCl또는 cadmium(Cd) 등 유해물질 처리 후 10 및 30% 정도 저해를 받고 있으나 생육 후기에 이를수록 처리된 유해인자에 의한 산화적 스트레스에 대하여 유식물은 자체의 SOD 활성 수준이 상향되는 방어능력을 보였으며 이러한 현상은 뿌리에서 더욱 현저하였다. SOD 활성도를 높이기 위해 항산화물질로 ascorbic acid를 처리한 유식물은 생육 후 최고 46%까지 높아짐을 보였으나 동시기의 스트레스 물질로 첨가한 cadmium 처리구에서 나타난 SOD 활성도에 비하여 유의할만한 상승효과의 차이를 보이지는 않았다. 이러한 결과는 참돌꽃 유식물 특히 뿌리부위는 항산화 기질의 처리 없이도 유해한 환경 인자에 노출되면 자체의 방어 수단으로 SOD 생성이 활발히 진행되고 있음을 시사하였다.

Effects of Hyperbaric Oxygen and $\alpha$-Tocopherol on Skin Antioxidant Enzymes Defence in Rats

  • In order to test the effects of hyperbaric oxygen (HBO) and $\alpha$-tocopherol on full-thickness skin grafts in rats, we peeformed full-thickness skin grafts bilaterally on rats. After surgery, we analyzed the tissue-concentrations of superoxide dismutase (SOD), catalase, and glutathione peroxidase(GPx)/reductase(GPr) on days 0, 2, 4, 7, 10, 14, 21 and 28. The four groups had similar patterns of change in SOD, catalase, GPx and GPr values. SOD increased initially, and was significantly increased at day 7, returning to the preoperative activity level on day 14 (control, HBO, and $\alpha$-tocopherol treated alone) and 28 (HBO plus $\alpha$-tocopherol). Catalase had a similar pattern of change as the SOD enzyme activity, except for the surgical control on day 2. Glutathione peroxidase/reductase activity in the four groups had a similar pat-tern of enzyme activity, with a significant increase from preoperative level on day 4, peaking during days 7 to 10, and returning to preoperative level on day 21(surgical control, HBO, and $\alpha$-tocopherol-treated alone) and 28 (HBO plus $\alpha$-tocopherol treated group). Hence, the clinical use of HBO and $\alpha$-tocopherol mixture can be recommended as an adjunctive treatment for free skin grafts in rats. But, the antioxidant used, its dose, and the timing of its administration, as well as, the exposure time and the pressure of HBO, should be the subject of further research.

  • Thiol peroxidase인 E.coli AhpC의 아미노산 서열을 database를 이용해 분석하여, TPx와 상동성이 있는 새로운 형태의 Thiol peroxidase를 찾아내었다. 그 중 병원성을 갖는 박테리아인 Haemophilus Influenzae에서 존재하는 TPx와 유사하고, GRX와 함께 fusion 되어있는 새로운 형태의 단백질의 유전자를 클로닝하여 E.coli에서 과발현시켜 분리정제 하였다. 정제된 TPx-GRX는 환원제로 thiol 성분을 갖는 MCO system(Fe, DTT, Oxygen)에 의하여 Glutamine Synthetase(GS)의 불활성화를 방어하는 티올 특이적 향산화활성을 갖고, peroxides를 제거하는 peroxidase 활성을 갖는 것을 밝혔다. 이 결과들로부터 TPx-GRX는 새로운 형태의 Thiol perosidase임을 알 수 있었다. 더 나아가서 이 결과들은 TPx-GRX가 병원성 박테리아에서 oxidative stress를 막는 생리적으로 중요한 역할을 할 것이라는 것을 시사한다.

운동량이 젊은 성인 남자의 비타민 C와 E 섭취량 및 혈장 비타민 C, $\alpha$-tocopherol 및 $\gamma$-tocopherol 수준에 미치는 영향 (The Effects of Exercise on the Vitamin C and E Intakes and Their Plasma Levels of Vitamin C, $\alpha$-tocopherol and $\gamma$-tocopherol in Young Male Adults)

  • We investigated the effect of physical exercise on the intakes and plasma levels of vitamin C and E in young male adults. The study population were divided into two groups: small-amount exercisers($\leq$ 30min/day, n=29) and moderate-amount exercisers(> 30min/day, n=32) according to their physical exercise habits measured by a questionnaire. Dietary intakes of vitamin C and E of the subjects were estimated by the 24 hour recall method. Plasma lipid profiles, vitamin C, $\alpha$-tocopherol and ${\gamma}$-tocopherol levels were analyzed. No significant differences were observed in total cholesterol, TG, HDL-C and LDL-C of plasma between two groups. Plasma vitamin C and $\alpha$-tocopherol levels in moderate-amount exercisers were significantly higher than those in small-amount exercisers, although dietary intakes of vitamin C and vitamin E were not significantly different between two groups. However, plasma ${\gamma}$-tocopherol level of moderate-amount exercisers did not significantly differ from small-amount exercisers. There was a positive correlation between the amount of exercise and plasma $\alpha$-tocopherol level. These results suggest that moderate amount of exercise over 30 min per day would lead to a healthy life with an activated antioxidant defence system against oxidative stress.(Korean J Nutrition 34(3) : 306~312, 2001)

유도선수들의 반복운동이 혈중 코티졸과 지질과산화 및 creatine kinase 활성에 미치는 영향 (The Effects of Repetitive Exercise on the Blood Cortisol, MDA, and Creatine Kinase Activity in Judoist.)

  • The purpose of the present study is to investigate the lipid peroxidation, creatine kinase activity and cortisol hormone levels following the training intensity in elite judo players. Six elite Judo players participated in the experiments (3h repetition judo program), which include stretching, judo skill practice and cool down without recess. Blood sampling were taken at the judo gymnasium at the time of resting, 1h training, 2h training, 3h training, 2h recovery, and 24h recovery time and this were analyzed for CK, MDA and Cortisol levels. The results obtained were analyzed via repeated measures of ANOVA using SPSS package program (ver.10.0) and a value of p<.05 was considered statistically significant. The results from this study were as follows. In the CK levels, which reflect the contribution of creatine phosphate and muscle damage degree, there was a significant difference (p<.05) after judo training in every period. Recovery 24h showed the highest level. In the MDA levels, which reflect lipid peroxidation, there was a significant difference (p<.05) after judo training. Recovery 2h showed the lowest level. In the cortisol hormone levels, which reflect stress status, there was a significant difference (p<.05). In this study, we can conclude that For the trained athletes, MDA level was lower at the time of exercise compare to the other period, this is caused by the increased antioxidant defence mechanism.

훈련방법의 차이가 SOD, Neutrophils 및 T세포에 미치는 영향 (The Effect of Different Type of Exercise on SOD, Neutrophils and T Lymphocytes)

  • Background: A physically active lifestyle and regular exercise training incurs many health benefits. One recently recognized benefit of regular moderate exercise is stress reduction and immune enhancement. Thus, a physical stress such as exercise may act at any number of points in the complex sequence of events collectively termed the immune response. Although exercise causes many propound changes in parameters of immune function, the nature and magnitude of such changes rely on several factors including the immune parameters of interest; type, intensity, and duration of exercise; fitness level or exercise history of the subject; environmental factors such as ambient temperature and humidity. Methods: This study was undertaken to investigate the effect of different type of exercise on superoxide dismutase (SOD), neutrophils, and T lymphocytes of Sprague-Dawley rats. Sprague-Dawley rats were randomly divided into three groups; a non-Trained group (NTG, n=6), a swim-Trained group (STG), and a treadmill-Trained group (TTG). The exercise regimen was designed in a treadmill (5 times/5 days/week) during 8-weeks for TTG, and swim training (5 times/5 days/week) during 8-weeks for STG, and the volume of exercise training was the same in both groups. Results: 8 weeks of regular swim and treadmill training significantly increased liver SOD concentration however, muscle SOD concentration was not statistically significant. In the level of neutrophils, TTG and STG showed significant difference, compared to NTG. TTG was the highest level of neutrophils. In the level of immune cell counts, there was significant difference among TTG, STG, and NTG both in the spleen and thymus. Conculsion: In conclusion, it can be stated that eight weeks swim and treadmill exercise training has beneficial effect in improving immune response and antioxidant defence capacity by augmenting immune cells and SOD activities of SD rats.

폐(肺) 조직(組織)에서 호도(胡桃) 추출액(抽出液)이 산화성(酸化性) 세포(細胞) 손상(損傷)의 방어기전(防禦機轉)에 미치는 영향(影響) (The beneficial effect of Juglandis Semen extraction on oxidant-induced lung cell injury)

  • 호도(胡桃)는 보양약(補陽藥)으로써 하초(下焦)를 온보(溫補)하고 원기(元氣)를 섭납(攝納)하며 윤폐신(潤肺腎)하는 효능(效能)이 있어 오래된 허한해수(虛寒咳嗽), 천식(喘息)의 치료(治療)에 사용되어 왔는데 폐(肺) 조직내(組織內)에서 산소유리기(酸素遊離基)들에 의(依)한 세포(細胞) 손상(損傷)의 방지(防止)여부를 알아보기 위(爲)하여 산화제(酸化劑)인 t-butylhydroperoxide(t-BHP)와 $H_2O_2$로써 세포(細胞) 손상(損傷)을 유발한 후 호도(胡挑) 추출액(抽出液)의 항산화(抗酸化) 효과(效果) 및 항산화(抗酸化) 효소(酵素)의 활성(活性)에 미치는 영향(影響)와 산소유리기(酸素遊離基)에 대(對)한 직접 소거효과(消去效果)를 조사(調査)하였다. 이 결과 호도(胡桃)는 폐(肺) 조직(組織)에서 지질(脂質)의 과산화(過酸化)를 억제(抑制)함으로 oxidant에 의(依)한 폐부종(肺浮腫) 유발(誘發)을 방지(防止)할 수 있음을 보여주었고 세포내(細胞內) glutathione의 농도(濃度) 및 항산화(抗酸化) 효소(酵素) 중(中) catalase와 superoxide dismutase의 활성(活性)에는 변화(變化)를 유발하지 못하였으나 glutathione peroxidase의 활성(活性)을 유의(有意)하게 증가(增加)시켰으며, superoxide radical과 hydroxyl radical의 생성(生成)을 감소(減少)시켰다. 따라서, 폐(肺) 조직(組織)에서 산화성(酸化性) 세포(細胞) 손상(損傷)에 대(對)한 호도(胡桃)의 보호(保護) 효과(效果)는 부분적으로 세포내(細胞內) 항산화(抗酸化) 효소(酵素)의 활성(活性) 증가(增加)와 산소유리기(酸素遊離基)들을 직접 소거(消去)시키는 작용(作用)에 기인(起因)하는 것으로 사료된다.
