Antioxidant Effects of Cranberry Powder in Lipopolysaccharide Treated Hypercholesterolemic Rats

  • This study was conducted to investigate the effects of cranberry power on antioxidant defense system in rats fed an atherogenic diet and injected with lipopolysaccharide (LPS). Sprague-Dawley rats were divided into the following 5 groups: normal diet+saline (NS), atherogenic diet+saline (AS), atherogenic diet+LPS (AL), atherogenic diet with 5% cranberry powder+LPS (AL-C5), and atherogenic diet with 10% cranberry powder+LPS (AL-C10). Total antioxidant status measured by ferric reducing ability of plasma (FRAP) was significantly reduced by LPS injection (24%) and was restored by the cranberry powder treatment (P<0.05). In addition, the mean level of plasma total phenolics was significantly decreased by LPS injection (P<0.05) and tended to be increased when cranberry powder was incorporated in to the diet. Activity of serum superoxide dismutase (SOD) tended to be lowered by LPS injection and declined further in cranberry powder fortified groups. Overall results indicate that dietary cranberry powder may provide appropriate antioxidants to counter the diminished antioxidant status induced by exposing hypercholesterolemic rats to LPS.

  • This study was conducted of investigate the antioxidant vitamins and serum lipids status of 23 healthy female subjects and 22 female patients suffering from cerebrovascular disease hospitalized at any of three of general hospitals in Taegu. The systolic blood pressure of patients suffering from cerebrovascular disease(151.4$\pm$30.2mmHg) was significantly higher than healthy controls(129.7$\pm$17.8mmHg). The frequency of drinking alcohol and smoking in patients was also higher. Analysis of patient and control dietary intakes revealed that all subjects were consuming the minimed recommended energy-nutrient levels. HDL-cholesterol(26.15 vs 37.74mg/dl), serum vitamin E(12.69 vs 17.74mg/l) and vitamin C(65.59 vs 152.50ug/dl) levels were significantly lower, but triglyceride(221.01 vs 87.30 mg/dl) and total-cholesterol / HDL-cholesterol levels(5.98 vs 4.89)were significantly higher in the patient group. According to the results of this study, it is expercted that refraining from smoking and drinking alcohol, cultivating healthy living habits which can decrease stress, and maintaining suitable levels of serum antioxidant vitamins can prevent the occurrence of cerebrovascular disease.

Oxidative Stress and Antioxidant Status during Transition Period in Dairy Cows

  • The study was conducted on 20 Holstein X Sahiwal cross bred dairy cows, with an average milk production of $2,752{\pm}113.79$ liters in $284{\pm}5.75$ days during a single lactation, that were divided in to two groups of 10 animals. We investigated the oxidative stress and antioxidant status during the transition period in dairy cows. In this study, plasma level of MDA was considered as an indicator of lipid peroxidation and SOD, catalase, GSH and GSHPx as antioxidants. The lipid peroxidation was significantly (p<0.001) higher in cows during early lactation as compared to the cows in advanced pregnancy. A significant positive correlation (r = +0.831, p<0.01) was determined between MDA and catalase in early lactating cows. In early lactating cows, blood glutathione was significantly lower than in advanced pregnant cows. However, early lactating cows showed non-significant negative correlation for all antioxidant enzymes with lipid peroxidation. In conclusion, dairy cows seemed to have more oxidative stress and low antioxidant defense during early lactation or just after parturition than advanced pregnant cows, and this appears to be the reason for their increased susceptibility to production diseases (e.g. mastitis, metritis, retention of fetal membranes etc.) and other health problems.

Effect of Supplemental Selenomethionine on Growth Performance and Serum Antioxidant Status in Taihang Black Goats

  • An experiment was conducted to evaluate the effect of different levels of supplemental selenomethionine (Se-Met) on growth performance and serum antioxidant status in Taihang Black goats. Fifty 16-week-old goats with an average body weight of 12.5${\pm}$0.5 kg were randomly assigned to five treatments fed a basal diet (0.049 mg Se/kg DM) supplemented with 0 (control), 0.10, 0.30, 0.50 and 1.00 mg of Se/kg DM (form Se-Met) for 80 days. Average daily gain and feed efficiency were higher (p<0.05) in the groups supplemented with 0.30 to 0.50 mg Se/kg DM compared with the control group. However, Se-Met supplementation had no influence on average daily feed intake (p>0.05). Se-Met supplementation significantly increased (p<0.01) the activity of glutathione peroxidase enzymes (GSH-Px) and superoxide dismutase (SOD) in serum. The group supplemented with 0.50 mg Se/kg DM had the highest activity of GSH-Px compared with other groups (p<0.05). Serum SOD activity was higher (p<0.05) in goats supplemented with both 0.30 and 0.50 mg Se/kg DM than in control goats and goats supplemented with 1.00 mg Se/kg DM. Serum glutathione-S-transferase (GST) activity and malondialdehyde (MDA) concentration were significantly decreased (p<0.05) in goats supplemented with 0.30, 0.50 and 1.00 mg Se/kg DM compared with control values. These results indicated that Se-Met supplementation markedly improved the antioxidant status in goats. Blood Se concentration increased linearly (p<0.001) and quadratically (p<0.001) as the level of supplemental Se-Met increased. The concentration of Se in the control diet (0.049 mg Se/kg DM) did not satisfy the Se requirement in goats as indicated by reduced growth rate, feed efficiency, activities of GSH-Px and SOD in serum, and blood Se concentrations. In conclusion, it is recommended that 0.30 to 0.50 mg of Se/kg DM from Se-Met (total diet Se of 0.349 to 0.549 mg/kg DM) be supplied in the diet of Taihang Black goats to enhance growth performance and improve antioxidant status.

Effects of N-acetylcysteine on the energy status and antioxidant capacity in heart and liver of cold-stressed broilers

  • Objective: Cold stress induces oxidative damage and impairs energy status of broilers. N-acetylcysteine (NAC) exhibits antioxidant properties and modulates energy metabolism of animals. This study was conducted to investigate the effects of NAC on energy status and antioxidant capacity of heart and liver in the cold-stressed broilers. Methods: The experiment consisted of 4 treatments in a 2×2 factorial arrangement with two diets (basal diet or plus 0.1% NAC) and two ambient temperatures (thermoneutral [conventional ambient temperature] or cold stress [10℃±1℃ during days 15 to 42]). Results: No ascites were seen in cold-stressed broilers. NAC did not attenuate the impaired growth performance of stressed birds. However, NAC decreased plasma asparagine but increased aspartate levels in cold-stressed birds (p<0.05). NAC reduced hepatic adenosine triphosphate (ATP) but elevated adenosine diphosphate contents in unstressed birds (p<0.05). The hepatic ratio of adenosine monophosphate (AMP) to ATP was increased in birds fed NAC (p<0.05). NAC decreased plasma malondialdehyde (MDA) level and cardiac total superoxide dismutase (T-SOD) activity in unstressed birds, but increased hepatic activities of T-SOD, catalase and glutathione peroxidase in stressed birds (p<0.05). NAC down-regulated hepatic AMP-activated protein kinase but up-regulated cardiac heme-oxigenase mRNA expression in stressed birds, and decreased expression of hepatic peroxisome proliferator-activated receptor coactivator-1α as well as hypoxia-inducible factor-1α in liver and heart of birds. Conclusion: Dietary NAC did not affect energy status but enhanced the hepatic antioxidant capacity by increasing the activities of antioxidant enzymes in cold-stressed broilers.

Effects of different selenium sources and levels on antioxidant status in broiler breeders

  • Objective: This study was conducted with the objectives to examine the impacts of inorganic selenium (Se) and different types and levels of organic selenium on the serum and tissues Se status and antioxidant capacity in broiler breeders. Methods: Five hundred and forty 48-wk-old Lingnan Yellow broiler breeders were randomly assigned to 6 dietary treatments, provided same basal diet (0.04 mg/kg of Se) with 0.15 mg/kg, or 0.30 mg/kg of Se from sodium selenite (SS) or from selenium-enriched yeast (SY) or from selenomethionine (SM). The broiler breeders were slaughtered after an 8-wk experiment. Results: The results showed that SM was better than SY and SS, 0.30 mg/kg level was better than 0.15 mg/kg level in Se deposition (p<0.05) in serum, liver, kidney, pancreas and muscle; in antioxidant status, organic selenium had better effects than SS in broiler breeders (p<0.05), but SM and SY had a similar result, and 0.15 mg/kg level was better than 0.30 mg/kg (p<0.05). Conclusion: The results demonstrated the evident advantage of supplementation of broiler breeders with 0.15 mg/kg SM, which improved tissue Se concentrations and antioxidant status, and can be considered as the best selenium source.

Effect of night light regimen on growth performance, antioxidant status and health of broiler chickens from 1 to 21 days of age

  • Objective: The study was conducted to evaluate the effects of night light regimen on growth performance, antioxidant status and health of Lingnan Yellow broiler chickens from 1 to 21 days of age. Methods: A completely randomized factorial design involved 2 photoperiods (constant lighting [CL], 24 L:0 D and intermittent lighting [INL], 17 L:3 D:1 L:3 D)${\times}2$ light intensities (10 lx and 30 lx). A total of one thousand six hundred and eighty 1-d-old Lingnan Yellow broiler chicks were randomly divided into 4 treatments with 6 replicates (70 birds per replicate). The experiment lasted for 21 d. Results: Photoperiods and light intensities had no effect on average daily gain, feed conversion ratio, and mortality of the broiler chickens (p>0.05). The INL had a significant effect on average daily feed intake (p<0.05) of broiler chickens compared with CL. Photoperiod and light intensity had an interactive effect on melatonin (MT) concentration (p<0.05). At CL, reducing light intensity increased MT concentration; INL birds had higher MT but MT concentration was not affected by light intensity. There was an interactive effect on glutathione peroxidase (GPx) and catalase (CAT) in serum and total antioxidant capability (T-AOC) in liver between photoperiod and light intensity. With the decrease of light intensity, the activities of GPx and CAT in serum and T-AOC in liver increased in CL group (p<0.05). Broiler chickens reared under INL had better antioxidant status and 10 lx treatments had higher activities of CAT in serum than 30 lx (p<0.05). Different photoperiods and light intensities had no effect on malondialdehyde. There was an interaction between photoperiod and light intensity on serum creatine kinase (CK) concentration (p<0.05). At CL, the elevated light intensity resulted in an increase in CK content; INL birds had lower CK concentration especially in low light intensity group. Besides, INL and low light intensity significantly reduced the concentration of serum corticosterone and heat shock protein 70 (p<0.05). Serum immunoglobulin M contents were increased in broiler chickens reared under the INL compared with CL group (p<0.05). Conclusion: Results above suggest that the night light regimen of INL and 10 lx could be beneficial to the broiler chickens from 1 to 21 days of age due to the better health status and electricity savings.

  • Glutathione S-transferase (GST) is a multigene family of phase II detoxifying enzymes that metabolize a wide range of exogenous and endogenous electrophilic compounds. GSTM1 and GSTT1 gene polymorphisms may account for inter-individual variability in coping with oxidative stress. We investigated the relationships between the level of lymphocyte DNA and antioxidative parameters and the effect on GST genotypes. GSTM1 and GSTT1 were characterized in 301 young healthy Korean adults and compared with oxidative stress parameters such as the level of lymphocyte DNA, plasma antioxidant vitamins, and erythrocyte antioxidant enzymes in smokers and non smokers. GST genotype, degree of DNA damage in lymphocytes, erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase (GSH-Px), and plasma concentrations of total radical-trapping antioxidant potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene, and cryptoxanthin were analyzed. Lymphocyte DNA damage assessed by the comet assay was higher in smokers than that in non-smokers, but the levels of plasma vitamin C, ${\beta}$-carotene, TRAP, erythrocyte catalase, and GSH-Px were lower than those of non-smokers (p < 0.05). Lymphocyte DNA damage was higher in subjects with the GSTM1- or GSTT1-present genotype than those with the GSTM1-present or GSTT1- genotype. No difference in erythrocyte antioxidant enzyme activities, plasma TRAP, or vitamin levels was observed in subjects with the GSTM1 or GSTT1 genotypes, except ${\beta}$-carotene. Significant negative correlations were observed between lymphocyte DNA damage and plasma levels of TRAP and erythrocyte activities of catalase and GSH-Px after adjusting for smoking pack-years. Negative correlations were observed between plasma vitamin C and lymphocyte DNA damage only in individuals with the GSTM1-present or GSTT1- genotype. The interesting finding was the significant positive correlations between lymphocyte DNA damage and plasma levels of ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. In conclusion, the GSTM1- and GSTT1-present genotypes as well as smoking aggravated antioxidant status through lymphocyte DNA damage. This finding confirms that GST polymorphisms could be important determinants of antioxidant status in young smoking and non-smoking adults. Consequently, the protective effect of supplemental antioxidants on DNA damage in individuals carrying the GSTM1- or GSTT1-present genotypes might show significantly higher values than expected.

폐경기 여성의 항산화 비타민 영양 상태와 혈중 지질 및 MDA 농도와의 관계 (Correlation of the Nutritional Status of Antioxidant Vitamins and Serum Lipids and MDA Levels in Postmenopausal Women)

  • This study investigated the correlation of the nutritional status of antioxidant vitamins and serum lipids and MDA levels in postmenopausal women. Data about general characteristics, dietary intakes and biochemical parameters, including serum lipids, MDA and antioxidant vitamins levels, were collected from 85 postmenopausal women. The subjects were classified into three groups according to their serum total cholesterol level: normocholesterolemia group (NC, < 200 mg/dL), moderate hypercholesterolemia group (MC, $200{\sim}239mg/dL$) and hypercholesterolemia group(HC, ${\geq}240mg/dL$). The results are as follows. 1) General characteristics and serum MDA levels were not significantly different among the three groups. 2) Daily nutrients intakes adjusted to energy intake were not significantly different among the three groups, and were compatible with dietary reference intakes (DRIs) for Koreans. 3) Dietary Vt. A, ${\beta}-carotene$, Vt. C and Vt. E intake were not significantly different among the groups, while Vt. E intake was positively related with serum TC (r=0.288, p<0.05) and triglyceride (r=0.341, p<0.001) levels. 4) Serum Vt. A level standardized by serum TC level was significantly low and serum Vt. E level was significantly high in the HC group. Serum Vt. E level was positively related with serum TC level (r=0.389, p<0.001). 5) Dietary Vt. E intake was negatively correlated to serum MDA level (r=-0.242 p<0.05). Serum Vt. C and Vt. E levels were also negatively correlated to serum MDA level (r=-0.312, p<0.001 and r=-0.299, p<0.05). When the correlation was analyzed only in the group with hypercholesterolemia, correlation coefficients between the antioxidant vitamin and serum MDA level were higher. We concluded that intakes of antioxidant vitamins can contribute to decreasing the risk of cardiovascular disease by decreasing the oxidative stress of body rather than by controlling serum lipid levels.

한국 정상성인의 항산화 관련 혈청 무기질 농도에 관한 연구 (A Study on Serum Concentrations of Antioxidant Minerals in Normal Korean Adults)

  • Studies on relationships between antioxidant nutrients and various chronic diseases are increasing. In this study , serum concentrations of antioxidant minerals(Se, Cu, Zn & Mn) wee measured and compared among normal Korean adults. The ICP -MS method was employed to measure mineral concentrations. The subjects consisted of 329 healthy adults(154 men & 175 women) aged 20-79 years. The mean serum concentration appeared to be 26.4$\pm$0.41 (Se), 89.6$\pm$1.53(Cu), 113.3$\pm$1.56(Zn) & 0.34$\pm$0.05(Mn)$\mu\textrm{g}$/㎗ for men and 24.5$\pm$0.32(Se), 95.1$\pm$1.58(Cu), 133.6$\pm$1.79(Zn) & 0.35$\pm$0.02(Mn) $\mu\textrm{g}$/㎗ for women respectively. Among antioxidant minerals measured, the mean serum levels of Se and Zn were higher in men than in women(p<0.001), whereas that of Cu was higher in women(p<0.01). No significant difference between sex was observed for the mean Mn concentrations. The mean serum levels of Se(p<0.01) and Zn(p<0.05) decreased significantly in the 60's women On the other hand, the Mn concentrations in the 60's of women showed a significant increase(p<0.01). This difference seems to be due to the changes in menopausal status . The Cu levels in men showed a consistent increase with advancing age. The mean Cu/Zn ratios were 0.83 $\pm$0.04 for men and 0.95$\pm$0.02(p<0.01) for women respectively. The fact that this ratio increased significantly at the 60's of men (1.26 ; p<0.001) requires further study in relation to chronic diseases. When analyzed by stepwise multiple regression, drinking positively influenced serum Se and Cu levels for both men and women , smoking, however, negatively influenced all serum mineral levels for women. Further studies are needed to clarify precise roles of antioxidant minerals, their interactions and their relation to disease status.
