Antifungal, Nematicidal and Antioxidant Activity of the Methanol Extracts Obtained from Medicinal Plants

  • The nematicidal, antifungal and antioxidant activities of methanol extracts from six Vietnamese native medicinal plants were evaluated by various assays in vitro. Of the plant extracts tested, Terminalia nigrovenulosa was found to possess the highest activity when compared to the others. The leaves and bark of T. nigrovenulosa showed strong inhibitory activity against Meloidogyne incognita and Fusarium solani. The DPPH (1,1-Diphenyl-2-picryl-hydrazyl) radical scavenging, reducing power and total antioxidant activities of T. nigrovenulosa bark were higher than that of the remainder plant extracts. Chitinase activity of these plants was also investigated using SDS-PAGE. The results obtained in the present study indicate that T. nigrovenulosa leaf extracts are the greatest potential source as nematicides and fungicides for the control of M. incognita and F. solani. In particularly, T. nigrovenulosa bark extracts could be used as a potential source of commercially viable levels of natural antioxidant.

Phytochemical and Antioxidant Properties of Korean Wheat Sprouts

  • Wheat is an economically important cereal worldwide and is a staple food in many countries. Extensive researches have been performed on the nutritional value and antioxidant potentials of sprouts of legumes like soybean, however very limited studies have been carried out on the sprouts of Korean wheat cultivar. The objective of this study was to investigate the phytochemical and antioxidant potentials of Korean wheat sprouts. The sprouts were grown for 10 (WS-10), 20 (WS-20), and 30 (WS-30) days at room temperature. The pH, color, chlorophyll, free amino acid, 1,1-diphenly-2-picrylhydrazyl (DPPH), and total polyphenol content of the sprout samples were determined. The pH of sprouts was increased in the older sprouts. Color values were significantly influenced by the age of sprouts. The chlorophyll content was lowest but the total free amino acid content was highest in WS-30. On the other hand, the DPPH free radical scavenging potential and total polyphenol content were lower in WS-30. The results suggested that Korean wheat sprouts could be a potential source of nutrients and natural antioxidants.

Effect of Berberis tinctoria leaf (Berberidaceae) extract on antidiabetic, antihyperlipidemic and antioxidant status in streptozotocin induced diabetes in rats

  • The present study was carried out to investigate the antidiabetic and antioxidant effect of methanolic extract of Berberis tinctoria leaves (MEBT), in streptozotocin induced diabetic rats. Oral administration of MEBT extract (150 mg/kg and 300 mg/kg) for a period of 14 days. Blood glucose levels, body weight food and liquid intake were measured on every $5^{th}$ day over a period of 14 days. In diabetic rats, MEBT at the dose of 150 mg/kg and 300 mg/kg body weight resulted in significant reduction in blood glucose levels. The study was further investigated to determine antioxidant and antihyperlipidemic potential of MEBT in streptozotocin (STZ) induced diabetic rats. These results suggest that the MEBT possess antidiabetic activity and is able to ameliorate biochemical damages in STZ induced diabetic rats and the results were found to be in a dose dependent manner.

Potential Antioxidant Activities of Enzymatic Digests from Fresh Water Microalgae, Pediastrum duplex and Dactylococcopsis fascicularis

  • In this study, we focused on natural water-soluble antioxidants from fresh water microalgae, Pediastrum duplex and Dactylococcopsis fascicularis from Jeju Island, Korea. They were prepared by enzymatic digestion using five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase), and the potential antioxidant activity of each was assessed. All enzymatic digests from P. duplex showed significant DPPH scavenging effects. Termamyl (60.6%) digest from P. duplex possessed the highest effects on hydrogen peroxide scavenging. Celluclast (58.1%) and Kojizyme digests (56.9%) from D. fascicularis possessed higher effects on superoxide anion radical scavenging. All enzymatic digests exhibited significant effects on both NO· scavenging and metal chelating. Lipid peroxidation was significantly in inhibited Viscozyme, Termamyl and Kojizyme digests from P. duplex and Ultraflo, Protamex, Kojizyme and Alcalase digests from D. fascicularis. These data suggest that enzymatic digests of the fresh water microalgae, P. duplex and D. fascicularis might be valuable sources of antioxidant which can be applied in food and pharmaceutical industry.

Potential Antioxidant Activites of Enzymatic Digests from Benthic Diatoms Achnanthes longipes, Amphora coffeaeformis, and Navicula sp. (Bacillariophyceae)

  • In this study, we focused on natural water-soluble antioxidants from the Jeju benthic diatoms, Achnanthes longipes, Navicula sp. and Amphora coffeaeformis. They were prepared by enzymatic digestion using five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) and their potential antioxidant activity was assessed. Among the enzymatic digests, Neutrase digest from A. coffeaeformis exhibited the highest effect in DPPH radical scavenging. Flavourzyme (48.7%), Viscozyme (47.4%) and Celluclast (45.7%) digests from Navicula sp. exhibited higher $O^{{\cdot}-}_2$ radical scavenging activity. Viscozyme digest from A. coffeaeformis (45.9%) possessed the highest effects in hydroxyl radical scavenging. Termamyl (89.3%) and Protamex (88.8%) digests from A. coffeaeformis had strong metal chelating activity. Lipid peroxidation was significantly inhibited in Termamyl and Kojizyme digests from A. longipes, AMG and Termamyl digests from Navicula sp. and Kojizyme digest from A. coffeaeformisi. These data suggest that enzymatic digests of the Jeju benthic diatoms might be valuable sources of antioxidant which can be applied in food and pharmaceutical industry.

Effects of Various Extracts from Turmeric (Curcuma longa L.) on Antioxidant Activity

  • The antioxidant activity of 70% methanol, 70% ethanol and chloroform-methanol (CM, 2:1, v/v) extracts from turmeric (Curcuma longa L.) was investigated in this study. The antioxidant potential of various extracts of turmeric was evaluated by using different antioxidant tests, namely DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, ABTS [2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ferric reducing antioxidant power and ${\beta}$-carotene bleaching assays as well as total phenol content and total flavonoid content. All extracts exhibited abundant total phenol content, total flavonoid content and strong antioxidant activity. Particularly, higher activity was exhibited by CM extract with $46.32{\pm}0.35mgCAE/g$ and $15.58{\pm}0.39 mgQE/g$ of total phenol content and total flavonoid content. Generally, antioxidant activity of various extracts from turmeric increased in the following order: 70% methanol extract, 70% ethanol extract and CM extract, respectively. Results suggested that turmeric could be widely used as a source of antioxidant and a kind of functional material in food.

Chlorogenic Acid, an Antioxidant Principle from the Aerial Parts of Artemisia iwayomogi that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.

  • 지방산화는 기본적으로 불포화지방과 산소의 결합이나 이 현상을 제대로 알고 실제 식품에 적용하기 위해서는 지방산화에 미치는 모든 인자들의 화학적 관점(one-electron reduction potential, bond dissociation enthalpy) 뿐만 아니라 물리적 현상(interface, antioxidant polar paradox)을 포괄적으로 이해해야 한다. Invitro실험법에 의해 도출된 산화방지제는 가능성을 보여주는 것이나 실제 식품과 bulk oil, 유화형태, 오르가노젤과 같은고체 상태의 다양한 매트릭스에서는 다른 활성을 나타낸다. 또한 산화방지제의 농도와 다른 물질의 존재에 따라 예상과 다른 활성을 나타내기도 한다. 산화방지제를 활용하여 최종제품의 유통기한을 증가시키기 위해서는 실제 활용될 제품의 매질로 사용해야 한다.

Antioxidant Activity of Vitex rotundifolia Seeds and Phytochemical Analysis Using HPLC-PDA

  • This study assessed in vitro antioxidant activity (ABTS+ and DPPH) of Vitex rotundifolia seeds collected from two different regions in Korea (Jungjang City and Sindu City). Three extraction methods using ethanol, methanol, and water were prepared separately and subjected to quantification by reverse-phase high-performance liquid chromatography-photodiode array (HPLC-PDA) analysis as well as antioxidant testing. Among them, the water-based extract exhibited superior activity in the ABTS+ compared with the ethanol- and methanol-based extracts, while the DPPH assay analysis, revealed that the methanol-based extract had very low antioxidant activity. The concentrations of vanillic acid (1), luteolin (2), vitexicarpin (3), and artemetin (4) were quantified using HPLC-PDA analysis. Vanillic acid (1) was identified as the main antioxidant in V. rotundifolia seeds. Combining the antioxidant activity and quantitative analysis results, the water-based extract was considered to have the highest antioxidant activity. Furthermore, vanillic acid (1) was detected in the leaves and stems of V. rotundifolia plants from different regions, indicating that this species has the potential for use in future antioxidant-applications.

Evaluation of the Antioxidant Potential of Natural Products Mediated by Inhibition of Xanthine Oxidase Activity

  • Since reactive oxygen species (ROS) play an important role in carcinogenesis and other several human diseases, antioxidants present in consumable fruits, vegetables, and beverages have received considerable attention as cancer chemopreventive agents. Thus, in order to identify antioxidants in plant extracts, potential activity was assessed by determining with inhibition of a xanthine/xanthine oxidase assay system. Approximately 170 plant extracts of Korean herbal medicines were primarily evaluated for the potential of antioxidant activity. As a result, 13 plant extracts were found to be active $(IC_{50}$<$100\;{\mu}g/ml)$. Especially, Juncus effusus, Selaginella tamariscina, Pueraria thunbergiana and Sedum albroseum showed strong inhibitory activity in this process. Further studies for the identification of active principles from these active lead plant extracts might be warranted.
