The antioxidant activity of hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) extracts was investigated by several in vitro antioxidants properties, including DPPH free radical scavenging activity, total phenolic and flavonoid contents, hydroxyl radical scavenging activity, reducing power activity, iron-chelating capacity and nitrite scavenging activity. Among the extracts in this study, the 70% EtOH extract showed higher antioxidant activity than the others. The $IC_{50}$ value of DPPH free radical scavenging activity was $99.26\;{\mu}g/mL$. Furthermore, the 70% EtOH extract also showed significantly high total phenolic and flavonoids contents and reducing power activity. However, the MeOH extract exhibited stronger effects on hydroxyl radical scavenging activity, iron-chelating capacity and nitrite scavenging activity. All the results implicated that, the hawthorn fruit may has the available potential to be utilize as a potential source of natural antioxidant.
Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.
Objective: The increasing consumer awareness of food, which can provide health benefits and potentially aid disease prevention, has become the driving force of the functional food market. Accordingly, the aim of this study was to investigate the effects of chicken genotype on the macronutrient content, bioactive peptide content, and antioxidant capacity within different breast meat. Methods: In this experiment, three genotypes of chicken, Thai indigenous, black-boned, and broiler (control), were reared with commercial feed under the same conditions. Thirty chickens were slaughtered at typical market age and the breasts were separated from the carcass to determine macronutrient content using the AOAC method. The antioxidant capacities of the chicken breasts were evaluated by in vitro antioxidant assays and the protein pattern was investigated using gel electrophoresis. Carnosine and anserine, which have antioxidant properties in animal tissue, were determined using high performance liquid chromatography. Results: The results showed that breast meat from Thai indigenous chickens had a greater macronutrient content and higher antioxidant capacity compared with the other genotypes (p<0.05). The protein pattern was similar between genotypes, however Thai indigenous chickens had the greatest myosin and actin content (p<0.05). In addition, carnosine and anserine values were greatest in the black-boned and Thai indigenous chickens compared with the broiler genotype (p<0.05). Conclusion: Thai indigenous chicken breast meat may be classified as a functional food as it has good nutritional value and is rich in antioxidant peptides.
A 60-d feeding trial was conducted to evaluate the effects of diets supplemented with two concentrations (0% and 0.3%) of black raspberry (Rubus coreanus Miquel) fruit by-product (RCFB) on the physicochemical characteristics, oxidative stability, antioxidant capacity, antioxidant enzyme activity, and fatty acid profile of M. longissimus dorsi (LL) porcine muscle from Berkshire finishing pigs meat. Results revealed that regardless of the sex, diets supplemented with 0.3% RCFB reduced (p<0.05) the thiobarbituric acid reactive substances (TBARS) expressed as malonaldehyde (MDA) content effectively. A higher antioxidant capacity [2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity] was found (p<0.05) in response to feeding supplemented with 0.3% RCBF for male or female pigs. Moreover, 0.3% RCFB dietary feed increased (p<0.05) the glutathione peroxidase enzyme activities (GPX1) in blood plasma for male or female pigs. However, no influences were observed (p>0.05) on meat color, WHC, shear force, and fatty acid contents while fed diet supplemented with 0% or 0.3% RCFB for male or female pigs. Overall, this study suggests that a diet supplemented with 0.3% RCFB may beneficially affect owing to better oxidative stability, higher antioxidant capacity, and antioxidant enzyme activity (blood plasma) in pigs which could be a promising natural antioxidant without affecting meat quality traits.
The objective of this study was to investigate antioxidant activities of freeze-dried, main root, and fine root of ginseng (Panax ginseng CA. Meyer), which were extracted with various solvents including ethanol, methanol, and water. Ethanol extracts in both parts showed the most powerful scavenging activities against DPPH radicals. Especially, ethanol extract of fine root had higher reducing power and antioxidant capacity than that of main root. The highest antioxidant activity in linoleic acid emulsion system was also observed in fine root extracted with ethanol, followed by methanol and water. Both ferrous ion chelating activity and ferric reducing antioxidant power (FRAP) of extracts were increased with the increase of extracts concentration. These results suggest that ethanol extract of fine root of ginseng has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus it can be applied for the effective extraction of functional material from ginseng for the usage of pharmaceutical and/or food industries.
Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.
The objective of this study was to evaluate the antioxidant capacity of Lactobacillus plantarum ZLP001 and its effects on growth performance and antioxidant status in weaning piglets. The survival in hydrogen peroxide and free radical-scavenging activity of Lactobacillus plantarum ZLP001 were analysed in vitro. The Lactobacillus plantarum ZLP001 showed high viability in 1.0 mmol/L hydrogen peroxide and high scavenging ability against hydroxyl, superoxide anion, and DPPH (1,1-diphenyl-2-picrylhydrazyl) radicals which was dose dependent. Ninety-six weaning piglets were selected ($7.45{\pm}0.79kg$) and divided into three groups comprising of negative control without any supplementation, treatment group with supplemented $6.8{\times}10^7$ Lactobacillus plantarum ZLP001 CFU/g of diet, and positive control with antibiotic treatment (chlorotetracycline, 80 mg/kg diet). The results showed that Lactobacillus plantarum ZLP001 supplementation enhanced feed conversion rates in piglets compared with control (p<0.05). Supplementation of Lactobacillus plantarum ZLP001 increased the concentration of superoxide dismutase (p<0.05), glutathione peroxidase (p<0.01) and catalase in serum (p<0.10), while decreased the concentration of malondialdehyde (p<0.05). The present study implies that the strain Lactobacillus plantarum ZLP001 had high antioxidant ability and its supplementation improved the growth performance and antioxidant status of weaning piglets, so it can be considered useful to alleviate oxidative stress and increase productive performance of pigs.
The purpose of this study was to determine the antioxidant effect of fucoxanthin. After rats were fed a normal fat diet (NF), high fat diet (HF), and high fat with 0.2% fucoxanthin diet (HF + Fxn) for 4 weeks, the markers of oxidative stress and antioxidant capacity like lipid peroxidation, plasma total antioxidant capacity (TAC), and activities of antioxidant enzymes (catalase, superoxide dismutase (SOD), and gluthathione peroxidase (GSH-Px)) were determined. mRNA expression of transcription factor, nuclear erythroid factor like 2 (Nrf2), and its target genes such as NAD(P)H quinone oxidoreductase1 (NQO1) and heme oxygenase-1 (HO-1) were also determined. Mean weight gain in the HF + Fxn group was lower, without statistical significance, and the total food intake in the HF + Fxn group was lower than that in the HF group (P < 0.05). The activity of GSH-Px (P < 0.05) in plasma was significantly higher in the HF + Fxn group than those in the HF group (P < 0.05). In the liver, the activities of catalase (P < 0.05) and GSH-Px (P < 0.05) in the HF + Fxn group were significantly higher than those in the HF group. Plasma TAC level was significantly higher in the HF + Fxn group than that in the HF group (P < 0.05). Lipid peroxidation in plasma tended to be lower without statistical significance. Fucoxanthin supplements were shown to have higher mRNA expression of Nrf2 and NQO1 than those in the high fat diet only group (P < 0.05). In conclusion, supplementation of fucoxanthin improved the antioxidant capacity, depleted by high fat diet, by activating the Nrf2 pathway and its downstream target gene NQO1. Therefore, supplementation of fucoxanthin, especially for those who consume high fat in their diet, may benefit from reduced risk of oxidative stress.
An active ingredient with hyaluronidase (HAse) inhibitory effect is one of the anti-aging approaches in cosmeceuticals. Here, red sea cucumbers (RSCs), Stichopus japonicus, from Jeju Island were evaluated to examine their HAse inhibitory and antioxidant activity effect. In this study, RSCs were extracted by six enzymatic hydrolysis (Alcalase; Al, Trypsin; Try, Neutrase; Neu, Pepsin; Pep, Alpha-chymotrypsin; Chy and Protamex; Pro). Alcalase hydrolysate (AlH) showed the highest antioxidant capacities for both of oxygen radical absorbance capacity (ORAC) and trolox equivalent antioxidant capacity (TEAC) methods, compared to those of other hydrolysates, at $66.59{\pm}0.78{\mu}M\;TE/mg$ and $135.78{\pm}3.24{\mu}M\;TE/mg$, respectively. Furthermore, AlH performed the highest capacity of HAse inhibitory with $IC_{50}$ value of 3.21 mg/ml. Thus, RSCs hydrolyzed by Al were chosen to determine the cellular antioxidant activity and hyaluronic acid (HA) production effect on Human immortalized keratinocyte cell line (HaCaT). The results showed that AlH improved the cell viabilities and intracellular reactive oxygen species (ROS) induced by 2,2'-Azobis(2-amidinopropane) dihydrochloride (AAPH) were significantly decreased. In addition, AlH increased HA amount by regulating HYAL2 and HAS2 expressions in the HaCaT cells. Taken together, AlH of RSCs collected from Jeju Island showed HAse inhibitory and antioxidant activities against skin-aging which shows its potentials can be an optional natural bioactive ingredient for novel cosmeceuticals.

Platycodon grandiflorum, Doraji as Korean name, is one of the most widely used traditional oriental medicine for bronchial diseases and also used as a folk remedy for geriatric diseases and inflammatory diseases. In recent studies, it has been reported that some effect of P. grandiflorum is derived from its antioxidant activity, although there is still a lack of evidence to establish its oxy-radical scavenging activity. In this study, total oxy-radical scavenging capacity (TOSC) assay was used to evaluate antioxidant activity of total extracts (T-PG), polysaccharide fraction (Po-PG), and saponin fraction (Sa-PG) isolated from P. grandiflorum against peroxyl radicals and peroxynitrites. And MTT assay was taken to assess cyto-protective effects of T-PG, Po-PG and Sa-PG in H4IIE cells treated with hydrogen peroxide and tert-butylhydroperoxide. In the TOSC assay, Sa-PG showed strong oxy-radical scavenging capacity compared with T-PG and Po-PG. In cell-based assay, T-PG and Po-PG protected cells from oxidative stress, but Sa-PG did not protect cells because of cytotoxicity of Sa-PG. These results suggest that the saponin components of P. grandiflorum have relatively strong antioxidant capacity and cytotoxicity in rat hepatoma cells.
