Purple rice (Oryza sativa L. var. indica) cv. Kum Doisaket is cultivated in northern Thailand. This study evaluated the mutagenic and antimutagenic properties of hydrophilic and lipophilic components of purple rice using the Ames test. The seed and hull of purple rice were extracted with hexane, methanol, ethanol, and water. The methanol extracts had the highest amounts of phenolic acids and flavonoids, while the hexane extracts contained large amount of tocols and ${\gamma}$-oryzanol. None of the extracts were mutagenic in Salmonella typhimurium strains TA98 and TA100. The hexane extract of rice hull and the methanol extract of rice seed were strongly effective against aflatoxin B1- and 2-amino-3, 4 dimethylimidazo (4, 5-f) quinoline-induced mutagenesis, while aqueous extracts showed weakly antimutagenic properties. All extracts with the exception of aqueous extracts enhanced the number of revertant colonies from benzo (a) pyrene induced-mutagenesis. None of the extracts inhibited mutagenesis induced by the direct mutagens 2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide and sodium azide. The hull extracts showed more potent antimutagenicity than the seed extracts. Based on a chemical analysis, ${\gamma}$-oryzanol and ${\gamma}$-tocotrienol in the hull and cyanidin-3-glucoside and peonidin-3-glucoside in the seed are candidate antimutagens in purple rice. The antimutagenic mechanisms of purple rice might be related to either modulation of mutagen metabolizing enzymes or direct attack on electrophiles. These findings supported the use of Thai purple rice as a cancer chemopreventive agent.
The present study was conducted to investigate the antimutagenic potential of the methanolic extract from the leaves of sweet potato (Ipomea batatas, IB) with the SOS chromotest (umu test) and Salmonella typhimurium TA 98 and TA 100. The anticarcinogenic effects were also studied by calculation of the $IC_{50}$ on human cancer cell lines and investigating the function of gap junction in rat liver epithelial cells. The IB extract inhibited dose-dependently the ${\beta}$-galactosidase activity induced spontaneously at concentration of more than 200 mg/ml in S. typhimurium TA 1535/pSK 1002, and decreased significantly (p < 0.01) the ${\beta}$-galactosidase activities induced by mutagen 6-chloro-9-[3-(2-chloroethylamino)proylamino]-2-methoxyacridine dihydrochloride (ICR) at dose of more than 0.4 mg/0.1 ml. The IB extract showed no effect on the spontaneous reversions of S. typhimurium TA 98 and 100 but benzo(${\alpha}$)pyrene (BaP)-stimulated reversions were decreased dose-dependently (p < 0.01) at the concentration of more than 100 mg/ml. The $IC_{50}$ value of stomach cancer cells was lower than that of normal rat liver epithelial cells, but the values of colon and uterine cancer cell lines were similar to those of normal rat liver epithelial cells. The transfer of dye through gap junctions was not affected by treatment of the IB extracts at any concentration during treatment periods. The simultaneously treatment of IB extract and 12-O-tetradecanoylphorbol-13-acetate (TPA) effectively prevented the inhibition of dye transfer induced by TPA 1 hour after treatment at all exposed concentrations. The number of gap junctions was significantly (p < 0.01) increased by the treatment with IB extract at concentrations of more than 40 ${\mu}g$/ml. The inhibition of the expression of gap junction proteins by TPA (0.01 ${\mu}g$/ml) was recovered dose dependently by the simultaneous treatment of IB extracts. Our data suggest that Ipomea batatas has antimutagenic and anticarcionogenic activity in vitro.

The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The physiological effects of natural color extracts from colorant plants(gardenia, beet and indigo) were studied. The methanol extract of indigo showed an inhibitory effect on the growth of E. coli and Staph. aureus, and also showed a strong antimicrobial effect on Trich. mentagrophytes compared to others. The methanol extract of indigo showed antimutagenic activities against aflatoxin B1(AFB1) in the Ames test using Salmonella typhimurium TA 100. The proliferation of Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells was inhibited by the methanol extract of indigo. So we decided to use natural indigo for dyeing the fabrics because of those effects. Dried indigo leaves were fermented at variouss temperature and the fermented indigo was reduced by using alkaline(NaOH, Ca(OH)2) and glucose to dye the fabrics. The values of K/S fermented indigo showed the highest value when it was fermented at 3$0^{\circ}C$. The indigo fermented at 3$0^{\circ}C$ had the greatest number of total bacterial counts and we identified one of the main microorganisms as Aspergillus niger. This microorganism was responsible for the indigo fermentation and accelerated indigo fermentation. So it can be supposed to reduce the fermentation period of indigo by inoculating Aspergillus niger into the indigo leaves at 3$0^{\circ}C$.
The antimutagenic activity of a methanol extract of Ecklonia stolonifera (Laminariaceae) against aflatoxin $B_1\;(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using S. Salmonella typhimurium TA100. The methanol extract also exhibited significant inhibitory effects on the micronuclei formation in mouse peripheral blood reticulocytes and the DNA damage in mouse spleen lymphocytes induced by N-methyl-N-nitrosourea (MMU) and benzo(a)pyrene (B(a)P). The MeOH extract was then sequentially partitioned with $CH_2Cl_2,\;CH_2Cl_2$ insoluble intermediate, EtOAc, n-BuOH, and $H_2O$. All fractions possessed antimutagenic activity but the $H_2O$ fraction was inactive. Among active fractions, the EtOAc and $CH_2Cl_2$ insoluble intermediate fractions showed the highest activity. Column chromatography using $SiO_2$ and Sephadex LH-20 yielded phloroglucinol from the EtOAc fraction. Phloroglucinol also demonstrated significant antimutagenic activity, and inhibitory effects on the micronuclei formation in mouse peripheral blood reticulocytes and DNA damage in mouse spleen lymphocytes induced by MMU and B(a)P.
In the present studies, decursinol angelate, decursin isolated from Angelica gignatis radix and oil fraction of Cnidii rhizoma was analyzed by normal phase HPLC and GC/MS respectively. The standardized water extracts of Angelica gignatis radix, Cnidii rhizoma and its complex named Gung-gui-tang was tested the anti mutagenic effects by in vitro genotoxicity using Salmonella reversion assay (Ames test) and micronucleus test in chinese hamster ovary(CHO) cells. Angelica gignatis radix, Cnidii rhizoma and Gung-gui-tang was not exhibited the antimutagenic effects in the Salmonella reversion assays with or without metabolic activation. However, the micronucleus test assays, Angelica gignatis radix and Gung-gui-tang was showed the antimutagenic effects significantly. The maximum inhibition observed with Gung-gui-tang was reduced by 59% in the micronucleus test without metabolic activation. In this paper, results are presented on the availability of potential antimutagenic activity of the water extracts of Gung-gui-tang.
The antimicrobial activities of horseradish ( Wasahia japonica) root extract against 4 kinds of food poisoning bacteria and 3 kinds of molfs were examined. The antimutagenic activity of horseradish ( Wasahia japonica) root extracts was also examined by Ames test with Salmonella tyhimurium TA 98 The antimicrobial activities of distilled water extracts from horseradish root were stronger than those of ethanol extracts, and stronger against molds than bacteria. Of the kinds of bacteria, Vibrio parahaemolyticus was best inhibited by the distilled water extracts from horseradish root. The antimicrobial activity of distilled water extracts from horseradish root were stronger against 3-Amino-1, 4-dimethyl-5H-pyrido{4,3-b}indole than 2-Amino-3,-8-dimethylimidazo-[4,5-f]quinoxaline.
We examined the in vitro mutagenic and antimutagenic effects of hexane-extracted hemp (Cannabis sativa L. subsp. sativa var. sativa) seed oil (HO) with and without S9-mediated metabolic activation, using the TA98 and TA100 Salmonella Typhimurium strains. The MTT assay revealed no cytotoxicity in HepG2 cells for HO quantities $\leq400g$/mL. In the mutagenicity test, revertant colonies did not exceed spontaneous colonies in number. Colony numbers did not increase in either strain after HO treatment, with or without metabolic activation. HO showed no mutagenic effects and did not induce a mutation in either strain. In the antimutagenicity test, HO reduced the number of mutated colonies induced by 4NQO in both strains. The inhibition rates of HO (TA98, 21-91%; TA100, 21-85%) indicated a potent reduction in mutagenicity induced by 4NQO. HO showed no significant mutagenicity and may have antimutagenic effects, as assessed by Ames testing.
Antidiarrheal, anticostive and antimutagenic effects of Bifidobacterium breve K-110, K-111 and B. infantis K-525 isolated from Korean were investigated in experimental animals. These Bifidobacteria were not significantly affected on the transport of barium sulfate in the small intestine. However, these Bifidobacteria significantly stimulated the transport of barium sulfate in the large intestine. Particularly, when Bifidobacterium breve K-110 (500 mg/kg) was orally administered, the transport of barium sulfate in the large intestine was increased 45%, compared to the control group. On the castor oil-induced diarrheal mice, Bifidobacterium breve K-111 had the antidiarrheal activity but the other Bifidobacteria did not had it. When the antimutagenicity of these Bifidobacteria and their peptidoglycans were examined using Salmonella typhimurium TA98/TA100 in an in vitro assay system, these Bifidobacteria and peptidoglycans showed inhibitory effect of $20{\sim}80%$. These results indicate that Bifidobacterium spp. had antidiarrheal, anticostive and antimutagenic activities as well as the inhibitory activity of harmful enzymes of intestinal bacteria in the intestine.
In this study, the antioxidant and antimutagenic activities of Cuscutae semen ethanol extract were evaluated. Antioxidant activity was measured by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and the Ames test was employed to determine the inhibition effect on mutagenicity in Salmonella typhimurium TA100. The extract showed significant free radical-scavenging activity towards the DPPH radical, and at a concentration of 400 ppm, its free radical-scavenging activity was similar to that of BHT. The $IC_{50}$ value of the extract was 89 ppm, indicating good antioxidant capacity. Moreover, at 5 mg/mL, the extract presented inhibitions of approximately 98.0% and 49.2% on mutagenicity induced by 4-nitroquinoline 1-oxide and sodium azide, respectively. The total polyphenols and flavonoid contents of the extract were 20.1 mg/g and 1.9 mg/g, respectively. Therefore, this study indicates that the ethanolic extract of Cuscutae semen has excellent antioxidative and antimutagenic potential.
It was investigated that the reduction of the intense brown color and increase of solubility, and depolymerization of polymerized melanoidins by ozonolysis affect their antioxidative and antimutagenic activities. Melanoidins was depolymerized and decolorized by ozonolysis. Ozone-treated melanoidins revealed a lower antioxidative activity and a higher antimutagenic activity than those of the untreated control. Fractions of ozone-treated melanoidins showed three peaks resemble to that of the melanoidins on Sephadex column chromatogram. Melanoidins of above MW 1,200 showed intense brown color. However, the strongest electron-donating ability was detected in the melanoidins of between MW 750 to 900. Ozone-treated and ozone-untreated melanoidins of between MW 900 to 1,000 revealed the higher antioxidative and antimutagenic activities.
