항균성물질이 함유된 세라믹 LDPE필름의 항균효과 및 식품의 저장성 (Antimicrobial Activity and Food Storage of LDPE Ceramic Film Containing Antimicrobial Agents)

  • 다양한 식품의 미생물에의한 변패를 억제하고 저장성을 높 일수 있는 포장필름을 개발하기위하여, 미생물이 생산하는 천연 항균성 물질을 홉착시킨 합성 세라믹올 첨가하여 세라 믹 LDPE 필륨을 제조하였다. 천연 항균제로는 methanol자화 방선균 MO-16 및 MO-17균이 생산한 항세균제와 새로이 분 리한 방선균 No. 31이 생산한 항진균제를 사용하였다. 방선 균 No. 31이 생산한 항진균성 불질은 배양 4일째 항균효과가 우수하였으며, $121^{\circ}C$에서 15분간 열처리시에도 항균력이 유 지되는 내열성이 확인되었다. 제조한 세라믹 LDPE 필름의 미생불 생육 억제효과를 검토한 결과 조분쇄한 돈육의 경우 포장하여 실온 및 $4^{\circ}C$에서 보존시 시판필름에 비해 미생물 생육 억제효과가 우수하였다. 고제배지에 제조필름을 첨가하 여 E. coli에 대한 항균효과를 검토한 결과 첨가량에 따라 항 균효과가 우수한 것이 입증되었다 제조펼름으로 포장한 6종 류의 식품에 대한 저장성을 검토한 결과 시판필름에 비해 저 장성이 우수한 것이 확인되었으며 특히 양송이에 대한 저장 효과가 우수하였다.

Prevalence and antimicrobial resistance of Salmonella spp. and Escherichia coli isolated from ducks in Korea

  • This study was conducted to investigate the prevalence and antimicrobial resistance of Salmonella spp. and Escherichia (E.) coli isolated from ducks in Korea. A total of 400 cecal content samples were collected from 40 duck farms in Korea. Isolated Salmonella spp. and E. coli strains were 83 and 364 of the 400 cecal samples, respectively. The most prevalent serotype among the 83 Salmonella isolates was Salmonella Typhimurium (51 isolates: 61.45%). Resistance to the tested antimicrobial agents by Salmonella isolates was low except for erythromycin, while the resistance of the E. coli isolates to the other tested antimicrobial agents was high and 90.9% (331/364) of E. coli isolates showed multi-antimicrobial resistance. Antimicrobial resistance in duck zoonotic pathogens should be of concern to the Korean duck industry, as these pathogens exhibit a high rate of antimicrobial resistance and pose a potential hazard to public health.

도계장과 돈육가공장에서 분리된 살모넬라 속의 특성 연구 (Characteristics of Salmonella Species Isolates from Poultry Slaughterhouses and Pork Meat Processing Plants)

  • This study was carried out to isolate and identify Salmonella species from poultry slaughterhouses and pork meat processing plants during the period from January 1997 to August 1998, and analyze resistance of antimicrobial agents and plasmid profiles of isolated Salmonella strains. A total of 15 Salmonella strains was isolated from poultry carcasses, swine carcasses, pork meats and cutting boards. Identified Salmonella strains were S. typhimurium, S. heidelberg, S. hilingdon, S. mbandaka, and S. virginia. Ten (66.7%) of 15 Salmonella strains showed resistance to antimicrobial agents and five strains (33.3%) of them were resistant to two or more antimirobial agents. Plasmids were isolated from three Salmonella isolates which had two or more plasmids.

축우(畜牛)에서 분리(分離)한 Corynebacterium renale의 각종(各種) 항균제(抗菌劑)에 대(對)한 감수성(感受性) (Sensitivity of Corynebacterium renale to Some Antimicrobial Agents)

  • In order to know the current status of sensitivity of Corynebacterium renale to some antimicrobial agents of current use, the sensitivity of 37 strains isolated from urine and vaginal cavity of healthy Korean native cattle and dairy cattle was studied by agar plate dilution method, and their sensitivity according to serological types was compared. The antimicrobial agents tested were streptomycin (SM), erythromycin (EM), oxytetracycline (OT), penicillin G (PC), gentamycin (GM), kanamycin (KM), ampicillin (AP), leucomycin (LM), chloramphenicol (CM), colistin (CL), sulfadimethoxine (SDM) and sulfadiazine (SDZ). Most strains were highly sensitive to PC, GM, AP, LM and KM, and moderate to CM and OT, but resistant to SM, EM, CL, SDM and SDZ. Serological type II was more sensitive to EM, SDM and SDZ than type I and II, but more resistant to SM.

Antimicrobial Agents That Inhibit the Outer Membrane Assembly Machines of Gram-Negative Bacteria

  • Gram-negative pathogens, such as Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumannii, pose a serious threat to public health worldwide, due to high rates of antibiotic resistance and the lack of development of novel antimicrobial agents targeting Gram-negative bacteria. The outer membrane (OM) of Gram-negative bacteria is a unique architecture that acts as a potent permeability barrier against toxic molecules, such as antibiotics. The OM is composed of phospholipids, lipopolysaccharide (LPS), outer membrane ${\beta}-barrel$ proteins (OMP), and lipoproteins. These components are synthesized in the cytoplasm or in the inner membrane, and are then selectively transported to the OM by the specific transport machines, including the Lol, BAM, and Lpt pathways. In this review, we summarize recent studies on the assembly systems of OM components and analyze studies for the development of inhibitors that target these systems. These analyses show that OM assembly machines have the potential to be a novel attractive drug target of Gram-negative bacteria.

Antimicrobial Activity of the Cell Organelles, Lysosomes, Isolated from Egg White

  • Lysosomes, as a cell organelle type, are safe biological control agents that may be possible replacements for chemical antimicrobial agents because they are simply isolated from egg white. In this study, it was found that the lysosomes isolated from egg white exhibited pH-dependent antimicrobial activity, with the optimal activity found at pH 6.0. The efficiency of lysosomes in inhibiting bacterial growth and activity was evaluated over a 12-h treatment period. Seven different microorganisms were used as bacterial strains, and the lysosomes showed a significant antimicrobial effect against all strains. In addition, the antimicrobial activity was maintained for 100 days, and there did not appear to be any resistance of E. coli to the lysosomal activity up to the eighth culture. However, the lysosomes did not affect the viability of mammalian cells, suggesting the biocompatibility of lysosomes. These highly effective lysosomes have a bright future in the application of novel antimicrobial sources as a cell organelle type.

Use of Bioluminescent Indicator Acinetobacter Bacterium for Screening and Characterization of Active Antimicrobial Agents

  • Because of the need for new antimicrobial substances with novel mechanisms of action, we report here the use of an Acinetobacter reporter system for high-throughput screening of active antimicrobial agents. The bioreporter Acinetobacter strain DF4/PUTK2 carrying luciferase genes luxCDABE was chosen because of its ecological importance and it is widespread in nature. This bioreporter is genetically engineered to emit light constitutively that can be measured in real time by luminometry. Hence, this reporter system was employed to determine the bacteriostatic actions of spent-culture supernatants derived from twelve bacterial isolates. Out of the results, the strongest bioluminescence inhibitory effect of the supernatants was recorded with Bacillus cereus strain BAC (S5). Subsequently, ethyl acetate extracts of extracellular products of strain BAC (S5) were separated by a thin-layer chromatography (TLC). Based on the bioluminescence inhibitory assay, three fractions were found to have antimicrobial activity. One fraction (C) having the strongest antimicrobial activity was further purified using TLC and characterized by IR, $^1H$ NMR, mass spectrometry, SDS-PAGE, and amino acid composition analysis. The results predicted the presence of 2-pyrrolidone-S-carboxylic acid (PCA) and the octadeconic-acid-like fatty acid. Fraction C also demonstrated a broad inhibitory activity on several Gram-negative and Gram-positive bacteria. In conclusion, the Acinetobacter reporter system shows great potential to be a reliable, sensitive, and real-time indicator of the bacteriostatic actions of the antimicrobial agents.

건강한 가축에서 분리한 Escherichia coli의 항생제 내성 조사(2010~2012년) (Antimicrobial resistance of Escherichia coli isolated from healthy animals during 2010-2012)

  • The purpose of present study was to investigate the antimicrobial resistance in Escherichia coli isolated from healthy animals in all provinces of the Republic of Korea. A total of 2,085 E. coli strains isolated from 11,336 fecal samples of healthy animals during 2010-2012 were examined for antimicrobial resistance. Comparison of average resistance rate through the years revealed that tetracycline (47.0% and 76.1%) and streptomycin resistance (42.6% and 64.6%) was most frequently observed in cattle and pigs, respectively. Whereas, in chicken isolates, resistance against nalidixic acid (90.9%) was highest among the antimicrobials tested. Percentage of E. coli that showed multidrug resistance (resistance against ${\geq}$ three subclasses of antimicrobial agents) was 17.6% (151/860) in cattle, 69.4% (506/729) in pigs, and 86.1% (427/496) in chickens. Overall, the rates of resistance are apparently different between animal species and, in particular, resistance was less prevalent in cattle than in pigs and chickens. In conclusion, this study showed higher prevalence of resistance in commensal E. coli strains to antimicrobial agents in Korean livestock and highlighted the urgent need for measures to regulate the abuse of antimicrobial agents.

Synthesis and Antimicrobial Screening of Pyrimidine Annulated Dihydropyrano[2, 3-c]pyrazole Derivatives

  • A series of pyrimidine annulated dihydropyrano[2, 3-c]pyrazole derivatives were synthesized and screened for their antimicrobial activity. The precursor, dihydropyrano[2, 3-c]pyrazole was synthesised under catalyst free condition using PEG-400 as reaction medium which facilitated improved yield compared to base catalyzed reaction. Wide scope of substrates, simple workup procedure and high yield even in the absence of catalyst are the major highlights of the protocol. Dihydropyrano[2, 3-c]pyrazoles on condensation with formic acid formed pyrimidine annulated dihydropyrano[2, 3-c]pyrazole derivatives. All the products are characterized using FTIR, $^1H-NMR$ and $^{13}C-NMR$ spectroscopic techniques. The molecules have shown good to moderate activity as antimicrobial agents when compared to the standard drug ciprofloxacin.

가축 및 하천에서 분리한 대장균의 항균제내성 (Isolation and Resistance Patterns of E. coli from Domestic Animals and Dranage in 1987)

  • One hundred and sixteen strains of E. coli isolated from drainage in Seoul city in 1987 and 104 strains of E. coli isolated from stools of domestic animals in suburb of Seoul in 1987 were examined for susceptibilities to 10 antimicrobial agents by the agar dilution method. The susceptibilities of the two groups to each antimicrobial agent were compared and correlations in the antimicrobial susceptibility of the 220 strains of E. coli among the 10 antimicrobial agents were also analyzed. The frequency of resistance strains was highest to tetracycline with 77%, and followed by chloramphenicol, streptomycin, ampicillin, kanamycin and cephalosporin in the descreasing order, tanging from 62% to 10%. Strains resistant to tobramycin, nalidixic acid, gentamicin and amikacin occupied 3 strains, 2 strains, 2 strains and 1 strain respectively.
