Plasmodium vivax is usually considered morbidity in endemic areas of Asia, Central and South America, and some part of Africa. In Thailand, previous studies indicated the genetic diversity of P. vivax in malaria-endemic regions such as the western part of Thailand bordering with Myanmar. The objective of the study is to investigate the genetic diversity of P. vivax circulating in Southern Thailand by using 3 antigenic markers and 8 microsatellite markers. Dried blood spots were collected from Chumphon, Phang Nga, Ranong and, Surat Thani provinces of Thailand. By PCR, 3 distinct sizes of $PvMSP3{\alpha}$, 2 sizes of $PvMSP3{\beta}$ and 2 sizes of PvMSP1 F2 were detected based on the length of PCR products, respectively. PCR/RFLP analyses of these antigen genes revealed high levels of genetic diversity. The genotyping of 8 microsatellite loci showed high genetic diversity as indicated by high alleles per locus and high expected heterozygosity ($H_E$). The genotyping markers also showed multiple-clones of infection. Mixed genotypes were detected in 4.8% of $PvMSP3{\alpha}$, 29.1% in $PvMSP3{\beta}$ and 55.3% of microsatellite markers. These results showed that there was high genetic diversity of P. vivax isolated from Southern Thailand, indicating that the genetic diversity of P. vivax in this region was comparable to those observed other areas of Thailand.
The binding affinity between the T-cell receptors (TCRs) and antigenic peptides mainly determines immunological recognition. It is not a trivial task that T cells identify the digital sequences of peptide amino acids by simply relying on the integrated binding affinity between TCRs and antigenic peptides. To address this problem, we examine whether the affinity-based discrimination of peptide sequences is learnable and generalizable by artificial neural networks (ANNs) that process the digital experimental amino acid sequence information of receptors and peptides. A pair of TCR and peptide sequences correspond to the input for ANNs, while the success or failure of the immunological recognition correspond to the output. The output is obtained by both theoretical model and experimental data. In either case, we confirmed that ANNs could learn the immunological recognition. We also found that a homogenized encoding of amino acid sequence was more effective for the supervised learning task.
탄저(anthrax)는 그람양성이고 포자형성 세균인 탄저균(Bacillus anthracis)으로부터 발명되어진다. 탄저독소는 세가지 요소로 구성되어 있으며, 방어항원(PA)은 숙주세포 표면에서 탄저 독소단백질 및 이종단백질을 세포질 내로 이동시키는 역할을 한다. 본 연구에서는 PA의 분자적 다양성과 국내에서 탄저균의 진화를 이해하고 확인하기 위해 국내외에서 발견된 탄저균 4 균주와 기존에 보고된 탄저균 26 균주로부터 2,294 bp의 PA유전자(pagA)의 DNA 염기서열을 분식하였다. 탄저균 30 균주으로부터 PA유전자의 염기서열을 비교 분식한 결과, 8개 부위에서 돌연변이를 확인 하였다. 돌연변이가 일어난 부위에 따라서 탄저균을 10종류의 PA 유전자형과 4 종류의 PA 표현형으로 구분하였다. 한국 경주에서 분류된 B. anthracis BAK는 600번째 아미노산 alanine이 valine으로 바뀌어서 B. anthracis ATCC 14185 보다 LF와 PA의 결합 위치를 근접하게 하였다. 탄저균의 Pag의 염기서열을 통한 계통분석학적인 분석 결과는 염색체상에서의 분류와 일치하여 탄저균사이에서 pXO1 플라스미드의 수평적인 이동은 없는 것으로 사료된다.
The vir genes are antigenic genes and are considered to be possible vaccine targets. Since India is highly endemic to Plasmodium vivax, we sequenced 5 different vir genes and investigated DNA sequence variations in 93 single-clonal P. vivax isolates. High variability was observed in all the 5 vir genes; the vir 1/9 gene was highly diverged across Indian populations. The patterns of genetic diversity do not follow geographical locations, as geographically distant populations were found to be genetically similar. The results in general present complex genetic diversity patterns in India, requiring further in-depth population genetic and functional studies.
Background: Feline calicivirus (FCV) is a common pathogen of felids, and FCV vaccination is regularly practiced. The genetic variability and antigenic diversity of FCV hinder the effective control and prevention of infection by vaccination. Improved knowledge of the epidemiological characteristics of FCV should assist in the development of more effective vaccines. Objectives: This study aims to determine the prevalence of FCV in a population of cats with FCV-suspected clinical signs in Hangzhou and to demonstrate the antigenic and genetic relationships between vaccine status and representative isolated FCV strains. Methods: Cats (n = 516) from Hangzhou were investigated between 2018 and 2020. The association between risk factors and FCV infection was assessed. Phylogenetic analyses based on a capsid coding sequence were performed to identify the genetic relationships between strains. In vitro virus neutralization tests were used to assess antibody levels against isolated FCV strains in client-owned cats. Results: The FCV-positive rate of the examined cats was 43.0%. Risk factors significantly associated with FCV infection were vaccination status and oral symptoms. Phylogenetic analysis revealed a radial phylogeny with no evidence of temporal or countrywide clusters. There was a significant difference in the distribution of serum antibody titers between vaccinated and unvaccinated cats. Conclusions: This study revealed a high prevalence and genetic diversity of FCV in Hangzhou. The results indicate that the efficacy of FCV vaccination is unsatisfactory. More comprehensive and refined vaccination protocols are an urgent and unmet need.
목 적 : Respiratory syncytial virus(RSV)는 소아 하기도 감염증의 중요한 원인의 하나이다. 연구자들은 8년간 분리된 RSV A 아군의 항원상의 다양성과 G 단백 유전자의 변이를 관찰하고자 하였다. 방 법 : 서울대학교 어린이병원에서 1990년부터 1998년까지 8년 동안 분리된 RSV A 아군 179주들을 대상으로 하였다. G 단백 유전자를 역전사 중합효소 연쇄 반응으로 증폭한 후 이 증폭 산물을 AluI, HineII, MboI, PstI, RsaI 및 TagI 등의 제한효소로 처리하여 이들의 절단 양상에 따른 유전자형들의 다양성을 파악하였다. 또, RSV에 대한 단클론 항체와의 반응 여부에 따른 항원형과 유전자형의 연관성을 검정하였다. 또한, 각 유전자형의 선택된 일부 주들의 염기 서열을 결정하여 이미 보고된 표준주 및 외국의 임상 분리주들과 비교하여 계통수 분석을 시행하였다. 결 과 : 8회의 RSV 유행기 중 A 아군은 7회에서 유행하였다. A 아군 179주의 G 단백 유전자의 역전사 중합효소 연쇄 반응 증폭 산물의 제한효소에 의한 절단 양상에 따른 유전자형은 23가지였으며 이 중 12가지는 2개 이상의 바이러스주에서 나타났다. 단클론 항체와의 반응 양상에 따른 항원형은 6가지로 분류되었으며 그 중 한가지 형이 91%를 차지하였다. 항원헝과 유전형간의 상관관계는 적었다. 우리나라에서 분리된 A 아군의 G 단백의 염기 서열은 표준주틀과 91~93%의 동질성을 보였으며, 아미노산 서열은 표준주들과 85~90%의 동질성을 보였다. G 단백 염기 서열을 토대로 한 계통수에서 우리나라에서 분리된 RSV는 각기 표준주와는 다른 부위에 속하였고 A 아군은 4개의 군을 형성하면서 1988년부터 1993년까지 영국과 스페인에서 분리된 주들과 부분적으로 연관 관계를 나타냈다. 결 론 : 결론적으로 RSV A 아군의 유전자형은 항원형보다 더욱 다양한 양상을 보이며, 우리나라에서는 표준주 및 외국의 임상 분리주와는 차이가 있는 다양한 유전자형의 RSV가 유행하고 있다. 향후 RSV의 연구에는 이러한 점이 고려되어야 할 것으로 생각된다.
Porphyromonas endodontalis is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections and this microorganism possesses a potential for pathogenicity. The purpose of this study was to compare the membrane components of Porphyromonas endodontalis and Porphyromonas gingivalis and to study the immune reaction patterns of Porphyromonas endodontalis with patients with periapical lesion. Porphyromonas endodontalis (ATCC 35406), Porphyromonas gingivals serotypea (381), serotype b(W50), serotype c(A7A1-28) were cultured in anaerobic condition. Rabbit antisera were prepared by intravenous injection of formalized whole cells and human sera were obtained from patients and dental students. Indirect immunofluorescence method was used to study on the cross reaction between Porphyromonas endodontalis and Porphyromonas gingivalis serotype a, b, c antigen. Total membrane protein profiles of Porphyromonas endodontalis antigen were studied by sodium dodecyl sulfate polyacrylamide gel electrophoresis and the reactivity of antigenic components of Porphyromonas endodontalis against sera of patients and rabbit anti-Porphyromonas endodontalis antisera were assessed by Immunoblotting method. The following results were obtained : 1. Antigens of Porphyromonas endodontalis has multiple antigenic components, and both patients with periapical lesion and normal healthy individual showed immune response to this. 2. Patients group and healthy individual group showed a diversity of immune reaction pattern but they showed immune response against 43kd protein. 3. Patients with periapical lesion showed more diverse immune response than healthy individual and in some patients, much more bands appeared to lower molecular weight protein. 4. According to indirect immunofluorescence and Immunoblotting study, Porphyromonas endodontalis did not share common antigen with Porphyromonas gingivalis serotype a, b, c.

Background: Carnivore protoparvovirus 1, also known as canine parvovirus type 2 (CPV-2), is the main pathogen in hemorrhagic gastroenteritis in dogs, with a high mortality rate. Three subtypes (a, b, c) have been described based on VP2 residue 426, where 2a, 2b, and 2c have asparagine, aspartic acid, and glutamic acid, respectively. Objectives: This study examined the presence of CPV-2 variants in the fecal samples of dogs diagnosed with canine parvovirus in Bogotá. Methods: Fecal samples were collected from 54 puppies and young dogs (< 1 year) that tested positive for the CPV through rapid antigen test detection between 2014-2018. Molecular screening was developed for VP1 because primers 555 for VP2 do not amplify, it was necessary to design a primer set for VP2 amplification of 982 nt. All samples that were amplified were sequenced by Sanger. Phylogenetics and structural analysis was carried out, focusing on residue 426. Results: As a result 47 out of 54 samples tested positive for VP1 screening, and 34/47 samples tested positive for VP2 980 primers as subtype 2a (n = 30) or 2b (n = 4); subtype 2c was not detected. All VP2 sequences had the amino acid, T, at 440, and most Colombian sequences showed an S514A substitution, which in the structural modeling is located in an antigenic region, together with the 426 residue. Conclusions: The 2c variant was not detected, and these findings suggest that Colombian strains of CPV-2 might be under an antigenic drift.
Theileria (T.) buffeli (formerly T. sergenti/T. orientalis) is the major hemo-protozoan distributed in the Far East Asian countries such as Korea, China and Japan. It is responsible for the clinical symptoms of anorexia, ateliosis, anemia, fever and icterus. It also causes abortion and sudden death under severe cases, resulting in economic losses for many livestock farms. The objective of this study was to analyze the genetic diversity of the major surface protein (Msp) gene in T. buffeli in Holstein in Korea, and we characterized the association of the diversification of the Msp gene and its relationship with the pathogenicity of Theileria. For this, complete blood counts and Theileria PCR sequence analysis were performed from 57 Holstein in Jeju Island. A total of 26 PCR positive Holstein (16 anemic and 10 non-anemic) were then randomly selected based on 18s rRNA sequence typing of the Theileria Msp gene. The DNA sequence of the T. buffeli Msp gene in Holstein showed 99.0%, 99.2%, 99.9%, 99.5%, 98.7%, 98.4% and 98.4% homology with T. sergenti, Theileria spp., T. sergenti, Theileria spp., Theileria spp., Theileria spp. and Theileria spp., respectively. The result showed a genetic variation of 57.7% (type I), 3.8% (type II), 15.4% (type III), 7.7% (type IV), 13.5% (type V) and 1.9% (type VI). Type I is the most frequent type in both anemic and non-anemic Holstein while type II was found in only non-anemic Holstein. This results of our study help confirm the diversity of Msp gene types and demonstrate that the gene type distribution of Msp genes varies among Theileria-infected Holstein in Jeju Island.
본 연구는 국내 타일레리아에서 주요항원단백질(major surface protein) 유전자의 다양성을 분석해 보고자 수행되었다. 나아가 Msp 유전자의 다양성과 타일레리아의 병원성과의 관계도 분석하였다. 제주에 있는 목장으로부터 총 177마리의 한우 혈액을 공시재료로 사용하여 혈액검사와 18S rRNA를 표적으로 하는 PCR을 실행하였다. 그 후, 타일레리아 18S rRNA에 양성인 28마리 (16마리 빈혈군과 12마리의 정상군)를 무작위로 선발하여 Msp유전자의 염기 서열을 반복하여 분석하였다. 총 56개의 염기서열 결과는 다변성 부위(517-571 bp)에 따라 크게 type I에서 type V까지 5가지 형태로 나눌 수 있었는데, 이는 유전자은행(GenBank)에 등록되어 있는 다음의 유전자와 98.9% 이상 일치하였다 (Theileria spp. from China-EU584237; T. sergenti from China-DQ078264; Theileria spp. from Thailand-AB081329; Theileria spp. from Japan-AB218442; T. sergenti from Japan-AB016280). 그 분포는 22, 15, 9, 8, 2개가 각각 type I에서 V까지 분포하였고 빈혈과 관계없이 type I이 가장 많이 나타나는 것으로 밝혀졌다(37.5%의 빈혈군과 41.7%의 정상군). 나머지 type중에서는 type II가 빈혈군에서 가장 많이(37.5%) 나타났으며, 반면 type IV는 정상군에서 많이 (25%) 나타났다. 본 연구는 국내 타일레리아 Msp유전자의 다양성을 밝히는데 좋은 자료로 활용될 수 있을 것이다.
