Detection of Aspergillus, Penicillium, and Fusarium Species by Sandwich Enzyme-Linked Immunosorbent Assay Using Mixed Monoclonal Antibodies

  • The antibody-mix sandwich enzyme-linked immunosorbent assay (Ab-mix sELlS A) system was developed in order to simultaneously detect the extracellular polysaccharide (FPS) of Aspergillus, Penicillium, or Fusarium species using one detection system. The detection limit and detection range of the Ab-mix sELISA towards EPS of Penicilliun citrinum were not changed, and those towards Fusarium moniliforme EPS were changed a little compared to that of individual sandwich ELISA [9, 10]. The fungal culture filtrates of Aspergillus and Penicillium species showed nearly similar reactivity towards Ab-mix sELISA as that of sELISA using the MAb lB8 alone [9]. Also, the fungal culture filtrates of Fusarium species showed nearly the same reactivity towards Ab-mix sELISA as that of sELISA using the MAb lB8 alone [10]. Thus, this ELISA system showed that the three genera of molds, Aspergillus, Penicillium, or Fusarium, which are three major important molds producing mycotoxins in food or agricultural commodities, could be detected at the same time, using one detection system.

Protective effects of mix-crude outer membrane protein Salmonella vaccine against salmonellosis in chickens and pigs (살모넬라 세포외막단백질 혼합백신을 이용한 돼지 및 닭에서의 살모넬라균감염증 방어효과)

  • We investigated the safety, immunogenicity and protectivity of mix-crude outer membrane protein (cOMP) vaccine against salmonellosis in animals. The mix-cOMP vaccine was extracted from Salmonella enterica serovar Typhimurium (ST) and Salmonella enterica serovar Enteritidis (SE) and Salmonella enterica serovar Braenderup (SB) isolated from pigs. The mix-cOMP vaccine gave significantly higher antibody response than ST-bacterin and ST-cOMP vaccine in guinea pigs. The survival rates of mix-cOMP vaccinated groups showed significantly higher (100%) than those (0-20%) of unvaccinated control group, challenged with 3 species of Salmonella (ST, SE and SB) in mice. Vaccinated groups in pigs showed reduction of clinical signs, increase of average weight gains, decrease of bacterial recovery rates, compared with unvaccinated groups. Especially, the survival rates (100%) of vaccinated groups in chickens showed higher than that (0%) of unvaccinated group. Based on these results, we suggest that the mix-cOMP Salmonella vaccine developed in this study will be effective for the protection against Salmonellosis caused by the various serotypes Salmonella species in animals.

Effects of Mix-1 on Anti-CD40 Antibody and Recombinant IL4- Induced Cytokine Production and Immunoglobulin E in Highly Purified Mouse B Cells

  • In the oriental medicine, a mixture of herbs has been commonly used as important components to control allergic and inflammatory diseases. In the present study, we prepared a mixture of Dictamni Radicis Cortex(Baiksunpee), Houttuyniae Herba(Uhsungcho), and Aurantii Immaturus Fructus(Jisil) to examine its anti-allergic effects in activated mouse splenic cells and found that Mix-1 is involved in regulating levels of B cell activating factors (CD23 and CD11a), IL-1β, IL-6, IL-10, TNF-α, and 1gE as well as HRF expression. It was observed that Mix-1 did not have cytotoxic effects on mLFC. Mix-1 showed inhibition of CD23 and CD11 alpaha expression in mouse B cells, and also decreased the production of IL-6, TNF-α, and 1gE. Both RT-PCR and ELISA analyses indicated that IL-6 and TNF alpha production were regulated at the gene expression level. In contrast, IL-10 mRNA and protein levels were increased in activated B cells by Mix-1 treatment. We also found that Mix-1 inhibited B cell proliferation and inhibited histamine releasing factor(HRF) expression, suggesting its inhibitory effect on histamine secretion. These data indicated that Mix-1 has an anti-allergic effect in activated macrophages and further suggest the possible application of Mix-1 as a therapeutic agent for the treatment of allergy-related diseases.

Application of mix LPS-ELISA for monitoring of antibodies to major serogroups of Salmonella in animal (Mix LPS-ELISA법을 이용한 살모넬라균 복합감염항체 동시 모니터링)

  • Salmonellosis caused by a number of serotypes of Salmonella is an infectious, acute or chronic, zoonotic disease and characterized by enteritis and diarrhea, septicemia in animal. In these studies we investigated the prevalent serotypes of Salmonella causing animal salmonellosis in Korea and the 71 strains of Salmonella spp. were isolated from materials such as mesenteric lymph nodes, fecal samples from slaughtered animal. With the identification test results, the most prevalent serotypes were, in order, S stanley 31 strains (43.7%), S typhimurium 19 strains (26.8%) and S montevideo 11 strains (15.5%), respectively. And we could establish the method for detection of antibodies to broad variety of Salmonella serotypes. Lipopolysaccharide(LPS) antigen extracted from Salmonella was more sensitive and specific than outer membrane protein antigen from that for detection of Salmonella antibody by using an indirect ELISA. The optimal concentration of antigen was 100ng/ml of LPS, the dilutions of conjugate and serum were 1 : 1,000~2,000 and 1 : 200~400, respectively. The mix LPS-ELISA which was used by mixing LPS from S typhimurium (group B), S choleraesuis (group C) and S enteritidis (group D) were more rapid and effective than that used LPS from individual strain for detection of Salmonella serogroup O4, O7 and O9 antibody at the same time. We could obtain the high values of optical density ($0.73{\pm}0.32$) by mix LPS-ELISA on the farm which had occurred salmonellosis, but very low values of $0.17{\pm}0.06$ on the negative farm of salmonellosis. So, the mix LPS-ELISA may be used to monitor the serological surveillance for the presence of infection with a number of serotypes of Salmonella and would be useful for prevention and control of salmonellosis in animal.

Prevalence of arthropod antibodies in Korean patients with allergic rhinitis

  • Arthropod antigens are main causative agents which induce allergic reponses in humans. However. little information is known about the prevalence of specific arthropod allergens in Koreans with allergic diseases. The current study was designed to determine the positive rates of arthropod antibodies by the Korean inhalant panel of MAST-CLA. One hundred sixty patients, who were diagnosed with allergic rhinitis from an out-patient center at the Soonchunhyang University Chunan Hospital, were studied between August 1998 to July 2000. The overall positive rate, at least more than one specific antibody of arthropods such as Dermatophagoides farinae (Df), Dermatophagoides pteronyssinus (Dp), and cockroach mix (Cm) , was 46.9%. Each positive rate of Df, Dp, and Cm was 45.0%, 43.1%, and 8.8%, respectively. A significant agreement among arthropod allergens was observed (Df and Dp: 95.6%, Kappa : 0.911, P < 0.001). Our data supported the fact that arthropods were the most common allergens in Korean patients with allergic rhinitis; however, the MAST-CLA should be modified to increase specificity of arthropod allergens .

Combinatorial Solid Phase Peptide Synthesis and Bioassays

  • Solid phase peptide synthesis method, which was introduced by Merrifield in 1963, has spawned the concept of combinatorial chemistry. In this review, we summarize the present technologies of solid phase peptide synthesis (SPPS) that are related to combinatorial chemistry. The conventional methods of peptide library synthesis on polymer support are parallel synthesis, split and mix synthesis and reagent mixture synthesis. Combining surface chemistry with the recent technology of microelectronic semiconductor fabrication system, the peptide microarray synthesis methods on a planar solid support are developed, which leads to spatially addressable peptide library. There are two kinds of peptide microarray synthesis methodologies: pre-synthesized peptide immobilization onto a glass or membrane substrate and in situ peptide synthesis by a photolithography or the SPOT method. This review also discusses the application of peptide libraries for high-throughput bioassays, for example, peptide ligand screening for antibody or cell signaling, enzyme substrate and inhibitor screening as well as other applications.

Prevalence of Antibodies to Human Herpesvirus 8 in Children (소아의 항 Human Herpesvirus 8 항체 양성률)

  • Purpose : Human herpsevirus 8(HHV-8), a gamma herpsevirus, was initially identified from Kaposi sarcoma(KS) lesions and has been known to be associated with several malignancies including Kaposi sarcoma. HHV-8 seroprevalence is variable by different geographic areas and populations. The prevalence of HHV 8 infection in Korean children is unclear. So, we investigated the prevalence of HHV-8 specific antibodies in healthy children in Seoul, Korea. Methods : Sera were obtained from 112 children(age 1~15 years, 64 males and 48 females) who visited our hospital for routine health checkup and used for investigating sero-prevalence of anti-HHV-8 antibodies. An indirect immunofluorescent assay was used to detect the IgG antibodies to the lytic viral antigen(Biotrin, Dublin, Ireland). A peptide mix ELISA kit was used to detect the IgG antibodies to peptides specific for HHV-8 open reading frame (ORF)(Biotrin, Dublin, Ireland). Results : Of 112 children, 4 children younger than 6 years of age were seropositive to HHV-8[all 4(3.5%) were positive by IFA and 2(1.8%) were positive by ELISA]. Conclusion : These results suggest that the prevalence of antibody to HHV 8 in children in Korea is very low.

Characteristics of Allergic Patients in Soonchunhyang University Cheonan Hospital (순천향대학교 천안병원에 내원한 알레르기 환자의 특성)

  • The purpose of this study was to evaluate the general features of allergic patients in northwestern Chungcheongnamdo who visited Soonchunhyang University Cheonan Hospital. The subjects in this study were 1692 suspected allergic patients. After their allergic symptoms were checked and a MAST-CLA test was conducted, the following results were obtained: 1. The mean age of the subjects was 23.1 years old. The male patients represented 56.2% and the female patients accounted for 43.8%. 1387 (82.0%) patients of those investigated suffered from allergic disease. 2. Among the 1387 patients, 1022 (73.7%) patients showed an increased total IgE level. The positive rate of those who were in their 40s (87.0%) was the highest, but their age made no difference to their positive rate of total IgE. 3. Concerning the positive rate for allergen specific antibody by age, those who were in their teens (73.5%) topped the list and similar in all age group except in their 40s. By gender, the positive rate of the male and female were 56.3% and 43.9% respectively. Regarding the positive rates by allergic disease, those who suffered from allergic rhinitis (60.4%) were most vulnerable, followed by the patients with allergic dermatitis (47.4%), with bronchial asthma (47.2%) and with urticaria (39.4%). 4. As for seasonal positive rates, they were most susceptible in April (77.2%) and May (71.1%). We discovered a significant difference according to seasons; Spring (60.1%), Winter (45.4%) and Summer (39.2%). 5. In case of Korean inhalent panel, the most dominant allergen-specific antibodies were "Cockroach mix" (31.1%), followed by "D. pteronyssius" (23.8%) and "Dog" (14.3%). In the event of food panel, the most popular allergen-specific antibodies were "D. farinae" (25.0%), followed by "D. pteronyssinus" (19.8%) and "Hose dust" (12.0%). 6. The residential type made no difference to the positive rates of "House dust", "Cockroach mix" and "Dog" as major antigen but compared with others, positive rates for "Tick" were somewhat higher in apartment buildings.

Study of the Most Common Allergic Foods in Korea (국내 주요 알레르기 원인 식품에 대한 조사)

  • Prevalence of food allergic disease was examined by identifying the most common foods implicated in allergic reactions in Korea. Patients were subjected to test determining the amount of specific IgE antibody in serum against food allergens by CAP system. A total 9054 CAP analyses on egg white, egg yolk, cow milk, ${\alpha}-lactalbumin,\;{\beta}-lactoglobulin$, casein, wheat, rice, buckwheat, soybean, peach, crab, shrimp, pork, beef, chicken, tuna, salmon, mackerel, and food mix were undertaken. The results were considered to be positive when CAP value was same and/or greater than +2 (0.7 U/mL). Positive results of CAP analyses were 11.3% (1022/9054 cases), consisting of 336 on egg white, 266 on cow milk, 95 on egg yolk, 76 on soybean, 69 on ${\alpha}-lactalbumin$, 61 on casein, 58 on ${\beta}-lactoglobulin$, 39 on buckwheat, 12 on wheat, 3 on beef, 2 on crab, and 1 on rice, shrimp, pork, chicken or mackerel, and 0 on peach, tuna or food mix. Egg, cow milk, soybean, buckwheat, and wheat were identified as the most common allergic foods in Korea, showing an average of two different food sources for allergy per patient.
