초음파 조사에 의한 항생제 내성균 불활성화 및 감수성 변화 (Comparison of inactivation and sensitivity of antibiotic resistance bacteria by ultrasound irradiation)

  • The 20-kHz ultrasonic irradiation was applied to investigate bacterial inactivation and antibiotic susceptibility changes over time. Applied intensities of ultrasound power were varied at 27.7 W and 39.1 W by changing the amplitude 20 to 40 to three bacteria species (Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus). By 15-min irradiation, E. coli, a gram-negative bacterium, showed 1.2- to 1.6-log removals, while the gram-positive bacteria, Enterococcus faecalis and Staphylococcus aureus, showed below 0.5-log removal efficiencies. Antibiotic susceptibility of penicillin-family showed a dramatic increase at E. coli, but for other antibiotic families showed no significant changes in susceptibility. Gram-positive bacteria showed no significant differences in their antibiotic susceptibilities after ultrasound irradiation. Bacterial re-survival and antibiotic susceptibility changes were measured by incubating the ultrasound-irradiated samples. After 24-hour incubation, it was found that all of three bacteria were repropagated to the 2- to 3-log greater than the initial points, and antibiotic inhibition zones were reduced compared to ones of the initial points, meaning that antibiotic resistances were also recovered. Pearson correlations between bacterial inactivation and antibiotic susceptibility showed negative relation for gram-negative bacteria, E. coli., and no significant relations between bacterial re-survival and its inhibition zone. As a preliminary study, further researches are necessary to find practical and effective conditions to achieve bacteria inactivation.

  • Antibiotic residues are undesirable in milk and milk products for a number of reasons. In particular, they can have harmful effects on public health and harm to the manufacturer of the cultured milk products, e.g. MRSA etc. Although government regulatory agencies and the dairy industry have been successful in decreasing the presence of high concentrations of antibiotic residues, violations still occur and lead to contaminated products. As a result, several rapid and reliable methods for the detection of antibiotic residues have been developed, including microbiological and instrumental analysis methods. The conventional methods are time consuming, but recent improvements have allowed for better detection time, sensitivity, and accuracy. An example of an advanced detection instrument is the biosensor, which has several applications in food and environmental science, e.g. food-born pathogen detection, antimicrobial residues etc. In the present review, the recent trends in the methods used to test for antibiotic residues in milk and dairy products, as well as their specific applications, have been discussed.

  • Porcine E coli infection is a disease caused by Enterotoxin produced by Enterotoxigenic Escherichia coli(ETEC). Enteric colibacillosis has become an economically important disease in pigs as a result of increasing intensification of farrowing management. The present study undertaken to obtain the antibiotic sensitivity and distribution of serogroups and pili producibility test of ETEC from E. coli isolates in Chonnam. The results obtained were as follows. 1. A total of 71 isolates identified as E, coli employing IMViC system from rectal specimens of 54 piglets with diarrhea. 2. In antibiotic sensitivity test, isolates showed high sensitivity to AN, CM, Fox, GM, but resistance to EM, NA TC. 3. The distribution of 25 Isolates of serogroups were 0141:K85(11.3%), 08:K87(8.5%), 064:K (5.6%), 0138:K8l (4.2%), 0139 :K82(2.8%), 0157:K88ac(1.4%) and 0149:K9l (1.4%). 4. MRHA of guinea pig erythrocytes was detected in 8 out of 25OK serotypes and 9 out of 46 unidentified serotypes. MRHA titers of serotypes showed from 64 to 128 in 0141: K85, 2 in 0138:K8l and no titers in 0139:K82. 5. The production of heat labile enterotoxin of ETEC was detect 39 out of 52 isolates showed $\beta$-hemolysin, 7 out of 52 isolates showed ${\gamma}$-hemolysin and 6 out of 52 isolates showed ${\gamma}$-hemolysin by $GM_1$ganglioside ELISA. The distribution of LT toxin were in 12 isolate showed $\beta$-hemolysin, 2 isolates showed ${\alpha}$-hemolysin and 3 isolates showed ${\gamma}$-hemolysin in 25 OK serotypes.

Augmentin 의 임상효과 (Clinical Study of Augmentin)

  • Augmentin is a formulation of amoxycillin trihydrate and potassium clavulanate, a fused beta-lactam molecule produced by the fermentation of Streptomyces clavuligerus. Most clinically important resistance is due to the production by bacteria of antibiotic destroying enzymes. In the case of penicillins and cephalosporins these enzymes are termed beta-lactamase as they destroy the beta-lectern ring of these antibiotics, completely inactivating them. The presence of clavulanic acid extends the spectrum of amoxycillin to include bet On clinical study of the intravenous Augmentin in the field of thoracic and cardiovascular surgical cases, we selected randomly 30 patients, 21 male and 9 female, age from 13 to 72, in the period from April to December 1985. Among the total 30 patients, 22 were preoperatively infected [11 thoracic empyema, 5 lobar pneumonia, 2 lung abscess, 2 bronchiectasis, one acute pyelonephritis with ureter stone and one rheumatic carditis], and 8 were not infected preoperatively [Table 1, 2]. Of the preoperatively infected group, 11 cases [50%] were culture positive [4 staphylococcus, 3 pseudomonas, 2 Serratia group, and one E. coli], and preoperatively non-infected group [8 cases] revealed expectedly negative findings on bacterial culture. All of the culture positive bacteria were sensitive to Augmentin on disc culture sensitivity test except one case of E. coli. Daily doses of intravenous Augmentin were 2.-1-6.0gm divided in 2-5 injections. Every injection administered [1.2gm at Augmentin dissolved in 20ml distilled water] slowly for more than 20 minutes. Duration of injection was variable according to the clinical conditions from minimum 5 to maximum 31 days. The results of antibiotic treatment with Augmentin and some other antibiotic combinations pre- and postoperatively were subgrouped as EXCELLENT, EFFECTIVE, and FAILURE. Clinical criteria of the therapeutic result were symptomatic, objective and laboratory improvement. 8 cases were excellent, 13 effective, and one failure among the preoperatively infected group, and all 8 cases of the preoperatively non-infected group were effective as pro;hylactive antibiotic therapy. Overall effective ratio was 97% in both subgroup. There was no side effect clinically and laboratory study including liver and kidney function test during and after the I.V. administration of Augmentin. Oral swallow tablets which were administered after discharge from hospital also revealed good effects with some degree of gastrointestinal trouble.

Dual Inoculation of Native Rhizobium spp. and Arbuscular Mycorrhizal Fungi: An Impact Study for Enhancement of Pulse Production

  • Fifteen Rhizobium spp. from nodules of 6 common pulses collected from 6 districts of Assam were studied for their infectivity, intrinsic antibiotic resistance, nitrogenase activity and effect of dual inoculation with two native Arbuscular Mycorrhizal Fungi viz. Glomus mosseae(GM) and Gigaspora gilmarie(GG). Out of the 15 isolates 9 were found nodulation positive and 6 of them(AR1, BR8, BR12, AR10, UR10 & GR21) were subjected to intrinsic antibiotic sensitivity test of which AR1 showed resistance against all the 9 test antibiotics. Isolates AR1 and GR21 showed the highest(4.25 mole, $gm^{-1}hour^{-1}$) and the lowest(1.05 mole, $gm^{-1}hour^{-1}$) nitrogenase activity respectively. In Most Probable Number count, the maximum Rhizobium population $5.8{\times}10^5$, was found in both Blackgram and Greengram variety of pulses. The maximum dry weight of nodules(3.14 g), dry weight of shoot(10.08 g), nitrogen content(7.68 mg, $plant^{-1}$), chlorophyll content(1.89 mg, $g^{-1}$), phosphorus content of shoot(6.17 mg, $g^{-1}$) and yield(535.67 kg, $Ha^{-1}$) were found when AR1 dually inoculated with GM in Blackgram.

항생제에 대한 세균학적 감수성시험방법에 관한 비교실험(Tetracycline, Neomycin 및 Colistin) (A Comparative Experiment on the Methods for Antibiotic Sensitivity Test In Vitro)

  • A comparative study was performed with 176 cultures of Salmonella organisms on tetracyline, neomycin and colistin in order to find out the relationship between the results obtained from the Ericsson's single disk method and the tube dilution method of antibiotic sensitivity tests which may be carried out in many hospital laboratories. With tetracycline, thirty-three out of 163 cultures of Salmonella typhi were found to be either sensitive or moderate sensitive by means of the disk method and thirty one(ca 94%) out of the thirty three cultures showed less than 1.0 ${\mu}g$ of the Minimal Inhibitory Concentretions(MIC) in the tube-dilution tests, which mean that there were a quite good agreement between the two methods. With neomycin, a hundred and five out of 163 S.typhi were appeared to be either sensitive or moderate sensitive by means of Ericsson's single disk method, among which 103 cultures showed less than 10.0 ${\mu}g$ MIC in the tubedilution method. And also there was a quite correlative pat. terns observed in the result of testing with 13 salmonella cultures other than S. typhi. With colistin, it was hard to observe any particular tendency in the distribution of plotting for 148 cultures showing less the 18 mm in the inhibiting zone diameters between MIC and disk sensitivity patterns except the fifteen, cultures out of 176 salmonella, which appeared to be sensitive in the single disk method and showed less than 1.0 ${\mu}g$ MIC in the tube dilution method.

Effect of Low Dose γ-Irradiation on the Fate and Cell Envelope of Bacillus cereus, Escherichia coli, and Salmonella Typhimurium

  • This study investigated the effect of low dose ${\gamma}$-irradiation on the damage of the cell envelopes and antibiotic sensitivity profiles of Bacillus cereus, Escherichia coli, and Salmonella Typhimurium. The bacteria suspension in tryptic soy broth was exposed to the ${\gamma}$-irradiation doses of 0, 1, 1.5, 3, and 5 kGy, and then stored at $0^{\circ}C$ for 24 h. A viability test, an antimicrobial sensitivity profile, and an electron microscopy were performed to observe the effects due to ${\gamma}$-irradiation treatment. B. cereus could survive the ${\gamma}$-irradiation up to 5 kGy while E. coli and S. Typhimurium were all deactivated at 1.5 kGy and 5 kGy, respectively. At 5 kGy, the cell count of B. cereus was significantly reduced, and the survived bacteria cells retained their important features. There were no significant changes observed in the antimicrobial sensitivity profile (p>0.05) for the recovered bacteria after irradiation treatment. Low dose ${\gamma}$-irradiation below 3 kGy was found to be insufficient to achieve decontamination of B. cereus and S. Typhimurium. Cell envelope damage and deactivation of different bacteria did not occur in the same manner; thus, deferent doses of ${\gamma}$-irradiation may be required for deactivation of different bacteria.

Microbiological Contamination and Sanitary Management of Aquarium Water in the Busan Area

  • This research has been performed by our own investigation, also cooperated with Health and Sanification Division and each of district offices in Busan metropolitan city. After choosing, we collected water samples five times for microbiological examination. As a result of investigating in 160 water samples from urban areas, we could detect 88 cases of Vibrio spp. Furthermore, there were four cases exceeding the acceptable limit of aquarium water (100,00/mL) and another four cases exceeding the limit of Coliform group (1,000 below/100 mL). As a result of investigating that we performed for 271 cases of water samples from coastal areas from April to November, we could detect 130 cases of vibrio species and 10 cases of Coliform group. After performing 17 kinds of antibiotic susceptibility test for 41 cases of isolated Vibrio parahaemolyticus, 27 cases showed tolerance to Amplicllin (AM), all of 31 cases showed intermediate resistance only to Cefazolin (CF) but had sensitivity to the rest of them. As a result of performing antibiotic susceptibility test, serum test and PFGE gene analysis on each 10 pair of Vibrio parahaemolyticus detected concurrently from intake-pipe water and, aquarium water, we couldn't get data showing that they are clearly same species in three kinds of test. In addition, UV sterilization, Ozonization and so on. Based on our research, intake pipe didn't have a problem with microbiological contamination so we are sure that the germ came from supplied fish had caused that kind of contamination. For effective management, UV sterilization or Ozonization which can be handled consistently should be adopted in aquarium.

Ga-67 골주사와 X선사진을 이용한 실험적 하악골 염증성 병변의 활성도 평가 (Ga-G7 SCINTIGRAPHIC AND RADIOGRAPHIC EVALUATION OF THE ACTIVITY OF EXPERIMENTALLY INDUCED MANDIBULAR INFLAMMATORY LESION)

  • The purpose of this study was to evaluate the usefulness of Ga-67 scintigram in predicting the disappearance of inflammatory activity in mandibular lesions of dogs. Inflammatory bony lesions were produced artificially by the injection of sclerosing solution and bacterial suspension through the distal root canals of the lower 4th premolars in 10 dogs. The serial Ga-67 scintigrams and periapical radiograms were obtained. After 3 to 6 weeks duration of antibiotic therapy, the experimental animals were killed, and the mandibles were dissected. Bacteriologic culture and antibiotic sensitivity test were done. 1. The following results were obtained. The negative Ga-67 scan has 100% predictive value, and is a strong evidence of the resolution of inflammatory activity. In 2 animals of positive Ga-67 scan, I animal had negative culture. In 14 cases of positive Ga-67 scan, radiographically there was increase in the size of lesion in 9 cases, no change in 5 cases. In 8 cases of negative Ga-67 scan, radiographically there was increase in the size of lesion in I case, no change in 6 cases and decrease in 1 case.
