Synthesis and Characterization of CdSe/CdS/N-Acetyl-L-Cysteine/Quercetin Nano-Composites and Their Antibacterial Performance

  • We have discovered that quercetin, once coated on the CdSe and CdSe-CdS quantum dots (QDs), becoming highly water soluble. In the present work, we have successfully synthesized CdSe/CdS/N-Acetyl-L-Cysteine(NAC)/Quercetin nano-composites in the aqueous solution. The products were characterized using UV-vis spectroscopy, X-ray powder diffraction, fluorescence spectroscopy, and Fourier transform infrared spectroscopy. The transmission electron microscopy (TEM) tests indicated that our nano-composite products are highly stable with homogeneous particle size and great monodispersity. Quercetin coated nano-composite CdSe/CdS/NAC/Quercetin showed different fluorescence behavior from that of CdSe/CdS/NAC. Most amazingly, the synthesized CdSe/CdS/NAC/Quercetin nano-composite exhibits strong antibacterial activity. The combination of the strong fluorescence and its antibacterial activity makes the quercetin modified quantum dots as a potential candidate for cancer targeted therapy and other cancer treatments.

Comparison of In Vitro, Ex Vivo, and In Vivo Antibacterial Activity Test Methods for Hand Hygiene Products (손 위생 제품에 대한 in vitro, ex vivo, in vivo 항균 시험법 비교)

  • Numerous methods have been applied to assess the antibacterial effectiveness of hand hygiene products. However, the different results obtained through various evaluation methods have complicated our understanding of the real efficacy of the products. Few studies have compared test methods for assessing the efficacy of hand hygiene products. In particular, reports on ex vivo pig skin testing are limited. This study aimed to compare and characterize the methodologies applied for evaluating hand hygiene products, involving in vitro, ex vivo, and in vivo approaches, applicable to both leave-on sanitizers and wash-off products. Our further aim was to enhance the reliability of ex vivo test protocols by identifying influential factors. We performed an in vitro method (EN1276) and an in vivo test (EN1499 and ASTM2755) with at least 20 participants, against Serratia marcescens or Escherichia coli and Staphylococcus aureus. For the ex vivo experiment, we used pig skin squares prepared in the same way as those used in the in vivo test method and determined the optimal treated sample volumes for sanitizers and the amount of water required to wash off the product. The hand sanitizers showed at least a 5-log reduction in bacterial load in the in vitro test, while they showed little antibacterial activity in the in vivo and ex vivo tests, particularly those with a low alcohol content. For the hand wash products, the in vitro test was limited because of bubble formation or the high viscosity of the products and it showed low antibacterial activity of less than a 1-log reduction against E. coli. In contrast, significantly higher log reductions were observed in ex vivo and in vivo tests, consistently demonstrating these results across the two methods. Our findings revealed that the ex vivo and in vivo tests reflect the two different antibacterial mechanisms of leave-on and wash-off products. Our proposed optimized ex vivo test was more rapid and more precise than the in vitro test to evaluate antibacterial results.

Effects of the Water Extract of Akebia (Akebia quinata Edcaisne) on the Growth of Clotridium perfringens and Some Intestinal Microorganisms (목통 (Akebia quinata Decaisne)의 물추출물이 Clostridium perfringens 및 주요 장내미생물의 생육에 미치는 영향)

  • As a result of screening the medicinal herbs which selectively control human intestinal microflora, water extract of Akebia quinata Decaisne was proved to have a strong inhibitory activity against the growth of Clostridium pefringens, a major harmful intestinal bacterium. The anti-bacte-rial activity was stable under the thermal treatment at 100$\circ$C for 120 min and in a range of pH 1 to 11. In addition, the water extract of Akebia quinata Decaisne showed the antibacterial activities against five different strains of Clostridia including C. perfringens. On the contrary, the extract did not inhibit the growths of Bifidobacterium, Lactobacillus, Escherichia coli, Enterococcus faecalis. The extract, however, suppressed markedly the growth of Bacteroides fragilis and Staphylococcus aureus in vitro. Alike in the mixed culture inoculated with human feces as starter, in vivo tests using rats showed that the extract tends to increase the numbers of Bifidobacteria and Lactobacilli in the intestinal microflora of rats, whereas those of Clostridia were attenuated.

Antibacterial and Deodorization Activities of Cotton Fabrics Dyed with Amur Cork Tree Extracts (황벽 추출액에 의한 면 염색물의 항균.소취성)

  • Using water and methanol, the colorant components were extracted from the heartwood of amor cork tree which is known as the oldest natural yellow dye. The dyeabilities of the colorant on cotton and the fastness of cotton fabrics dyed with amur cork tree extracts were investigated. According to the dye uptake and mordant concentration, antibacterial activities and deodorization properties of dyed fabrics were also investigated. As a results, K/S values of dyed fabrics were increased with concentrations of mordants and amur cork tree extracts. Washing fastness of dyed fabrics was very low, light fastness was fair and the other fastness were good. Deodorization rates of dyed fabrics were increased in proportion to concentrations of amur cork tree extracts. The reduction rates of bacteria measured by Bioassay tests were also increased in proportion to concentrations of mordants and amur cork tree extracts.

Incorporation of silver nanoparticles on the surface of orthodontic microimplants to achieve antimicrobial properties

  • Objective: Microbial aggregation around dental implants can lead to loss/loosening of the implants. This study was aimed at surface treating titanium microimplants with silver nanoparticles (AgNPs) to achieve antibacterial properties. Methods: AgNP-modified titanium microimplants (Ti-nAg) were prepared using two methods. The first method involved coating the microimplants with regular AgNPs (Ti-AgNP) and the second involved coating them with a AgNP-coated biopolymer (Ti-BP-AgNP). The topologies, microstructures, and chemical compositions of the surfaces of the Ti-nAg were characterized by scanning electron microscopy (SEM) equipped with energy-dispersive spectrometer (EDS) and X-ray photoelectron spectroscopy (XPS). Disk diffusion tests using Streptococcus mutans, Streptococcus sanguinis, and Aggregatibacter actinomycetemcomitans were performed to test the antibacterial activity of the Ti-nAg microimplants. Results: SEM revealed that only a meager amount of AgNPs was sparsely deposited on the Ti-AgNP surface with the first method, while a layer of AgNP-coated biopolymer extended along the Ti-BP-AgNP surface in the second method. The diameters of the coated nanoparticles were in the range of 10 to 30 nm. EDS revealed 1.05 atomic % of Ag on the surface of the Ti-AgNP and an astounding 21.2 atomic % on the surface of the Ti-BP-AgNP. XPS confirmed the metallic state of silver on the Ti-BP-AgNP surface. After 24 hours of incubation, clear zones of inhibition were seen around the Ti-BP-AgNP microimplants in all three test bacterial culture plates, whereas no antibacterial effect was observed with the Ti-AgNP microimplants. Conclusions: Titanium microimplants modified with Ti-BP-AgNP exhibit excellent antibacterial properties, making them a promising implantable biomaterial.

Adsorption Capacity and Antibacterial Activity of Porous Feldspar Porphyry (다공성 구조가 발달한 장석반암의 흡착과 항균성 연구)

  • As weathering processes, micro-cavities are formed on the surface of rocks, and in particular, the porous structure is increased in feldspar. Adsorption and antibacterial tests were carried out to clarify the environmental function of porous feldspar porphyry. Almost all the heavy metals were adsorbed in the feldspar filter and the adsorption rate could be controlled by changing the filter length. The shake flask method of fabric coated with 5% and 7% feldspar powder showed very high antibacterial activity of 98% and 99.9%, respectively. The cation exchange capacity at a particle size of $10{\mu}m$ was 114.63 meq/100g probably due to the porous structure. The potential value of porous feldspar porphyry as a resource is sufficient based on the results of the experiment.

Antibacterial effect of urushiol on E. faecalis as a root canal irrigant

  • Objectives: The purpose of this study was to compare the antibacterial activity of urushiol against Enterococcus faecalis (E. faecalis) to that of NaOCl. Materials and Methods: The canals of thirty two single rooted human teeth were instrumented with Ni-Ti files (ProTaper Next X1, X2, X3, Dentsply). A pure culture of E. faecalis ATCC 19433 was prepared in sterile brain heart infusion (BHI) broth. The teeth were submerged in the suspension of E. faecalis and were incubated at $37^{\circ}C$ for 7 days to allow biofilm formation. The teeth were randomly divided into three experimental groups according to the irrigant used, and a negative control group where no irrigant was used (n = 8). Group 1 used physiologic normal saline, group 2 used 6% NaOCl, and group 3 used 10 wt% urushiol solution. After canal irrigation, each sample was collected by the sequential placement of 2 sterile paper points (ProTaper NEXT paper points, size X3, Dentsply). Ten-fold serial dilutions on each vials, and 100 µL were cultured on a BHI agar plate for 8 hours, and colony forming unit (CFU) analysis was done. The data were statistically analyzed using Kruskal-Wallis and Mann-whitney U tests. Results: Saline group exhibited no difference in the CFU counts with control group, while NaOCl and urushiol groups showed significantly less CFU counts than saline and control groups (p < 0.05). Conclusions: The result of this study suggests 10% urushiol and 6% NaOCl solution had powerful antibacterial activity against E. faecalis when they were used as root canal irrigants.

Anti-inflammatory Effect on RAW 264.7 Cells and Antibacterial Effect on Cutibacterium acnes of Compounds Isolated from Sedum takesimense (섬기린초(Sedum takesimense)에서 분리된 화합물들의 RAW 264.7 Cell에 대한 항염증 효과와 여드름 원인균에 대한 항균 효과)

  • This study investigated anti-inflammatory and antibacterial activities of Sedum takesimense ethanolic extract, and 3 isolated compounds. To confirm anti-inflammatory and anti-acne activities, a nitric oxide (NO) inhibition assay, pro-inflammatory factor (TNF-α, IL-1β, and IL-8) inhibition assays, and minimum inhibitory concentration (MIC) tests were performed. The 3 isolated compounds were identified as 4,6-di-O-galloylarbutin (OGA), 2,4,6-tri-O-galloyl-glucose (OGG), and 1,2,4,6-tetra-O-galloyl-β-glucose (TOGG). The ethanolic extract and isolated compounds (OGA, OGG, TOGG) effectively inhibited production of NO and pro-inflammatory cytokines (TNF-α, IL-1β, and IL-8). Furthermore, OGG and TOGG exhibited MIC values toward Cutibacterium acnes of 12.5 ㎍/mL and 3.2 ㎍/mL, respectively. These results suggest that S. takesimense extract exerts an anti-inflammatory effect on LPS-induced RAW264.7 cells, and an antibacterial efficacy against C. acnes.

Comparative Study of the Biological Activity of Propolis Extracts with Various Countries of Origin as Cosmetic Materials (원산지별 프로폴리스 추출물의 화장품 소재로서의 생리활성 비교연구)

  • Propolis is a sticky resinous substance that is formed by the combination of honeybee secretions and resin of plants, which serves to protect from bacteria and viruses. This study aims to evaluate the efficacy of propolis extract from Korea (KPE), China (CPE), and Brazil (BPE) through antioxidant, antibacterial, whitening, and anti-inflammatory tests, and to examine their potential as cosmetic materials. KPE, CPE, and BPE showed significant antioxidant activities on flavonoid/polyphenol content and free radical scavenging activity. The antibacterial effect of propolis on skin flora was determined by measuring the minimal inhibitory concentration (MIC). KPE showed better antibacterial efficacy than CPE and BPE in C. acnes (KPE, CPE, and BPE: (62.5, 250, and 500) ㎍/mL, respectively). Furthermore, KPE inhibited the melanin synthesis in human epidermal melanocytes and production of nitric oxide and PGE2 induced by lipopolysaccharide (LPS) in mouse macrophages, which showed better than did CPE or BPE. Taken together, the propolis extracts can be applied to antioxidant, antibacterial, and anti-inflammatory ingredient for cosmetics, while KPE showed superior potential in antibacterial, anti-inflammatory, and whitening efficacies.

  • This paper presents an invetigation into the cause of deterioration of wet surrounding RC structures subjected to checmical attacks such as sewage. The antibacterial-reforming agent is developed after determining the permeability of the RC structure. After application of the anitbacterial-reforming agent through SEM, the permeability, compressive strength properties and the micro-structure of the concrete were evaluated for durability. In addition, the antibacterial-reforming agent was combined with a protective coating for the wet surrounding RC structure and evaluated for durability. The combined effect of the antibacterial-reforming agent and the protective coating were evaluated in field tests in both sewer system and tunnel sites.
