비자의 항균 및 알레르기성 접촉 피부염 개선 작용 연구 (Study on the Improvement Effect of Antibacterial and Allergic Contact Dermatitis on Torreyae Semen Extracts)

  • The purpose of this study was to investigate the efficacy of Torreyae semen extract against antimicrobial and allergic contact dermatitis to explore its utility as a herbal resource. To this end, the antibacterial effects of Torreyae Semend extracted with hot water and ethanol (30%, 60%, 90%) were investigated using Staphylococcus aureus, Staphylococcus epidermidis, pseudomonas aeruginosa, and Propionibacterium acnes. And observed the anti-atopic dermatitis efficacy with a mouse model that allergic contact dermatitis induced by DNCB. Experimental results showed that the antibacterial activity against S. epidermidis and P. aeruginosa was significantly higher in the hot water extract of Torreyae Semen. We observed that the extracts of 30% ethanol and hot water of Torreyae Semen inhibited growth in a concentration-dependent manner in P. acnes & S. epidermidis. We observed that the administration of hot water extracts and 30% ethanol extract of Torreyae Semen significantly reduce the thickness of ear & back and gross skin condition compared to the control group in the mice with allergic contact dermatitis. As a result of H & E histological staining, it was observed that the Torreyae Semen extraction group had markedly reduced epidermis and skin thickness, granules, and keratinization compared to the control group. The groups administrated with Torreyae Semen hot water extracts had decreased TNF-α and IL-10 contents compared with the control group. As a result, Torreyae Semen may be used as a useful herbal resource because it has a certain level of antibacterial and anti-atopic effects.

  • Rhizomes of Kaempferia parviflora (Zingiberaceae) have been used in traditional Thai medicine for health promotion. In this study, the antibacterial activity of ethanol extract of K. parviflora against Cronobacter spp. and enterohemorrhagic Escherichia coli (EHEC) was investigated using paper disc dilution method. The results revealed that the ethanol extract exhibited antibacterial activity against Cronobacter spp. and EHEC. With an increasing concentration of K. parviflora ethanol extract, larger zones of inhibition of Cronobacter spp. and EHEC strains tested were observed. Therefore, its antibacterial activity suggested that K. parviflora could be used as a natural additive to ascertain food safety of various dairy products.

Antibacterial Effects of Galla Rhois Extract against Streptococcus suis Infection in Mice

  • Streptococcus suis (S. suis) is a major swine pathogen and an emerging zoonotic agent and is an increasing public health problem across Asia. The present study was undertaken to estimate the antibacterial effect of GR extract and therapeutic effect of GR extract against S. suis infection in mice. At the concentration of GR extract 2.5 mg/ml, the antibacterial effect was not shown on S. suis. However, the antibacterial effect against S. suis was observed at the concentration of GR extract 5.0 mg/ml. Oral administration of GR extract at the dose of 10 mg/kg showed a therapeutic effect for S. suis infected BALB/c mice. The mortality of GR extract-treated mice at the concentration of 5, 10 and 20 mg/kg was 80%, 70%, and 50% at 12 days, respectively, while that of untreated mice was 100% at 8 days after a lethal dose of S. suis infection. The results of our study strongly indicate that GR extract has potential as an effective for S. suis infection in mice.

만응환(萬應丸) 에탄올 추출물의 메티실린 내성 포도상구균에 대한 항균활성 및 내성억제 효과 (Antibacterial Activity and Inhibition of Resistance in Methicillin-resistant Staphylococcus aureus by Maneung-hwan Ethanol Extract)

  • Objectives In this study, we investigated the antimicrobial activity of a 70% ethanol extract of Maneung-hwan (MEH), which is prescribed by practitioners of oriental medicine for use against methicillin-resistant Staphylococcus aureus (MRSA). Methods The antibacterial activity of MEH against MRSA strains was evaluated using the disc diffusion method, broth microdilution method (minimal inhibitory concentration, MIC), checkerboard dilution test, and time-kill test. The mechanism of action of MEH was investigated by bacteriolysis using detergents or ATPase inhibitors Additionally, mRNA and protein expression were investigated by quantitative reverse transcription-polymerase chain reaction and western blot assay, respectively. Results The MIC of MEH was 25~1,600 ㎍/mL against all the tested bacterial strains. We showed that MEH extract exerts strong antibacterial activity. In the checkerboard dilution test, the fractional inhibitory concentration index of MEH in combination with antibiotics indicated synergism or partial synergism against S. aureus. The time-kill study indicated that the growth of the tested bacteria was considerably inhibited after a 24-h treatment with MEH and selected antibiotics. To measure the cell membrane permeability, MEH (3.9 ㎍/mL) was combined with Triton X-100 (TX) at various concentrations N,N-dicyclohexylcarbodimide (DCCD) was also tested as an ATPase inhibitor. TX and DCCD cooperation against S. aureus exhibited synergistic action. Accordingly, the antimicrobial activity of MEH in the context of cell membrane rupture and ATPase inhibition was assessed. Additionally, the expression of genes and proteins associated with resistance was reduced after exposing MRSA to MEH. Conclusions These results suggest that MEH possesses antibacterial activity and acts as a potential natural antibiotic against MRSA.

  • 본 연구에서는 가격이 저렴하고 우수한 결과를 갖는 항균제인 견운모(sericite)를 사용하였다. 견운모의 입자 크기는 $15{\mu}m$로 고정하였다. 박리강도의 효과는 접착제의 종류에 영향을 받지 않았다. 박리강도는 접착제 함량이 증가함에 따라 증가하였고, 접착제 함량이 $20g/m^2$ 일 때 기준 값을 만족하였다. 항균제로 처리된 면직물의 한국공업규격(KS) 세탁견뢰도를 만족하는 항균제 함량은 8%이다. 신발용 면직물에 있어 항균처리를 위한 연속적인 공정 조건들은 다음과 같다: 용융온도 $120^{\circ}C$, 냉각시간 20 s, 압착온도 $130^{\circ}C$, 압착시간 30 s.

항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과 (Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

Comparative Analysis of Anticancer and Antibacterial Activities among Seven Trametes Species

  • Species in the genus Trametes (Basidiomycota, Polyporales) have been used in natural medicine for a long time. Many studies reported that mycelia or fruiting bodies of Trametes spp. exhibited effects of antioxidant, anti-inflammatory, anticancer, and antimicrobial activities. However, comparative analysis in this genus is scarce due to limitation of morphological identification and the sample number. In this study, the 19 strains of seven Trametes species were chosen to generate a five-gene-based phylogeny with the 31 global references. In addition, 39 culture extracts were prepared for 13 strains to test for anticancer and antibacterial activities. Strong anticancer activities were found in several extracts from T. hirsuta and T. suaveolens. Anticancer activities of T. suaveolens, T. cf. junipericola and T. trogii were first described here. The antibacterial ability of T. versicolor and T. hirsuta extracts has been confirmed. The antibacterial activities of T. suaveolens have been reported at the first time in this study. These results suggest an efficient application of the genus Trametes as the drug resources especially for anticancer agents.

Preparation and Performance Evaluation of a Zinc Oxide-Graphene Oxideloaded Chitosan-Based Thermosensitive Gel

  • This study aimed to develop and assess a chitosan biomedical antibacterial gel ZincOxide-GrapheneOxide/Chitosan/β-Glycerophosphate (ZnO-GO/CS/β-GP) loaded with nano-zinc oxide (ZnO) and graphene oxide (GO), known for its potent antibacterial properties, biocompatibility, and sustained drug release. ZnO nanoparticles (ZnO-NPs) were modified and integrated with GO sheets to create 1% and 3% ZnO-GO/CS/β-GP thermo-sensitive hydrogels based on ZnO-GO to Chitosan (CS) mass ratio. Gelation time, pH, structural changes, and microscopic morphology were evaluated. The hydrogel's antibacterial efficacy against Porphyromonas gingivalis, biofilm biomass, and metabolic activity was examined alongside its impact (MC3T3-e1). The findings of this study revealed that both hydrogel formulations exhibited temperature sensitivity, maintaining a neutral pH. The ZnO-GO/CS/β-GP formulation effectively inhibited P. gingivalis bacterial activity and biofilm formation, with a 3% ZnO-GO/CS/β-GP antibacterial rate approaching 100%. MC3T3-e1 cells displayed good biocompatibility when cultured in the hydrogel extract.The ZnO-GO/CS/β-GP thermo-sensitive hydrogel demonstrates favorable physical and chemical properties, effectively preventing P. gingivalis biofilm formation. It exhibits promising biocompatibility, suggesting its potential as an adjuvant therapy for managing and preventing peri-implantitis, subject to further clinical investigations.

볏붉은잎 추출물의 돼지 유래 병원성 세균에 대한 항균효과 (Antibacterial activity of Callophyllis japonica-methanol extracts against the pathogenic bacteria from swine)

  • Interest in marine organisms as potential sources of bioactive agents has increased in recent years. The red seaweed, Callophyllis (C.) japonica, is abundant in the coastal regions of Jeju Island in South Korea. A previous study shows that C. japonica extracts have antioxidant activity and radioprotective effects. In this study, an methanol extract of C. japonica was tested whether it has antibacterial effects against the bacteria from swine. In vitro antibacterial activities of the crude extracts prepared from the C. japonica using 80 % methanol were tested for inhibitory activity against the Escherichia (E.) coli (S175), Enterococcus (E.) faecium (ATCC 51558), Salmonella (S.) Typhimurium and Staphylo-coccus (S.) aureus (ATCC 25923) by using broth dilution method. All organisms were incubated in brain heart infusion medium containing 1% extract at 0, 4, 8, 12 and 24 hrs. The 3 days-old piglets were fed an experimental diet supplemented with 1% C. japonica for 1 week. And the change of the coliform bacteria in feces were examined after supplement of C. japonica for 1 week. When the inocula containing $10^2{\sim}10^3$CFU/ml of each organism were used the extracts of C. japonica showed various degrees of antibacterial effects on all bacteria tested. The CFU value ($6.3\times10^8$CFU/ml) of C. japonica for E. coli was decreased 30% compared with vehicle controls ($9.0\times10^8$CFU/ml) after 8 hrs incubation. The proliferation rate of E. faecium was inhibited about 68% at 4 hrs, 81% at 8 hrs and 76% at 12 hrs after incubation, respectively. The proliferation rate of S. Typhimurium was inhibited about 96% at 4 hrs, 90% at 8 hrs and 72% at 12 hrs after incubation with extracts of C. japonica. The proliferation rate of S. aureus was inhibited more than 90% each time courses. Conclusively, a red seaweed extract of C. japonica was found to be effective against a number of gram negative and gram positive bacteria such as E. coli, E. faecium, S. Typhimurium, and S. aureus. The number of coliform bacteria was increased in the 1% C. japonica-treated group, as compared to those of controls. This result suggests that C. japonica extracts be added as an effective natural antibacterial agent. The precise mechanism of antibacterial effects and its application on swine industry remains to be further studied.

다양한 약물과 영양제가 장내세균에 미치는 영향 (The influence of various medicines and nutritional supplements against bacteria with gastrointestinal relevance)

  • 최근 우리나라는 서구화된 식생활과 생활양식의 변화로 다양한 대사성 질환과 만성 질환이 나타나고 있다. 이러한 질환들은 지속적으로 약물을 복용해야 한다. 또한 많은 사람들이 건강한 삶을 유지하기 위해 건강기능식품과 각종 비타민, 영양제를 복용한다. 하지만 복용하는 약물들이 장내 세균에는 어떠한 영향을 주는 지에 대한 연구는 많이 이뤄지지 않고 있다. 이에 본 연구에서는 흔하게 사용되며 장기간 복용하는 약이 항균활성시험으로 위장 관련 박테리아에 영향을 미치는 지를 조사하였다. 그 결과 비타민 및 미네랄, 중추신경계에 작용하는 약들이 장내 세균과 유산균에 대해 항균력을 나타냈으며, 그 중 중추신경계에 작용하는 약 중 항현훈제의 dexibuprofen는 장내 유익균 인 Lactobacillus casei과 Lactobacillus rhamnosus와 장내 유해균 인 Staphylococcus aureus에 대해 항균력이 높게 작용하였다. 또한 심혈관계, 조혈기계에 작용하는 약 중 항이뇨제 의 fenofibric acid는 장내 유익균인 Lactobacillus casei에 대해 항균력이 나타났다. 비타민 및 미네랄은 대부분의 장내 균에서 항균력을 보이지만, 특히 Vitamin B-Complex/with C와 vitamin C가 유익균인 Bifidobacterium infantis과 장내 유해균 인 Escherichia coli, Enterobacter aerogenes, Shigella flexneri, Salmonella Typhimurium, Staphylococcus aureus에서 높게 나타났다. 따라서 본 연구결과 장내 유해균과 유익균에 다양하게 복용되는 약들이 대체적으로 항균활성을 보이는 것으로 판단된다.
