With continuing demand for the development of new, effective and safe therapies, an investigation was carried out to test the efficacy of an antibacterial agent derived from marine edible seaweed. The methanolic extract of Ecklonia cava from marine edible seaweed evinced potential antibacterial activity against Enterococcus faecalis. Among five solvent-soluble fractions of E. cava methanolic extract, the ethyl acetate soluble extract (EtOAc) exhibited the strongest antibacterial activity, with a MIC value of $128{\mu}g/mL $ against E. faecalis strains. Furthermore, a synergistic antibacterial effect between an antibiotic and the EtOAc fraction was assessed using fractional inhibitory concentration (FIC) indices. A combination of ciprofloxacin and the EtOAc fraction resulted in a ${\sum}FIC_{min}$ range of 0.188 and ${\sum}FIC_{max}$ of 0.508 to 563, suggesting that the ciprofloxacin-EtOAc fraction of E. cava combination resulted in an antibacterial synergy effect against E. faecalis.

The purpose of this study was to investigate the efficacy of Torreyae semen extract against antimicrobial and allergic contact dermatitis to explore its utility as a herbal resource. To this end, the antibacterial effects of Torreyae Semend extracted with hot water and ethanol (30%, 60%, 90%) were investigated using Staphylococcus aureus, Staphylococcus epidermidis, pseudomonas aeruginosa, and Propionibacterium acnes. And observed the anti-atopic dermatitis efficacy with a mouse model that allergic contact dermatitis induced by DNCB. Experimental results showed that the antibacterial activity against S. epidermidis and P. aeruginosa was significantly higher in the hot water extract of Torreyae Semen. We observed that the extracts of 30% ethanol and hot water of Torreyae Semen inhibited growth in a concentration-dependent manner in P. acnes & S. epidermidis. We observed that the administration of hot water extracts and 30% ethanol extract of Torreyae Semen significantly reduce the thickness of ear & back and gross skin condition compared to the control group in the mice with allergic contact dermatitis. As a result of H & E histological staining, it was observed that the Torreyae Semen extraction group had markedly reduced epidermis and skin thickness, granules, and keratinization compared to the control group. The groups administrated with Torreyae Semen hot water extracts had decreased TNF-α and IL-10 contents compared with the control group. As a result, Torreyae Semen may be used as a useful herbal resource because it has a certain level of antibacterial and anti-atopic effects.
The development of new antimicrobial agents is essential for the effective treatment of diseases such as sepsis. We previously developed a new short peptide, Pap12-6, using the 12 N-terminal residues of papiliocin, which showed potent and effective antimicrobial activity against multidrug-resistant Gram-negative bacteria. Here, we investigated the antimicrobial mechanism of Pap12-6 and a newly designed peptide, Pap12-7, in which the 12th Trp residue of Pap12-6 was replaced with Val to develop a potent peptide with high bacterial selectivity and a different antibacterial mechanism. Both peptides showed high antimicrobial activity against Gram-negative bacteria, including multidrug-resistant Gram-negative bacteria. In addition, the two peptides showed similar anti-inflammatory activity against lipopolysaccharide-stimulated RAW 264.7 cells, but Pap12-7 showed very low toxicities against sheep red blood cells and mammalian cells compared to that showed by Pap12-6. A calcein dye leakage assay, membrane depolarization, and confocal microscopy observations revealed that the two peptides with one single amino acid change have different mechanisms of antibacterial action: Pap12-6 directly targets the bacterial cell membrane, whereas Pap12-7 appears to penetrate the bacterial cell membrane and exert its activities in the cell. The therapeutic efficacy of Pap12-7 was further examined in a mouse model of sepsis, which increased the survival rate of septic mice. For the first time, we showed that both peptides showed anti-septic activity by reducing the infiltration of neutrophils and the production of inflammatory factors. Overall, these results indicate Pap12-7 as a novel non-toxic peptide with potent antibacterial and anti-septic activities via penetrating the cell membrane.

This thesis tried applying Ayurvedic hair and scalp care program and analyzing its effect. Ayurvedic treatment strengthens our immunity through natural healing power. I measured with MIC test and Halo test in order to examine the antibacterial efficacy against colonies which reside in head, when Ayurvedic base oils and essential oils were used on hair and scalp. Finally, I reached the results as follows. I examined the antibacterial efficacy of base oils and essential oils against Bacillus subtilis and Propionibacterium acnes, based on the MIC test with 9 kinds of base oil and 18 kinds of essential oil. By way of experiment, base oils did not show the antibacterial efficacy from all colonies, and in case of essential oils, such as Chamomile, Clary sage, Jasmine, Neroli and Rose oil, the antibacterial efficacy against two colonies did not appear by the measure of below 5%. It turned out that Thyme, Geranium, Lavender and Tea tree had an high effect on two colonies. According to the Halo test which experimented on 7 kinds of bacteria with the essential oils like Geranium, Lavender and Thyme and with the base oils like Sesame and Coconut, it went to prove that base oils was inefficient on antibiosis. It was examined that all of essential oils had the effect on antibiosis from 7 kinds of bacteria. Thyme showed the best efficacy of antibiosis in the MIC and the Halo test.
A study on quinolone antibacterial (pefloxacin methanesulfonate) was performed to use for the drug of fisheries. Petloxacin was proved excellent in antibacterial activity and resistance against fish pathogens when compared with the existing antibacterials. And any side effect was not observed during the period of indicated use. An outline of MIC(Minimal Inhibitory Concentration) was $1.6{\sim}6.4{\mu}g/ml$, $TLm_{48h}$ value were 380~420 ppm in Israeli carp(Cyprinus caprio) and 2100~2300 ppm in flounder(Paralichthys olivaceus). The residual time of fish body was less than 15 days. So we can treat some bacterial disease of fish by the dosage of 100 g/day/ton of fish body weight for 3 days and pefloxacin is thought to be used effectively and widely against most bacterial fish pathogens.
Combination therapy of antibiotics is leading to improved efficacy or safety profiles with decrease emergence of bacterial resistance. Because of this benefit, many of antibacterial combinations have been used in veterinary practice for the past few decades. The purpose of this study was to examine the in vitro activity of an amoxicillin alone and in combination with other antibiotics against pathogenic bacteria of fish origin. Based on the fractional inhibitory concentration (FIC) index (FIC $$\leq_-$$ 0.5), a synergistic interaction was shown in combination of florfenicol with amoxicillin or cefuroxime. The combination of florfenicol and amoxillin showed higher antibacterial activity than that of florfenicol and cefuroxime. Ratio of amoxicillin and florfenicol in combination was 1 : 1, which showed the antibacterial activity against bacterial isolates of fish as compared with other ratios. A synergetic effect of the combination (amoxicillin and florfenicol) was further confirmed in the time-kill curve study. The study showed a better in vitro antibacterial activity of a 1 : 1 combination of amoxicillin and florfenicol than the individual antibacterial against bacterial isolates of fish. In conclusion, the combination of florfenicol and amoxicillin may serve as a potential antibacterial therapy in fishes infected pathogenic bacteria.

Norfloxacin was proved excellent in antibacterial activity and sensitivity against fish bacterial pathogens when compared with the existing antibacterials and antibiotics. And any side effect was not observed during the period of indicated use. An outline of minimal inhibitory concentration was $0.03{\sim}0.1{\mu}g/ml$, $TLm_{48h}$ value was 3,500mg/l. The residual time of the day in fish body was less than 17 days and any pathological changes were not observed. The study has revealed that norfloxacin can be applied to treat some fish bacterial disease by the dosage of 100g/day/ton of fish body weight for about 3 days perorally. Further, norfloxacin may be used for the control of bacterial pathogens in eel.
The antibacterial efficacy of a few filling materials were studied in vitro. Zinc oxide eugenol cement. dycal, cavitec, hipol, cervident, clearfill and microfill were tested against Staphylococcus albus and Lactobacillus acidophilus on thioglycolate agar in order to determine whether the materials are effective inhibitors of bacterial growth. 1. Of these materials tested, zinc oxide eugenol cement was the most effective inhibitor of bacterial growth and revealed about 5.5mm in width of inhibition zone against Staphylococus albus and about 5.0mm against Lactobcillus acidophilus. 2. Dycal was moderate inhibitor of the both bact$\={e}$ria showing 2.5mm of the zone in width. 3. Cavitec was the slightest effect against the both bacteria 4. Composite resins tested in this study showed no evidence of inhibiting bacterial growth against the both bacteria.
Background: Florfenicol might be ineffective for treating Staphylococcus aureus small colony variants (SCVs) mastitis. Objectives: In this study, florfenicol-loaded chitosan (CS)-sodium tripolyphosphate (TPP) composite nanogels were prepared to allow targeted delivery to SCV infected sites. Methods: The formulation screening, the characteristics, in vitro release, antibacterial activity, therapeutic efficacy, and biosafety of the florfenicol composite nanogels were studied. Results: The optimized formulation was obtained when the CS and TPP were 10 and 5 mg/mL, respectively. The encapsulation efficiency, loading capacity, size, polydispersity index, and zeta potential of the optimized florfenicol composite nanogels were 87.3% ± 2.7%, 5.8% ± 1.4%, 280.3 ± 1.5 nm, 0.15 ± 0.03, and 36.3 ± 1.4 mv, respectively. Optical and scanning electron microscopy showed that spherical particles with a relatively uniform distribution and drugs might be incorporated in cross-linked polymeric networks. The in vitro release study showed that the florfenicol composite nanogels exhibited a biphasic pattern with the sustained release of 72.2% ± 1.8% at 48 h in pH 5.5 phosphate-buffered saline. The minimal inhibitory concentrations of commercial florfenicol solution and florfenicol composite nanogels against SCVs were 1 and 0.25 ㎍/mL, respectively. The time-killing curves and live-dead bacterial staining showed that the florfenicol composite nanogels were concentration-dependent. Furthermore, the florfenicol composite nanogels displayed good therapeutic efficacy against SCVs mastitis. Biological safety studies showed that the florfenicol composite nanogels might be a biocompatible preparation because of their non-toxic effects on the renal tissue and liver. Conclusions: Florfenicol composite nanogels might improve the treatment of SCV infections.
Lactobacillus plantarum and Leuconostoc mesenteroides are crucial functional starters and predominant isolates in a wide range of fermented foods, particularly kimchi, whose constituents exhibit bioactive properties. We previously developed a methodology using anion exchange resins to purify peptidyl compounds from Lb. plantarum LBP-K10. Antibacterial cultures of Lb. plantarum LBP-K10 were obtained from the respective cultures' supernatants and filtrates. However, conclusive evidence of the efficacy of kimchi filtrates in eradicating pathogenic bacteria is lacking. We aimed to simulate the potential effects of antibacterial filtrates that contained antibacterial compounds which were derived from cultures of Lb. plantarum LBP-K10. We acquired the kimchi filtrates using a combination of centrifugation and filtration methodologies, without the requirement for inoculation. The filtered liquid from Chinese cabbage kimchi, inoculated with Lb. plantarum LBP-K10 as a starter culture, and the non-inoculated liquid from Chinese cabbage kimchi (referred to as CCK and CCKRef, respectively) were were examined. CCK demonstrated greater inhibitory activity and a more significant bactericidal effect against the bacterial indicator strains. The minimum inhibitory concentration demonstrated comparable outcomes in tests against both Gram-positive and Gram-negative bacteria. This research offers a groundbreaking examination that displays the effectiveness of profiling peptidyl compounds within kimchi filtrates for curing bacterial infections.
