  • Purpose : In recent years, essential oils have been produced using natural extracts for various uses. Their functionality is currently being tested not only for cosmetics and perfumes but also for other categories of products. Therefore, this study verified their antibacterial effects on S. mutans, P. gingivalis, and L. gasseri which are the representative strains that cause oral diseases. Methods : Eighteen types of natural essential oils were made at a concentration of 50 % (v/v) using Tween 20, and their antibacterial effects were verified by applying S. mutans, P. gingivalis, and L. gasseri. The antibacterial effects were measured with the disc diffusion method. All the experiments were repeated three times, and the mean value of three measurement values for each variable was used for data analysis. A one-way analysis of variance was conducted using these mean values. Results : Of the eighteen types of essential oils tested, sixteen types showed antibacterial effects on S. mutans, and sixteen and fifteen types exhibited antibacterial effects on P. gingivalis and L. gasseri respectively. The types of essential oils with high-level antibacterial activities were geranium, may chang, and bergamot for S. mutans, lemongrass, bergamot, and eucalyptus for P. gingivalis, and lemongrass, machan, and geranium for L. gasseri in order of antibacterial effect. This result was statistically significant (p<.001). In addition, in the case of mandarin oil, it was found that there was no antibacterial activity in all three strains. Conclusion : This study proved the antibacterial activities of essential oils, which are used for various purposes in daily life, against dental caries and periodontal diseases. The study results will likely be applied to different prevention programs for oral health and broadly used to develop products such as oral care items and dentifrices.

Potential of Erythrosine-Mediated Photodynamic Therapy as a Cavity Disinfectant: Antibacterial Efficacy and Bonding Ability

  • This study aimed to evaluate the antibacterial efficacy of erythrosine-mediated photodynamic therapy (PDT) against Streptococcus mutans (S. mutans) in dentin and its effect on the shear bond strength (SBS) of composite resin to dentin. Eighty extracted human noncarious premolars were used in this study. Forty teeth were used for the antibacterial activity test, while the remaining 40 were used for the SBS test. Both experiments were conducted with 4 experimental groups (n = 10): control (distilled water), sodium hypochlorite (NaOCl, 6%), chlorhexidine (CHX, 0.12%), and erythrosine-mediated PDT. Antibacterial effects were evaluated by counting S. mutans colony-forming units (CFUs). The SBS of composite resins to dentin was measured using a universal testing machine. All treatments (NaOCl, CHX, and PDT) demonstrated statistically significant differences in antibacterial activity compared with the control group (p < 0.05). The antibacterial effects were ranked from strongest to weakest as follows: NaOCl, PDT, and CHX. In the SBS test, the NaOCl group exhibited a statistically significant difference compared with the CHX, PDT, and control groups (p < 0.05), with the lowest bond strength. No statistically significant differences were found among the CHX, PDT, and control groups (p > 0.05). Erythrosine-mediated PDT exhibited significant antibacterial effects against S. mutans, with higher antibacterial activity than CHX but lower than NaOCl. Only NaOCl negatively affected the bond strength of composite resin to dentin. In conclusion, erythrosine-mediated PDT shows potential as a cavity disinfectant due to its significant antibacterial effects against S. mutans and lack of adverse effects on bond strength.

Antioxidant and Antibacterial Effects of Mixed Extracts of Phyllanthus emblica, Geranium (Pelargonium graveolens) and Commiphora myrrha: Possibility of Natural Materials for Acne Treatment

  • Various skin diseases are occurring due to external factors such as urbanization and increase in environmental pollution and wearing masks due to COVID-19. Accordingly, various functional cosmetics are being released, but as some side effects are reported, research on functional cosmetics materials using natural plants is necessary. Therefore, in this study, the antioxidant, antibacterial and anti-inflammatory effects of Phyllanthus emblica, Geranium (Pelargonium graveolens), and Commiphora myrrha mixed extracts (PGC) that pharmacological efficacy has been verified were analyzed and their potential as functional cosmetics raw materials was examined. Four extracts (PGC-1~4) were prepared according to the extraction method. ABTS and DPPH radical scavenging activity experiments were conducted for the antioxidant efficacy of the extracts. In addition, paper disc experiments and LPS inflammation-inducing cytokine experiments were conducted to examine the antibacterial and anti-inflammatory effects. In addition, a cell viability test was performed to confirm cytotoxicity. As results of the study, all extracts showed antioxidant, antibacterial, and anti-inflammatory effects without cytotoxicity, and in particular, PGC-4, a fermentation and ultrasonic extract, showed the best efficacy. This means that the extraction yield of useful components varies depending on the extraction method.

연교김패전(連翹金貝煎)의 Escherichia coli에 대한시험관내 항균력 평가 (In Vitro Antibacterial Effects of Yeonkyokeumpae-jeon against Escherichia coli)

  • Objectives: The object of this study was to observe the in vitro antibacterial effects of Yeonkyokeumpae-jeon (YKKPJ) have been used for treating various gynecological diseases including mastitis in Korea, and individual six kinds of herbal composition aqueous extracts - Forsythiae Fructus (FF), Millettiae Caulis (MC), Lonicerae Flos (LF), Fritillaria Thunbergii Bulb (FT), Taraxci Herba (TH) and Prunellae Spica (PS) against E. coli. Methods: Antibacterial activities against E. coli of YKKPJ, FF, MC, LF, FT, TH and PS aqueous extracts were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.7 and MCF-7. The results were compared with ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts against E. coli were detected as $0.039{\pm}0.013mg/ml$, $0.064{\pm}0.033mg/ml$, $0.108{\pm}0.053mg/ml$, $0.078{\pm}0.027mg/ml$, $16.250{\pm}8.385mg/ml$, $15.625{\pm}9.375mg/ml$, $0.254{\pm}0.131mg/ml$, repectively. YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts showed antibacterial effects against to E. coli, except for FT and TH, which were showed negligible antibacterial effects, respectively. In addition, ciprofloxacin with YKKPJ, FF, MC, LF and PS aqueous extracts also showed marked dosage-dependent inhibition of bacterial growth, and favorable inhibitory effects on the both bacterial invasion and intracellular killing assays using MCF-7 and Raw 264.7 cells were detected in this experiment. Conclusions: The results obtained in this study suggest that traditional polyherbal formula YKKPJ aqueous extracts showed more favorable antibacterial activities as compared to individual six kinds of herbal composition aqueous extracts. The antibacterial effects of YKKPJ against E. coli considered as results of complicated synergic effects of their six kinds of herbal components rather than simple antibacterial effects of single herbal components. It means, YKKPJ aqueous extracts may show potent anti-infectious effects against E. coil for mastitis.

투농산(透膿散) 및 과루우방탕(瓜蔞牛蒡湯)의 Staphylococcus aureus에 대한 In Vitro 항균력 평가 (In Vitro Antibacterial Effects of Tunong-san and Kwaruwoobang-tang Aqueous Extracts against Staphylococcus aureus)

  • Objectives: Recently Ciprofloxacin, used in the treatment of mastitis, showed many serious side effects. The object of this study was to recognize whether TNS and KWT can be used in the treatment of mastitis by observing the in vitro antibacterial effects of TNS and KWT aqueous herbal extracts against S. aureus. Methods: Antibacterial activities of TNS and KWT aqueous extracts against S. aureus ATCC 25923 were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.6 and MCF-7. The results were compared with Ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of aqueous extracts of TNS and KWT against S. aureus were detected as ($0.313{\pm}0.107$) and ($0.137{\pm}0.053$) mg/ml, respectively. MIC of Ciprofloxacin was detected as ($0.469{\pm}0.297$) ${\mu}g/ml$ at same conditions. In addition, TNS, KWT aqueous herbal extracts and Ciprofloxacin were also showed marked dosage-dependent inhibition of bacterial growth, and dramatical inhibitions on the both intracellular killing assays and bacterial invasion using Raw 264.6 and MCF-7 cells were detected. Conclusions: The results obtained in this study suggest that TNS and KWT aqueous herbal extracts showed antibacterial effects against S. aureus, and they also showed dosage-dependent inhibitory effects on the bacterial growth. And they showed the significant intracellular killing and bacterial invasion effects. It means, KWT and TNS may show more potent anti-infectious effects against S. aureus in vivo.

농산물용 기능성 골판지 제조를 위한 신규 항균재료 개발에 대한 연구 (Development of new antibacterial materials for manufacturing functional corrugated board for agricultural products)

  • In this study, new antibacterial materials were developed to manufacture a functional corrugated board. Sulfur solution, a new antibacterial solution made from inorganic sulfur in the laboratory, and other antibacterial mat erials were adopted to treat the surface of a linerboard. We measured the antibacteriocidal and bacteriostatic activities, as well as the fungal resistance of the surface-treated linerboards, to identify the antibacterial properties. The mechanical properties of the surface-treated linerboard were also determined in order to identify the effects of the antibacterial materials on linerboard properties. Linerboard treated with sulfur solution, PVOH, and sodium metasulfite showed the highest antibacterial activity, while linerboard treated with sulfur solution and nano sulfur showed the highest fungal resistance. It was identified that sulfur solution has effective antibacterial properties. The antibacterial materials did not affect the mechanical properties of the surface-treated linerboard, but the binder showed significant effects in terms of the burst strength, the compressive strength, and the stiffness of the linerboard.

Effects of Storage Temperature and pH on the Stability of Antibacterial Effectiveness of Garlic Extract against Escherichia coli B34

  • The effect of long-term storage on garlic antibacterial activity was investigated. A concentration of 5% or more garlic was found to be necessary to completely inhibit Eschrichia coli growth in tryptic soy broth. This value is substantially higher than the minimum inhibitory concentration of 1% for E. coli reported previously. pH-modified garlic extract was stored at different temperatures to investigate the impact of storage conditions (i.e., temperature, pH, period of storage) on the stability of the antibacterial activity of the garlic extract used against E. coli B34. The antibacterial effectiveness of the garlic extract against E. coli remained stable when both the storage temperature and the pH of the extract were kept low. When the garlic extract was stored at $40^{\circ}C and above, most or all of the garlic antibacterial activity disappeared after a 24-h storage period, regardless of the storage pH. The antibacterial activity was weakened when the pH of the garlic extract was adjusted to 8, and at low temperatures.

Antibacterial Activity and Synergism of Hydnocarpi Semen Extracts with Ampicillin or Oxacillin against Methicillin-resistant Staphylococcus aureus

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a serious clinical and an urgent problem worldwide. Few new drugs are available against MRSA, because MRSA has the ability to acquire resistance to most antibiotics, which consequently increases the cost of medication. In the present study, the antibacterial activity of Hydnocarpi Semen was investigated. The most effective method is to develop antibiotics from the natural products without having any toxic or side effects. Therefore, there is a need to develop alternative antibacterial drugs for the treatment of infectious diseases. Five Clinical isolates (MRSA) were obtained from five different patients at Wonkwang University Hospital (Iksan, South Korea). The Other 2 strains were ATCC 33591 (Methicillin-resistant strain) and ATCC 25923 (Methicillin-susceptible strain). Antibacterial activity (Minimal Inhibitory Concentrations, MICs) was determined by broth dilution method, disk diffusion method, MTT test, and checkerboard dilution test. Antibacterial activity of n-hexane fraction was remarkable, and had a MICs ranging from $31.25-125{\mu}g/m{\ell}$. FICI values for HFH+AM and HFH+OX were 0.13-0.19 and 0.04-0.29, showing the increase of synergistic effect. When combined together, these antibacterial effects were dramatically increased.

Antibacterial Phytosterols and Alkaloids from Lycoris radiata

  • This research aimed to investigate the antibacterial activity of Lycoris radiata. The methanol extract and solvent fractions from L. radiata exhibited antibacterial activities against Escherichia coli, Staphylococcus aureus, and Helicobactor pylori. Open-column chromatography was used to isolate phytochemical constituents from L. radiata; spectroscopic analysis elucidated their structures as ${\beta}$-sitosterol (1), daucosterol (2), O-methyllycorenine (3), lycorenine (4), lycoricidinol (5), lycorine (6), and lycoricidine (7). Further testing of compounds 1 - 7 revealed antibacterial effects against E. coli, S. aureus, and H. pylori, which suggested the potential of these substances as antibacterial agents. We determined that compounds 1 and 2, isolated from the n-hexane fraction, were more effective against S. aureus and H. pylori. Compound 4, isolated from the methylene chloride fraction, exhibited noticeable antibacterial effects against E. coli. This study is the first report on the antibacterial activities of phytochemical constituents from L. radiata against E. coli, S. aureus, and H. pylori.

광촉매를 이용한 셀룰로오스섬유의 기능화에 관한 연구 - 항균·소취성 및 자외선 차폐성을 중심으로 - (The Functional Properties of Cellulose Fabric Treated with TiO2 - Focusing on Antibacterial activity, Deodorization & UV cut ability -)

  • In this study, we measured the antibacterial activities, deodorization, UV cut ability, whiteness and SEM, according to the size($5{\mu}m$, $15{\mu}m$) of $TiO_2$, concentration(3%, 5%, 10%) and dipping temperature($50^{\circ}C$, $70^{\circ}C$) with using anatase type of $TiO_2$ photocatalyst. Photocatalyst is the substance which carries out functions, such as decomposition, removal, deodorization, antibacterial, etc. of a contaminant, in a place with light based on an oxidation-reduction reaction. The results of this study were as follow. Antibacterial activities are increased with increasing of the $TiO_2$'s concentration, and $TiO_2$ has high antibacterial activities for Staphylococcus aureus but it has low antibacterial activities for Klebsiella pneumoniae. The deodorization and UV cut ability is very good, therefore be able to get good effects with using only 3% of $TiO_2$. Every effects are increased by using small size of $TiO_2$ and high dipping temperature.
