본 연구는 백량금 추출물의 화장품 원료로서의 특성을 알아보기 위하여 항산화, 미백 및 항염 효과와 관련된 다양한 실험을 실시하였다. 70% 메탄을 추출물을 대상으로 실험한 결과 1,1-diphenyl-2-picryl-hydrazyl (DPPH) 라디칼 소거 효과는 0.01% 이상의 농도에서 90%의 효과를 나타내었고, 세포내 멜라닌 생성 억제 효과는 0.05% 이상의 농도에서 50% 이상의 효과를 나타내었다 이 후 70% 메탄을 추출물을 대상으로 MPLC를 사용하여 성분 분리 실험을 실시하여 5개의 분획들을 얻었고, 이들을 대상으로 실험한 결과 3번, 4번 그리고 5번 분획들에서 항산화(DPPH 라디칼 소거, Mn-SOD 생성 억제), 미백(멜라닌 생성 억제)그리고 항염($IL-1{\alpha}$, IL-6, COX-2, total NO 생성 억제)효과를 나타내었다.
The flowers and buds of Lonicera Flower (LF), are used in Korean herbal medicine for latent-heat-clearing, antipyretic, detoxicant and anti-inflammatory ailments. This plant is used worldwide for the treatment of many types of inflammatory disease including respiratory infections, diabetes mellitus, rheumatoid arthritis and play an important role in immune reaction. These pharmaceutical effects of LF looks like to be related to its antioxidant capacity and phytochemicals containing in LF. In this study, the antioxidant activity of extract from LF was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and the inhibitory effect on collagen induced platelet aggregation. The LF extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on platelet aggregation. In conclusion, the LF extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.
Objectives The object of this study was to assess the effect of Gyejibokryunghwan (GBH) on anti-oxidant and anti-inflammatory activities in RAW 264.7 cells and on factors associated with fracture union in tibia-fractured rats. Methods The 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity was measured to assess anti-oxidant activity. The production of nitric oxide (NO), interleukin-6 (IL-6), interleukin-$1{\beta}$ ($IL-1{\beta}$) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) in the RAW 264.7 cells were measured to assess anti-inflammatory activity. The production of osteocalcin, calcitonin, carboxy-terminal telepeptides of type II collagen (CTXII), transforming growth factor-${\beta}$ ($TGF-{\beta}$), bone morphogenetic protein-2 (BMP-2) in serum of tibia-fractured rats were measured to assess the effects of fracture union. X-rays were taken every two weeks from 0 to 4th week to assess fracture union effect. Results DPPH radical scavenging activity of GBH was increased according to concentration of GBH in RAW 264.7 cell. NO, prostaglandin $E_2$ ($PGE_2$), IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ were significantly decreased, indicating anti-inflammatory effect. Osteocalcin, calcitonin, $TGF-{\beta}$ were significantly increased in the experimental groups. CTXII was significantly decreased in the experimental groups. BMP-2 was not significantly changed in the experimental groups. The X-ray showed that the experimental group has better healing effects on tibia-fractured rats than control group. Conclusions From above result, GBH has an effect on anti-oxidant, anti-inflammatory activities in RAW 264.7 cells. GBH showed significant results in factors related with fracture union and radiologic examination. In conclusion, GBH can help fracture union and it well be expected to be used actively in clinics.
Objectives : Antioxidant, Anticancer and Antibacterial Activities of Naesohwangryntang and its composition oriental medicines. Methods : We were experimented anti-oxidation effect and growth inhibition ability on cancer cells and antibacterial activity on various kinds of bacteria of skin. Results : The results were obtained as follows : Electron donating ability(EDA) of water extract Naesohwangryntang and ethanol extract Naesohwangryntang was 60% and 70% at 1000 ppm concentration. In the test of SOD-like activity, ethanol extract showed more activity with 27.4% in 700 ppm, while water extract was low in 19.6%. Clear zones formed by sample against the human skin-resident microflora indicated that anti-microbial activity of ethanol extract Naesohwangryntang was higher than that of water extract Naesohwangryntang. The growth inhibition rates of each sample on lung-cancer(A549), at 1000 ppm cancer cell was over 40%. The growth inhibition rate of the each sample melanoma-cancer(B16F10, G361), at 1000 ppm was over 80%. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidation and antibacterial effect is useful to be applied in cosmetic industry.
There is currently increased interest in the identification of antioxidant compounds that are pharmacologically potent and have low or no side effects. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. FARFARE FLOS has been frequently used on the respiratory system including bronchitis, phthisis. In this study, the antioxidant activity of extract from FF was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on Cu2+-induced human LDL oxidation. The FF extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation. And this study was designed to evaluate whether FFEA may ameliorate oxidative stress and inflammatory status through the antioxidative mechanism in LPS-stimulated RAW 264.7 murine macrophage cell line. Treatment of RAW 264.7 cells with FFEA significantly reduced LPS-stimulated inflammatory response in a dose-dependent manner. In conclusion, the FF extracts have anti-oxidative and anti-inflammatory effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. Potatoes have a short growing season, high production per unit area, relatively strong environmental adaptability, and are cultivated in more than 130 countries around the world. It is the world's fourth-largest crop, following rice, wheat, bean and maize. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by cell based anti-oxidation assay. The smallest amount of ROS(Reactive oxygen species) was generated when the compound was derived from 'Haryung' and 'hongyoung' and strong SOD(Superoxide dismutase) activity was observed in 'Sumi' and 'Jayoung'. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers
Objectives : This paper aimed to verify the applicability of mixed extract ofAngelica gigasNakai,Cnidium officinaleMakino,Paeoniala ctifloraPall,Rechmannia glutinosaLibosch,Scutellaria baicalensisGeorgi, which were prescribed for improving inflammation in Donguibogam, as the materials for beauty food and functional medicinal herb cosmetics by manufacturing such mixed extract and evaluating the biological activity of the extract.Methods : The mixed medicinal herb water extract(MMW) and ethanol extract(MME) were freeze-dried to be used as the specimen. We performed electron donating ability, lipid acidification inhibitory activity, anti-inflammatory activity against skin flora, MTT assay, NO inhibitory activity and the protein expression inhibitory activity of iNOS and COX-2.Results : For anti-oxidation experimentation, the electron donating abilities of MMW and MME were above 60.0% and 90.0% at 500 μg/ml, respectively. In the inhibition rate of lipid peroxidation, MMW and MME showed 43.1% and 52.1% at 1,000 μg/ml, respectively. As a result of antimicrobial activity, both the MMW and MME showed significant clear zones forPropionibacterium acnesat 4 mg/disc, but did not indicated the clearzones forStaphylococcus aureus, Escherichia coliandStaphylococcus epidermidis. Anti-inflammatory activity by NO assay showed LPS-induced NO was significantly inhibited in a concentration-dependent manner. Also, the expression of iNOS and COX-2 proteins were significantly inhibited following treatment with MMW and MME of 50 μg/ml.Conclusions : Accordingly, it can be concluded that mixed medicinal herb extract has the potential to beused as a functional food and cosmetic material.
본 연구에서는 홍차 흑효모 발효물의 기능성화장품 소재로서의 가능성을 규명하고자 항산화, 미백, 항주름 효능을 확인하였다. 홍차 흑효모 발효물의 결과는 홍차 추출물, 흑효모 발효물과 비교하여 나타내었다. 항산화능 평가 결과 홍차 흑효모 발효물은 흑효모 발효물보다 뛰어난 DPPH 라디칼 소거능을 나타냈으며 홍차 추출물과는 유사한 결과를 나타냈다. 과산화수소로 유도된 세포 내 활성산소종 실험에서도 홍차 흑효모 발효물은 유의적 제거 활성을 보였다. 또한 홍차 흑효모 발효물은 tyrosinase 저해 활성을 보이며 Melan-a, B16F10 세포를 이용한 멜라닌 형성이 억제됨을 확인하였다. 항주름 효능 확인을 위해 콜라겐 분해 효소(MMP-1)를 측정한 결과 홍차 흑효모 발효물이 홍차 추출물, 흑효모 발효물보다 높은 항주름 활성을 보였다. 본 연구 결과를 통해 홍차 흑효모 발효물은 항산화능 효능을 보이며 미백, 항주름 효능을 보아 기능성 화장품 소재로 개발될 수 있음을 확인하였다.
고추에서 추출한 수용성 조다당(CAP-0)에 높은 항보체 활성이 존재함을 발견하였으며, 활성은 pronase 처리에 의해 변하지 않았으나 periodate 산화에 의해 활성이 현저히 감소하였다. CAP-0는 cetyltrimethylammonium bromide 처리에 의해 4획분(CAP-1,2,3,4)으로 분리되었으며, CAP-1에서 가장 높은 황보체 활성이 나타났다. CAP-1은 음이온 교환 chromatography에 의해 활성과 수율이 높은 CAP-1-IV와 V로 분리되었으며, III은 수율은 높았으나 활성이 매우 낮았다. CAP-1-III와 IV는 gel 여과에 의해 고분자량 획분$(M.W.\;III_a\;70,000,\;IV_a\;195,000,\;V\;140,000)$과 저분자량 획분 $III_a,\;IV_b$으로 나뉘어졌다. 이 4획분과 V는 gel 여과와 전기영동상에서 순수하였으며 고분자량 획분에서 항보체 활성이 높았다.
Objectives: Bletillae Rhizoma, the roots of Bletilla striata, is used to restrain the leakage of blood and stop bleeding. It can cure the sores, ulcers, and chapped skin. This study was designed to investigate the collagen metabolism, elastase and tyrosinase activity of Bletillae Rhizoma extract (BR). Methods : The effects of BR on type I procollagen production and collagenase activity in human normal fibroblasts Hs68 after UVB (312 nm) irradiation were measured by ELISA method. The elastase activity, tyrosinase activity, and L-DOPA oxidation after treatment of BR were measured as well. Results : In the present study, the collagen production (type I procollagen) was significantly increased to $15.7{\pm}1.8$ ng/ml at a concentration of BR 100 ${\mu}g/ml$ in UVB damaged Hs68 cells. The increased collagenase activity after UVB damage was significantly recovered to $42.7{\pm}0.7%$, $54.5{\pm}3.5%$, and $38.4{\pm}0.9%$ by BR 10, 30, and 100 ${\mu}g/ml$. The activities of BR 10 mg/ml on tyrosinase activity was significantly reduced to $45.1{\pm}8.4%$ as well. However, there were no significant effects on the elastase activity and the L-DOPA oxidation. Conclusion : BR showed the promoting effects of collagen synthesis and inhibitory effects of collagenase activity in Hs68, human normal fibroblast cells. And these could be thought to have the anti-wrinkle effects and whitening effects in vitro. These results suggest that BR may have potential as an anti-aging ingredient in cosmetic treatment.
