의이인탕 구성약물의 비만관련 연구 분석 (Analysis of the Obesity-Related Research for Each Constituent Herb of Euiiin-tang)

  • The purpose of this study is analyzed of exploratory research potential as anti-obesity agents of Euiiin-tang. Four Korean databases and 2 Korean Journals (Riss4U, KISS, OASIS, DBPIA, and Journal of Korean Rehabilitation Medicine, Journal of Korean Medicine of Obesity Research) were searched using search word 'individual herbs' and 'obesity', 'weight loss', 'fat', 'hypertension', 'hyperlipidemia', 'diabetes'. Clinical and Experimental Research published in the journal were analyzed, review research, studies of pharmacopunctures and studies of mixed herbal medicine were excluded. We collected 23 studies. Seven studies of Coicis Semen, 10 stdies of Ephedra Herba, 2 study of Angelica gigas Nakai, 3 studies Atractylodis Rhizoma Alba, 1 studies of Glycyrrhiza inflata Batal. Most studies were experiment researches which were composed of in vivo or in vitro, and clinical trial was 5 studies of Ephedra Herba. Main constituent herb, Coicis Semen, Ephedra Herba were thought to represent an anti-obesity effect. Through the result, we can assume to be likely effect of Euiiin-tang as obesity medicine.

고지방식이로 유도된 C57BL/6J 마우스 비만모델에서 참마홍국발효의 비만 억제 및 지질 저하 효과 (Anti-Obesity and Lipid Lowering Effect of Discorea japonica Thunb. Fermented with Monascus in High-Fat Diet Induced Obese C57BL/6J Mice Model)

  • This study was conducted to investigate the anti-obesity effect of Discorea Japonica Thunb. fermented with Monascus After inducing obesity by feeding hight fat diet (diet induced obesity model: DIO) for four weeks, each 8 rats were assigned to normal (Nor), high fat diet (HF), high fat diet containing orlistat (PC), high fat diet containing different concentration of Discorea Japonica Thunb. fermented with Monascus (UPDM_L, UPDM_H) and Discorea Japonica Thunb. (UPD) extract. Although the UPD, UPDM_L (ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 400 mg/kg) and UPDM_H (DIO oral administration ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 800 mg/kg) showed weight gain inhibition effects, the results of poor obesity inhibition rather than PC were confirmed. However, it showed a more effective weight loss effect in UPDM_H than UPD, and significantly reduced the weight of epididymal fat and subcutaneous fat. Furthermore, the possibility of anti-obesity effects of Discorea Japonica Thunb. fermented with Monascus can be confirmed by observing the effects of reducing serum total cholesterol, triglyceride and LDL concentrations, reducing ALT and AST levels, and inhibiting fat build-up in liver tissue. It is believed that Discorea Japonica Thunb. fermented with Monascus can be expected to utilize as a functional material that is important to improve anti-obesity and metabolic syndrome.

Inhibition of Adenovirus 36 Replication and Lipid Accumulation by Distylium racemosum

  • Obesity is a worldwide disease and one of the major risk factors. Virus among many factors can lead to obesity. Adenovirus 36 (Ad-36) is the adipogenic virus linked with human obesity. Nevertheless, there is no drug to treat both Ad-36 infection and obesity associated with virus. For the precedent study on anti-cholesterol test, Distylium racemosum (D. racemosum), Quercus salicina (Q. salicina) and Raphiolepis indica (R. indica) were selected. This study was carried out to evaluate the anti-cholesterol effects, anti-lipid effects and inhibition of Ad-36 replication from three extracts. D. racemosum ($50{\mu}g/mL$) inhibited lipid accumulation on 3T3-L1 adipocyte. D. racemosum inhibited adipocyte differentiation through suppression of regulator peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR{\gamma}$) genes and adipocyte-specific genes such as adipocyte protein 2 (aP2). D. racemosum inhibited replication of Ad-36 at $50{\mu}g/mL$ of concentration. Therefore, the extract of D. racemosum could be a candidate for development of anti-Ad-36 and anti-obesity drugs.

고수 에탄올 추출물의 고지방식이 비만 동물모델에서의 항비만효과 (Ant-Obesity Effect of Coriandrum sativum L. Ethanol Extract in High Fat-Induced Obesity Animal Model)

  • This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.

  • Objectives : Obesity is a public health concern associated with chronic diseases including hyperlipidemia, diabetes, fatty liver, atherosclerosis and cancer. As several anti-obesity drugs have been limited owing to their side effects, the development of new anti-obesity drugs through herbal medicines has been increasing. Cynanchum Wilfordii Radix (CW) traditionally is consumed for various health benefits including immune enhancing, anti-inflammation and anti-tumor activities. The aim of the present study is to evaluate the effects of CW on High fat diet (HFD)-induced obese mice. Methods : The mice were randomly divided into four groups (n=7). The mice were respectively fed a normal diet (ND), a high-fat diet (HFD), HFD plus CW (50 mg/kg/day), HFD plus CW (100 mg/kg/day). All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : HFD-fed mice showed an increase in the body weight and serum biochemistry parameters levels (total cholesterol and triglycerides) as well as organic tissue weights. However, the administration of CW to obese mice induced a reduction in their body weight, food efficiency ratio, blood biochemistry parameters and weight of liver and fat compared with the HFD fed mice. Additionally, we observed that CW inhibited the lipid accumulation in liver and serum lipid parameter induced by HFD. Conclusions : Taken together, the findings of this study suggest that CW may be a potential agent for use in the treatment of obesity and obesity-related metabolic diseases.

몽골산 약용식물 수종의 항비만 활성 탐색을 위한 기초 연구 (Basic research for exploring anti-obesity activity of several medicinal plants from Mongolia)

  • Objective : Obesity in modern society has a significant impact on pathological, psychological and social problems. Therefore, many studies on obesity treatment using herbal extracts with fewer side effects have been conducted. This study was designed to investigate the effect of inhibiting fat accumulation in vitro in order to Mongolian medicinal plants find anti-obesity candidate among. Methods : We measured pancreatic lipase inhibitory activity in Mongolian medicinal plants extract. Cytotoxicity of these extracts was monitored in 3T3-L1 cells by MTT assay. In addition, the anti-obesity effects was confirmed at concentrations of 0.2, 0.1 and 0.05 mg/㎖ through Oil red O staining. Results : Among Mongolian medicinal plants, Rheum undulatum roots in September (RURS), Paeonia anomala L. (PAL), and Fragaria orientalis (FO) showed the highest pancreatic lipase inhibitory activity. As a result of the MTT assay, more than 80% was judged to be non-toxic, and the concentration was determined, and as a result of evaluating the lipid accumulation inhibitory effect, 6 types were selected as candidates. Conclusion : Based on these results, the top 7 species expected to be used as anti-obesity functional materials were selected. However, additional efficacy verification and mechanism of action need to be established in the future. So, it is expected that the medicinal plants verified through this will be used as functional materials for the prevention and treatment of obesity.

Anti-Obesity and Inhibitory Effect of Lipid Accumulation of The Cone of Pinus rigida × Pinus taeda in 3T3-L1 Cells

  • With the COVID-19 pandemic, there is increasing interest in anti-obesity strategies. According to the National Statistical Office, the obesity rate in Korea was 38.3% in 2020 and 37.1% in 2021. Obesity is a risk factor for several severe diseases, including stroke, heart disease, type 2 diabetes, and certain types of cancer. Pinus rigida × Pinus taeda is a hybrid of Pinus rigida Mill and Pinus taeda Linn, and its cones are considered a by-product. Although previous studies have investigated their pharmacological effects on antioxidant activity and protection against oxidative DNA damage, few researchers have explored their potential as functional natural materials. Therefore, we evaluated the anti-obesity effects of the cone of ethyl acetate fraction of P. rigida × P. taeda (ERT), specifically its ability to inhibit lipid accumulation. Our analysis showed that ERT contains phytochemicals (catechin and caffeic acid) which are known to improve immune function and inhibit cell damage. ERT inhibited lipid droplet accumulation at the cellular levels through Oil Red O staining. Furthermore, ERT suppressed the expression of adipogenic transcription factors (PPARγ and CEBP/α) as well as downstream lipogenic target genes (FAS and SREBP-1) thereby inhibiting adipogenesis. ERT also down-regulated key adipogenic markers, including aP2α, while inducing the phosphorylation of AMPK. It has been reported that PPARγ and CEBP/α are expressed in the early stages of adipose differentiation, while SREBP-1 is expressed in the late stage. Therefore, our findings suggest that ERT activates AMPK signaling pathways, which inhibits adipogenic transcription factors (PPARγ, C/EBPα, and SREBP1) and lipogenic genes (FAS and aP2α), thereby blocking lipid accumulation and preventing obesity and related disorders. ERT showed potential as a new resource for developing a functional material for anti-obesity agents.

Anti-obesity Effects of Safflower Seeds (SS) on the Differentiation of 3T 3-L1 Pre-adipocytes and Obese Mice Fed a High-fat Diet

  • Safflower seeds, classified as a member of the Asteraceae family, a dicotyledonous plant, contain linoleic acid as a major component, known for its pharmacological effect of strengthening bones. Additionally, safflower seeds have been reported to have pharmacological effects on vascular diseases such as atherosclerosis. In this study, we investigated the anti-obesity effect of safflower seed extract by examining its impact on adipocyte differentiation using Oil Red O staining, triglyceride quantification, and GPDH activity measurement. The results showed that safflower seed extract significantly inhibited adipocyte differentiation. Furthermore, we confirmed that safflower seed extract improved body weight, liver weight, adipose tissue size, glucose, and triglyceride levels in a high-fat diet-induced mouse model. These findings suggest that safflower seed extract exhibits potent anti-obesity activity both in vitro and in vivo and has the potential to be developed as a material for future anti-obesity therapies.

  • Obesity increases the risk of developing metabolic diseases such as hypertension, type 2 diabetes, hyperlipidemia, and cardiovascular diseases, as well as some cancers. To prevent the occurrence of these diseases and death, it is essential to manage obesity. Though there are several treatments for obesity, lifestyle interventions, such as diet and exercise, and drug therapy are most widely used in clinical practice. Among the anti-obesity drugs available, the weight loss effect of naltrexone/bupropion has been well-proven. We present a case study in which naltrexone/bupropion, a glucagon-like peptide-1 agonist, and a sodium-glucose transporter 2 inhibitor showed significant weight loss and improved metabolic parameters. Additionally, the management of type 2 diabetes and hypertension, which are common diseases in patients with obesity, was also included.

Anti-obesity Effect of Black Vinegar Fermented with Herbal Extracts

  • Vinegar has been widely produced for a variety of industrial and domestic use as well as medicinal use. For sale of the commercial vinegar with herbal extracts, we produced an experimental black vinegar through sequential fermentation of alcohol, followed by acetic acid according to the manufacturer's procedure. To investigate the effect of anti-obesity of black vinegar on biochemical values, we evaluated enzyme activities via acetyl-CoA carboxylase (ACC), which plays a critical role in the lipid metabolism. We found that increased phosphorylated adenosine monophosphate (AMP) activated protein kinase (AMPK) and ACC in L6 mouse muscle cells treated with the manufactured vinegar. Based on the results, supplementation of experimental herbal black vinegar inactivates ACC, enhancing the phosphorylation of AMPK. Thus, the lipid oxidation and inhibitory effect of fatty acid synthesis by the black vinegar expects to facilitate the anti-obesity activity.
