HaCaT 세포주에서 캐모마일 (Matricaria chamomilla) 추출물의 항병원 성 및 항염 효과에 관한 연구 (Study of Anti-microbial Activities and Anti-inflammatory Effects of Chamomile (Matricaria chamomilla) Extracts in HaCaT cells)

  • Chamomile (Matricaria chamomilla), a member of the Asteraceae family, is a well-known for medicinal plant and can be found in India and Europe. Chamomile is an effective sedative and various medical effects. But, the effects of acne treatment by chamomile were not investigated. Therefore, we assessed the anti-oxidant effects, anti-microbial activity and anti-inflammatory effects by chamomile extracts in HaCaT keratinocyte cells. Anti-oxidant effects of chamomile extracts were investigated by DPPH assay. Also, results of MTT assay was demonstrated that chamomile extracts did not have a cytotoxic effect in HaCaT cells. To assess the antimicrobial activity, we determined formation of inhibition zone of Propionibacterium acnes by extracts from chamomile. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) induces production of inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6 and IL-8 and expression of COX-2. Chamomile extracts could inhibit TNF-${\alpha}$-induced mRNA expression levels of IL-$1{\beta}$, IL-6, IL-8 and COX-2 gene. These results demonstrated the possibility of chamomile for prevention and treatment of skin inflammatory diseases such as acne.

비방탈명산(秘方奪命散)의 항산화 ${\cdot}$ 항암 ${\cdot}$ 항균 효과 연구 (Study on the Anti-oxidative, Anti-microbial and Anti-cancer effect of Bibangtalmyungsan.)

  • Objectives : This study was carried out to evaluate anti-oxidative, anti-microbial and anti-cancer effect for clinical application of Bibangtalmyungsan (BTS) Results : 1. The oxidative effects were measured by polyphenol, DPPH radical scavenging activity. BTS water extract was showed more effective than ethanol extract and also various solvent fractions from BTS water extract showed effective in the following order : ethyl acetate fraction > butanol fraction > hexane fraction ${\fallingdotseq}$ chloroform fraction ${\fallingdotseq}$ aqueous fraction 2. The results of anti-microbial effects were as follows. 1) Antibacterial activities of BTS extracts against Gram's negative and positive bacteria were ineffective 2) Antifungal activities of the BTS extracts against Aspergillus spp. , Trichohyton mentagrophyte KTCC 1077 were not effective. 3. The result of anti-cancer effects were as followings: 1) BTS ethanol extract was more effective than water extract against Caco-2, Calu-6, but it had cytotoxic effect against NIH3T3. 2) On the Caco-2, effective only in ethyl acetate $fraction(IC_{50}:$ 35.81 ${\mu}g/ml)$. 3) On the Calu-6, the most effective in ethyl acetate $fraction(IC_{50}:$ 189.65 ${\mu}g/ml)$ and effective In butanol $fraction(IC_{50}:$ 299.74 ${\mu}g/ml)$ and hexane $fraction(IC_{50}:$ 345.13 ${\mu}g/ml)$ 4) On the SUN-601, $IC_{50}$ value was within 80 ${\mu}g/ml$ in ethyl acetate fraction and hexane fraction. 5) On the HCT-1l6, the most effective in ethyl acetate $fraction(IC_{50}:$ 82.94 ${\mu}g/ml)$ and effective in hexane $fraction(IC_{50}:$ 374.56 ${\mu}g/ml)$. 6) On the AML-2/WT, the most effective in ethyl acetate $fraction(IC_{50}:$ 41.44 ${\mu}g/ml)$ and effective In hexane $fraction(IC_{50}:$ 303.01 ${\mu}g/ml)$. 7) On the NIH3T3, effective only in ethyl acetate $fraction(IC_{50}:$ 203.42 ${\mu}g/ml)$, but it was more ineffective than other cancer cells. Conclusion : These result suggest that BTS has antioxidative, antifungal activities and cytotoxic effects against Caco-2, Calu-6, SUN-601, HCT-116, AML-2/WT and NIH3T3, especially ethyl acetate fraction from water extract has more effective in antioxidative and anticancer effects.

복분자주 주박의 항균, 항산화 및 항혈전 활성 (Anti-microbial, Anti-oxidant, and Anti-thrombosis Activities of the Lees of Bokbunja Wine (Rubus coreanus Miquel))

  • 복분자는 당도가 낮고 신맛이 강해 과실주로 주로 제조되고 있으며, 복분자주 제조 후 부생되는 주박은 대부분 폐기되고 있다. 본 연구에서는 현재까지 생리활성이 보고된 바 없는 복분자주 주박을 대상으로 열수 추출물, ethanol 추출물 및 이들의 다양한 분획물을 조제하여 항균, 항산화 및 항혈전 활성을 평가하였다. 그 결과, 복분자주 주박의 ethylacetate (EA) 분획은 413~459 mg/g의 높은 polyphenol을 함유하고 있으며, 그람양성세균에 대한 강력한 성장억제 활성과, DPPH 음이온, ABTS 양이온에 대한 우수한 radical 소거능, nitrite 소거능 및 환원력을 나타내었다. 또한 혈액내의 thrombin, prothrombin, coagulation factor 에 대한 저해를 통한 혈액응고저해 활성이 우수하였다. 복분자주 주박의 butanol 분획은 우수한 항산화력과 함께 내인성 혈전 생성에 관련된 aPTT 연장활성을 나타내었으며, aspirin에 필적하는 강력한 혈소판 응집저해 활성을 나타내었다. 한편 복분자주 주박의 열수 추출물의 순차적 유기용매 분획 후의 물 잔류물은 aspirin보다 강력한 혈소판 응집저해를 나타내어 새로운 항혈소판제 개발 소재로 이용 가능함을 확인하였다. 본 연구결과는 폐기되고 있는 복분자주 주박을 이용한 고부가가치 식품 및 향장소재 개발이 가능함을 제시하고 있다.

고구마 소주 주박의 항균 및 항혈전 활성 (Anti-microbial and Anti-thrombosis Activities of Lees of Sweet Potato Soju)

  • 고구마 소주는 전분과 섬유질이 많은 고구마를 발효시킨 후 상압증류하여 제조되어 특유의 부드러운 맛과 향미를 가진다. 본 연구에서는 별도의 용도가 없어 폐기되고 있는 고구마 소주 주박의 효율적인 이용을 위해, 주박의 ethanol 및 열수 추출물을 조제하고 이들을 순차적 유기용매 분획하여 각각의 hexane, ethylacetate (EA), butanol 분획물과 물 잔류물을 조제하여 각 시료의 항균 및 항혈전 활성을 평가하였다. 그 결과, 주박의 ethanol 추출 효율이 열수 추출 효율보다 1.36배 높았으며, 분획물 중에서는 EA 분획물이 가장 높은 total polyphenol 함량을 나타냄을 확인하였다. 고구마 소주 주박의 추출물 및 분획물(0.5 mg/ml)은 인간 적혈구에 대한 용혈활성을 나타내지 않았으며, 열수 및 ethanol 추출물의 EA 분획물에서만 광범위 항세균 활성과 강력한 혈액응고 저해 활성이 나타났다. 또한 혈소판 응집저해 활성의 경우 열수 추출물의 EA 분획에서만 아스피린과 유사한 응집저해를 나타내었다. 본 연구결과는 폐기되고 있는 고구마 소주 주박의 열수 추출물의 EA 분획이 신규의 항세균 및 항혈전제로 개발될 수 있음을 제시하고 있다.

미생물 성장을 억제하기 위하여 수용성 절삭유에 과다하게 첨가한 붕소와 아민 사례 연구 (Ethanolamine and boron abuse to limit microbial growth in water-synthetic metalworking fluids)

  • This study was conducted to examine whether a specific synthetic metalworking fluid (MWF), "A", in use for 10 months without replacement, displayed microbial resistance and to identify the additives associated with the control of microbial growth. Three synthetic MWF products ("A", "B", and "C") were studied every week for two months. Microbial deterioration of the fluids was assessed through evaluation by endotoxin, bacteria and fungi levels in the MWFs. In addition, formaldehyde, boron, ethanolamine, and copper levels were also studied to determine whether they influence microbial growth in water-based MWFs. Throughout the entire study in the sump where MWF "A" was used, bacteria counts were lower than 103 CFU/mL, and endotoxins never exceeded 103 EU/mL. These levels were significantly lower than levels observed in sumps badly deteriorated with microbes. Boron levels in MWF "A" ranged from 91.7 to 129.6 ppm, which was significantly higher than boron levels found in other MWF products. The total level of ethanolamine (EA) in MWF "A" ranged from 35,595 to 57,857 ppm (average 40,903 ppm), which was over ten times higher than that found in other MWFs. Monoethanolamine (MEA), diethanolamine (DEA) and triethanolamine (TEA) concentrations in MWF "A" were also significantly higher than seen in other MWFs. However, although EA and boron might improve anti-microbial performance, their abuse can pose a serious risk to workers who handle MWFs. From an industrial hygiene perspective, our study results stress that the positive synergistic effect of boron and EA in reducing microbial activity in MWF must be balanced with the potentially negative health effects of such additives. Our study also addresses the disadvantage of failing to comprehensively report MWF additives on Material Safety Data Sheets (MSDS). Future research in MWF formulation is needed to find the best level of EA and boron for achieving optimal synergistic anti-microbial effects while minimizing employee health hazards.

삼황세제가감방(三黃洗劑加減方)의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구 (A Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of Samhwangseje gagambang(SHB))

  • Herbal mixture water extract of (Phellodendron amurense, Scuellaria baiklensis, Spphora flavescens, Lithospermum erythrorhizon, Mellaphis chinesis, Alumite, Zanthoxylum schinifolium, Glycyrrhiza uralensis), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur. The growth of Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur was inhibited allergy and LPS induced cyclooxygenase-2(COX-2)gene expression in RAW24.7macrophage. The results indicated th ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 11-38% and 11-56%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.7 cells. The cytotoxicity of the extract using M7T assay showed the cytotoxicity of 7 and 18% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.

대황(大黃), 황금(黃芩), 황백(黃柏) 복합 추출물의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구 (A Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of mixture of Rheum coreanum Scutellaria baikalensis Phellodenron amurense)

  • Herbal mixture water extract of (Rheum coreanum Scutellaria baikalensis, Phellodendron amurese), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis and Propionbacterium acne. The growth of Stapylococcus epidermis and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 18-36% and 10-61%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.1 cells. The cytotoxicity of the extract using MTT assay showed the cytotoxicity of 6% and 13% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to this acne and skin diseases therapy.

Anti-Oral Microbial Activity and Anti-Inflammatory Effects of Rosmarinic Acid in Lipopolysaccharide-Stimulated MC3T3-E1 Osteoblastic Cells on a Titanium Surface

  • Background: The purpose of this study was to investigate the anti-oral microbial activity and anti-inflammatory effects of rosmarinic acid (RA) in lipopolysaccharide (LPS)-stimulated MC3T3-E1 osteoblastic cells on a titanium (Ti) surface during osseointegration, and to confirm the possibility of using RA as a safe natural substance for the control of peri-implantitis (PI) in Ti-based dental implants. Methods: A disk diffusion test was conducted to confirm the antimicrobial activity of RA against oral microorganisms. In order to confirm the anti-inflammatory effects of RA, inflammatory conditions were induced with 100 ng/ml of LPS in MC3T3-E1 osteoblastic cells on the Ti surface treated with or without 14 ㎍/ml of RA. The production of nitric oxide (NO) and prostaglandin E2 (PGE2) in LPS-stimulated MC3T3-E1 osteoblastic cells on the Ti surface was confirmed using an NO assay kit and PGE2 enzyme-linked immunosorbent assay kit. Reverse transcription polymerase chain reaction and western blot analysis were performed to confirm the expression of interleukin (IL)-1β and tumor necrosis factor (TNF)-α in total RNA and protein. Results: RA showed weak antimicrobial effects against Streptococcus mutans and Escherichia coli, but no antimicrobial activity against the bacteria Aggregatibacter actinomycetemcomitans and the fungus Candida albicans. RA reduced the production of pro-inflammatory mediators, NO and PGE2, and proinflammatory cytokines, TNF-α and IL-1β, in LPS-stimulated MC3T3-E1 osteoblastic cells on the Ti surface at the protein and mRNA levels. Conclusion: RA not only has anti-oral microbial activity, but also anti-inflammatory effects in LPS-stimulated MC3T3-E1 osteoblasts on the Ti surface, therefore, it can be used as a safe functional substance derived from plants for the prevention and control of PI for successful Ti-based implants.

  • MeOH extract of twenty medicinal herbs were screened for their effects against protein kinase C (PKC) using bleb-forming assay and PKC enzyme assay. Smilax china and Sanguisorba officinalis showed potent anti-PKC activity. Campsis grandiflora and Galla Halepensis showed moderate inhibitory effect on PKC.
