Anti-Inflammatory Effects of Bee Venom on Phthalic Anhydride-Induced Atopic Dermatitis

  • Background: Atopic dermatitis (AD) is a chronic inflammatory condition which can be studied using phthalic anhydride (PA) to induce AD. Anti-inflammatory properties of bee venom (BV) wereinvestigated to determine whether it may be a useful treatment for AD. Methods: AD was induced by applying to pical PA to 8-week-old HR-1 mice (N = 50), then treating with (0.1, 0.25, and 0.5 ㎍) or without topical BV. Body weight, ear thickness histology, enzyme-linked immune sorbent assay (serum IgE concentrations), Western blot analysis [inducible nitric oxide synthase, cyclooxygenase-2, IκB-α, phospho-IκB-α, c-Jun N-terminal kinase (JNK), phospho-JNK, p38, phospho-p38, extra cellular signal-regulated kinase (ERK), and phospho-ERK], and the pull down assay for immunoblotting (p50), were used to measure inflammatory mediators. Results: PA + BV (0.1, 0.25, and 0.5 ㎍) significantly decreased ear thickness without altering body weight. IgE concentrations decreased in the PA + BV (0.5 ㎍)-treated groups compared with PAtreatment. Tumor necrosis factor-α, interleukin-1β, inducible nitric oxide synthase, cyclooxygenase-2, phospho-IκB-α, phospho-JNK, p38, phospho-p38, and phospho-ERK, all decreased following treatment with PA + BV compared with the PA-treatment alone. p50 was upregulated in the PA + BV-treated groups compared with the PA-treated group. Furthermore, the number of mast cells decreased in the PA + BV-treated groups compared with the PA-treated group. Epidermal thickness was significantly lower in the PA + BV-treated group compared with PA treatment alone. Conclusion: BV maybe a useful anti-inflammatory treatment for AD.

  • The experiment was conducted to validate anti-inflammatory effects of pinitol from bean. It was evaluated for some molecule targets by wound healing assay and RT-PCR. The results of wound healing assay was shown dose-dependent inhibition of cell migration in cancer cells and inhibited RNA expression of ICAM-1, CD 44, MMP-17, MMP-14 and ARF2. Immune suppression activity in a mouse provoked by DNFB observed that inflammatory reaction with pinitol were reduced ear swelling and inflammatory cells infiltration in mouse atopic models. The result confirmed that pinitol have the effect of dose-dependent immune suppression activity.

Anti-inflammatory Activity of Fucoidan with Blocking NF-κB and STAT1 in Human Keratinocytes Cells

  • Fucoidan, a sulfated polysaccharide is found in several types of edible brown algae. It has shown numerous biological activities; however, the molecular mechanisms on the activity against atopic dermatitis have not been reported yet. We now examined the effects of fucoidan on chemokine production co-induced by TNF-α/IFN-γ, and the possible mechanisms underlying these biological effects. Our data showed that fucoidan inhibited the TNF-α/IFN-γ-induced production of thymus and activation-regulated chemokine (TARC) and macrophagederived chemokine (MDC) mRNA in human keratinocytes HaCaT cells. Also, fucoidan suppressed phosphorylation of nuclear factor kappa B (NF-κB) and activation of signal transducer and activator of transcription (STAT)1 in a dose-dependent manner. In addition, fucoidan significantly inhibited activation of extracellular-signal-regulated kinases (ERK) phosphorylation. These data indicate that fucoidan shows anti-inflammatory effects by suppressing the expression of TNF-α/IFN-γ-induced chemokines by blocking NF-κB, STAT1, and ERK1/2 activation, suggestive of as used as a therapeutic application in inflammatory skin diseases, such as atopic dermatitis.

고욤잎, 감잎 및 뽕잎 복합추출물의 항아토피 효과 (Anti-Atopic Effects of Mixed Extracts from Date Plum, Persimmon, and Mulberry Leaves)

  • 본 연구는 고욤잎, 감잎 및 뽕잎을 단독으로 사용하기보다 혼합하여 사용할 경우 고욤잎, 감잎 및 뽕잎 복합추출물이 아토피 피부염(AD) 증상을 억제하는지 효과를 밝히고, 항염 효능을 나타내는 기능성 소재로서의 이용 가능성을 알아보고자 하였다. 그 결과 육안 평가를 통해 피부의 건조 상태, 스케일링, 미란, 찰과상 및 홍반 같은 아토피 피부염 증상이 AD 모델에서 증가하였지만, 고욤잎, 감잎 및 뽕잎 각각의 추출물과 복합추출물을 투여하였을 경우 완화되는 것을 확인할 수 있었으며, 복합추출물이 고욤잎, 감잎 및 뽕잎 각각의 추출물보다 더 뛰어난 효과를 나타내었다. 피부 두께와 염증세포 및 비만세포의 침윤은 AD 모델에서 크게 증가하였지만, 고욤잎, 감잎 및 뽕잎 단독추출물과 복합추출물을 투여하였을 경우 감소하는 것을 확인할 수 있었으며, 복합추출물이 단독추출물보다 더 뛰어난 효과를 나타내었다. 혈청중의 IgE와 IL-4의 수치를 측정한 결과 AD 모델에서 많이 증가하였으나, 고욤잎, 감잎 및 뽕잎 단독추출물과 복합추출물을 투여하였을 경우 감소하는 것을 확인할 수 있었으며, 복합추출물이 단독추출물보다 더 뛰어나게 억제하는 효과를 나타내었다. HMC-1 세포에 복합추출물을 처리하였을 경우 염증성 사이토카인인 $TNF-{\alpha}$와 IL-6의 생성량이 유의적으로 감소하였으며, 또한 RAW 264.7 세포에 복합추출물을 처리하였을 경우 염증 매개 인자인 $TNF-{\alpha}$, IL-6, $PGE_2$ 및 NO 생성량이 감소하였고, 이러한 염증 매개 인자 억제 효능은 복합추출물이 단독추출물보다 더 뛰어나게 억제하는 것을 확인하였다. 이러한 결과로 미루어볼 때 고욤잎, 감잎 및 뽕잎을 혼합한 복합추출물을 사용할 때 효능이 더 우수한 것을 확인하였고, 아토피 피부염 증상 개선 및 염증관련 질환 치료를 위한 기능성 천연물 소재 및 제품 개발에 응용이 가능할 것으로 생각한다.

An Atopic Preventive Drink (APD) reduces Th2 cytokines in LPS-treated RAW 264.7 cells

  • We analyzed the effects of an Atopic Preventive Drink (APD) on the regulation of Th2 cytokines using RAW 264.7 macrophage cells. In the evaluation of nitric oxide (NO) production in cells, NO production levels were shown to be elevated only in the APD-treated group in a dose-dependent manner. In the lipopolysaccharide (LPS) with APD-treated group, NO production significantly decreased as APD concentration increased. Further, mRNA expression levels and protein concentrations of pro-inflammatory cytokines in cells were determined. Th2 stimulatory cytokine ($IL-1{\beta}$) and Th2 cytokine (IL-6 and IL-10) levels were significantly reduced in the LPS with APD-treated group compared to the only LPS-treated group. mRNA expression levels of inflammatory-related genes (COX-2 and iNOS) were significantly reduced in the LPS with APD-treated group compared to the only LPS-treated group. These results suggest that APD has an anti-atopic effect by reducing mRNA and proteins expressions of Th2 cytokines and inflammatory-related genes.

아토피 피부염에 대한 황토가미방(黃土加味方)과 자초복합방(紫草複合方) 겸용 연구 (A Study on Combination of Topical Jachobokhabbang(JBB) and Internal Hwangtogamibang(HTGMB) for the Treatment of Atopic Dermatitis)

  • In order to investigate the efficacy of a combination of JBB as topical and HTGMB as internal treatment method, changes in various immune related factors and histological changes in NC/Nga induced animal model was studied. Combined treatment of topical JBB and internal HTGMB significantly reduced the atopic dermatitis clinical index, the number of immune cells such as CD19+, CCR3+, B220+/IgE+, and Gr-1+/CD11b+ in DLN and dorsal skin, compared to the control group. Otherwise increased CD3+, CD4+/CD25+, CD8+ and CD4+ cells in the DLN. And also combined treatment of topical JBB and internal HTGMB suppressed the lymphocytes and mast cells from infiltrating into the skin tissues when stained with H&E and toluidine blue. Based on the results above, it is strongly suggested that the combined treatment of topical JBB and internal HTGMB significantly induced anti-allergic activities through immune modulation. The findings can be applied to developing a more sustainable treatment for atopic dermatitis and be helpful in practicing combined treatments in clinical treatments in the future.

Immune modulation and possible pathological implications mediated by naturally produced immunoglobulin G idiotypes: from historical to recent experimental and clinical studies focused on atopic dermatitis

  • Since the 1950s decade, it has been suggested that a naturally produced or induced repertoire of immunoglobulin G (IgG) idiotypes may exert some immunoregulatory functions. In the last decades, some more advanced theories have suggested that the repertoire of IgG idiotypes may influence the development or control of some atopic diseases. In atopic dermatitis (AD), some evidence indicated that the IgG repertoire obtained from these patients could effectively mediate regulatory functions on thymic and peripheral CD4+ and CD8+ T cells. Furthermore, some recent clinical trials have corroborated the hypothesis that IgG from AD patients can exert regulatory functions in vivo. Here, we revised some historical aspects that yield current approaches developed in vitro and in vivo to elucidate a recently proposed theory termed "hooks without bait" that can strengthen the broad spectrum of research about evaluating different sets of IgG idiotypes and determine their immunological effects.

시험관내에서 천연물제제 BSASM의 항염증 및 면역억제 효능 평가 (Evaluation of the Anti-inflammatory and Immunomodulatory Effects of BSASM Using in vitro Experiments)

  • For effective management of atopic dermatitis, it is important to introduce a therapeutic agent although having the fewest side effects, has the greatest anti- inflammatory effect. In the course of screening anti-inflammatory agents, we obtained BSASM composed of several plant extracts. This study was designed to investigate anti-inflammatory and immunomodulatory effects of BSASM. As a first step, $NF-{\kappa}B$ luciferase reporter assay was performed to know the involvement of BSASM in the production of proinflammatory cytokines because $NF-{\kappa}B$ element has been known to play a major role in expression of cytokine genes such as interleukin-8 (IL-8) or tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$. LPS (lipolysaccharide)-induced $NF-{\kappa}B$ activation was inhibited by BSASM. In addition, we found the fact that BSASM inhibits LPS-induced produced production of IL-8 and $TNF-{\alpha}$ proinflammatory cytokines, indicating BSASM has anti-inflammatory effect. In interleukin-2 (IL-2) luciferase reporter assay in Jurkat T cells, BSASM reduced PHA (Phytohemagglutinin)-induced IL-2 luciferase activity, suggesting the possibility that BSASM might also have an immunomodulatory function in T cell-mediated immune response. Based on these results, we suggest the possibility that BSASM can be introduced to improve symptom of immune-related skin diseases, namely, atopic dermatitis.

DNCB로 아토피가 유도된 NC/Nga생쥐에서 천연 탄산 온천수의 온도별 항아토피 효과 (The anti-atopic effect of natural carbonated hot spring water on DNCB-induced NC/Nga mice under different temperature)

  • Objectives : The aim of this study is to investigate the effect of natural carbonated hot spring water (NCHW) on DNCB-induced NC/Nga mice, an atopic dermatitis (AD) model. Methods :At first, NC/Nga mice were prepared and induced to have atopic lesion on their back skin by dinitrochlorobenzene (DNCB). They swam for 20 minutes everyday in tanks where the one is filled with $28^{\circ}C$ NCHW, the other is with $25^{\circ}C$ carbonated hot spring water and another is with artificial carbonated hot spring water (ACHW). After 3 weeks, We assessed the skin clinical score and macroscopic appearance, total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and histological changes. Results : There are meaningful results of improving atopic lesion-state by relieving the count of total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and recovering skin clinical score in the group with $28^{\circ}C$ NCHW in comparison with the other groups. Conclusions : The NCHW may have potential as an effective treatment for atopic dermatitis.

참모자반 물 추출물의 항아토피 효과 (Inhibitory Effect of Sargassum fulvellum Water Extract on 2,4-Dinitrochlorobenzene-induced Atopic Dermatitis-like Skin Lesions in Mice)

  • 본 연구는 BALB/c mice의 등 쪽 부위에 반복적인 DNCB 도포를 통해 유발시킨 아토피 피부염에 있어서 참모자반 물 추출물의 효능에 대해 연구하였다. 참모자반 물 추출물을 2주 동안 지속적으로 처리하고 IL-4 및 IFN-γ cytokine의 분비량, 비장세포 증식능, 혈청 중 총 IgE 함량, 육안 평가 및 skin clinical severity score, 조직관찰을 실시하였다. 그 결과 참모자반 물 추출물을 지속적으로 도포함으로써 IL-4 cytokine과 총 IgE 함량이 현저히 감소하는 것을 확인할 수 있었으며 IFN-γ cytokine은 유의적으로 증가하는 것을 확인하였다. 비장세포 증식능에는 참모자반 물 추출물의 도포처리가 큰 영향을 미치지 않는 것으로 나타났다. 육안평가 및 skin clinical severity score 결과, 참모자반 물 추출물을 2주간 지속적으로 도포 처리하였을 때, 그 증상이 눈에 띄게 완화되는 것을 확인할 수 있었으며 score 또한 DNCB 단독 처리군과 비교 시 유의적으로 감소하였다. 등 조직 관찰에서도 참모자반 물 추출물에 의해 DNCB로 유발된 아토피 동물 실험군의 경피 및 진피 조직 두께 및 진피에서의 mast cell의 침윤 정도가 감소되었음을 확인하였다. 이상 결과를 종합 해볼 때, 참모자반 물 추출물이 아토피 피부염 치료제로서 이용 가능성이 있을 것으로 사료된다.
