The Effects of Sunbanghwalmyung-eum Extract on Anti-oxidant, Anti-allergic and Anti-inflammatory ability in mouse cell (Mouse cell에서 선방활명음(仙方活命飮)의 항산화작용과 항알러지 및 항염증 효과)

  • Background and Objectives : The aim of this study was to investigate the anti-oxidant, anti-allergic and anti-inflammatory ability of the Taglisodog-eum(SHE) extract on the RAW 264.7 and EL4 cells Materials and Methods : Three types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of SHE using Riboflavin; second, in vitro experiment to investigate the inhibition of Th 2 cell differentiation by SHE using EL4 cells (IL-4 mRNA expression); third, the suppression of $NF-{\kappa}B$ activation using RAW 264.7 cells (iNOS and COX-2 mRNA expression). Results : The anti-oxidant ability of SHE were dose-dependantly increased. From in vitro, the LPS-induced iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with SHE and the PMA-induced IL-4 mRNA expression were also dose-dependantly decreased in EL4 cells. $NF-{\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE Conclusion : The results suggest that SHE has dose-dependant anti-oxidant ability, and has anti-allergic and anti-inflammatory effects through the suppression of $NF-{\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

  • Experimental studies were done to research the clinical effects of Yongdamsagantang and Yongdamsagantang-gamibang (Yongdamsagantang with Lonicerae Flos and Forsythiae Fructusadded) on the Anti-allergic effect in rats and mice. The results obtained as follows; 1. In the effects of Yongdamsagantang and Yongdamsagantang-gamibang on vascular permeability responses to intradermal histamine in rats, Yongdamsagantang group revealed none significant affect, but Yongdamsagantang-gamibang group revealed significant effect. 2. In the effects of Yongdamsagantang and Yongdamsagantang-gamibang on vascular permeability responses to intradermal serotonin in rats, Yongdamsagantang and Yongdamsagantang-gamibang group revealed significant effect. 3. In the 48hrs homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody against egg albumin, Yongdamsagantang group revealed none significant effect, but Yongdamsagantang-gamibang group revealed significant effect. 4. In the delayed type hypersensitivity responses to Picryl Chloride in mice, Yongdamsagantang and Yongdamsagantang-gamibang group revealed none significant effect, in the delayed type hypersensitivity responses to SRBC in mice, Yongdamsagantang group revealed none significant effect, but Yongdamsagantang-gamibang group revealed significant effect. According to above-stated results, Yongdamsagantang is none significant Anti-allergic effect. But Yongdamsagantang-gamibang is concluded to be effective as immediated type hypersensitivity and recommended to be used for the treatment of allergic diseases. (Asthma, Allergic urticaria, Allergic rhinitis, etc.)

  • Since the main symptoms of COVID-19 involve the respiratory system, the infection rate of this disease is predicted to be higher in patients with other respiratory conditions such as allergic rhinitis. In such a situation, it will be meaningful to conduct research on an allergy treatment that has fewer side effects and can effectively reduce allergy symptoms. Here, we prepared experimental samples under various fermentation conditions with mixed extracts of six medicinal plants. To examine the anti-allergic efficacy of these samples, an egg albumin-induced allergic rhinitis animal model experiment, a serum histamine and IgE experiment, and a COX and LO inhibitory activity experiment were conducted. As a result of animal experiments, OVA+SP-4 showed superior efficacy compared to OVA+SP-1 in nasal rubbing and sneezing experiments and had anti-allergic efficacy similar to that of OVA-cetirizine. The serum histamine concentration of OVA+SP-4 was also 1.3 times higher than that of the OVA+cetirizine group, showing a high histamine reduction ability, and IgE showed the same trend. An analysis of COX inhibitory efficacy also confirmed that COX-1 and COX-2 inhibitory efficacy is high, and the longer the fermentation time, the higher the antiallergic efficacy. The composition proposed by this study is expected to have a significant effect on sustainable allergy prevention and treatment in the future by applying it to human patients.

Experimental Study on Anti-allergic and Anti-inflammatory Effects of Soeumin-Googwihyangso-san Methanol Extract in Vitro (소음인(少陰人) 궁귀향소산(芎歸香蘇散)의 항(抗) 알레르기 및 항(抗) 염증에 미치는 실험적 연구)

  • Soeumin-Googwihyangso-san(SGGHSS) has been used for the prevention or treatment of Soeumin-allergic rhinitis. This study was performed to demonstrate anti-allergic and anti-inflamatory effects of SGGHSS methanol extract in HMC cell lines and activated mouse B cells and CD4+ T cells. SGGHSS inhibited the production of TNF-$\alpha$ in PMA plus A23187 activated HMC-1 cells but not that of IL-6, as measured by ELISA. SGGHSS inhibited the expression of CD23 and surface IgE in B cells as determined by flowcytometry. It also inhibited secretion of IFN-$\gamma$ and IgG1, the Th1 related IgG type, but increased that of IL-4 in anti-CD40 and IL-4 treated B cells as measured by ELISA. As for Th cell differentiation, SGGHSS did not much affect IL-4 or IFN-$\gamma$. Taken together, our data showed that SGGHSS exerted an anti-inflammtory effect by inhibiting TNF-$\alpha$ in mast cells and has anti-allergic activity not via inhibition of CD4+ T cell, but via inhibition of B cells. These results suggest some evidence that SGGHSS can be applied to allergic disease.

  • This experimental study was done to research effects of a number of extract on the anti-allergic effect. The results were obtained as follows: 1. In effect of herb-extract on compound $\frac{48}{80}$-mediated histamine release from Evans blue skin assay, Isatis Japonica Miquel, Dictamnus dasycarpus Turcz, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum chinense, Magnolia liliflora, Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(MtOH), Trichosanthes kirilowii, Phellodendron amurense Rupr, Schizonepeta tenuifolia Var, Betula platyphylla show considerable visible anti-allergic effect. In the result of quantification of histamine induced compound $\frac{48}{80}$, Spirodela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively. 2. In effect of Herb-Extract on compound $\frac{48}{80}$-mediated histamine release from RPMC, Spirodcla polyrhiza, Cimicifuga heracleifolia inhibit histamine release effectively. 3. In effect of Herb-Extract on anti-DNP IgE-mediated histamine release from Evansblue skin assay. Spirodela polyrhiza, Cimicifuga heracleifolia(0.1mg/ml). Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(0.1mg/ml), Trichosanthes kirilowii(0.1mg/ml) show considerable visible anti-allergic effect. In the result of quantification of histamine induced anti-DNP IgE, Spiradela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively 4. In the result of genetic manifestative inhibition about the human mast cell-1(HMC-1), Cimicifuga heracleifolia has considerable effect in IL-4 in IL-5, and Tussilage farfara L. has in IL-4. According to the above results, it is suggested that several Herb-Extract have anti-allergic effect.

  • In a bioassay-guided search for anti-allergic compounds from higher plants of Korea, poly-methoxyflavones, 3',4',5,6,7,8-hexamethoxyflavone (I), 5-hydroxy-3',4',6,7,8-pentamethoxyflavone (II) and 3',4',5,7,8-pentamethoxyflavone (III) have been isolated from the immature peels of Citrus unshiu. Structures of these compounds were elucidated on the basis of spectroscopic techniques. Compounds I and II inhibited dose-dependent histamine release from the rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE.

Inhibitory Effect of Rehmannia Glutinosa Pharmacopuncture Solution on β-hexosaminidase Release and Cytokine Production via FcεRI signaling in RBL-2H3 Cells (RBL-2H3세포에서 생지황약침액의 FcεRI 신호전달을 통한 β-hexosaminidase분비와 Cytokine생성 억제 효과)

  • Background: Type I allergy is involved in allergic asthma, allergic rhinitis, and atopic dermatitis which are accompanied by an acute and chronic allergic inflammatory responses. Rehmannia glutinosa is a traditional medicine in the East Asian region. This study examined whether a Rehmannia Glutinosa pharmacopuncture solution (RGPS) had anti-allergic or anti-inflammatory effects in antigen-stimulated-RBL-2H3 cells. Methods: We determined the effect of RGPS on cell viability using the 3-[4,5-dimethylthiazolyl]-2,5-diphenyltetrazolium bromide (MTT) assay. We also examined the effect of RGPS on the release of ${\beta}$-hexosaminidase and the secretion of IL-4 and TNF-${\alpha}$ using ELISA. In addition, we evaluated the effect of RGPS on the mRNA expression of various cytokines; IL-2, IL-3, IL-4, IL-5, IL-13 and TNF-${\alpha}$ using RT-PCR. Furthermore, we assessed the activation of mitogen-activated protein kinases (MAPKs) and nuclear factor (NF)-${\kappa}$B using Western blotting after RGPS treatment. Results: We found that RGPS ($10^{-4}$ to $10^{-1}$ dilution) did not cause any cytotoxicity. We observed significant inhibition of ${\beta}$-hexosaminidase release and suppression of the protein secretion of IL-4 and TNF-${\alpha}$ and mRNA expression of multiple cytokines in antigen-stimulated-RBL-2H3 cells after RGPS treatment. Additionally, RGPS suppressed not only the phosphorylation of MAPKs, but also the transcriptional activation of NF-${\kappa}$B in antigen-stimulated-RBL-2H3 cells. Conclusions: These results suggest that RGPS inhibits degranulation and expression of cytokines including IL-4 and TNF-${\alpha}$ via down-regulation of MAPKs and NF-${\kappa}$B activation in antigen-stimulated-RBL-2H3 cells. In conclusion, RGPS may have beneficial effects in the exerting anti-allergic or anti-inflammatory activities.

Human Skin Safety Test of Green Tea Cell Extracts in Condition of Allergic Contact Dermatitis

  • Various kinds of positive effects of green tea extracts had been studied for long time which included anti-inflammation, anti-aging, and cardiometabolic effects. Although topical steroid and non-steroidal calcineurin inhibitors may control clinical symptoms of allergic contact dermatitis, some of patients also present allergic reaction to these topical agents. Therefore, we have tried green tea extracts for managing this skin disorder with expectation of anti-inflammatory effect without potential side effects including skin irritation and toxic responses. The toxicity test of green tea extract also did not show any sign of irritation in the skin throughout the test period. Moderate severity of allergic contact dermatitis presented satisfactory clinical outcome at second week follow-up which was final visit of outpatient. This result mean that green tea extract has a positive effect for managing allergic contact dermatitis but its potency and efficacy seem to be so not strong enough to control moderate severity allergy skin lesion. In this pilot study, we were able to conclude that green tea cell extracts might be applied for potential anti-inflammatory soaking without skin toxicity.

Anti-Allergic Effect of Oroxylin A from Oroxylum indicum Using in vivo and in vitro Experiments

  • Oroxylum indicum has long been used in Asian traditional medicine to prevent and treat respiratory diseases, diabetes, diarrhea and other conditions. Oroxylin A is a flavone that is present in Oroxylum indicum and in Scutellaria baicalensis. Because the root extracts of both plants have been shown to have anti-allergic effects, the authors investigated whether oroxylin A is likely to have beneficial effects on allergic asthma using female Balb/c mice and rat RBL-2H3 mast cells. Antigen-induced degranulation was measured in vitro by measuring b-hexosaminidase activity. A murine ovalbumin-induced allergic asthma model was used to test the in vivo efficacy of oroxylin A. Sensitization and challenge of ovalbumin induced allergic asthma responses, the accumulations of eosinophils and Th2 cytokine levels in bronchoalveolar lavage fluid and lung tissues. Oroxylin A administration decreased numbers of inflammatory cells, especially eosinophils, and reduced the expression and secretion of Th2 cytokines, including IL-4 and IL-13, in lung tissues and bronchoalveolar lavage fluid. Histologic studies showed oroxylin A reduced inflammatory signs and mucin production in lungs. These findings provide evidence that oroxylin A has potential use as an anti-allergic therapeutic.

Effect of Salvia plebeia on IgE antibody mediated allergic reaction in rats

  • The effect of aqueous extract of Salvia plebeia R. Br. (Labiatae) (AESP) on immunoglobulin (lgE) antibody mediated allergic reactions in rats was investigated. AESP inhibited passive cutaneous anaphylaxis (PCA) when intravenously, intraperitoneally, and orally administered. AESP dose-dependently inhibited histamine release from rat peritoneal mast cells activated by anti-DNP IgE antibody. Moreover, AESP had an inhibitory effect on anti-DNP IgE antibody induced $TNF-{\alpha}$ production from RPMC. These results suggest that AESP inhibit the IgE-mediated allergic reaction in rats.
