야생 산삼 지상부 및 지하부의 화학성분 분석과 피부 항노화 효능 평가 (Analysis of Chemical Components for Aerial and Underground Parts of Wild Ginseng and Evaluation of Skin Anti-aging Efficacy)

  • Objectives : This study was intended to reveal the chemical profiles of aerial(leaf, stem) and underground(rhizome, radix) parts of wild ginseng, and to investigate their anti-aging effects on human skin cells. Methods : Wild ginseng, estimated for over 20 years, was divided into the aerial and underground parts. Total phenolic contents of each extracts were measured using a Folin-ciocalteu method. The contents of 18 amino acids, 8 minerals and 27 ginsenosides were determined by GC-FID, ICP-MS and LC-MS, respectively. The anti-aging effects, including the radical scavenging activity, the activation of mitochondrial function on human fibroblasts, and the proliferation activity on human keratinocyte progenitor cells, for the whole plant and underground part of wild ginseng were evaluated. Results : The total phenolic acids, amino acids, and minerals in the aerial part were more than twice as high as in the underground part. Compared to the cultivated ginseng root, there were various types of ginsenosides in both parts of wild ginseng, and the total amount was more than twice as high. In particular, the aerial part significantly contained ginsenoside F1, F2, C-Mc1, and C-O, and the distinctive patterns that distinguish each parts of wild ginseng from the cultivated ginseng root were derived. The whole plant and underground part of wild ginseng exhibited significant antioxidant effect(14.3-45.6%), activation of mitochondrial membrane potential(105.5-120.1%), and cell proliferation(112.1-125.4%). Conclusions : The entire plant and underground part of wild ginseng are high value-added plants and have beneficial effects on skin anti-aging properties through its abundant metabolites.

추출용매에 따른 단삼 추출물의 항노화 활성 (Effects of solvents on the anti-aging activity of Salvia miltiorrhiza extract)

  • 본 연구에서는 단삼 뿌리 추출물의 세포 안정성, 수렴 활성, NO 소거능, iNOS 단백질 발현량, 염증성 cytokine 분비, elastase 효소 저해능, type I pro-collagen 합성능 등을 조사하여 기능성 화장품 소재로서의 가능성을 평가하고자 하였다. 단삼 80% 에탄올추출물(SE)과 열수추출물(SW)을 대상으로 NHDF 및 RAW264.7 세포에서 0.05~0.5 mg/mL 농도 처리에 따른 독성을 보이지 않아 수렴, 염증, 주름 개선 효능을 진행하였다. 그 결과 수렴활성 측정 결과 SE는 10 mg/mL에서 74.6%의 활성을 나타냈다. SE는 농도 의존적으로 LPS에 의해 유도된 NO, iNOS 단백질, TNF-α 및 IL-6 cytokine의 생성을 유의미하게 감소시켰다. 게다가 SE와 SW는 elastase 효소 활성을 억제할 뿐만 아니라 TNF-α에 의해 감소된 pro-collagen의 생성을 증가시켰다. 따라서 단삼 에탄올추출물은 수렴제, 염증 관련 피부 질환, 주름 생성을 효과적으로 예방할 수 있는 천연 화장품 소재로서의 활용 가능성이 높을 것으로 기대된다.

API X70 라인파이프강의 인장 특성에 미치는 변형 시효의 영향 (Effect of Strain Aging on the Tensile Properties of an API X70 Linepipe Steel)

  • The effect of strain aging on the tensile properties of API X70 linepipe steel was investigated in this study. The API X70 linepipe steel was fabricated by controlled rolling and accelerated cooling processes, and the microstructure was analyzed using optical and scanning electron microscopes and electron backscatter diffraction. Strain aging tests consisting of 1 % pre-strain and thermal aging at $200^{\circ}C$ and $250^{\circ}C$ were conducted to simulate U-forming, O-forming, Expansion(UOE) pipe forming and anti-corrosion coating processes. The API X70 linepipe steel was composed of polygonal ferrite, acicular ferrite, granular bainite, and bainitic ferrite whose volume fraction was dependent on the chemical composition and process conditions. As the thermal aging temperature increased, the steel specimens showed more clearly discontinuous type yielding behavior in the tensile stress-strain curve due to the formation of a Cottrell atmosphere. After pre-strain and thermal aging, the yield and tensile strengths increased and the yield-to-tensile strength ratio decreased because yielding and aging behaviors significantly affected work hardening. On the other hand, uniform and total elongations decreased after pre-strain and thermal aging since dislocation gliding was restricted by increased dislocation density after a 1 % pre-strain.

모발 노화에 따른 물성변화와 외인성 노화모델의 개발 (A New Attempt to Establish the Extrinsic Aging Hair Model to Evaluate The Response to Aging in Physical Property)

  • 인체 조직은 노화 현상을 겪으면 산화로 인한 손상이 진행되어 구조적인 변화가 일어나고 물성 저하를 겪는다. 피부가 겪는 노화 과정은 잘 알려져 있고 많은 평가가 수행되고 있지만 상대적으로 모발(머리카락)의 노화에 의한 평가는 잘 진행되어 있지 않아 모발의 노화를 케어하는 연구가 어려운 실정이다. 이에 본 연구는 모발의 노화 현상을 케어할 효능 기작을 적용할 수 있도록 노화된 모발 샘플을 제작하고 물성을 평가하여 안티에이징 효능을 확인하는 것을 목적으로 진행하였다. 먼저 내인성 노화가 진행된 모발의 지질량을 외인성 노화를 진행시킨 시료들과 비교한 결과, 펌 1회 처리 모발이 60대의 모발 지질량과 가장 유사함을 발견하였고, 이 모발을 통해서 원자 현미경으로 나노 스케일의 거칠기와 매크로 스케일의 인장강도를 평가하여 내인성 노화 모발과 유사한 물성 값을 가지고 있음을 확증하였다. 실제로 펌 처리를 통해 외인성으로 노화시킨 모발에 카르보디이미드와 펩타이드 결합을 적용하여 DSC로 구조적 변화와 거칠기 및 인장강도를 확인한 결과, 외인성 요인으로 노화된 모발을 에이징 이전의 상태의 물성 쪽으로 개선시키는 효과를 확인할 수 있었다. 뿐만 아니라 내인성 노화에 의해서 소실 된 내부 구성 성분을 프로폴리스를 이용하여 보강한 경우에도 외인성 노화가 유도된 모발의 물성이 에이징 이전 상태의 물성으로 회복되는 안티 에이징 효능이 발생함을 확인하였다. 본 연구에서 사용한 외인성 노화 유발 모발 시료 및 평가는 모발의 외인성 노화 연구에 유용하게 적용될 수 있음을 최초로 확인하였다.

Photoprotective Effects of Soybean Extract against UV-Induced Damage in Human Fibroblast and Hairless Mouse Model

  • Soy isoflavones have been reported to possess many physiological activities such as antioxidant activity and inhibition of cancer cell proliferation. This study investigated the photoprotective effects of soybean extract in human fibroblast cell line and hairless mice model. Human fibroblast was treated with soybean extract before and after ultraviolet B (UVB; 290-302 nm) irradiation. In the soybean extract treated group, the cells showed better resistance to ultraviolet (UV) than control group. The amount of type I collagen recovered from the soybean treated group was higher than the vehicle group exposed to UV-induced damage. Moreover, increased expression of metalloproteinases-1 as a result of UV irradiation was suppressed by the soybean extract. Female mice were orally administered soybean extract and irradiated with UVB light for 8 weeks. The effects of the soybean extract on the skin appearance, collagen deposition and epidermal thickness in the UV-damaged mouse skin were analyzed using histopathological methods. In soybean extract treated group, the skin had a better morphology than that of the control group. Furthermore, the amount of type I collagen was increased and overexpression of MMP-1 was reduced in the soybean extract group compared to vehicle group. Additionally, up-regulation of pro-inflammatory cytokines induced by UV irradiation was suppressed by dietary soybean extract treatment. It appears that soybean extract had a photoprotective effect, including anti-aging and anti-inflammatory effect, from UV-induced damage in not only human fibroblast, but also hairless mice. We confirmed that these effects were possibly due to promotion of collagen synthesis and inhibition of MMP-1 expression.

공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향 (An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

  • 세리신은 누에고치에서 추출한 단백질로 많은 건강상의 이점을 가지고 있다. 본 연구는 화장품 소재로서 누에고치에서 유래된 세리신 표준품의 항주름 활성 및 항염증 활성을 평가하기 위해 수행되었다. 세리신의 항산화 효과는 DPPH 및 ABTS 측정법에 의해 측정되었다. 또한 대식세포인 Raw 264.7 cell에서의 세포 생존율을 확인하였으며, lipoplyscaccharide를 이용하여 유도된 염증반응을 이용하여 세리신의 항염증 효과를 조사하였다. 그 결과 세리신은 DPPH, ABTS에서 항산화 활성을 보였으며 세포 독성을 가지지 않은 1,000 ㎍/mL의 농도에서 NO를 억제하였다. 종합하여 세리신은 항산화 활성을 가지며 항노화 및 항염증 화장품의 우수한 소재가 될 수 있음을 보여준다.

The Effect of Anti-UV Agents on UV Shielding and Photolytic Aging of Paper

  • The behaviors of deterioration in the mechanical and optical properties of paper during sunlight exposure are mainly attributed to UV irradiation. The effect of different additives (anti-UV agents) on paper properties during exposure was studied. The results showed negligible changes in the mechanical properties of samples prepared with two different ways, whereas the shielding ability was increased when the additives were applied to the surface of hand sheets. Also, the paper which is treated with anti UV agents can keep freshness more effectively of agricultural products from UV irradiation.

진생베리 발효추출물의 화장품 약리활성 (Study on Cosmeceutical Activities from Fermented Ginseng Berry Extracts)

  • 진생베리는 진세노사이드 Re을 다량함유하고 있고 항염, 항암, 혈당강하 및 미백효과가 있다. 본 연구에서는 Rhizopus Oligosporus 균주를 이용하여 진생베리 발효공정을 확립하였으며 진생베리 발효물에 대한 화장품 약리활성을 분석하였다. 발효에 의한 인삼 베리 추출물의 전자 공여 능력은 1,000 ㎍/mL 농도에서 81%를 나타냈다. ABTS+ 라디칼 소거능 1,000 ㎍/mL 농도에서 100.2% 효과를 나타내었다. 피부 미백과 관련된 티로시나 아제 억제 효과는 1,000 ㎍ /mL 농도에서 57%였다. 피부 주름과 관련된 엘라스타제 억제효과는 1,000 ㎍/mL 농도에서 47%였다. 또한, 콜라게나제 억제 효과는 1,000 ㎍/mL 농도에서 33%였다. 이상의 결과로부터 진생베리 발효물이 항염과 미백, 주름개선 효능을 가진다고 판단된다. 따라서 진생베리 발효물이 향우 항염 및 항노화 화장품 원료로서의 이용 가능성이 매우 높을 것으로 판단된다.

한방 기능성 입욕제 SP1과 SP2의 항산화, 항노화, 미백효과에 관한 연구 (Study on the Antioxidative, Antiaging and Whitening Effects of Bathing Aid SP1 and SP2)

  • The purpose of this study is to develop bathing aids as a strategic products to promote the medical tourism in Sancheong Jirisan Oriental medicinal herbs special district using medicinal herbs produced in Sancheong province, and to verify the effect of the bathing aids in vitro. We investigated the cytotoxicity activity, antioxidant activity, antiaging and whitening effects of Sanchung-PNU 1 (SP1) and Sanchung-PNU 2 (SP2) made with traditional medicinal herbs. The cytotoxicity activity was measured by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. Antioxidant activities were determined by DPPH (1.1-diphenyl-2-picrylhydrazyl) radical scavenging capacity assay. We measured the inhibitory effect against tyrosinase activity to prove the whitening effect, and the inhibitory effect against elastase activity to prove the anti-aging effect. Two proposed prescriptions, SP1 and SP2, showed not significant cytotoxicity but significant (p<0.001) improvement in anti-oxidation, anti-wrinkle, and whitening effects compared to the control group. The result shows that these bathing aids have excellent DPPH radical scavenging effect and significant inhibitory effect against elastase and tyrosinase activity. These findings suggest that these bathing aids have a strong antioxidant, anti-aging, and whitening effect.
