Anti-stress Activities of Ginsenoside Rb1 is Related with GABAnergic Neuron

  • The main aim of this study was to investigate stress related activities of ginsenosides and their action mechanism. Control group and ginsenoside supplemented groups were exposed to stress while no-stress group was not done. Animals of each group (n=$8\~10$) were orally administerd 100 mg red ginseng extract (R-G), or 10 mg ginsenosides/kg body weight once a day. Animals were given materials for 5 days without stress, and then were given supplements for 5 days with restraint and electroshock stress. Mice were given materials for 5 days for experiments on anti-fatigue effect. After loading final stress, stress-related behavioral changes of experimental animals were examined and plasma corticosterone levels were measured. R-G and ginsenoside $Rb_{1}$ supplementation partially blocked the stress effects on locomotion and elevated plus-maze test in rats and mice. They also partially blocked stress induced behavioral changes such as freezing, smelling, face-washing, rearing behavior in rats. R-G and $Rb_{1}$ decrease adrenal gland size and plasma corticosterone level, which were increased by stress in rats. R-G increased enduring time on the Rota rod, cold water and horizontal wire, but $Rb_{1}$ didn't. Effects of $Rb_{1}$ on plusmaze test were inhibited by administration of flumazenil. These results suggest that $Rb_{1}$ is the main antistress principle in ginseng and it's effect is modulated by GABAnergic nervous system.

Four active monomers from Moutan Cortex exert inhibitory effects against oxidative stress by activating Nrf2/Keap1 signaling pathway

  • Paeonol, quercetin, β-sitosterol, and gallic acid extracted from Moutan Cortex had been reported to possess anti-oxidative, anti-inflammatory, and anti-tumor activities. This work aimed to illustrate the potential anti-oxidative mechanism of monomers in human liver hepatocellular carcinoma (HepG2) cells-induced by hydrogen peroxide (H2O2) and to evaluate whether the hepatoprotective effect of monomers was independence or synergy in mice stimulated by carbon tetrachloride (CCl4). Monomers protected against oxidative stress in HepG2 cells in a dose-response manner by inhibiting the generation of reactive oxygen species, increasing total antioxidant capacity, catalase and superoxide dismutase (SOD) activities, and activating the antioxidative pathway of nuclear factor E2-related factor 2/Kelch-like ECH-associated protein 1 (Nrf2/Keap1) signaling pathway. We found that the in vitro antioxidant capacities of paeonol and quercetin were better than those of β-sitosterol and gallic acid. Furthermore, paeonol apparently diminished the levels of alanine transaminase and aspartate aminotransferase, augmented the contents of glutathione and SOD, promoted the expressions of Nrf2 and heme oxygenase-1 proteins in mice stimulated by CCl4. In HepG2 cells, paeonol, quercetin, β-sitosterol, and gallic acid play a defensive role against H2O2-induced oxidative stress through activating Nrf2/Keap1 pathway, indicating that these monomers have anti-oxidative properties. Totally, paeonol and quercetin exerted anti-oxidative and hepatoprotective effects, which is independent rather than synergy.

  • A yeast hydrolysate SCP-20 with anti-stress and mitigative effects for premenstrual syndromes (PMS) was composed with crude protein (57.5%) and carbohydrates (28.1%). For the investigation of anti-stress effect in the immobilization stressed rats, the weights of spleen, thyroid and kidney in the group of SCP-20 sdministration were significantly different from those in the group not given SCP-20. Total power (TP), suppression indicator of stress, was obtained from frequency of heart rate variability. TP was changed by the administration of SCP-20 suggesting that SCP-20 has the autonomous nervous control effect. In premenstrual assessment form(PAF), the administration of SCP-20 reduced the intensity of somatic symptoms, emotional symptoms, and behavioral symptoms.

Inhibitory Effect of Mori Ramulus on Oxidative Stress Induced by High Glucose in LLC-$PK_1$ Cells (고농도 포도당에 노출된 마우스 신장상피세포에서 상지(桑枝)의 산화 스트레스 억제 효과)

  • Objectives : Recent etiological studies show that oxidative stress might play a major role in the diabetes and its complications. Mori Ramulus (MR) has been known to have antioxidative, anti-inflammatory and antidiabetic effects. The methanol extract of MR was tested for its effectiveness in LLC-PK1 cells exposed to high glucose. Methods : The cytoprotective effect of MR was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The antioxidative effect was measured in terms of generation amount of ${\cdot}O_2^-$ by 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), NO by 4,5-diaminofluorescein (DAF-2), $ONOO^-$ by dihydrorhodamine 123 (DHR 123) in the high glucose -treated LLC-$PK_1$ cells. Western blotting was performed using anti-AGE, anti-RAGE, anti-MAPKs(ERK1/2, JNK, p38), anti-PI3K, anti-Akt, and anti-NF-${\kappa}$B (p50, p65) respectively. Results : MR extract reduced cell death and inhibited the generation of ${\cdot}O_2^-$, NO, $ONOO^-$ in the high glucose-treated LLC-$PK_1$ cells. MR inhibited the expression of AGE, RAGE, MAPKs, PI3K, and Akt by means of decreasing NF-${\kappa}$B activation. MR also inhibited NF-${\kappa}$B activation itself. Conclusions : These results indicate MR has cytoprotective, antioxidative, and anti-inflammatory effects. Therefore it is suggested that MR might prevent and cure diabetes and its complications.

An Experimental Study on the Anti-stress Effect by Taeŭmin chŏngsimyonjat'ang (태음인(太陰人) 청심연자탕(淸心連子湯)의 항(抗)스트레스 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • In order to, study the anti-stress effect of $Tae{\breve{u}}min$ $Ch{\breve{o}}ngsimy{\breve{o}}njat^{\prime}ang$, several measures of stress, including catecholmine changes, body weight changes, and gastric ulcers were used.the following result were observed: 1. Plasma epinephrine contents of stress by food in rats, in the controlled group were $219.3{\pm}18.4pg/ml$, but in the experimental group the value was decreased to $169.0{\pm}16.6pg/ml$. 2. Plasma norepinephrine content of stress by food in rats in the controlled group was $303.0{\pm}18.9pg/ml$ but in the experimental group the value was decreased to $243.6{\pm}17.6pg/ml$. 3. Plasma dopamine contents of stress by food in rats in the controlled group was $16.9{\pm}1.9pg/ml$ but in the experimental group the value was derceased to $12.8{\pm}1.5pg/ml$, but it was nonsignificant. 4. Effects of body weight of stress by food in the Shay rats in controlled group was $175.9{\pm}3.0g$, but in the experimental grou the value was increased to $187.7{\pm}4.8g$. 5. Gastric ulcers of stress by food in Shay Rats, in the experimental group, the glade was decreased more than in the control group. 6. It is considered that the anti-stress effect of Taeumin Chongsimyonjat' depends on its concentration rate. Based on the results above, it may be concluded that Taeumin chongsimyonjat' aug is effective to prevent the harmful effects of stress.

  • Objectives : Anti-oxidation activities of Malvae Semen and Abutili Semen were compared and evaluated. Methods : We tested the anti-oxidant effect through in vitro experiment and in vivo experiment that induced oxidative stress using ethanol. Results : 1. In the anti-oxidative activities of Malvae Semen and Abutili Semen they had high effects in case of the scavenging activitie of DPPH and superoxide anion radical, and inhibitory effect of linoleic acid oxidation. 2. Malvae Semen had large quantities of phenolic compound than Abutili Semen, but the anti-oxidative activity of Malvae Semen was weaker than that of Abutili Semen. 3. The anti-oxidative effect of Abutili Semen was more strong than that of Malvae Semen in the rat stressed by ethanol. Conclusion : Anti-oxidation activities of Abutili Semen were stronger than those of Malvae Semen. Therefore, the complementary usage of Malvae Semen and Abutili Semen can be considered.

Preventive Effects of Peony Root Extracts on Oxidative Stress, Thrombosis and Atherosclerosis (백작약 추출물이 항산화활성, LDL 산화 억제 및 혈전용해에 미치는 영향)

  • Objectives: There is currently increased interest in the identification of natural antioxidant compounds derived from various plants. Peony Root (PR) is used worldwide for the treatment of many types of cardiovascular disease including atherosclerosis and hypertension. It has been used in Korean traditional medicine for the treatment of glycosuria, hypertension and cancer. However, to date, no studies concerning the antioxidant properties of PR have been conducted. Therefore, this study was conducted to evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species and anti-thrombosis effect in response to treatment with PR using various screening methods including biological and non-biological oxidants. Methods: In this study, the antioxidant activity of extract from PR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on collagen-induced platelet aggregation. Results: The PR extracts were found to have a potent scavenging activity of oxidative stress [DPPH, superoxide anion, hydroxyl radical, nitric oxide and peroxynitrite, etc.] as well as an inhibitory effect on LDL oxidation and on platelet aggregation. Conclusions: The PR extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

Anti-oxidative and anti-inflammatory effects of aerial parts of Rumex japonicus Houtt. in RAW 264.7 cells (양제엽(羊蹄葉) 메탄올 추출물의 항산화 및 항염증 효과)

  • Objectives : The aerial parts of Rumex japonicus Houtt. (RF) is used by traditional clinics to treat parasite infection in East asia. This study aims a verification of anti-oxidative and anti-inflammatory effects of RF methanol extract. Methods : Anti-oxidative effects of RF were measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were measured. The inhibitory effects against the production of inflammatory mediators including NO, prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and the translocation of nuclear factor (NF)-${\kappa}B$ in LPS-stimulated RAW 264.7 cells by RF were tested. Results : RF scavenged DPPH, superoxide, NO and peroxynitrite radicals, and RF (at $200{\mu}g/m{\ell}$) reduced the inflammatory mediators definitely. Conclusions : These results indicate that RF may be a potential drug source for oxidative stress related inflammatory diseases.

Sonchus asper extract inhibits LPS-induced oxidative stress and pro-inflammatory cytokine production in RAW264.7 macrophages

  • BACKGROUND/OBJECTIVES: Sonchus asper is used extensively as an herbal anti-inflammatory for treatment of bronchitis, asthma, wounds, burns, and cough; however, further investigation is needed in order to understand the underlying mechanism. To determine its mechanism of action, we examined the effects of an ethyl acetate fraction (EAF) of S. asper on nitric oxide (NO) production and prostaglandin-E2 levels in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. MATERIALS/METHODS: An in vitro culture of RAW264.7 macrophages was treated with LPS to induce inflammation. RESULTS: Treatment with EAF resulted in significant suppression of oxidative stress in RAW264.7 macrophages as demonstrated by increased endogenous superoxide dismutase (SOD) activity and intracellular glutathione levels, decreased generation of reactive oxygen species and lipid peroxidation, and restoration of the mitochondrial membrane potential. To confirm its anti-inflammatory effects, analysis of expression of inducible NO synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$, and the anti-inflammatory cytokines IL-$1{\beta}$ and IL-6 was performed using semi-quantitative RT-PCR. EAF treatment resulted in significantly reduced dose-dependent expression of all of these factors, and enhanced expression of the antioxidants MnSOD and heme oxygenase-1. In addition, HPLC fingerprint results suggest that rutin, caffeic acid, and quercetin may be the active ingredients in EAF. CONCLUSIONS: Taken together, findings of this study imply that the anti-inflammatory effect of EAF on LPS-stimulated RAW264.7 cells is mediated by suppression of oxidative stress.

In vitro Anti-inflammation Effect of Adventitious Shoots of Toona sinesis in Propionibacterium acnes-induced Skin Dermatitis

  • Toona sinensis (TS) leaf is known to antinociceptive, antioxidative stress and skin moisturizing effects. Acnes vulgaris is a chronic skin disease with various symptoms including itchiness, pain and interruption of normal skin function. Propionibacterium acnes (P. acnes) is a major factor in the occurrence of inflammatory acnes. This study evaluated the antioxidant and anti-inflammation effects by TS extract from adventitious shoots. TS extract showed anti-inflammatory activities by suppression of pro-inflammation mediators (iNOS and COX-2) in LPS-stimulated RAW264.7 cells. TS extract also has anti-inflammatory activities by inhibiting the secretion of pro-inflammatory cytokines on P. acnes-stimulated HaCaT cells. These effects were regulated by MAPK signaling pathway. Therefore, we suggest that TS extract from adventitious shoots might have applications as a medicine for treating P. acnes-induced skin diseases.
