Effect of Medicinal Plant Water Extracts on Glucose-regulating Enzyme Activities in Goto-Kakizaki Rat Liver Cytosol (약용식물 물 추출물이 Goto-Kakizaki 흰쥐의 간 세포액에서 당대사 관련효소 활성에 미치는 영향)

  • We have studied the anti-diabetic effects of medicinal plant water extracts on hepatic glucose-regulating enzymes such as glucokinase (GK) and acetyl-CoA carboxylase (ACC). $\alpha$-Glucosidase inhibitor is usually used to prevent and treat type II diabetes; thus, anti-$\alpha$-glucosidase activity of medicinal plant water extracts was assayed. The hepatic cytosol faction of a type II diabetic animal (Goto-Kakizaki rat) was used in GK and ACC activity assays. The medicinal plants were Lycium chinense (JGP), Discorea japonica Thunb. (SY), Pyrus pyrifolia (YSB), Cornus officinalis (SSY), Paeonia suffruticosa ANDR. (MDP), Cordyceps militaris (DCH), and Acanthopanax senticosus (GSO). JGP, SY, YSB, and SSY water extracts increased the hepatic GK activity and all medicinal plant water extracts led to an increase in hepatic ACC activity. YSB, SSY, MDP, and GSO water extracts showed significantly higher anti-$\alpha$-glucosidase activity than control samples. The highest anti-$\alpha$-glucosidase activity was observed in GSO water extract and the anti-$\alpha$-glucoside activity was higher than that of Acarbose (reference $\alpha$-glucosidase inhibitor). We suggest that JGP, SY, YSB, and SSY water extracts may exert an anti-diabetic effect by enhancing the glucose metabolism and that YSB, MDP and GSO may be used as natural $\alpha$-glucosidase inhibitors in type II diabetic conditions. Increased ACC activity by plant water extracts may provide additional anti-diabetic effect.

Anti-diabetic Activity of Polysaccharide from Salicornia herbacea (함초 다당체의 항당뇨 활성)

  • The present study investigated the effect of physiologically active polysaccharide (SP1) isolated from Salicornia herbacea on streptozotocin-induced diabetic rats. Male Spraque-Dawley rats were divided into four groups which were normal control group (NC), diabetic control group (DC), diabetic CSP group (DCSP), and diabetic SP1 group (DSP1). Animals were administrated with 2% experimental drinks for 6 weeks. The levels of glucose, triglyceride, total cholesterol, and high density lipoprotein (HDL)-cholesterol in the serum were measured before and after intake of test compounds. The levels of glucose and triglyceride in the DSP1 were significantly lower than those in the DC by 25% and 20%, respectively. The levels of total cholesterol and high density lipoprotein (HDL)-cholesterol in the DSP1 were similar to those in the DC. These results suggest that SP1 substantially exhibit anti-hyperglycemic and anti-hypertriglyceridemic activity in diabetic rats. Therefore SP1 is believed to show remarkable anti-diabetic effect on streptozotocin-induced diabetic rats.

Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits (산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

Antidiabetic Activity of IH-901 in db/db Mice (db/db 마우스에서 IH-901의 항 당뇨 활성)

  • The pharmacological properties of ginseng are mainly attributed to ginsenosides, the active constituents that are found in the extracts of different species of ginseng. Lately; the studies on ginsenosides are mainly focused on IH-901, a major intestinal bacterial metabolite of ginsenosides. In this study; we examined the anti-diabetic activity of IH-901 in C57BU61 db/db mice model. IH-901 was administrated orally at a dose of 20 mg/kg for 5 weeks. During the experimental period, body weight and blood glucose levels were measured every week. After 5 weeks, db/db mice were sacrificed and diabetic parameters were analyzed. IH-901 treated group showed a significant decrease in fasting blood glucose levels (from 10.5 mM to 9.4 mM), insulin resistance index (from 163.6 to 100.2) and triglyceride levels (from 115.3 to 70.1) compared to the diabetic control. In Pancreatic islets morphology; IH-901 treated group revealed much less infltrated mononuclear cells, indicating that IH-901 recovered ${\beta}$-cell damage due to hyperglycemia. In addition, IH-901 upregulated expressions of glucose transporter 4 (GLUT4) and PPAR-${\gamma}$ in skeletal muscle and adipose tissue, respectively. Taken together IH-901might be a potential anti-hyperglycemic agent with insulin sensitizing effect.

Effects of Herb-Combined Remedy(HCR) on Blood Glucose Levels and Diabetic Nephropathy in STZ-induced Diabetic Rats (Streptozotocin 에 의해 유발된 당뇨병 쥐의 혈당과 당뇨병성 신증에 한약복합처방이 미치는 효과)

  • Diabetes mellitus is a worldwide epidemic. Global projections suggest that most nations will have a doubling of the incidence of Diabetes mellitus(DM) within 20years. The Herb-Combined Remedy(HCR) of DM was known as one of anti-hyperglycaemic agents. But it is unclear whether HCR can be used to treat patients with Diabetic nephropathy or not. The present study was designed to investigate the anti-hyperglycaemic and preventive effects from diabetic nephropathy of HCR in STZ-induced diabetic rats. Treatment with HCR regulated blood glucose levels, but did not affect body weights. In addition, HCR was able to decrease blood glucose levels after 120min. Serum BUN levels were not effective but, serum Creatinine levels were lowered by treatment with HCR or MF. In histopathological observation of renal tissue, both HCR and MF groups showed decreased tendency of inflammation and renal injury. In conclusion, these results demonstrate that HCR is effective to treat patients with DM and also has preventive activity on diabetic nephropathy.

Anti-lipase and Lipolytic Activities of EtOH Extract from Juniperus rigida (노간주나무 에탄올추출물의 지방 흡수 억제 및 지방분해 효능)

  • Obesity is an important risk factor that significantly increases mortality and disease rates in the cardiovascular disease, diabetes, and various diseases. So far, the most powerful way to inhibit fat absorption is pancreatic lipase inhibitors. In this study, we investigated the anti-obesity effect of the extract of Juniperus rigida. Juniperus rigida extract (JRE) had a inhibitory effect on pancreatic lipase activity ($IC_{50}$=8.63 ${\mu}g$/ml). In in vivo oil-emulsion loading test, this extract also inhibited the intestinal fat absorption. In addition, we measured inhibitory effects of JRE on activity of phosphodiesterase (PDE) and hormone sensitive lipase (HSL) among the important enzymes associated with lipolysis. JRE strongly inhibited PDE activity ($IC_{50}$=4.56 ${\mu}g$/ml), whereas inhibitory effect on HSL activity was very weak compared with orlistat. As a result, JRE inhibited the absorption of fat by inhibiting the activity of pancreatic lipase and induced lipolysis through inhibition of PDE activity. Therefore, we suggest that Juniperus rigida may be a potential therapeutic agent improving obesity.

Effects of Gamma-irradiation on Anti-diabetic and Cytotoxic Activities of Heat-treated Mistletoe (Viscum album) (감마선 조사가 열처리 겨우살이의 항당뇨 및 세포독성에 미치는 영향)

  • Mistletoe (Viscum album) has been widely used as a functional food material for various therapeutic purposes from ancient time. In this study, we examined anti-diabetic and cytotoxic activities of heated-treated mistletoe and the effects of gamma-irradiation on its activities. Heat-treated mistletoe extract was prepared by heating during different time (3, 6, 9 and 12 h) and gamma-irradiated with different doses of 0, 10, 30, 50, 70 and 100 kGy. Heat-treated mistletoe extracts showed a concentration-dependent cytotoxicity on rat insulinoma RINm5F cells and the effect was gradually decreased as heating time increased up to 12 h. 12 h heat-treated extract was no cytotoxic. Gamma-irradiation enhanced the reduction of heat-treated mistletoe-induced cytotoxicity and the decreasing effect was an irradiating dose-dependent. In particular, all of 70 kGy irradiated and heat-treated mistletoe extracts did not showed the cytotoxicity and the effect was comparable to 12 h heat-treated mistletoe extract. Among those extracts, 3 h heat-treated mistletoe extract gradually increased the insulin secreting activity by gamma-irradiation and the effect was the best at 70 kGy, whereas 12 heat-treated extract was no effect. On the test of ${\alpha}$-glucosidase inhibitory activity, 3 h heat-treated mistletoe extract showed the concentration dependent effects and gamma-irradiation induced more activity at 70 kGy, compared to non-irradiated 3 h and 12 h heated mistletoe extracts. These results suggest that the combination of heat treatment and gamma-irradiation might be more effective than only heat-treatment for improving the anti-diabetic activity of mistletoe extract and reducing its cytotoxicity.

  • White jelly fungus (Tremella fuciformis Berk; TF) has been used as a traditional medicine in Asia; it is known to prevent hypertension, aging, cancer, and arteriosclerosis. This study aimed to investigate the anti-diabetic effects of fermented Tremella fuciformis Berk (FTF) ethanol extracts fermented with L. rhamnosus BHN-LAB 76. We show that FTF increases the ${\alpha}$-glucosidase inhibitory activity and suppress the adipogenesis of 3T3-L1 adipocytes. These inhibitory effects of FTF are accompanied by the regulation of the phosphorylation of AMPK, JNK, and Akt. These data demonstrate that FTF not only inhibits adipogenesis by affecting the adipogenic signaling, but also increases the anti-diabetic effects by regulating the insulin signaling pathway. Therefore, we suggest that the FTF can be used for developing functional food and cosmetics materials.

  • Cudrania tricuspidata have been used for anti-inflammatory, anti-hepatotoxic, anti-hypertensive and anti-diabetic activities. In this study, in order to investigate the efficacy of antioxidant activity, the bio-activity guided fraction and isolation of physiologically active substance were performed. H$_2$O, 30%, 60%, 100% MeOH and acetone fractions were examined antioxidant activity by DPPH method. It was revealed that 30%, 60%, 100% MeOH frations have significantly antioxidant activity. Anti-diabetic Effects of CCCA, CMESS, and Cordycepin from Cordyceps militaris and the Immune Responses in Streptozotocin-induced Diabetic Mice

  • Anti-diabetic effect of various fractions of Cordyceps militaris (CM), CCCA (crude cordycepin containing adenosine), CMESS (ethanol soluble supernatant), and cordycepin were evaluated in streptozotocin (STZ) induced diabetic mice, CMESS showed potent inhibitory activity of 34.7% in starch-loaded mice (2 g/kg) while acarbose as a positive standard exhibited 37.8% of inhibition rate. After 3 days administration (50 mg/kg), cordycepin (0.2 mg/kg), and acarbose (10 mg/kg) dramatically reduced blood glucose level (inhibition ratio: 46.9%, 48.4% and 37.5% respectively). CCCA that has high contents of cordycepin (0.656 mg/4 mg) did not influence on reducing blood glucose level. The proliferation of splenocytes and peritoneal macrophages derived from STZ-induced diabetic mice administered samples were evaluated out by addition of mitogens to see the stability of the usage of these herbal medicines. Proliferation of T-lymphocyte was significantly decreased; while NO production was increased more than two fold to STZ control in the cordycepin-administered group. Changes of serum enzyme levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were also evaluated. Cordycepin administered group was appeared to acarbose. We conclude that CMESS and cordycepin may be useful tools in the control of blood glucose level in diabetes and promising new drug as an anti-hyperglycemic agent without defects of immune responses and other side effects.
