Occurrence and Distribution of Crop Diseases Caused by Invasive Alien Pathogens in Korea (외래침입 병원체에 의한 작물 병 발생 및 분포)

  • Occurrence and distribution of crop diseases caused by invasive alien pathogens were surveyed in Korea from 2001 to 2003. Of 22 species of invasive alien pathogens surveyed, 18 species were found on their host plants but 4 species were not. Scientific names of the alien plant pathogens introduced into Korea in the past were reviewed. Revised scientific names of 10 species including anthracnose pathogens were amended in the list. Host ranges of invasive alien pathogens were investigated. Eight species of the invasive alien pathogens were found on new host plants as well as their host plants recorded in the past.

Identification, Mycological Characteristics and Response to fungicides of Anthracnose Pathogen Isolated from Pepper and Boxthorn in Cheongyang (청양 지역 고추와 구기자에서 분리한 탄저병균의 동정, 균학적 특징 및 살균제 저항성)

  • It was conducted to identify all 47 isolates obtained from infected fruits of pepper and boxthorn, and to investigate the mycological characteristics and the response to fungicides. All of 11 isolates from pepper were identified as Colletotrichum acutatum included into A2 group. Among 36 isolates from boxthorn, 14 isolates were identified as C. gloeosporioides, and the others were done as C. acutatum, which were composed as A1 group with 15 isolates and A3 with 7 isolates. After incubating the isolates on PDA at $25^{\circ}C$ for 10 days, the colony color of C. acutatum was greyish white, while that of C. gloeosporioides was orange at center of colony and was gradually turned into an greyish white to the periphery. The rate of conidia showing ellongated ellipsoidal shape with round ends was over 95% in C. acutatum isolated from pepper. However, C. acutatum isolated from boxthorn produced ellongated ellipsoidal conidia with the rate of 75%, and the others were pointed at one or both ends. Regardless of species of Colletotrichum, all isolated used in this study was showed an optimal temperature at $25^{\circ}C$. $EC_{50}$ values of all isolates of Colletotrichum spp. to 2 fungicides as carbendazim and the mixture of carbendazim and diethofencarb was investigated by an agar dilution method. With C. acutatum isolates from pepper belonged to A2 group, the mean of $EC_{50}$ value to carbendazim and the mixture of carbendazim and diethofencarb was 0.68 and $3.16{\mu}g/ml$, respectively. In the case of C. acutatum isolates from boxthorn, which were divided into 2 groups as A1 and A3 group, that to carbendazim was 0.21 at A1 and $0.24{\mu}g/ml$ at A3, while that to the mixture was 1.52 and $3.35{\mu}g/ml$. Isolates of C. gloeosporioides showed the mean of $EC_{50}$ value was $0.12{\mu}g/ml$ to carbendazim and $0.92{\mu}g/ml$ to the mixture. The value of resistant factor was higher in the isolates of C. acutatum obtained in boxthorn than those from pepper.

Studies on the Host Range of Colletotrichum dematium Isolated from Anthracnose of Pepper and Toxic Metabolites Produced by the Pathogen (고추 탄저병균(炭疽病菌) Colletotrichum dematium의 기주범위(寄主範圍) 및 대사독소(代謝毒素)에 관(關)한 연구(硏究))

  • This studies were conducted to investigate pathgenicity and host range of Colletotrichum dematium isolated from anthracnose of pepper, and phytoxicity of its culture filtrate and the partially purified toxin. The results obtained were as follows. 1. Investigation on the host range of C. dematium has revealed that pepper as well as soybean, tomato, spinach, and beet were highly susceptible, egg plant and water melon were moderately susceptible and stone leek was slightly suceptible, but no symptoms were produced on carrot, tabacco, cucumber and melon. 2. The culture filtrates of C. dematium in Czapeck dox liquid media were toxic to leaves of pepper and caused necrosis and wilting of the plant. The toxicity of culture filtrates was most active at 15 days after fungal growth in Czapeck dox liquid media and the toxin productivity in still culture was higher than that in shaking culture. 3. The partially purified toxic substance was isolated from the culture filtrates by the acetone precipitation method. When cuttings of various pepper cultivars were placed in the toxin solutions, suceptible cultivars and resistant cultivars were equally toxic and showed necrosis and wilting of the leaves. 4. Several other plants such as soybean, tomato and carrot were also affected with the toxin solution by shoot cutting bioassay and showed veinal necrosis, leaf spots and wilting of the shoots. 5. The acetone precipitation toxin affected seed germination of pepper, cucumber, sesame and egg plant and inhibited the growth of root and hypocotyl of the seedlings.

Effective Heat Treatment Techniques for Control of Mung Bean Sprout Rot, Incorporable into Commercial Mass Production

  • Seedlot disinfection techniques to control mung bean sprout rot caused by Colletoricum acutatum and C. gloeosporioides were evaluated for commercial production scheme. Soaking seedlots in propolis (100 X) and ethanol (20% for 30 min) appeared promising with control values of 85.5 and 80.8 respectively, but still resulted in up to 20% rot incidence. None of the C. acutatum conidia survived through hot water immersion treatment (HWT) for 10 min at temperatures of 55, 60 and $65^{\circ}C$, whereas the effective range of the dry heat treatment (DHT) was $60-65^{\circ}C$. Tolerance of mung bean seedlot, as estimated by hypocotyl elongation and root growth, was lower for HWT than for DHT. Germination and growth of sprouts were excellent over the range of $55-65^{\circ}C\;at\;5^{\circ}C$ intervals, except for HWT at $65^{\circ}C$ for 5 min. At this marginal condition, heat damage appeared so that approximately 2% of seeds failed to sprout to normal germling and retarded sprouts were less than 5% with coarse wrinkled hypocotyls. These results suggested that DHT would be more feasible to disinfect mung bean seedlots for commercial sprout production. Heat treatment at above ranges was highly effective in eliminating the epiphytic bacterial strains associated with marketed sprout rot samples. HWT of seedlot at 55 and $60^{\circ}C$ for 5 min resulted in successful control of mung bean sprout rot incidence with marketable sprout quality. DHT at 60 and $65^{\circ}C$ for 30 min also gave good results through the small-scale sprouting system. Therefore, we optimized DHT scheme at 60 and $65^{\circ}C$ for 30 min, considering the practical value of seedlot disinfection with high precision and accuracy. This was further proved to be a feasible and reliable method against anthracnose incidence and those bacterial strains associated with marketed sprout rot samples as well, through factory scale mung bean sprout production system.

Antifungal Activity of Decursinol Angelate Isolated from Angelica gigas Roots Against Puccinia recondita (당귀로부터 분리한 decursinol angelate의 밀 붉은녹병에 대한 항균활성)

  • Rust causes significant losses in the yield and quality of various crops. The development of new effective and environmentally benign agents against the pathogen is of great interest. In the course of searching a natural antifungal compound from medicinal plants, we found that the methanol extract of Angelica gigas roots had a potent control efficacy against wheat leaf rust (WLR) caused by Puccinia recondita. The antifungal substance was isolated from the methanol extract by silica gel column chromatography, alumina column chromatography and $C_{18}$ preparative HPLC. It was identified as decursinol angelate by EI-MS and $^1H$-NMR data. In in vivo test, decursinol angelate effectively suppressed the development of WLR and red pepper anthracnose (RPA) among the 6 plant diseases tested. In addition, the wettable powder-type formulation of ethyl acetate extract of A. gigas roots significantly suppressed the development of WLR. The crude extract containing decursinol angelate and the chemical appear to be a potential candidate for control of WLR. In addition, this is the first report on the in vivo antifungal activity of decursinol angelate against WLR as well as RPA.

History of Disease Control of Korean Ginseng over the Past 50 Years (과거 50년간 고려인삼 병 방제 변천사)

  • In the 1970s and 1980s, during the nascent phase of ginseng disease research, efforts concentrated on isolating and identifying pathogens. Subsequently, their physiological ecology and pathogenesis characteristics were scrutinized. This led to the establishment of a comprehensive control approach for safeguarding major aerial part diseases like Alternaria blight, anthracnose, and Phytophthora blight, along with underground part diseases such as Rhizoctonia seedling damping-off, Pythium seedling damping-off, and Sclerotinia white rot. In the 1980s, the sunshade was changed from traditional rice straw to polyethylene (PE) net. From 1987 to 1989, focused research aimed at enhancing disease control methods. Notably, the introduction of a four-layer woven P.E. light-shading net minimized rainwater leakage, curbing Alternaria blight occurrence. Since 1990, identification of the bacterial soft stem rot pathogen facilitated the establishment of a flower stem removal method to mitigate outbreaks. Concurrently, efforts were directed towards identifying root rot pathogens causing continuous crop failure, employing soil fumigation and filling methods for sustainable crop land use. In 2000, adapting to rapid climate changes became imperative, prompting modifications and supplements to control methods. New approaches were devised, including a crop protection agent method for Alternaria stem blight triggered by excessive rainfall during sprouting and a control method for gray mold disease. A comprehensive plan to enhance control methods for Rhizoctonia seedling damping-off and Rhizoctonia damping-off was also devised. Over the past 50 years, the initial emphasis was on understanding the causes and control of ginseng diseases, followed by refining established control methods. Drawing on these findings, future ginseng cultivation and disease control methods should be innovatively developed to proactively address evolving factors such as climate fluctuations, diminishing cultivation areas, escalating labor costs, and heightened consumer safety awareness.
