Antagonistic Activity of Polyfermenticin SCD Against Helicobacter pylori KCTC 2948

  • Bacillus polyfermenticus SCD and polyfermenticin SCD, named tentatively as the bacteriocin produced by B. polyfermenticus SCD, showed antimicrobial activity against Helicobacter pylori KCTC 2948 growth. When crude polyfermenticin SCD was added to the growing H. pylori cells, viable cell numbers were reduced, indicating antimicrobial action. The antimicrobial effect was increased remarkably at higher concentrations of polyfermenticin SCD and longer exposure. Morphological changes were observed in the bacteriocin-treated cells; in the exponential phase, they appeared as shrunken rods, while in the stationary phase, they showed coccoid forms.

  • Antibiotic and physico-chemical properties of an active compound from actinomycetes isolate G-37, of which the culture filtrate had an inhibitory effect against tobacco mosaic virus(W) infection, were examined. The active compound, which was purified by ethylacetate extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography, showed strong antibacterial activities especially against Gram-positive bacteria including Bacillus subtillis, Sarcina lutea and Staphylococcus aureus. From the IH-NMR, FAB/RfS, UV spectral data, and physicochemical properties, the active compound of G-37 appears to belong to a peptide antibiotic group. Among the known peptide antibiotics in the antibiotic group, No. 280, A-30912, and Taitomycin showed molecular weights and ultra violet spectrum similar to those of the active compound from G-37, but was not identical to the compound, which suggests that it may be a new peptide antibiotics.

Antagonistic Effect of Chitinolytic Bacteria on Soilborne Plant Pathogens (토양전염성 식물병원균에 대한 Chitin 분해세균들의 길항효과)

  • One hundred and thirty bacterial isolates with high chitinolytic activity on chitin agar media were isolated and identified. Most of the isolates were Aeromonas hydrophila (110 isolates), and the others were Serratia marcescens (11 isolates), Aeromonas caviae (3 isolates), Chromobacterium violaceum strain C-61 (2 isolates), Chromobacterium violaceum strain C-72 (1 isolate) and unknown species (3 isolates). Among them, C. violaceum strain C-61 had highest chitinolytic activity and fungal growth inhibition on PDA. This bacterium also inhibited the growth of Rhizoctonia solani, Sclerotinia scelrotiorum, Phytophthora capsici and Pythium ultimum, but it didn't inhibit the growth of Fusarium oxysprum and Fusarium solani. C. violaceum strain C-61 suppressed damping-off of eggplant caused by R. solani. Populations of the chitinolytic bacteria such as Aeromonas hydrophila, Serratia marcescens, Aeromonas caviae, Chromobacterium violaceum strain C-61 and Chromobacterium violaceum strain C-72 introduced into R. solani-infested soil were continuously decreased until 20 days after treatment, but their populations except A. caviae were not changed significantly and maintained over 5$\times$104 CFU per g of soil thereafter.

  • A novel series of aryl sulfonylpiperazine derivatives (5-15) were synthesized as 5-$HT_6$ ligands. In vitro assay was evaluated by measuring the 5-HT-induced $Ca^{2+}$ increases using HeLa cell line expressing the cloned human 5-$HT_6$ receptor, and the compound 13 showed potent 5-$HT_6$ receptor antagonistic effect with $IC_{50}$ value of 3.9 ${\mu}M$. Compound 13 also showed good selectivity on the 5-$HT_6$ over 5-$HT_4$ and 5-$HT_7$ receptors.

Antifungal Activity of an Endophytic Fungus Aspergillus versicolor DYSJ3 from Aphanamixis grandifolia Blume against Colletotrichum musae

  • An endophytic fungus strain DYSJ3 was isolated from a stem of Aphanamixis grandifolia Blume, which was identified as Aspergillus versicolor based on the morphological characteristics, internal transcribed spacer (ITS) and calmodulin gene sequences analyses. A. versicolor DYSJ3 exhibited strong antagonistic activity against Colletotrichum musae, C. gloeosporioides and Fusarium oxysporum f. sp. cubense with the inhibition rates of 61.9, 51.2 and 55.3% respectively. The antifungal metabolites mainly existed in the mycelium of A. versicolor DYSJ3, and its mycelial crude extract (CE) had broad-spectrum antifungal activities against plant pathogenic fungi. The CE had a good thermal stability, and the inhibition rate of 100 mg/mL CE against C. musae was above 70.0% after disposing at 120 ℃ for 1 h. Five secondary metabolites were isolated from the CE and identified as averufanin, ergosterol peroxide, versicolorin B, averythrin and sterigmatocystin. Activity evaluation showed versicolorin B exhibited inhibitory effects on the mycelial growth and conidial germination of C. musae, and sterigmatocystin had a weak inhibitory effect on the mycelial growth of C. musae.

Research Trend and Prospect in Ferromagnetic Superconductor (강자성 초전도체의 연구동향과 전망)

  • The findings of ferromagnetic superconductor have attracted much attention not only for fundamental research to investigate how the antagonistic properties of ferromagnetism and superconductivity coexist peacefully but also for potential technological applications. Firstly, in order to help for understanding the ferromagnetic superconductor, I have explained the orbital and paramagnetic pair-breaking effects of magnetic field, which breaks the superconducting Cooper pairs. In addition to such effects of magnetic field, the singlet Cooper pairs become unstable upon going through the ferromagnetic materials by the proximity effect. The proximity effect occurs at the interface of thin films composing of superconductor and ferromagnet and leads to have very short penetration depth of Cooper pairs. However, a type of odd-frequency triplet in comparison with the singlet could be very stable and has a longer effective depth. It needs to be explored for the innovative spintronic devices. Finally, various ferromagnetic superconductors coexist and the lower-dimensional materials under the Quantum confinement effect have been introduced.

Effect of Probiotic Clostridium butyricum NCTC 7423 Supernatant on Biofilm Formation and Gene Expression of Bacteroides fragilis

  • Enterotoxigenic Bacteroides fragilis (ETBF) is the main pathogen causing severe inflammatory diseases and colorectal cancer. Its biofilm plays a key role in the development of colorectal cancer. The objective of this study was to determine the antagonistic effects of cell-free supernatants (CFS) derived from Clostridium butyricum against the growth and biofilm of ETBF. Our data showed that C. butyricum CFS inhibited the growth of B. fragilis in planktonic culture. In addition, C. butyricum CFS exhibited an antibiofilm effect by inhibiting biofilm development, disassembling preformed biofilms and reducing the metabolic activity of cells in biofilms. Using confocal laser scanning microscopy, we found that C. butyricum CFS significantly suppressed the proteins and extracellular nucleic acids among the basic biofilm components. Furthermore, C. butyricum CFS significantly downregulated the expression of virulence- and efflux pump-related genes including ompA and bmeB3 in B. fragilis. Our findings suggest that C. butyricum can be used as biotherapeutic agent by inhibiting the growth and biofilm of ETBF.

  • Experimental studies were made to evaluate clinical efficacy and validate Oriental medicine description of 'Banhabaikchulchenma-Tang' which has been used for controlling of headache, heartburn and giddiness, etc. The results of these studies were as follows; 1) Suppressive actions were not shown on convulsion due to spine and diencephatic causes. 2) Analgesic action was recognized in mice and rats. 3) Sedative actions due to prolongation of hypnosis time, muscle relaxation and decreasing locomotive function were noted. 4) Relaxing action was noted in the ileum of mice, also antagonistic antispasmodic action was recognized on contraction of the ileum induced by acteylcholine and $BaCl_2$. In connection with results of these studies, efficacy based on the Oriental medical reference were consistant with actual experimental results. It is considered that 'Banhabaikchulchenma-Tang' has the therapeutic effect on the headache and giddiness, etc.

Effect of STEL-toluene on Metabolism of Benzene in Rats (단시간 허용농도의 toluene이 benzene대사에 미치는 영향)

  • Benzene and toluene, which are widely used aromatic hydrocarbons in workplace, are recently proved to cause health hazards due to their toxic effects. This study investigated the influence of toluene on the urinary excretion of benzene metabolite by administering short term exposure limit(STEL) of these compounds(i.e., 13.8mg/kg of benzene and 108.8 mg/kg of toluene) intraperitoneally into Sprague-Dawley rats. After administration, urinary phenol concentration of rat was measured by gas chromatography for every three hours. Data were analyzed by non-parametric statistical methods using Kruskal-Wallis multi-sample test and Mann-Whitney U test. The following results were obtained : 1. Administration of STEL-benzene increased urinary phenol concentration in lats. 2. Urinary phenol concentration was increased logarithmically according to the dosage of benzene. 3. Excretion of phenol in urine was decreased when benzene and toluene were administered simultaneously compared with administering benzene alone. In summary, these results reveal that administration of STEL of toluene has antagonistic effect of urinary excretion of benzene metabolite in rats.
