Determination of Glyphosate in Whole Blood by HPLC-fluorescence Detection (HPLC 형광검출법에 의한 Glyphosate의 혈중농도 측정)

  • A rapid and sensitive method for the determination of glyphosate, a phosphated amino acid herbicide, in whole blood is presented. After removal of protein, the whale blood was purified by using the anion exchange resin (Dowex 1), and derivatized with 9-fluorenylmethyl chloroformate (FMCL). Derivatized glyphosate from blood sample was injected onto a Whatman partisil 10SAX column and separated with 0.1M phosphate buffer (pH 2.5) and acetonitrile (ratio=3:1). The high performance liquid chromatography-fluorescence detection gave the detection limit of 86pg and linearity of 0.9999 in the range of 0.25 $\mu$g/ml and 25 $\mu$g/ml. The recoveries of glyphosate added to the blood samples were ranged from 75.3% to 100.4% compared to the samples prepared in water. The derivatized glyphosate was stable at various acidity and temperature. This method has been successfully applied to the blood samples of lethal intoxication with the herbicide glyphosate.

  • The anion exchange separation of a number of benzoic acid and its derivatives has been studied in various concentrations of $NiCl_2-methanol$ solvent medium. The volume distribution coefficients with Amberlite CG-400, $Cl^-$form have been measured in these medium. Several synthetic mixtures of organic acids were separated quantitatively by elution with a suitable concentration of $NiCl_2-MeOH$ solution suggested by the distribution data. The concentration of organic acids were determined by UV spectrophotometry.

Analysis of Angiotensin I Converting Enzyme Inhibitory Activity of Oligosacchride Extracted from Capsosiphon fulvescens (매생이 유래 올리고당의 추출 분리 및 Angiotensin I Converting Enzyme 저해능 분석)

  • The hydrolysates prepared with various enzyme digestion of Capsosiphon fulvescens were used to measure the inhibitory effects against angiotensin I converting enzyme (ACE). The commercially available enzymes such as Celluclast, Viscozyme, Lysing enzyme, Flavourzyme, Alcalase and Pectinex were used to digest C. fulvescens and produce hydrolysates. The maximum ACE inhibitory activity was observed using Alcalase hydrolysis (72.9%). The optimal conditions of Alcalase extraction were pH 8.0 and extraction time for 12 hr. The hydrolysates were fractionated using preparative-LC and anion-exchange chromatography on DEAE-cellulose and the fraction B and B-2 were isolated. The ACE inhibitory activity of fraction B-2 by anion-exchange chromatography was 82.6%. The molecular weight of fraction B-2 estimated using size exclusion chromatography was about 1 kDa. The monosaccharide composition of the fraction B-2 was determined to be mannose (1.1%), glucuronic acid (1.3%), galactose (1.3%) and glucose (96.3%).

Anionic Indicators on the Surface of Submicrospheres Consisting of Ionic Palladium(II) Complex

  • Ionic palladium(II) complex, [($Me_4en)Pd(L)]_2(ClO_4)_4\;(Me_4$en = N,N,N',N'-tetramethylethylenediamine; L = bis-(4-(4-pyridylcarboxyl)phenyl)methane), allows to form a monodisperse submicrosphere without any template or additive. Surface-perchlorates on the submicrosphere have been exchanged by anionic pH indicators such as thymol blue, bromothymol blue, and bromocresol green. The ionic and amphiphilic properties of the palladium(II) complex appear to be primarily associated with the formation and easy surface-anion exchange of submicrosphere. The surface-anion exchange through the electrostatic interaction is a very good tool for the surface-modification. The color of the pH indicator-exchanged submicrospheres is very sensitive to pH and Hg$^{2+}$ cation, and thus they are promising to submicrospherical indicators.

Modeling of continuous diffusion dialysis of aqueous solutions of sulphuric acid and nickel sulphate

  • At steady state, the simultaneous transport of sulphuric acid and nickel sulphate through an anion-exchange membrane Neosepta-AFN (Astom Corporation, Tokyo, Japan) was investigated in a two-compartment counter-current dialyzer with single passes. The transport was quantified by the recovery yield of acid, rejection of salt and four phenomenological coefficients, which were correlated with the acid and salt concentrations in the feed. The phenomenological coefficients were determined by the numerical integration of the basic differential equations describing the concentration profiles of the components in the dialyzer. This integration was combined with an optimizing procedure. The experiments proved that the acid recovery yield is in the limits from 63 to 91 %, while salt rejection is in the limits from 79 to 97 % in the dependence on the volumetric liquid flow rate and composition of the feed.

  • Interactions between thiamine.HCl and its disulfide derivatives TTFD. TPD and .alpha., .betha. cyclodextrins were investigated. By measuring the H-NMR, C-NMR chemical shifts, the assumption that cyclodextrin may from a inclusion complex with thiamines was supported qualitatively. To calculated the stability constants of them, anion exchange chromatography was applied. The simple, rapid HPLC method was proved to be pertinent thiamine/cyclodextrin system which was chemically unstable and less soluble.

Direct Observation of an Antihomoaromatic Bicyclooctadienyl Cation

  • The question of the bicyclohomoaromatic stabilization and destabilization is examined. The chemistry of bicyclo(3.2.1)octa-3,6-dienide anion has been studied on order to test these concepts. The bicyclooctadienide anion is shown to be stable delocalized ion which undergoes a facile proton-deuterium exchange reaction. The solvolysis of bicyclo(3.2.1)octa-3,6-dienyl p-nitrobenzoate is much slower than the monoene analog. We have made direct observation of the bicyclooctadienyl and octenyl cations by $^{19}F$-nmr spectroscopy, and were able to demonstrate that the bicyclooctadienyl cation was bishomoantiaromatic.
