A Study on the Hypotensive Action of Astragali Radix Water Extract in the Rabbit (황기의 혈압강하 작용에 관한 연구)

  • Effects of Astragali Radix Water Extract (ARWE) on the blood pressure were investigated in the whole rabbit and the spinalized rabbit. ARWE, when administered into the ear-vein or lateral ventricle, produced a fall in the blood pressure in the whole rabbit, but intravenous ARWE in the spinalized rabbit did not elicite the hypotensive action. Pretreatments with chlorisondamine, guanethidine, phentolamine and cyproheptadine in the whole rabbit weakened the depressor action of ARWE. The hypotensive action of the whole rabbit to ARWE was not influenced by the pretreatment of the animals with diphennylhydramine, propranolol, and vagotomization, whereas inhibited by atropine. ARWE did not affect the pressor response by angiotensin. However, it enhanced the hypertensive action by norepineprine and reduced the elevation in the blood pressure by carotid occlusion in the whole rabbit. These experimental observations suggest that ARWE may cause the depressor response via mechanisms of the central sympathetic blocking action, cholinergic action by peripheral origin and serotonin-like action.

Comparison of serum and pancreatic enzyme activities in serval vertebrates (수종 척추동물의 췌장및 혈청효소의 비교생화학적연구)

  • The levels of activities of amylase, lipase and trypsin in both the pancreatic tissue and serum of 18 species of vertebrate animals were measured and enzymologically compared with each other. 1) The value of amylase in the pancreas of experimental mammalia has been found decreasing in the order pig, rat, dog, cat, rabbit and cow; that of pancreatic lipase has been found decreasing in the order of pig, dog, cat, rat, rabbit and cow; and that of trypsin has been found decreasing in the order of pig, cow, dog, rat, rabbit. Thus the value of all the above three kinds of enzymes were observed highest in pig, but in cow amylase and lipase were observed lowest while trypsin were observed considerably high. 2) In view of diets, the comparatively high values of pancreatic enzyme were observed in the ommivorous animals such as pig, rat, dog, while the values observed low in the herbivorous animals, such as cow and rabbit. 3) In the bovine, the values were observed moderately high except lipase which were found comparatively low. 4) In the Reptilia and Amphibia such a mud turttle and frog, the values were shown in similar measure with each other, that is, the pancreatic amylase and trypsin were observed considerably high while the lipase was found low. 5) In the species of Reptilia such as a viper and snake, the activities of pancreatic enzymes were not detected. But in the tissue of liver, stomach, activities of the enzymes were found considerably high. Lacertilia animals such as lizard the values of pancreatic enzymes were little observed. 6) In the fish in which the pancreatic tissue is scattered in the liver, the pancreatic enzymes were found in the liver tissue considerably higher than in the other tissues but lower than in the warm-blooded animals, especially the lipase was lower. 7) In generally the values of serum amylase and lipase were observed higher than those of man; and even in the cold-blooded animals in which the values of pancreatic enzymes were shown low or none, the values were also observed high. 8) The above three kinds of pancreatic enzyme values of those experimental animals have shown a tendency of higher degree in higher taxa than in lower taxa according to taxonomical order. 9) In view of tissue, the pancreatic cell was observed large in the mammalian animals such as rat and pig and cytoplasm was also abundantly contained in the acinous cell; and the bovine and the snake haave the pancreatic cells of the similar rosette form the comparatively large acinous cells of long rhombic form in the comparatively large acinous cells of long rhombic form in which the spindle shaped neucleus and the abundant cytoplasm were contained. In the fish the pancreatic cell were found scattered in the liver in which the very large pancreatic islet were found.

  • The study of irritation and toxicity of honeybee(Apis mellifera. L) venom collected by a bee venom collector applied topically to the skin and mucous membrane were carried out to prove the safety of honeybee venom in clinical use. Animal for the research was the rabbit and the solution for the test was made from honeybee venom. Six animals were used for the skin test and nine animals were used for the eye mucous membrane test. In results, both tests proved that honeybee venom makes no irritable reaction on skin and eye mucous membrane of rabbit. We consider that this result is helpful for saying about the safety of honeybee venom in clinical use.

Comparative Study of Histological Structure in the Tongue Root of Domestic Animals (각종 가축 설근부의 조직학적 구조에 대한 비교 연구)

  • In order to know the comparative histological structure of the tongue of some domestic and laboratory animals, the root of the tongue has been investigated by light microscopy. The filiform papillae were mainly distributed in the root of the tongue of most animals and many foliate papillae were also found in the dog and rabbit. On the other hand, only filiform papillae were found in the fowl. The very well-developed mucosa was found in the cattle and in the dog, fowl, rat and mouse was fair. The lamina propria was well developed in the cattle, dog and pig, The muscle layer was well developed in the cattle. The adipose tissue which distributed between the muscle fibers was mainly found in the dog. The lingual glands were well developed in the pig, fowl and rabbit. The properties of these glands were shown strung acid mucopolysaccharide in the dog and pig, weak acid mucopolysaccharide in the fowl, rabbit and rat and neutral or acid mucopolysaccharide in the mouse. The species differences on the histological structure of the tongue were also ascertained by light microscopical observations.

Immunocytochemical Localization of Nitric Oxide Synthase-containing Neurons in Mouse and Rabbit Visual Cortex and Co-Localization with Calcium-binding Proteins

  • Nitric oxide (NO) occurs in various types of cells in the central nervous system. We studied the distribution and morphology of neuronal nitric oxide synthase (NOS)-containing neurons in the visual cortex of mouse and rabbit with antibody immunocytochemistry. We also compared this labeling to that of calbindin D28K, calretinin, and parvalbumin. Staining for NOS was seen both in the specific layers and in selective cell types. The densest concentration of intense anti-NOS immunoreactive (IR) neurons was found in layer VI, while the weak anti-NOS-IR neurons were found in layer II/III in both animals. The NOS-IR neurons varied in morphology. The large majority of NOS-IR neurons were round or oval cells with many dendrites coursing in all directions. Two-color immunofluorescence revealed that only 16.7% of the NOS-IR cells were double-labeled with calbindin D28K in the mouse visual cortex, while more than half (51.7%) of the NOS-IR cells were double-labeled with calretinin and 25.0% of the NOS-IR cells were double-labeled with parvalbumin in mouse. By contrast, 92.4% of the NOS-IR neurons expressed calbindin D28K while only 2.5% of the NOS-IR neurons expressed calretinin in the rabbit visual cortex. In contrast with the mouse, none of the NOS-IR cells in the rabbit visual cortex were double-labeled with parvalbumin. The results indicate that neurons in the visual cortex of both animals express NOS in specific layers and cell types, which do not correlate with the expression of calbindin D28K, calretinin or parvalbumin between the two animals.

  • Effects of Akebiae Lignum, whose scientific name is Akebia quinata Decaisne, on the blood pressure were investigated with EtOH extract in whole and spinal rabbits. Akebia Lignum EtOH extract (AEE), when given intravenously, produced a fall in blood pressure not only in whole rabbit but also in spinal rabbit and AEE administered into a lateral cerebral ventricle of whole rabbit did not elicit a fall in blood pressure. The depressor responses of the whole rabbit to intravenous AEE were weakened by treatment of the animals with atropine and chlorisondamine but not by vagotominijation, phentolamine avil, and then the depressor action causing by AEE in the whole rabbit was not affected by pretreatment of physostigmine which is cholinesterase inhibitor and of hemicholinium which blocks acetylcholine synthesis by interfering with choline uptake in nerves. These observations suggest that the hypotensive action of AEE of which component is not affected by cholinesterase is due to direct action at parasympathetic receptor.

Pathological Studies on Experimental Poisoning of Organic Phosphate Insecticide (유기인제 중독에 관한 병리학적 연구)

  • The authors believe that farm livestok will be greatly affected by the marked increasing use of organic phosphate. This study was carried out to observe the clinical signs and histopathological changes of mouse, guinea pig, hamster and rabbit that were orally administered with diazinon used usually as agricultural insecticide, and cholinesterase (ChE) activity was histochemically examined in the liver, heart, kidney, adrenal gland, duodenum and salivary gland of these experimental animals administered with diazinon. The results obtained were as follows: 1. Clinical signs such as dullness, severe salivation, ataxia, dyspnea, irregular slight convulsion and inappetance and as the histopathological changes cloudy swelling, congestion and hemorrhage of parenchymal organs, catarrh or local necrsois of the gastrointestinal tract, congestion or hemorrhage of the other organs were observed. Especially, hemorrhage of adrenal glands (rabbit, guinea pig) and pulmonary congestion and hemorrhage were necessarily constant. 2. In the histochemical study, ChE activity appeared intensely in the liver, heart, medulla of adrenal glands and salivary glands (submaxillary and parotid) of control animals, but ChE activity was negative or markedly decreased in experimental animals administered with diazinon. There was no marked difference between the control and experimental animals in ChE activity of the kidney. 3. Histochemical observation of ChE activity was helpful to explain the clinical signs and histopathological changes and was regarded as a diagnostic method for organic phosphate poisoning.

Intraosseous Phlebography for Determination of the Effect of Vitamin D$_3$ and Calcium Phosphate on Fractured Bone Healing in Rabbit (비타민 D$_3$와 인산칼슘의 토끼 대퇴골 골절치유 효과에 대한 골수강내 정맥 조영술)

  • Intraosseous phlebography was performed to determine the healing effect of the dosing treated with vitamin-D$_3$ and/or calcium phosphate on the fractured bone in male rabbits. Femoral shafts of 16 adult male rabbits were fractured and these animals were divided into 3 treated groups and 1 control group l. The 1st treated group of rabbits was dosed with vitamin-D$_3$(VD), the 2nd treated group of rabbits was given calcium phosphate(CP), and the 3.d treated group of rabbits received vitamin-D$_3$ and calcium phosphate. The control group of rabbits was dosed with saline alone. Radiographs were taken weekly in each rabbit over 6 weeks period and the results obtained were as follows. 1. Sinusoidal vein was visualized at 3rd week in eve animal received CP and VC, at 5 week in animals received VD and in control animals. 2. Main nutrient vein was observed more clearly in treated groups than in control group of animals. 3. These results indicate that venous reconstruction of the fractured bone in groups of rabbits received CP and VC was earlier and more complete than those of other groups of rabbits. 4. Therefore, intraosseous phlebography is suitable for determination of the bone healing effect of vitamin-D$_3$ and/or calcium phosphate in femoral fractured rabbits.

Effects on intraventricular norepinephrine on blood pressure and heart rate of rabbits (측뇌실내(側腦室內) Norepinephrine의 가토심박(家兎深博) 급(及) 혈압(血壓)에 미치는 영향(影響))

  • Effects of intraventricular norepinephrine (NE) on rabbit blood pressure and heart rate were investigated. 1) Blood pressure was little affected by small doses of NE (below $500{\mu}g$) but showed marked rise by 1 mg. 2) Heart rate was decreased by intraventriccular NE $(200{\sim}500{\mu}g)$. One mg of NE caused less pronounced bradycardia than with smaller doses. The bradycardia could not be observed in vagotomized or atropinized animals. 3) Intraventricular NE potentiated reflexive bradycardia produced by 5-hydroxytryptamine. 4) Cord-sectioned rabbit showed different responses; the smaller doses $(100{\sim}200{\mu}g)$ produced transitory bradycardia and depression of blood pressure, which followed by tachycardia and pressure rise. The transitory bradycardia and depressor effects were not observed in cord-sectioned and vagotomized rabbit. 5) Treatment of animals with reserpine, guanethidine and hexamethonium changed the effects of intraventricular NE on blood pressure, i.e., in these cases the smaller doses of NE caused maked elevation of blood pressure. 6) From these observations it was inferred that central NE caused stimulation of cardioinhibitory and vasomotor center. The former seemed to be more sensitive to NE than the latter. Susceptibility of the vasomotor center to NE seemed to be influenced by peripheral sympathetic tone.

Effect of Sodium Hyaluronate on Osteoarthrosis in Rabbit Model (골관절증이 유발된 토끼에서 Sodium Hyaluronate의 효과)

  • Osteoarthrosis was induced in rabbit knees through resecton of anterior cruciate ligament. Sodium hyaluronate (1%) was administerated into articular space at the dose levels of 0.1 mg/kg once a week using 1ml sylinge and 26G needle for test groups. Saline was administrated for control groups. After 6 and 12 weeks, animals were sacrificed to conduct macroscopic observation and histopathologic examination. The articular lesions were evaluated and test groups were compared with control groups. No significant differencies were showed between test groups and control groups at macroscopic observation in both 6- and 12- week groups. In histopathologic examination, control groups showed higher osteoarthrosis than test groups. The articular surfaces of control groups showed fibrillation, denudation, pannus formation and hypocellularity. The articular surfaces of test groups showed fibrillaton and hypercellularity. These indicate that sodium hyaluronate has inhibitory effects on osteoarthrosis at least in rabbit and could be used for treatment of osteoarthrosis.
