Oral administration of the Herbal Medicines in animal models for treatment of Allergic Rhinitis: a Review of Animal Study Reports Published in Korea (알레르기성 비염 동물모델에서 한약제재 경구투여에 대한 국내 연구보고 고찰)

  • Objective : This study is to review the effect of herbal medicines on treatment of Allergic Rhinitis in animal models reported in Korean domestic journals.Methods : The Databases(NDSL, Koreantk, OASIS, KISS) are searched with terms as Allergic Rhinitis. among them, only animal study reports were reviewed. Animal model, intervention, and observation methods of measuring indicators were extracted.Results : 27 articles were reviewed. All studies used OVA to induce Allergic Rhinitis. 7 studies used simple herbal medicines, 20 studies used complex herbal medicines, To evaluate the effect of the Herbal medicines, 23 studies used immunological examination, 18 studies used histological examination, 16 studies used hematologic examination, 3 studies used macroscopy The evidences of 5 studies is insufficient to show that Herbal medicines are effective for treatment of Allergic Rhinitis.

Trends in Animal Experiments for Herbal Medicine Treatment of Atopic Dermatitis: During the 10 year's results (한약의 아토피피부염에 대한 국내외 동물실험 연구 동향: 최근 10년을 중심으로)

  • Objectives The aim of this study was to analyze the research trends in herbal medicine as a treatment for atopic dermatitis (AD) in an animal model. Methods We conducted a search targeting papers published from 2013 onwards in Korean and English databases. The search terms were focused on "AD", "dermatitis", and "Korean medicine". Only animal experimental studies involving the oral administration of single or complex prescriptions of Korean medicine were included, whereas in vitro studies, local administration studies, and studies of specific components of Korean medicine were excluded. We searched the PubMed and Embase databases for English databases and the Science on Research Information Sharing Service (RISS) and Korean Studies Information Service System (KISS) databases for Korean studies. Results Among the herbal medicine intervention groups, the most commonly used single herbs were baekseonpi, danggui, and douchi, whereas a mixture of hwangryun and gamcho was used in three studies, making it the most frequently used combination. There were significant improvements in indicators related to the symptoms of AD and various pro-inflammatory markers. Conclusions These results indicate the efficacy of herbal medicines in the treatment of AD in animal models. However, this study had several limitations. Therefore, future systematic reviews that address these limitations and provide more comprehensive analyses are required.

Estimation of genetic parameters for temperament in Jeju crossbred horses

  • Objective: Temperament can be defined as a type of behavioral tendency that appears in a relatively stable manner in responses to various external stimuli over time. The aim of this study was to estimate genetic parameters for the records of temperament testing that are used to improve the temperament of Jeju crossbred (Jeju${\times}$Thoroughbred) horses. Methods: This study was conducted using 205 horses (101 females and 104 males) produced between 2010 and 2015. The experimental animals were imprinted and tamed according to the Manual for Horse Taming and Evaluation for Therapeutic Riding Horses and evaluated according to the categories for temperament testing (gentleness, patience, aggressiveness, sensitivity, and friendliness) between 15 months and 18 months of age. Each category was scored on a five-point linear scale. Genetic parameters for the test categories were analyzed using a multi-trait mixed model with repeated records. The ASReml program was used to analyze the data. Results: The heritability of gentleness, patience, aggressiveness, sensitivity and friendliness ranged from 0.08 to 0.53. The standard errors of estimated heritability ranged from 0.13 to 0.17. The test categories showed high genetic correlations with each other, ranging from 0.96 to 0.99 and high repeatability, ranging from 0.70 to 0.73. Conclusion: The results of this study showed that the test categories had moderate heritability and high genetic correlations, but additional studies may be necessary to use the results for the improvement programs of the temperament of Jeju crossbred horses.

Chronic Paraspinal Muscle Injury Model in Rat

  • Objective : The objective of this study is to establish an animal model of chronic paraspinal muscle injury in rat. Methods : Fifty four Sprague-Dawley male rats were divided into experimental group (n=30), sham (n=15), and normal group (n=9). Incision was done from T7 to L2 and paraspinal muscles were detached from spine and tied at each level. The paraspinal muscles were exposed and untied at 2 weeks after surgery. Sham operation was done by paraspinal muscles dissection at the same levels and wound closure was done without tying. Kyphotic index and thoracolumbar Cobb's angle were measured at preoperative, 2, 4, 8, and 12 weeks after the first surgery for all groups. The rats were sacrificed at 4, 8, and 12 weeks after the first surgery, and performed histological examinations. Results : At 4 weeks after surgery, the kyphotic index decreased, but, Cobb's angle increased significantly in the experimental group (p<0.05), and then that were maintained until the end of the experiment. However, there were no significant differences of the kyphotic index and Cobb's angle between sham and normal groups. In histological examinations, necrosis and fibrosis were observed definitely and persisted until 12 weeks after surgery. There were also presences of regenerated muscle cells which nucleus is at the center of cytoplasm, centronucleated myofibers. Conclusion : Our chronic injury model of paraspinal muscles in rats shows necrosis and fibrosis in the muscles for 12 weeks after surgery, which might be useful to study the pathophysiology of the degenerative thoracolumbar kyphosis or degeneration of paraspinal muscles.

Comparative Analysis of 3 Experimental Mouse Model for Blood Hematology and Chemistry

  • The immune system and neuroendocrine systems are the two key components that maintain bodily homeostasis. Peripheral blood specimens can indicate abnormalities in a body, which often cause various threats to human health, including devastating autoimmune or metabolic diseases. To develop a treatment regimen for such diseases, experimental animal models are indispensable to researchers in academic fields. In this study, we examined the peripheral blood of 3 representative mouse strains (ICR, Balb/c, and C57Bl/6), which are widely used, to investigate whether there is a difference in reference range according to animal model. We performed hematological and chemistry analysis on individuals of both genders. The results of hematology analysis showed that the number of most types of blood cells was lower in ICR than in the other two strains. The results of chemical analysis revealed no specific pattern, but different patterns according to the individual indicator. Although the distinction between ICR and B6 was prominent, differences between Balb/c and B6 were also observed for several indicators. For some indicators, totally different patterns existed between females and males. Conclusively, this study provides the information that 3 experimentally representative mouse models have their own basal levels of blood components, suggesting the importance of a careful choice of a proper mouse model in research into immune or metabolic diseases, to exclude any biases.

The Effect of Tongkyu-tang on the Ovalbumin-inhalation Rat Model with Allergic Rhinitis (통규탕(通竅湯)이 알레르기 비염 모델 횐 쥐에 미치는 영향)

  • Background and Objectives: Allergic rhinitis is an allergic reaction characterized by sneezingm itchy nose, mouth and throat, congestion and/or nasal discharge. The offending allergenes are usually pollens, molds, dust mites and animal allergen. Recently, the incidence of infectious nasal diseases tend to decrease. However, allergic rhinitis has increased and treatment in most cases has only deat with the symptom. Tongkyu-tang was composed of sixteen crude drugs. The Oriental Medical References mention therapeutic effects of Tongkyu-tang on nasal obstruction, watery nasal discharg. And Tongkyu-tang has clinically been used for the treatment of common cold, headache, sternutation, rhinitis etc. Speacially Tongkyu-tang is one of the most frequently used medical treatment for the allergic rhinitis. Experimental studies were conducted to investigate for the effect of Tongkyu-tang on the changes of neutrophil segment, lymphocyte, total IgE and nassal tissue in allergic rhinitis of ovalbumin-inhalation rat. Meterial and Methods: Fifteen Sprague-Dawley rats were divided into three group: normal group, control group, experimental group. To induce the allergic rhinitis in control group and experimental group, rats were sentitized intraperitoneally with 0.1 % ovalbumin solution 3 times at intervals of I week. Then intranasal sensitization was performed by diffusing 0.1 % ovalbumin solution 3 times at intervals of 2 days. After that time, rat in the experimental group were oral administration treated by Tongkyu-tang for 28 days. We observed changes in nasal tissue; changes in the number of white blood cell, red blood cell and total Ig E; also changes in the segment of neutrophil and lymphocyte in blood. And we observed the changes of AST, ALT of three group. We used anova test statistically. Result: The number of leucocyte remained unchanged between three group. The number of erythrocyte was increased in experimental group and control group when compared with the normal group. The segment of neutrophil, in blood was decreased in experimental group when compared with the control group but, that was not significant statistically(p<0.05). The promotion of lymphocyte in blood was significantly decreased in experimental group when compared with the Control group(p<0.05). Total IgE was decreased in experimental group when compared with the control group but, that was not significant statistically(p<0.05). The cilium be well preserved in experimental group: the nasal tissue in experimental group was similar to in the normal group. Congestion and expantion of grandular cell in nasal submucosa, hypertropy of epithelium in nasal mucosa, acid mucus in epithelium and neutral mucus in subepithelium were decreased in experimental group when compared with the control group. Effect of Tongkyu-tang on the liver function were also studies in rats. Treatment of Tongkyu-lang did not affected on AST and ALT. Conclusion: Considering the above experimental results, it is suggested that oral administration treatment using Tongkyu-lang, without worry about liver function injury, decreased response on an Animal model with Allergic Rhinitis.

Deformation prediction by a feed forward artificial neural network during mouse embryo micromanipulation

  • In this study, a neural network (NN) modeling approach has been used to predict the mechanical and geometrical behaviors of mouse embryo cells. Two NN models have been implemented. In the first NN model dimple depth (w), dimple radius (a) and radius of the semi-circular curved surface of the cell (R) were used as inputs of the model while indentation force (f) was considered as output. In the second NN model, indentation force (f), dimple radius (a) and radius of the semi-circular curved surface of the cell (R) were considered as inputs of the model and dimple depth was predicted as the output of the model. In addition, sensitivity analysis has been carried out to investigate the influence of the significance of input parameters on the mechanical behavior of mouse embryos. Experimental data deduced by Fl$\ddot{u}$ckiger (2004) were collected to obtain training and test data for the NN. The results of these investigations show that the correlation values of the test and training data sets are between 0.9988 and 1.0000, and are in good agreement with the experimental observations.

  • Apoptosis is now widely recognized as a common form of cell death and represents mechanism of cell clearance in many physiological situations where deletion of cells is required. In vivo administration of bacterial lipopolysaccharide (LPS) to Balb/c mice induced DNA fragmentation in the thymus. DNA fragmentation in the thymus was roughly dependent on the dose of LPS injected and reached the peak 18 hours after injection. This apoptosis in the thymus might be mediated due to LPS stimulant. DEN (diethylnitrosamine) has been shown to cause liver cancer in experimental animals and humans. The hepatic tumorigenesis was induced by ad libitum feeding of DEN only. It was suggested that DEN induced hepatic tumorgenesis in rat is a good reproducible model for studying biochemical and pathophysiological changes associated with the development of hepatic tumorigenesis and apoptosis.

Effect of electrical stimulation and vitamin AD3E on fracture healing in a rat model (전기자극과 vitamin AD3E가 랫드 골절치유에 미치는 영향)

  • Electrical stimulation and vitamin $AD_3E$ administration have been shown to enhance the repair of biological tissues such as bone, ligament and tendon, The objective of this study were (a) to investgate the therapeutic effects of different levels of electrical stimulation and vitamin $AD_3E$ administration on fracture healing in a rat model and (b) to identify the most effective voltage level. Ninety Sprague-dawley rats were divided into electrical stimulation group and vitamin $AD_3E$ administration group. Electricla stimulation group was divided into four groups on the basis of the level of current delivered. The experimental groups received current (which varied by group), while the controls, with identical electrodes, received no current. In vitamin $AD_3E$ administration group, experimental group was injected 2,500 IU of vitamin A, 1,250 IU of vitamine $D_3$, 1mg of vitamine E intramuscularly. And in control group 0.1ml of saline was injected intramuscularly. After time periods (7-day, 14-day, 21-day for stimulation all rats were tested with combination of biochemical, roentgenologic and histomorphological methods. The results obtained were as follows ; In electrical stimulation groups, serum calcium and inorganic phosphorus level of experimental and control groups showed non specific change within normal physiological ranges. In vitamin $AD_3E$ administration group, serum calcium level of experimental and control groups showed non specific changes within normal physiological ranges, while experimental group showed slightly higher serum inorganic phosphorus level. According to roentgenologic and histomorphological examination, 2V 25Hz electrical stimulation group and vitamine $AD_3E$ administration group showed statistically significant improvements in bone density and ossification reaction until day 14. The terapeutic effect of stimulation on fracture healing was similiar to that of vitamin $AD_3E$ administration. In this study stimulation of 2V 25Hz was the most effective level of electrical stimulation for the healing of fracture of rats.

  • A simple mathematical model of simultaneous combined action of environmental agents has been proposed to describe the synergistic interaction of microwave and high ambient temperature treatment on animal heating. The model suggests that the synergism is caused by the additional effective damage arising from an interaction of sublesions induced by each agent. These sublesions are considered to be ineffective if each agent is taken individually. The additional damage results in a higher body temperature increment when compared with that expected for an independent action of each agent. The model was adjusted to describe the synergistic interaction, to determine its greatest value and the condition under which it can be achieved. The prediction of the model was shown to be consistent with experimental data on rabbit heating. The model appears to be appropriate and the conclusions are valid.
