An osteological study of animal bones excavated from Jeju Jongdali shell-mount (제주 종달리패총 유적에서 출토된 동물 유물의 해부학적 연구)

  • The classification of bony pieces which were excavated from Jongdali archaeological site in Jeju was studied. The total number of bone remains were 81 pieces, in which 31 pieces were classified into animal bones. The animal species consisted of Cervus spp., Sus scrofa, Bos taurus and Equus caballus. This finding suggests that the major fauna in this peroid(B.C. 100 - A.D. 100) is wild boar, deer, horse and cattle.

The Effect of Dietary Protein Source and Sulfur Amino acid Content on bone Metabolism in Growing Rats (식이 단백질의 종류와 함황아미노산 함량이 성장기 쥐의 골밀도에 미치는 영향)

  • This study was performed to evaluate the effect of dietary protein source and sulfur amino acid content on bone metabolism in ra. Thirty male rats (body weight 145$\pm$2g) were divided into three groups. The rats in the first group were fed on casein 20% diet as animal protein source and those in the second group were fed on soy 20% diet as plant protein source. Sulfur amino acid ratio of these group was 1.07:1. The rats in the third group were fed on soy 20% diet and the sulfur amino acid were supplemented with the amount contained as much in the soy 20% diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks, The total body, spine, femur bone mineral density and bone mineral content were measured using Dual Energy X-ray Absorptiometry Calcium, phosphate, pyridinoline, creatinine in urine and calcium, phosphate, alkaline phosphatase, osteocalcin in serum were measured. During the experimental period, plant protein (soy protein) group had a lower urinary Ca excretion, urine pyridinoline & crosslinks value and had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein) group. There were no significant differences in serum calcium, phosphate, alkaline phosphatase and osteocalcin among the three groups of the rats. The findings from this study demonstrated that plant protein (soy protein) is beneficial of bone mineral density because it had a higher Ca efficiency in total bone and femur bone mineral density than animal protein (casein). However, the supplementation of sulfur amino acid on soy results were consistent with prior studies that dietary sulfur amino acid load had a negative effect on calcium balance. The rats fed sulfur amino acid supplementation diet increased urinary calcium excretion and decreased calcium efficiency for total and femur mineral density. Anti-Ionizing Radiation Effect of Selenium on Osteoporosis Model during Bone Repair Process (골다공증 모델의 뼈 재생기에 있어 셀레늄(Selenium)의 방사선 보호작용)

  • Selenium (Se) is an essential trace element for humans and animals, and several findings suggest that dietary Se intake may be necessary for bone health. Accumulating evidence indicates that Se compounds possess anticancer properties. Se is specifically incorporated into proteins in the form of selenocysteine and non-specifically incorporated as selenomethionine in place of methionine. This study evaluated protection by Se in the bone repair process in ovariectomized rats after irradiation. For such purpose, 80 ovariectomized female Sprague-Dawley rats were randomly divided into 4 experimental groups: ovariectomized (Ov), Ov/Se, Ov/irradiated (Irr) and Ov/ Se/Irr. A bone defect was created on the tibia of all animals 40 days after ovariectomy. Two days after surgery, only the Ov/Se and Ov/Se/Irr rats received 0.8 mg Se/kg. Three days after surgery, only the Ov/Irr and Ov/Se/Irr rats received 10 Gy of X-rays on the lower limb region. The animals were euthanized at 7, 15, 22 and 29 days after surgery to assess the repair process, which was evaluated by analysis of trabecular bone number (Masson Trichrome) and birefringence analysis (Picrosirius). It was possible to observe a delay in the bone repair process in the ovariectomized/irradiated group and similarity between the ovariectomized, Ov/ Se and Ov/Se/Irr groups. Our findings suggest that sodium selenite may influence a radioprotective effect in the bone repair of tibia of ovariectomized rats without toxicity.

Effects of Dietary Combinations of Vitamin A, E and Methionine on Growth Performance, Meat Quality and Immunity in Commercial Broilers

  • The experiment was conducted to study the effect of dietary combinations of vitamin A (VA), vitamin E (VE) and methionine (Met) on growth performance, meat quality and immunity in commercial broilers. Ross chicks (n=3,630) were allocated to five experimental treatments with three replicates per diet. The dietary treatments were: VA 8,000 IU, VE 10 IU (diet 1); VA 12,000 IU, VE 10 IU (diet 2); VA 8,000 IU, VE 100 IU (diet 3); VA 12,000 IU, VE 100 IU (diet 4) and; VA 12,000 IU, VE 100 IU/kg diet and 20% Met higher than other groups (diet 5). The Met content in diet 1 to diet 4 were as per the requirement suggested by NRC. Separate vitamin premixes were prepared for each treatment diet as per the requirement of study. The 35 d study revealed significantly (p<0.0001) higher weight gains in broilers fed diet 3 and diet 5, than in the rest of the groups during starter phase (0-3 weeks) only. The feed intake did not vary significantly at all phases of study, but feed efficiency was significantly (p<0.05) lower in diet 1 during starter and overall phase (4-5 weeks). The bone strength and bone composition, except bone calcium, remained unaffected due to experimental diets studied after 35 d of experimental feeding. The thio-barbituric acid reactive substances (TBARS) were significantly (p=0.0013) lower in the breast meat in group 5, followed by group 3, than in the rest of the groups. The immune studies conducted, antibody titers to sheep red blood cells, thickness index to phytohaemagglutinin-P, and heterophil: lymphocyte ratio, did not show any significant difference among treatments. It could be concluded that supplementation of VA, VE and Met at higher levels could be beneficial to broilers only during the starter phase.

A Study on the Factors Affecting Bone Mineral Density in Adult Women - Based on the Mothers of Elementary School Students- (일부 초등학생의 어머니를 대상으로 한 성인 여성의 골밀도에 영향을 미치는 요인에 관한 연구)

  • The bone mineral density(vertebrae Ll-4, femoral neck, Ward's triangle and trochanter) of 160 healthy adult women aged 29-45 was measured and general characteristics, anthropomentric measurements, and dietary intake were also studied to determine the main factors affecting the bone mineral density(BMD) of adult women. The nutrient intake of the subjects was adequate to RDA level except energy, Ca, Fe, and vit A. The BMD of vertebrae L24 showed significant positive association with nutrient intake such as animal protein, P, Ca(animal) and Fe(animal). The BMD of the femoral neck correlated significantly with anthropometric measurements such as height, weight, waist circumference, hip circumference, body fat and body mass index. The BMD of both sites(vertebrae L24 and femoral neck) were significantly related to vertebrae L24 was daily Ca intake and that of femoral neck was daily energy expenditure. In conclusion, adequate nutrient intake, especially protein, P, Ca and Fe as well as increases in physical activity were suggested to prevent the loss of bone mineral density in adult women.

Functionality of Dairy Foods on Osteoporosis

  • Osteoporosis is a systemic skeletal disease characterized by decreased bone mass, which results in a markedly increased risk of traumatic fractures. This disease is a worldwide health problem with a high prevalence. Recently, various dietary components have been found to minimize the risk of developing osteoporosis through their ability to stimulate bone formation and optimize bone health. Among them, probiotics and fermented milk can have beneficial effects to human health. Bioactive compounds derived from probiotics in fermented milk can especially modulate physiological functions related to bone health. Here, we review the evidence to support these insights into newly found functionality of dairy foods for osteoporosis prevention.

Effects of Capsaicin on Adipogenic Differentiation in Bovine Bone Marrow Mesenchymal Stem Cell

  • Capsaicin is a major constituent of hot chili peppers that influences lipid metabolism in animals. In this study, we explored the effects of capsaicin on adipogenic differentiation of bovine bone marrow mesenchymal stem cells (BMSCs) in a dose- and time-dependent manner. The BMSCs were treated with various concentrations of capsaicin (0, 0.1, 1, 5, and $10{\mu}M$) for 2, 4, and 6 days. Capsaicin suppressed fat deposition significantly during adipogenic differentiation. Peroxisome proliferator-activated receptor gamma, cytosine-cytosine-adenosine-adenosine-thymidine/enhancer binding protein alpha, fatty acid binding protein 4, and stearoyl-CoA desaturase expression decreased after capsaicin treatment. We showed that the number of apoptotic cells increased in dose- and time-dependent manners. Furthermore, we found that capsaicin increased the expression levels of apoptotic genes, such as B-cell lymphoma 2-associated X protein and caspase 3. Overall, capsaicin inhibits fat deposition by triggering apoptosis.

Trabecular bone Thickness Measurement of Rat Femurs using Zoom-in Micro-tomography and 3D Fuzzy Distance Transform (Zoom-in Micro-tomography와 3차원 Fuzzy Distance Transform을 이용한 쥐 대퇴부의 해면골 두께 측정)

  • Micro computed tomography (micro-CT) has been used for in vivo animal study owing to its noninvasive and high spatial resolution capability. However, the sizes of existing detectors for micro-CT systems are too small to obtain whole-body images of a small animal object with $\sim$10 micron resolution and a part of its bones or other organs should be extracted. So, we have introduced the zoom-in micro-tomography technique which can obtain high-resolution images of a local region of an live animal object without extracting samples. In order to verify our zoom-in technique, we performed in vivo animal bone study. We prepared some SD (Sprague-Dawley) rats for making osteoporosis models. They were divided into control and ovariectomized groups. Again, the ovariectomized group is divided into two groups fed with normal food and with calcium-free food. And we took 3D tomographic images of their femurs with 20 micron resolution using our zoom-in tomography technique and observed the bone changes for 12 weeks. We selected ROI (region of interest) of a femur image and applied 2D FDT (fuzzy distance transform) to measure the trabecular bone thickness. The measured results showed obvious bone changes and big differences between control and ovariectomized groups. However, we found that the reliability of the measurement depended on the selection of ROI in a bone image for thickness calculation. So, we extended the method to 3D FDT technique. We selected 3D VOI (volume of interest) in the obtained 3D tomographic images and applied 3D FDT algorithm. The results showed that the 3D technique could give more accurate and reliable measurement.

Optimization of a Multi-Step Procedure for Isolation of Chicken Bone Collagen

  • Chicken bone is not adequately utilized despite its high nutritional value and protein content. Although not a common raw material, chicken bone can be used in many different ways besides manufacturing of collagen products. In this study, a multi-step procedure was optimized to isolate chicken bone collagen for higher yield and quality for manufacture of collagen products. The chemical composition of chicken bone was 2.9% nitrogen corresponding to about 15.6% protein, 9.5% fat, 14.7% mineral and 57.5% moisture. The lowest amount of protein loss was aimed along with the separation of the highest amount of visible impurities, non-collagen proteins, minerals and fats. Treatments under optimum conditions removed 57.1% of fats and 87.5% of minerals with respect to their initial concentrations. Meanwhile, 18.6% of protein and 14.9% of hydroxyproline were lost, suggesting that a selective separation of non-collagen components and isolation of collagen were achieved. A significant part of impurities were selectively removed and over 80% of the original collagen was preserved during the treatments.
