A case of using a vibrating saw to collect brain tissue for BSE (bovine spongiform encephalopathy) test in Holstein (홀스타인에서 BSE (bovine spongiform encephalopathy) 검사용 뇌 조직 채취를 위한 진동톱 사용례)

  • BSE (bovine spongiform encephalopathy)-like symptoms salivation, gait disorder, and the downer syndrome occurred to 8 heads of cattle at six cattle farms in Gyeonju, east eastern Gyeongsang province in 2013. Eight brain tissue samples using a vibration saw (Multimaster$^{(R)}$ FEIN, Germany) were collected referring to "The brain removal technique of transmisible spongiform encephalopathy sampling DVD" by Animal And Plant Quarantine Agency. Brain removal techinque using a vibration saw on the thick, solid frontal bone of cattle is more convenient for incision and removal than an ax for BSE testing.

Effect of Chlorine Dioxide Gas Application to Egg Surface: Microbial Reduction Effect, Quality of Eggs, and Hatchability

  • Controlling of microorganisms in the industrial process is important for production and distribution of hatching and table eggs. In the previous study, we reported that chlorine dioxide ($ClO_2$) gas of a proper concentration and humidity can significantly reduce the load of Salmonella spp. on eggshells. In this study, we compared microbial reduction efficacy on egg's surface using hatching eggs and table eggs, internal quality of table eggs, and hatchability after both the conventional method (washing and UV expose, fumigation with formalin) and $ClO_2$ gas disinfection. Application of 40 ppm $ClO_2$ gas to the table and hatching eggs, respectively, reduced the aerobic plate count (APC) with no statistical difference compared with the conventional methods. Additionally, we didn't observed that any significant difference in albumin height, Haugh unit (HU), and yolk color, this result confirms that 40 ppm $ClO_2$ had no effect on the internal quality of the table eggs, when comparing with the UV treatment method. The hatchability of hatching eggs was not statistical different between formaldehyde fumigation and 80 ppm $ClO_2$ gas treatment, though the value was decreased at high concentration of 160 ppm $ClO_2$ gas. From these results, we recommend that $ClO_2$ gas can be used as a safe disinfectant to effectively control egg surface microorganisms without affecting egg quality.

Comparison of Culture, Conventional and Real-time PCR Methods for Listeria monocytogenes in Foods

  • We compared standard culture methods as well as conventional PCR and real-time PCR for the detection of Listeria monocytogenes (L. monocytogenes) in milk, cheese, fresh-cut vegetables, and raw beef that have different levels of background microflora. No statistical differences were observed in sensitivity between the two selective media in all foods. In total, real-time PCR assay exhibited statistically excellent detection sensitivity (p<0.05) and was less time consuming and laborious as compared with standard culture methods. Conventional culture methods showed poor performance in detecting L. monocytogenes in food with high levels of background microflora, generating numerous false negative results. While the detection of L. monocytogenes in fresh cut vegetable by culture methods was hindered only by L. innocua, various background microflora, such as L. innocua, L. welshimeri, L. grayi, and Enterococcus faecalis appeared on the two selective media as presumptive positive colonies in raw beef indicating the necessity of improvement of current selective media. It appears that real-time PCR is an effective and sensitive presumptive screening tool for L. monocytogenes in various types of foods, especially foods samples with high levels of background microflora, thus complementing standard culture methodologies.

  • 본 연구는 넓은잎 딱총나무 잎으로부터 유도된 캘러스의 형태학적 특성과 효능에 관한 것이다. 딱총나무에서 유도된 캘러스 내의 세포들을 촬영한 표면주사현미경 사진에서 분열능이 높은 세포의 특성을 보여주었다. 유도된 넓은잎 딱총나무 캘러스를 생물반응기 내에서 배양시켜 얻은 딱총나무 식물세포배양 추출물은 DPPH 제거 및 $PPAR{\alpha}$ 활성 실험결과에서 자유라디컬 제거능 및 항염증 효능이 높게 나타났다. 이러한 딱총나무 캘러스 세포 배양 추출물은 화장품의 항노화 신소재로서 응용될 수 있다.

Prevalence of Dirofillaria immitis in Raccoon Dogs (Nyctereutes procyonoides) in Korea (한국 너구리(Nyctereutes procyonoides)의 심장사상충감염에 대한 유병률)

  • Heartworm infection is a fatal disease causing heart failure and pulmonary diseases in dogs. This heartworm infection can also occur in wild carnivores including Raccoon dogs. Recent study found that relatively high prevalence rate in wild Raccoon dog population. Therefore, this study was designed to evaluate the prevalence rate of D. immitis in free-roaming Raccoon dogs and the recovery rate of microfilariae in infected Raccoon dogs in Korea. Overall prevalence rate of D. immitis in Korean Raccoon dogs was 17.8%. Prevalence rate in male Raccoon dogs was 21.8%, while that in female Raccoon dogs was 12.8%. Microfilariae were not detected in 17 Raccoon dogs having positive in heartworm antigen test. Our study result suggested that the prevalence rate of D. immitis in Korea is twice higher than that of Japan. In addition, microfilaremia is rare in Raccoon dogs as commonly noticed in cats.

Toxicity of Several Insecticides Against Halyomorpha halys (Hemiptera: Pentatomidae) and Gymnosoma rotundatum (Diptera: Tachinidae) (몇 가지 살충제의 썩덩나무노린재와 뚱보기생파리에 대한 독성)

  • Toxicity of several insecticides was tested against the brown marmorated stink bug, Halyomorpha halys (Hemiptera: Pentatomidae), and adult Gymnosoma rotundatum (Diptera: Tachinidae) in a sweet persimmon orchard. H. halys was captured using black light traps and G. rotundatum adults were collected using sticky traps lured with an aggregation pheromone, methyl-(E,E,Z)-2,4,6-decatrienoate of Plautia stali. Five insecticides, namely, bifenthrin 2 WP, buprofenzine+dinotefuran (20+15) WP, clothianidin 8 SC, dinotefuran 10 WP, and thiamethoxam 10 WP, were moderately toxic to G. rotundatum, resulting in 55.3% to 74.3% mortality. Bifenthrin 2 WP caused 93.1% mortality at 72 h within cages as a residual toxicity, and others caused varied mortalities.

A New record of Elaeagnus (Elaeagnaceae) to Korean Flora: E. pungens Thunberg (한국 보리수나무속의 미기록 식물: 통영볼레나무)

  • Elaeagnus pungens Thunb., belongs to the family Elaeagnaceae, was recently recorded for the first time in Mt. Miruk, Tongyeong-si, Gyeongsangnam-do. The native distribution range of this species includes China and Japan in East Asia, while it is considered as an invasive species in North America. Elaeagnus pungens is related to E. glabra, but the species can be distinguished from its congeners by having revolute and repand leaf blades, silvery white and brown scales on the abaxial surface of the leaf, and a campanulate calyx. We propose the new Korean name 'Tong-yeong-bol-re-na-mu,' based on its first collecting site. A detailed description of vegetative and floral characteristics, trichome scanning electron micrographs, dichotomous keys, and the habitat of this species are also provided.

Incidence of Viral Diseases and Occurrence of Three Unreported Viruses in Yams in Korea

  • During 2012 to 2014, a survey for the presence of viral diseases in yam plants was carried out in a field of the Institute for Bioresources Research in Gyeongsangbuk-do, Korea. A total of 88 leaf samples were collected and tested by reverse transcription polymerase chain reaction using specific primer sets. Eighty-one samples were positive for Broad bean wilt virus 2 (BBWV2), Chinese yam necrotic mosaic virus (ChYNMV), Cucumber mosaic virus (CMV), Japanese yam mosaic virus (JYMV), and Yam mild mosaic virus (YMMV), whereas Yam mosaic virus (YMV) was not detected. Additionally, seven samples were negative for all viruses. Several samples exhibited mixed (double and triple) infections. Three viruses (CMV, JYMV, and YMMV) were detected for the first time in yam plants in Korea. A BLAST search showed that three viruses shared nucleotide identities with CMV-Ca (98%), JYMV-O2 (91%), and YMMV-TG_NH_1 (86%). Thus, our findings confirmed that yam plants cultivated in Korea were infected with multiple viruses with three of these viruses reported for the first time in Korea.

Relationship Between Degree Centrality of Livestock Facilities in Vehicle Movement Network and Outbreak of Animal Infectious Disease (차량이동 네트워크에서의 축산시설 연결중심성과 가축 전염병 발생 사이의 관계)

  • The national loss caused by the periodic livestock epidemic is very large. In addition, vehicle movement is the main cause of livestock epidemics in Korea. In this context, this study analyzed the relationship between the degree centrality of livestock facilities and the outbreak of infectious diseases. For this purpose, a livestock vehicle movement network was constructed using the facility entrance data provided by KAHIS. Afterwards, the centrality index was derived for each facility in the vehicle movement network and the mean centrality index of the epidemic and non-epidemic facilities were compared. The analysis results are summarized as follows. First, the degree centrality of epidemic facilities is significantly greater than non-epidemic facilities. As a result of the analysis of the entire period data and the period-based data, in most data, the degree centrality of facilities where livestock infectious diseases occurred was significantly greater than most non-occurrence facilities. Second, in the entire period data, the difference in degree centrality between the epidemic and non-epidemic facilities was smaller for HPAI than for FMD. On the other hand, no significant difference was found in the results of the analysis according to the divided period. The policy implications of the results are as follows. First, proactive management of facilities based on centrality is needed. Second, in the case of cloven-hoofed animal facilities, it is more urgent to introduce a management policy based on the degree centrality.

Survey on Nematodes in Cymbidium and Chemical Control of Ditylenchus sp. (심비디움에 발생하는 선충 조사 및 줄기선충류(Ditylenchus sp.)에 대한 약제방제 효과)

  • Surveys were conducted on the occurrence of nematodes in the root systems of 1-3-year old Cymbidium hybida Swartz cultivated for export in Korea. The most frequently detected plant-parasitic nematode was Ditylenchus sp. with 4.7, 43.7, and 49.7/200 cc growth medium in 1, 2, and 3 year-old cymbidiums, respectively. And the densities of non-parasitic nematodes, dorylaimids, were 35.3, 70.5, and 155.8/200 cc growth medium in 1, 2, and 3-year-old cymbidiums, respectively. Three-year-old cymbidiums collected from Siheung, Ansan, and Gimpo had low densities of Aphelenchus sp. and Aphelenchoides sp. with under 40 individuals/pot, and the dorylaimid densities were 56-824/pot. To evaluate the effects of nematicides on Ditylenchus sp. in cymbidium, Emamectin benzoate EC, Fosthiazate SL, and Cadusafos CS were tested at two farms in Ansan and Gimpo. Emamectin benzoate EC showed control effects of 75.7 and 89.5%, whereas Fosthiazate SL and Cadusafos CS showed 27.2 and 65.3% and 30.1 and 90.5% control effects in the tests.
