Effect of Dietary Monascus Pigment on the Liver Damage Induced with $CCl_4$in Rats

  • In the biological world, there are a number of ecological fights for survival between each organism such as plants, animals and microorganism In such events, an organism can use its natural bioactive products as defence agent against other organisms. Furthermore, natural bioactive products can be utilized for medicine or functional food. Recently, we investigate the effect of Monascus pigment extracted from a fungus, Monascus anke, on the alcohol metabolism and blood lipid profile. In the present study, it is observed that Monascus pigment supplemented dietary may have a hepatoprotective effect on rat's liver damage induced with $CCl_4$ . By treatment with $CCl_4$(3 times, I.P), liver damage was reduced more in the rats fed 2% Monascus pigment extract supplemented diet than those fed standard diet, based on the serum levels of alanine aminotransferase, microsomal glucose-6-phosphatse activity and hepaic malondialdehyde content. On the other hand, oxygen free radical generating enzymes, hepatic P-450 dependent aniline hydroxylase, xanthine oxidase, and oxygen free radical scavenging enzymes, hepatic glutathione S-transferase, catalase, superoxide dismutase activities were generally higher both in $CCl_4$ treated group and control fed 2% Monascus pigment extract supplemented diet than those fed standard diet. In conclusion, the rats fed 2% Monascus pigment extract supplemented diet showed more reduced liver damage than those fed standard diet, which may be due to the acceleration of oxygen free radical metabolism.

Evaluation of a Schzandrin C Derivative DDB-mixed Preparation(DWP-04) on Acetaminophen Detoxification Enzyme System in the Animal Model (오미자 Schizandrin C 유도체 DDB 복합물 DWP-04가 Acetaminophen 해독계에 미치는 영향)

  • The effects of the DWP-04 [DDB:selenium yeast:glutathione (31.1 : 6.8 : 62.1 (w/w%)] on acetaminophen detoxification enzyme system were studied in rats. Treatment with DWP-04 was prevented againt acetaminophen-induiced hepatotoxicity in rat as evidenced by the decreased formation of lipid peroxide. Effect of DWP-04 on the activities of free radical-generating enzymes, free radical scavenging enzymes and glutathione-related enzymes as well as detoxification mechanism of DWP-04 against acetaminophen-treated was investigated in rat. Activities of cytochrome p450, cytochrome b5, aminopyrine demethylase and aniline hydroxylase as free radical-generating enzymes activities were decreased by the treatment with DWP-04 against acetaminophen treated. Although acetaminophen-induced hepatotoxicity results in the significantly decrease in the level of hepatic glutathione and activities of glutathine S-transferase, quinone reductase, glutathione reductase and ${\gamma}-glutamyl-$cysteine synthetase, these decreasing effects were markedly lowered in the DWP-04-treated rat. Therefore, it was concluded that the mechanism for the observed preventive effect of DWP-04 against the acetaminophen-induced hepatotoxicity was associated with the decreased activities in the free radical-generating enzyme system.

Biotransformation of Theophylline in Cirrhotic Rats Induced by Biliary Obstruction

  • The object of this work was to study the pharmacokinetic differences and the cause of these differences in cirrhotic rats induced by biliary obstruction when aminophylline (8 mg/kg as theophylline, i.v.) was administered. The concentrations of theophylline and its major metabolite (1,3-dimethyluric acid) in plasma were determined by HPLC. In addition, formation of 1,3-dimethyluric acid from theophylline in microsomes and the changes in the activity of drug metabolizing enzymes, which are suggested to be involved in theophylline metabolism, were determined. In cirrhotic rats, the systemic clearance of theophylline was reduced to 30% of the control value while AUC (area under the palsma concentration-tie curve) and (t1/2)$\beta$ were increased 1.3 fold and3.5 fold, respectively. The formation of 1,3-dimethyluric acid was decreased to 30% of the control value in microsomes of cirrhotic rat liver. In cirrhotic rat liver, activities of aniline hydroxylase (CYP2E1 related), erythromycin-N-demethylase (CYP3A related), and methoxyresorufin-O-demethylase (CYP1A2 related), which were reported to be related with theophyline metabolism, were decreased to 67%, 53%, and 76% that of normal rat liver, respectively. From the results, it can be concluded that in cirrhotic rats induced by biliary obstruction, the total body clearance of theophylline is markedly reduced and it may be due to decreased activity of drug metabolizing enzymes in liver.

Protective effects of alginate-free residue of sea tangle against hyperlipidemic and oxidant activities in rats

  • The antihyperlipidemic and antioxidant activities of dietary supplementation of sea tangle from Goseong and the alginate-free residue of sea tangle were investigated in Sprague Dawley rats treated with a high-fat diet, streptozotocin, poloxamer 407, and bromobenzene. The alginate-free residue of Goseong sea tangle induced a significant reduction in triglycerides and total cholesterol levels, as well as a significant increase in high-density lipoprotein cholesterol levels. Alginate-free Goseong sea tangle residue reduced the activities of the phase I enzymes aminopyrine N-demethylase and aniline hydroxylase, which had been increased by intraperitoneal injection of bromobenzene. Pretreatment with Goseong sea tangle residue prevented a bromobenzene-induced decrease in epoxide hydrolase activity. Bromobenzene reduced hepatic glutathione content and increased hepatic lipid peroxide levels. Pretreatment with alginate-free Goseong sea tangle residue prevented lipid peroxidation induced by bromobenzene, but pretreatment with Goseong sea tangle did not. These results suggest that Goseong sea tangle residue exerted antihyperlipidemic and antioxidant activities that were higher than those induced by alginate-containing sea tangle. Therefore, the alginate-free residue may contain physiologically unknown active components, other than alginic acid, which may potentially be used to prevent hyperlipidemic atherosclerosis.

Effect of SAENGCHINYANGHYOLTANG on the hepatic metabolic enzyme system in streptozotocin-induced diabetic rats (고혈당(高血糖) 쥐의 간(肝) 대사효소계(代謝酵素系)에 미치는 생진양혈탕(生津養血湯)의 영향(影響))

  • SANGNYANGHYOLTANG(SYT) is one of the most important prescription that has been used in oriental medicine for diabetes mellitus. The sudy was done in order to elucidate the anti-diabetic effect of SYT. After pretreatment of SYT(1,000mg/kg) for 6 weeks, the effect of of SYT was prevented on serum liver function test and hepatic lipid peroxide content in rats i.v. injected with streptozotocin(STZ, 50mg/kg, tail vein) 5 weeks after pretreatment of SYT. The hepatic microsomal cytochrome P-450 and aniline hydroxylase were significantly decreased, and aminopyrine N-demethylase activity was significantly increased in SYT-STZ group as compared with control group. Changes in aldehyde oxidase, xanthine oxidase, superoxide dismutase, catalase, epoxide hydrolase, UDP-glucuronyltransferase and sulfotransferase activities were not significantly different in any of the group. The cytosolic glutathione S-transferase activity was significantly decreased in SYT-STZ group as compared with control group. The selenium-independent glutathione peroxidase was significantly increased in SYT-STZ group as compared with control group, but there was no significant difference in selenium-dependent glutathione peroxidase in any of the groups. The hepatic glutathione concentration was significantly increased in SYT-STZ group as compared with control group, and ${\gamma}-glutamylcystein$ synthetase and glutathione reductase activities were not significantly different in any of the groups. The hepatic lipid peroxide content, serum aminotransferase and sorbitol dehydrogenase activities were slightly decreased in significantly in SYT-STZ groups.

In vivo and In vitro Anti-lipid Peroxidative Effect of the Extract Complex of Korean Anti-thirst Drugs (한방 소갈약 추출물 및 복합물에 의한 In vivo 및 In vitro 지질과산화 저해효과)

  • In Oriental medicine, the prescriptions composed of several herb medicines have been used. It is still unclear how the sum of several extracts of anti-thirst drugs represents the anti-lipid peroxidative action. Three anti-thirst herb medicines, Kalopanax pictus (K), Pueraria thunbergiana (P) and Rhus verniciflua (R), were extracted with MeOH and $H_2O$, respectively, and the former one was fractionated into the resultant EtOAc extract. Each extract was reconstituted to give KPR311, KPR131 and KPR113 where, for example, KPR311 represents the complex of K-P-R {3:1:1 (w/w/w)} of the three extracts. The order of the inhibitory effect in bromobenzene-induced lipid peroxidation in rats was as follows: EtOAc extract>$H_2O$ extract>MeOH extract. Extract complexes were found to be more potent than the extracts of individual crude drugs. The KPR131 of EtOAc extract was found to be the most potent among the tested samples. These anti-lipid peroxidative effects were also supported by the decrease of aniline hydroxylase activity and aminopyrine N-demethylase activity, on the other hand by the increase of epoxide hydrolase activity. All the tested samples were assayed in vitro antioxidative effects such as DPPH assay, ADP/NADPH/Fe$^{3+}$ assay and ascorbic acid/Fe$^{2+}$ assay. The EtOAc extracts also showed the most significant antioxidative effects. These results suggest that the sum of anti-thirst drugs could reflect the effects of respective crude drugs.s.s.

Effects of Boganhwan Decoction on the Liver Lipid Peroxide Content and Metabolic Enzyme System (노화과정(老化過程)의 흰쥐에서 보간환(補肝丸)이 간장(肝臟)의 대사효소계(代謝酵素系)에 미치는 영향(影響))

  • Aging in the life form occurs due to a gradual progression of the body growth and degeneration. Morphological and functional changes in the body decreases the adaptation and prevention capacity leading into the decline of a life force. Various studies have been released to examine the anti-aging effects of herbal prescriptions. This experiment has chosen Boganhwan which is used for the deficiency of the liver function and studied the anti-aging factors by examining the biotransformation enzymes. The following results were obtained in this study: 1. Hepatic lipid peroxide activity was significantly suppressed in the experimental group treated with Boganhwan for 2 weeks at the dosage of 350mg/kg, while other dosage groups did not present much changes. 2. Insignificant changes were shown for the cytochrome P-450 level, aminopyrine demethylase, and aniline hydroxylase (AH) activities. Cytochrome P-450 do not appears to be a part of the detoxification scheme. 3. Boganhwan decoction treated group showed most significant increase of superoxide dismutase (SOD), catalase, superoxide, and glutathione activities at the concentration of 350mg/kg and 500mg/kg. 4. Glutathione S-transferase and glutathione made most significant increase at the decoction concentration of 350mg/kg and 500mg/kg compared to the control group. 5. Hepatic glutathione concentration, protein bound-SH, and nonprotein bound-SH made most significant increase at the decoction concentration of 350mg/kg and 500mg/kg compared to the control group. From the above results, Boganhwan decoction played an important role in eliminating foreign substances in the body excluding cytochrome P-450 enzyme system. Thus, Boganhwan decoction can provide substantial aid in preventing and treating senile related illnesses.

  • Bobitang(BBT) is one of the most important prescription that has been used in oriental medicine(dongyibogam) for recovering spleen condition. The study was done to evaluate effects of BBT water extract on the spleen lipid peroxide content and metabolic enzyme system changes. After pretreatment of BBT I (100mg/kg), BBT II(250mg/kg), BBT III(350mg/kg), BBT IV(500mg/kg) for 1 week, lipid peroxide content and metabolic enzyme system changes of the spleen was measured in 8 months rats. The results were obtained as follows : 1. The content of spleen lipid peroxide was significantly decreased in all experimental groups as compared with control, and best in BBT III IV treated groups. 2. The activity of spleen superoxide generation was significantly decreased in all experimental groups as compared with control, and best in BBT IV III treated groups. 3. The activity of cytochrome P-450 and aminopyrine demethylase wasn't significant change. 4. The activity of aniline hydroxylase was significantly decreased in BBT IV II treated groups, xanthine oxidase was significantly decreased in all experimental groups, aldehyde oxidase was significantly decreased in BBT IV treated group as compared with control. 5. The activity of antioxidant enzymes as superoxide dismutase, catalase, glutathione peroxidase was significantly increased in all experimental groups as compared with control. 6. The activity of glutathion S-transferase was significantly increased in all experimental groups, the concentration of spleen glutathione was significantly increased in BBT IV treated group as compared with control. 7. The activity of ${\gamma}$ -glutamylcystein synthetase was significantly increased in BBT III IV I treated groups as compared with control, the activity of glutathione reductase wasn't significant change. From the above results, BBT is cosidered to have effect of remove peroxide content and free radical that was made during ageing process. It is expected that treatment of BBT can be applied in future clinical study of delaying the ageing process.

Effect of p-Phenylenediamine Application to Rat Skin on the Liver Oxygen Free Radical Systems

  • To evaluate the influence of hepatic oxygen free radical systems on liver injury by topical p-phenylenediamine (PPD) application on rat skin, PPD (25 mg/16.5 $\textrm{cm}^2$) was topically applied to the abdominal region 5 times every other day and sacrificed. By PPD treatment, increasing rate of liver weight/body weight (%), serum activities of alanine aminotransferase and aspartate aminotransferase and decreasing rate of microsomal glucose-6-phosphatase activity were higher in the rats fed tungstate supplemented diet than those fed a standard diet. These findings indicate that group fed tungstate supplemented diet have more severe liver injury compared with group fed standard diet on topical PPD application. However, the activities of oxygen free radical generating enzymes such as xanthine oxidase (XO) and cytochrome P450 dependent aniline hydroxylase and those of oxygen free radical scavenging enzymes were not found to be different between these two animal groups. In the present study, a novel monitoring method to detect the generating of oxygen free radicals in liver extract was devised. Throughout this method, the oxidized PPD produced by oxygen free radicals was determined colorimetrically. The increasing rate of PPD oxidation by liver homogenate was higher in tungstate fed animals than in standard diet fed ones. Among the fractionations of liver extract, the mitochondrial and postmitochondrial fractions in the liver extract of tungstate fed animals led to a higher availability of PPD oxidation by PPD treatment compared with standard diet fed ones. In conclusion, these results suggest that an enhanced liver injury in tungstate fed animals treated with PPD may be due to oxygen free radicals produced in other systems except oxygen free radicals generating from cytosolic XO system. Especially, oxidative availability by PPD can be used for oxygen free radical detection in some tissue.

The Anti-hepatotoxic Effect of Ginseng in Rats: Meta-analysis

  • The aim of this meta-analysis was to systematically investigate the anti-hepatotoxic effect of ginseng in rats induced toxicity which damage to liver. Primary researches were gained on the ScienceDirect database, the DBpia, and the KISS, and the data about the effect factors in plasma and in enzyme were listed as many as possible. The effect factors were alanine transaminase (ALT), aspartate transaminase (AST), liver aminopyrine N-demethylase (AD), liver aniline hydroxylase (AH), liver 3,4-Methylenedioxyamphetamine (liver MDA), cytochrome P450 (P450), serum alkaline phosphatase (ALP), serum lactate dehydrogenase (LDH), cytochrome b5 (Cyto b5), glutathione reductase (GR), Liver glutathione S-transferase (GST), liver glutamyltransferase (GT), Liver (${\gamma}-GCS$), serum liver 3,4-Methylenedioxyamphetamine (serum MDA), serum sorbitol dehydrogenase (SDH), serum total protein (TP), serum ${\gamma}-glutamyltransferase$ (${\gamma}-GT$). To investigate the effect of ginseng, the mean difference (MD) between the group of rats induced by toxicity (RH) and the group of rats induced by toxicity with ginseng (RHG) were combined, and the significance of MDs were tested. The combined MDs were checked the biases caused by heterogeneity among studies and the publication biases, and adjusted by using random effect model and trim and fill method, respectively. The effect about ALT, AST, ALP, LDH, SDH, TP and ${\gamma}-GT$ in plasma factors were significant, and about AD, liver MDA, P450, Cyto b5, GR, GST, GT and ${\gamma}-GCS$ in enzyme factors were significant. The treatment with ginseng supplementation was significantly effected on plasma and enzyme factors of damaged-rats.
