Vertebrate Fauna, Speciation and Geological History in the Cheju Island (제주도의 척추동물상과 종분화 및 지사학적 역사)

  • Cheju island had that a fresh water Pisces composed of 9 Orders, 12Families, 24 Species, Amphibians composed of 2 Orders, 6 Families, 9 Species, Reptiles composed of 2 Suborders, 5 Families, 10 Species, Aves composed of 18 Orders, 49 Families, 236 Species and Mammalian composed of 6 Oredrs, 9 Families, 16 Species. So, total vertebrate's fauna were 35 Oredrs, 2 Suborders, 80 Families, 4 Subfamilies and 295 Species. Endemic species of the Cheju island were Mustela sibirica quelpartis, Apodemus agrarius vhejuensis, Micromys minutus hertigi and Crocidura russula quelpartis, Ageithalos caudatus trivirgatus, Sitta europaea bedfordi, Eophona personata personata and Dendrocopos oeucotos quelpartis, Troglodytes troglodytes fumigatus, Parus major minor, Cettia diphone cantans and Hynobius leechii quelpartis. Especially, Sibynoghis collaris and Anguilla mauritiana were only habitated in the Cheju island. And the Cheju island was formed in extending from Plieocene to Pleistocene. Differentiation of species was continued by geological isolation 0.3 million years that repeating glacial epoch and interglacial epoch.

Isolation and Characterization of Antibiotic Resistant Vibrio Strains from Japanese Eel (Anguilla Japonica) Cultured in Korea (국내산 양식 뱀장어에서 항생제 내성 비브리오 세균 분리 및 특성)

  • Continuous mortality in commercially cultured Japanese eel (Anguilla japonica), showing symptoms of dermal ulcerations and focal hemorrhages on the body, occurred on a private farm in November, 2019 in Korea. A series of mortality had been described in one local eel culture farm from November to December in 2019. From the three cases, three isolates of Vibrio spp. were recovered from the blood, ascitic fluid, and kidney of the dead fish, respectively. Based on the 16S rRNA sequence comparisons, the Vibrio isolates from the 1st and 3rd cases (strain named 1E1-2 and 3K1-2) were identified as V. fluvialis and the isolate from the 2nd case was identified as V. plantisponsor (strain named 2A3-1). Moreover, the 16S rRNA-based phylogenetic analysis revealed that strain 1E1-2 and 3K1-2 were most similar to V. fluvialis NBRC 103150T, and strain 2A3-1 was most similar to V. plantisponsor NBRC103148T. According to the results of the antibiotic resistance determination, V. fluvialis 1E1-2 showed intermediate resistance to tetracycline and chloramphenicol, and was resistant to trimethoprim-sulfamethoxazole. V. plantisponsor 2A3-1 showed intermediate resistance to ciprofloxacin and levofloxacin, and was resistant to trimethoprim-sulfamethoxazole. V. fluvialis 3K1-2 showed intermediate resistance to tetracycline, and was resistant to ampicillin and trimethoprim-sulfamethoxazole. These results have provided the evidences on the occurrence of antibiotic-resistant Vibrio infection in commercially cultured Japanese eels are present in Korea.

Comparative study on growth of leafy vegetables grown in a hybrid BFT-aquaponics using Japanese eel, Anguilla japonica and hydroponics

  • Aquaponics is a cultivation system that combines aquaculture and agricultural hydroponics. This study investigated the productivity of leafy vegetables cultivated in the hybrid biofloc technology-aquaponics (HBFT-AP) using Japanese eel fed two kinds of diets (Mash, commercial powdered feed and extruded pellet [EP]) and hydroponics (HP).The mash was fed to the fish in a type of a dough mixed with water for 6 weeks (Exp1) and switched to the EP containing 2.7% monobasic potassium phosphate (MKP) for subsequent 6 weeks (Exp2). Leafy vegetables of 8 cultivar were employed in the experiment and water quality [dissolved oxygen (DO, mg/L), pH, water temperature (℃), electrical conductivity (EC; ㎲/cm), turbidity (NTU), TAN (NH3 + NH4+) (mg /L), NO2-N (mg/L), NO3-N (mg/L) and PO4-P (mg/L)] was measured 6 times a week. Leafy vegetable productivity (HBFT-AP vs HP) was compared in respective experiment, which was similar or somewhat higher in HBFT-AP. During the 12-week feeding trial, concentrations of nitrite (NO2-N) and phosphorus (PO4-P) were kept low in variability while total ammonia nitrogen (TAN) and NO3-N levels increased with time in HBFT-AP. At the end of two feeding trials, values of weight gain (WG, %), feed efficiency (FE, %), specific growth rate (SGR, %) and protein efficiency ratio (PER) were higher in Exp2 than in Exp1. As well, higher values in hematocrit (PCV, %), plasma K (mEq/L) and inorganic phosphorus (mg/dL) were found (p < 0.05) in Exp2 where fish were fed the EP (EP: 38.60%, 2.80 mEq/L and 7.04 g/dL; Mash: 33.20%, 1.95 mEq/L and 5.50 g/dL). Leafy vegetables in HBFT-AP using Japanese eel fed the EP with MKP 2.7% had a productivity similar (4 kinds of cultivar) or somewhat higher (4 kinds of cultivar) compared to those in HP. Also, Japanese eel fed the EP showed higher values of Pi (m/dL) and K (mEq/L) in plasma compared to those fed commercial powder diet.

Assessment of Neuronal Cell-Based Cytotoxicity of Neurotoxins from an Estuarine Nemertean in the Han River Estuary

  • A heteronemertean, Yininemertes pratensis, was collected in Han River Estuary, South Korea. This estuarine nemertean has been known by the local fishermen for harmful effects to the glass eels, juveniles of Japanese eel Anguilla japonica, migrating to fresh water. The present study confirmed the neurotoxic effects of this heteronemertean ribbon worm at the cellular level. Derivative types of neurotoxic tetrodotoxin (TTX), 5,11-dideoxy TTX (m/z 288) and 11-norTTX-6(S)-01 (m/z 305.97), were identified through HPLC and MALDI-TOF MS. However, significant neurotoxicity was confirmed in the fraction containing an undefined molecule corresponding to the 291.1 (m/z) peak, when tested in rat primary astrocytes and dorsal ganglion cells. This study is the first to report neurotoxins of the estuarine nemertean, fairly abundant in the Han River estuary, and suggests the long-term monitoring of population dynamics and surveillance of the toxicity in this river estuary.

Comparative Efficacy of Antifungal Agents for Aquaculture Fish and their Eggs (양식 어류와 이들 난에 대한 항곰팡이성 약물들의 효과 비교)

  • In fresh water fish hatcheries and farms, Saprolegniales often cause serious mortality to the fish and their eggs. Malachite green is an effective antifungal agent, but is carcinogenic to fish and humans. Alternative antifungal agents are needed. Presently, we tested various concentrations of MBT-01108 (Opuntia ficus-indica extracts) alone and in combination with bronopol, formalin and sodium chloride (MBT-01108 mixture) on in vitro mycelial growth and in vivo remediation of adult eel, Anguilla japonica, infected with Saprolegnia sp. and fertilized eggs of chum salmon, Oncorhynchus keta, to evaluate the compounds' antifungal efficacy on eyed-egg and hatching rates. MBT-Oll08 mixtures incorporating bronopol and formalin at respective concentrations of 50 and 30 parts per million (ppm), and 100 and 20 ppm were most effective in controlling Saprolegnia in vitro and in vivo (P<0.05). Repeated daily exposures to 50 ppm and 100 ppm MBT-01108 were more effective than exposure every 2-3 days post-fertilization for the inhibition of Saprolegnia infection of rainbow trout, Oncorhynchus mykiss eggs as compared with control (0 ppm).

Effect of TBT and PAHs on CYP1A, AhR and Vitellogenin Gene Expression in the Japanese Eel, Anguilla japonica

  • Gene expressions of cytochrome P4501A (CYP1A), aryl hydrocarbon receptor (AhR) and vitellogenin (Vg) by endocrine disruptors, benzo[${\alpha}$]pyrene (B[a]P) and tributyltin (TBT) were examined in cultured eel hepatocytes which were isolated from eels treated previously with B[a]P (10 mg/kg) or estradiol-$17{\beta}$ (20 mg/kg) in vivo, and the relationship between CYP1A, AhR and Vg genes were studied. When the cultured eel hepatocytes were treated with B[a]P ($10^{-6}-10^{-5}M$) the gene expressions of CYP1A and AhR were enhanced in a concentration-dependent manner. However, when treated with TBT ($10^{-9}-10^{-5}M$) the gene expressions of CYP1A and AhR were suppressed at high concentrations ($10^{-6}-10^{-5}M$), while having no effects at low concentrations ($10^{-9}-10^{-7}M$). Gene expression of Vg was also suppressed by TBT in a concentration-dependent manner in cultured eel hepatocytes which was previously treated in vivo with estradiol-$17{\beta}$.

  • A study on efficacy and safety of Bio-Gammamix was performed to use for the preventive drug against bacterial diseases to mirror carp(Cyprinus carpio), rainbow trout(Salmo gairdneri), eel(Anguilla japonica) and flounder(Paralichthys olivaceus). The main ingredients of Bio-Gammamix were ferrous fumarate and mixed extrated powders of licorice root, ox bile and garlic. The test drug was proved to improve feeding habit and feed efficiency and decrease cumulative mortality remarkably. After administration, no side effects and histopathological changes were observed. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1kg/ton of fish body weight perorally once or two times in a day for 7 days and Bio-Gammamix is thought to be used effectively an widely against most bacterial fish pathogens.

  • To prevent food-poisoning outbreaks by Vibrio vulnificus the antimicrobial efficacy of grapefruit seed extract (GFSE) was examined. Minimal inhibitory concentration (in vitro) for the microorganism was found to be 50∼100 ppm. Transmission electron micrographs of Vibrio vulnificus showed the biocidal action of this natural antimicrobial agent would be related to specific respiratory effect coupled with the destruction of permeable function of microbial cell membrane. After Anguilla japonica GFSE-injected to the body was incubated in the seawater contaminated by Vibrio vulnifiucs the fish meats were taken up, mixed with control diet and used as a diet in the feeding experiment. In this experiment the effect of GFSE treated with fish muscle on body weight, protein efficiency ratio, serum enzymes and serum blood components of broiler chicks was investigated. It is proved from this study that there is neither Vibriosis nor toxicity associated with GFSE itself an fish meats treated with it when it is injected to the fish body at a level of 250 ppm or less.

On the Ichthyofauna of the Tamjin River System (탐진강 수계의 어류상에 관하여)

  • The ichthyofauna was investigated at the thirteen sites in the Tamjin River from August 1994 to February 1995. The collected fishes were classified into thirty-nine species belonging to 29 genera and 10 families. Among these 12 species were classified as endemic species or subspecies in Korea. Major species in this river system appeared to be Zacco temminchi, Acanthorhodeus gracilis, Zacco platypus, Rhodeus uyekii, Puntungia herzi and Squalidus gracilis majimae. Odontobutis obscura interrupta was collected for the first time in this area.

Antibacterial efficacy and safety of copper sulfate pentahydrate to cultured fish (양식 어류에 있어서 황산동(copper sulfate pentahydrate)의 안전성과 살균효과에 대한 연구)

  • Antibacterial efficacy and safety of copper sulfate pentahydrate (5% solution) was evaluated in laboratory and field studies for disinfection of some fish bacterial pathogens in cultured fish, flounder(Paralichthys olivaceus), mirror carp(Cyprinus carpio), gold fish(Carrassius auratus) and eel(Anguilla japonica). Dipping of fishes for acute toxicity was performed for a period of 48h, and the TLm value(median tolerance limit) were 4.0ppm in mirror carp and flounder, 4.4ppm in rainbow trout, and 10.0ppm in gold fish. After exposure below than TLm value for 1 month, no side effects and histopathological changes were observed. The test drug were shown high antibacterial activities against most bacterial pathogens by exposure in the laboratory and field trials. However, Aeromonas salmonicida showed a resistance to the dosage. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1ppm for 6hr and the test drug is thought to be used effectively and widely as a disinfectant against most bacterial fish pathogens.
