To investigate the potential therapeutic value of a plant extract against amyloid ${\beta}-peptide-induced$ cell damage, we first screened extracts of 250 herbs, and finally selected a water extract of Spinacia oleracea for further study. This extractshowed the potential to inhibit the reactions of oxidants. We measured the angiotensin-converting-enzyme (ACE) inhibitory activity of the extract, and assessed the ability of the extract to protect neuronal cells from chemical-induced cell death. SH-SY5Y neuroblastoma cells were used in this assay. The extract exerted protective effects on $H_2O_2-induced$ cell death, when $H_2O_2$ was used at 100 M, 200 M, and 500 M (protection of 87%, 73%, and 58%, respectively). When 50 M of amyloid ${\beta}-peptide$ was added to the test cells, however, the extract had no protective effect on cell death. The extract inhibited ACE activity in a dose-dependent manner, and exhibited potent protection against the deleterious effects of $H_2O_2$. In sum, these results suggest that a water extract of Spinacia oleracea has the potential to afford protection against chemical-induced neuronal cell death, and the extract may be useful in the treatment of neurodegenerative diseases. The precise molecular mechanism of neuroprotection is under investigation.

This study evaluated the protein recovery and functional properties and biological activity of boiled and steamed process water (BPW and SPW, respectively) generated from the preparation of concentrated roe of bastard halibut (BH; Paralichthys olivaceus), skipjack tuna (ST; Katsuwonus pelamis), and yellowfin tuna (YT; Thunnus albacares) using the cook-dry process. The protein loss from the water extracts (EXT) of 100 g of roe protein was 15.05-19.71% and was significantly (P<0.05) higher than that of BPW (5.47-10.34%) and SPW (3.88-8.18%). The foam capacity of BPW (166-203%) and SPW (15-194%) was better than that of EXT. The emulsifying activity index of the original samples was lower than those ($15.40-107.86m^2/g$) of diluted protein samples. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and the reducing power of BPW and SPW were stronger than those of EXT. The 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid ($ABTS^+$) radical scavenging activity of EXT (0.028-0.045mg/mL) was significantly higher those of BPW and SPW. The angiotensin I-converting enzyme (ACE) inhibitory activity of SPW was the highest for BH (1.04 mg/mL), followed by YT and ST. The predominant amino acids in SPW were Glu, Ala, Leu, and His. These results demonstrate that processing water containing diluted organic components, including protein, can be consumed directly by humans as a functional reinforcing material after appropriate concentration processes.

This study was conducted to investigate the physiological activities of the ethanol extracts from 10 different strains of Pholiota nameko. In addition, the ${\beta}$-glucan and polyphenol contents from these strains were also measured. Total polyphenol contents from all the strains were more than 40 mg% with the highest content of $61.50{\pm}0.59$ mg% and ${\beta}$-glucan contents were 30% with the highest content of $37.20{\pm}1.12%$. The highest inhibitory activities on ${\alpha}$-amyloglucosidase activity and nitric oxide production were $13.78{\pm}0.56%$ and $56.59{\pm}7.11%$, respectively. However, ethanol extracts from all the strains have little effects on DPPH radical and nitrite scavenging rates and the activity of angiotensin I converting enzyme. The cytotoxic activity of ethanol extracts from all the strains on A549 cell was shown upto 30% with the highest effect of $47.96{\pm}8.46%$ treated at 1 mg/mL concentration.

Fresh onions were extracted by two methods of conventional extraction(CE) and microwave assisted extraction(MAE) under different extraction conditions. Onion extracts obtained by CE and MAE were examined in oleoresin yield and physiological activities. The optimal extraction time of MAE was about 5 minutes, whereas that of CE was 2 hours. Therefore extraction time was shortened drastically by MAE but there was no significance in oleolesin yield. The electron donating abilities also showed negligible difference between two extracts obtained by CE and MAE, and 45% level in CE and 50% level in MAE. The nitrite scavenging effect was diminished while pH was increasing, and showed a high elimination effect over 85% at pH 1.2. The tyrosinase inhibitory effect was 20% level in both CE and MAE when water was used and 40% level when ethyl alcohol was used. The angiotensin I converting enzyme inhibitory effect showed higher activity with 70% level in MAE than 60% level in CE. The pyruvic acid content was 6.8 and 6.4 moles per 1g of fresh onion by CE and MAE when water was used, and was 4.3 and 5.6 moles by CE and MAE when ethyl alcohol was used.
The phenolic compounds extracted from Aster scaber were examined for their biological activities owing to their potential use in health and beauty food products. The phenolic content in water and 60% ethanol extracts were $11.1{\pm}0.11$ and $4.18{\pm}0.05mg/g$, respectively. The DPPH radical scavenging activities of the water and ethanol extracts were 87% and 91% at $50{\mu}g$ phenolics/mL, respectively. At the same phenolics concentration, the respective extracts showed 84% and 95% for ABTS radical decolorization activities and 95% and 97% for TBARs. The antioxidant protection factors for the water and ethanol extracts at $200{\mu}g$ phenolics/mL were 1.87 and 2.22 PF, respectively. Enzyme inhibitory activities of the water and ethanol extracts ($50{\mu}g$ phenolics/mL) were 50.8% and 69.4% on angiotensin converting enzyme, 91% and 80% on xanthine oxidase, and 24% and 89% on ${\alpha}$-amylase, respectively. The tyrosinase inhibitory activities indicating skin-whitening were 47% and 25% for the water and ethanol extracts, respectively. Anti-wrinkle effect of the water extract was relatively higher than that of the ethanol extract. These results suggest that the water and ethanol extracts of Aster scaber can be used as an ingredient in health and beauty food products.

Physiological properties of Sarcodon aspratus extracts were investigated. Yields of water, 30, 60, and 90% ethanolic extracts were 52.10, 46.90, 41.50, and 32.20%, respectively. The 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of 60% ethanolic extract was 40.40% at a concentration of 4 mg/mL, which was highest of all the extracts. The polyphenol content of 60% ethanolic extract was the highest at 16.23 mg/g at a concentration of 1 mg/mL. Superoxide anion radical scavenging activities were 56.30~77.05% at concentrations of 1, 2, and 4 mg/mL, which were better than the ascorbic acid activity (56.19% at 10 mg/mL) (P<0.05). Nitrite-scavenging ability was the most effective in 30% ethanolic extract. A pH level of 3.0 was the most effective for all extracts. Angiotensin-converting enzyme inhibitory activities of Sarcodon aspratus extracts were from 52.67 to 89.52%.

This study was conducted to investigate the changes of major components and bioactivities of tea produce. The tea produce were made by various methods, with different degree of fermentation during manufacturing process. Except green tea, degrees of fermentation in Wizo Tea, Ilsoae Tea, and Hwang Tea were $5{\sim}10%,\;50{\sim}60%$ and $70{\sim}80%$ respectively. The result are as follows : The general component(moisture, crude ash, crude lipid, and crude protein) and the content of total polyphenol in tea products were not shown significant difference during the fermentation process. The content of caffeine in tea extracts decreased sharply as degree of fermentation of tea. In comparison of hunter values in tea extract, lightness was decreased as fermentation redness (a) was all (-), and yellowness(b) was increased sharpy with degree of fermentation. Radical scavenging activity using DPPH of tow kinds of tea was potent and decreased generally with degree of feimentation. Inhibitory effects of tea extracts against angiotensin I converting enzyme were also potent.
Barley brans, divided into fractions I $(pearlers\;1{\sim}4)$, II $(pearlers\;5{\sim}20)$, and III $(pearlers \;21{\sim}24)$, germ, and broken kernels were collected as pearling by-products produced by an industrial process consisting of 24 consecutive barley pearlers. The pearling by-products were extracted with 75% ethanol, and polyphenol extracts were separated using Sepabeads SP-850 resin. Total polyphenol content was the highest (43.68%) in the polyphenol extract separated from fraction I. Polyphenol compounds analyzed by HPLC were tentatively identified as proanthocyanins and prodelphinidin polymers, which were known to have antioxidative and antimutagenic activities. Antioxidative abilities were estimated to be in the following order: fraction I>germ>fraction II>fraction III. At 0.01% polyphenol extract, angiotensin I-converting enzyme inhibitory activity was the highest in the germ fraction (40.04%). Nitrite-scavenging activity was over 70% in all fractions. Superoxide dismutase-like activity was in the range of $64{\sim}78%$ in fractions I and II.
The efforts were made to optimite ethanol extraction from persimmon leaf with the time of extraction$(1.5{\sim}2.5\;hrs)$, the temperature of extraction$(70{\sim}90^{\circ}C)$, and the concentration of ethanol$(0{\sim}40%)$ as three primary variables together with several functional characteristics of persimmon leaf as reaction variables. The conditions of extraction was best fitted by using response surface methodology through the center synthesis plan, and the optimal conditions of extraction were established. The contents of soluble solid and soluble tannin went up as the concentration of ethanol went up and the temperature of extraction went down, and the turbidity went down as the concentration of ethanol went down. Electron donation ability was hardly affected by the extraction temperature and had the tendency to go up as the concentration of ethanol went up. The inhibitory activity of xanthine oxidase(XOase) had the tendency to go up as both the concentration of ethanol and the temperature of extraction went up. The inhibitory activity of angiotensin converting enzyme(ACE), the significance of which still was not recognized, showed the maximum when the concentration of ethanol was 27%. In result, the optimal conditions of extraction was the extraction time of two hours, the extraction temperature of $75{\sim}81^{\circ}C$, and the ethanol concentration of $33{\sim}35%$.

This study was conducted to improve the yield of extracts from Alaska pollock Theragra chalcogramma head and sea tangle Laminaria japonica byproducts using various commercial enzymes, such as Alcalase, Flavourzyme, Neutrase (NH), and Protamex. Among the enzymatic hydrolysates, the yield was highest in hydrolysate incubated with NH for 4 h. NH-treated hydrolysates (NHH) also improved functional properties, such as angiotensin-I converting enzyme (ACE) inhibitory activity and 2,2-diphenyl-1-picryldrazyl (DPPH) radical scavenging activity, as compared to extracts from Alaska pollock head and sea tangle byproducts. Total free amino acid and taste values of NHH were 379.7 mg/100 mL and 24.03, respectively, after digestion for 4 h. These values are 2.2-fold and 1.9-fold higher compared with those of water soluble fractions extracted from Alaska pollock head and non-forming sea tangle, respectively. According to the taste value results, the major taste-active compounds among free amino acids of NHH were glutamic acid and aspartic acid. These results suggest that NHH can be used as an ingredient for natural seasoning preparation.
