Physiological Activities of Bioconversion Products Using Bacillus Subtillis KJ-3 and Their Mixtures (Bacillus Subtilis KJ-3를 이용한 생물전환물 및 그 혼합물의 생리활성)

  • This research was performed to develop a new material consisting of a mixture of Red Allium cepa (RA) Cucurbita moschata duch (CM), and Angelica gigas Nakai (AG). RA and CM have low storage stability because of their high moisture content. Therefore, their major components were extracted and used for the research after a content analysis. In order to overcome these limitations, the quercetin from RA, ${\beta}-carotene$ from CM, and decursin/decursinol angelate (D/DA) from AG were separately extracted, and the biochemical activity of each extract and mixture was compared. RA was bioconverted by the Bacillus subtillis KJ-3 (BS3) after ethanol extraction. After bioconversion, the quercetin content of RA was increased by 128.9%. ${\beta}-carotene$ was detected in the CM ethanol extract and its content was very low concentrations at 0.2 mg/g. The AG ethanol extract (1 mg) contained 0.4146 mg and 0.3659 mg of D/DA, respectively. The purity of the D/DA was found to be about 78%. The flavonoid and polyphenol content of each extract and their mixtures (mixture 1 (RA:CM:AG = 5:2:3), mixture 2 (RA:CM: AG = 3:5:2), and mixture 3 (RA:CM:AG = 3:2:5)) were measured. In addition, the cell survival rate, anti-inflammatory activity, and antioxidant ability were also evaluated. In all the results, the antioxidant activity of mixture 3 was most effective. Therefore, these findings provide basic data for future food development using a 3:2:5 mixture of RA, CM, and AG.

A Study of the Transdermal Permeation of Lotion Formulations Containing Angelica gigas Nakai Extracts in Franz Diffusion Cells (Franz diffusion cell을 이용한 참당귀 추출물 함유한 로션제형의 피부 투과 연구)

  • Angelica gigas Nakai (AGN) has been used in Korean herbal medicine for various pharmacological activities, such as to create antioxidant and skin whitening effects. Decursin and decursinol angelate of AGN extracts can be used as potential active drugs and cosmetic ingredients. This study investigated the possibility of topical delivery of AGN extracts using a manufactured emulsion system. Lotion was formulated by using Tefose® and paraffin for the oil phase, Kolliphor RH 40 for the surfactant and solubilizing agent-which showed high solubility in water (0.82 mg/ml)-and a water phase with a carbomer. In vitro skin permeation of decursin and decursinol angelate was determined using a Strat-M® membrane in Franz diffusion cells. Lotion samples as the experimental group (248.08±19.72 ug/cm2) significantly increased the permeation of decursin and decursinol angelate for up to 24 hr compared to the control group (119.18±19.23 ug/cm2). The permeability was also characterized by the flux (penetration rates) and Kp (permeability coefficient) values. The experimental group (17.20±1.23 ug/h/cm2 and 5.73±1.39 cm/h*10-3) had higher flux and Kp than the control group (8.22±1.24 ug/h/cm2 and 2.74±0.51 cm/h*10-3). Lotion with decursin and decursinol angelate of AGN extracts could be used for the topical application of drug and cosmetic products.

The Growth Characteristics of Angelica gigas Nakai according to Cultivation Regions (재배지별 참당귀의 생육특성)

  • 참당귀(Angelica gigas Nakai)는 중국의 동북부지역, 일본 및 한반도 전역에 자생하고 있으며, 예로부터 보혈, 활혈지통, 윤장등의 효능으로 심신혈허, 월경불순, 통경경폐 등의 병증에 주로 사용되었고, 8월부터 뿌리가 급격히 비대하여 10월 말에서 11월 초에 수확을 한다. 본 연구는 참당귀의 주산지인 봉화, 평창을 비롯한 영양, 인제 등 4지역을 선정하여 미세기상장비를 설치 및 토양의 이화학성 분석을 통해 지역별 생육환경을 측정하였고, 또한 참당귀의 생육특성을 조사하였다. 그 결과 5월에서 10월까지 평균 대기온도와 일사량은 인제지역에서 다른지역에 비해 비교적 높은 경향을 나타냈다. 또한 토양온도는 5월에서 8월까지는 영양지역이, 9월과 10월은 봉화지역이 다른지역에 비해 비교적 높은 것으로 조사되었다. 지역별 토양 성분의 이화학적 특성은 봉화지역이 유기물(5.17%), 전질소(0.19%), 유효인산(2187.76mg/kg), 칼륨(1.53cmol+/kg)의 함량이 다른 지역에 비해 높은 경향으로 확인되었고, 인제지역이 칼슘(12.63 cmol+/kg), 마그네슘(3.7cmol+/kg)등 미량원소의 함량이 높은 것으로 분석되었다. pH는 지역별 차이가 확인되지 않았으며, 영양지역의 토성이 사질식양토로 배수성의 차이는 확인되었다. 이에 따른 참당귀의 생육특성은 초장과 줄기직경은 영양지역에서 각각 70.95cm와 35.72mm로 가장 높게 측정되었으며, 잎 길이및 너비는 평창지역에서 각각 42.84cm, 52.39cm로 가장 높게 측정되었다. 뿌리직경은 영양에서 50.49mm, 뿌리길이는 봉화에서 38.05cm, 생중량은 평창에서 328.23g으로 다른지역에 비해 높은 경향을 나타냈다. 상기 결과를 바탕 재배환경과 생장량간의 상관관계를 분석하였다. 재배환경 중 대기 및 토양온도, 대기 습도는 실험이 수행되어진 전 지역에서 이상고온이나 저온과 같은 특이점 없이 비슷한 양상으로 측정되었고, 이에 따라 참당귀생장에 대해 유의한 상관관계가 분석되지 않았다. 토양습도는 지상부(초장, 줄기직경, 잎길이, 잎너비)의 생육과 음의 상관관계에 있다고 사료되나 유의성이 검증되지 않았다.

Effects of Gamma Irradiation on Antimicrobial Activity of Korean Medicinal Herb Extracts (한약재 추출물의 향균활성에 대한 감마선 조사의 영향)

  • Irradiated and non-irradiated Korean medicinal herbs were extracted by water and 70% ethanol. Antimicrobial activity of these extracts were invesigated against selected food hygiene microoganisms. The ethanol extracts of the non-irradiated Agrimonia pilosa ledebour japonica Nakai, Curcuma longa Linne and Angelica gigas Nakai were completely inhibited on four species of bacteria, such as Vibrio parahaemolyticus, Clostridium perfringenes, Bacillus subtilis and Staphylococcus aureus. Also, above four strains did not have antibacterial activity in the water-ethanol mixtures. Futhermore, the ehtanol extracts of the non-irradiated Agimonia pilosa ledebour Japonica Nakai, Curcuma Iedoaria Roscoe, Curcuma longa Linne and Scutellaria baikalensis George were shown inhibitory effects against Aspergillus flavus and Pen icillium islandicum. And the water extract of Scutellaria baikalensis George was the same effect to these molds. Essentially the same results were observed when samples irradiated at a dose of 10 kGy.

Toxicity and Antitumor Activities of Korean Medicinal Plants (I) (한국산 생약의 독성 및 항암 작용 (제1보))

  • Thirty species of Korean medicinal plants which have been frequently used in Oriental herb prescriptions were evaluated on their acute toxicity and potential antitumor activities against P-388 lymphocytic leukemia. The criteria for toxicity evaluation of measuring weight loss, toxicity day survivors and computing log cell kill indicated that 11 species possessed acute toxicity according to the doses administered. No significant antitumor activities were observed while the root of Angelica gigas Nakai (Umbelliferae) exhibited only 24% increased life span.
