Effect of Root Head Diameter of Seedling on Growth and Bolting Response in Angelica gigas Nakai (묘두직경(苗頭直徑)에 따른 참당귀의 생육(生育) 및 추태반응(抽苔反應))

  • This study was to determine optimal sizeof root top using Suwon 2 selected from the Jinbu local population though pure line selection and five root head diameters of $0.11{\sim}0.30,\;0.31{\sim}0.50,\;0.51{\sim}0.70,\;0.71{\sim}0.90\;and\;0.91{\sim}1.10cm$. Emergence rate, characteristics of top, and root were increased with increasing diameterof seedling root. In the group of root head diameter from 0.51cm to 0.70cm, bolting rate was the lowest being less than 3.3% and root yield was the highest being more than 300kg/l0a. Bolting rate was positively correlated with plant height while showed negatively correlated with root yield. The study showed that the optimum of root head diameter raised seedlings in the open field appeared to be from 0.31 to 0.70cm.

  • For standardization of LMK02 quality, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelica gigas Nakai in the constituents of LMK02 were estimated as indicative components. From LMK02 water extract, has been used in vitro test for its beneficial effects on neuronal survival and neuroprotective functions, particularly in connection with APP-related dementias and Alzheimer's disease (AD). $A{\beta}$ oligomer derived from proteolytic processing of the ${\beta}$-amyloid precursor protein (APP), including the amyloid-${\beta}$ peptide ($A{\beta}$), play a critical role in the pathogenesis of Alzheimer's dementia. We determined that oligomer amyloid-${\beta}$ ($A{\beta}$) have a profound attenuation in the increase in rat hippocampus H19-7 cells from. Experimental evidence indicates that LMK02 protects against neuronal damage from cells, but its cellular and molecular mechanisms remain unknown. Using a hippocampus cell line on $A{\beta}$ oligomer-induced neuronal cytotoxicity, we demonstrated that LMK02 inhibits formation of $A{\beta}$ oligomer, which are the behavior, and possibly causative, feature of AD. In the Red Ginseng, the average amounts of Ginsenoside Rg3 were $47.04{\mu}g/g$ and $42.3{\mu}g/g$, 90 % of its weight were set as a standard value. And, in the Angelica gigas Nakai, the average amounts of Decursin were 2.71 mg/g and 2.44mg/g, 90 % of its weight were also set as a standard value. The attenuated $A{\beta}$ oligomer in the presence of LMK02 was observed in the conditioned medium of this $A{\beta}$ oligomer-induced cells under in vitro. In the cells, LMK02 significantly activated antiapoptosis and decreased the production of ROS. These results suggest that neuronal damage in AD might be due to two factors: a direct $A{\beta}$ oligomer toxicity and multiple cellular and molecular neuroprotective mechanisms, including attenuation of apoptosis and direct inhibition of $A{\beta}$ oligomer, underlie the neuroprotective effects of LMK02 treatment.

Antioxidative Activities and Contents of Polyphenolic Compound of Medicinal Herb Extracts (한약재 추출물의 폴리페놀 화합물과 항산화 활성)

  • The antioxidative activity was measured on the substances of water and ethanol soluble extract from Astragalus membranaceus Bunge, Chrysanthemum morifolium Ramat, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Zizyphus jojoba Miller, Paeonia lactiflora Pallas, Cnidium officinale Makino by four different in vitro experimental models of DPPH (a,a'-diphenyl-$\beta$-picrylhydrazyl) method, superoxide dismutase like activity, thiocyanate method, and TBARS (thiobarbituric acid reactive substances) method. The Lycium chinensis Miller contained the highest amount of polyphenolic compounds. The electron donating ability of water extract from Glycyrrhiza uralensis Fischer and ethanol extract from Chrysanthemum were higher than those of the others. The superoxide dismutase-like activity of water extract from Astragalus membranaceus Bunge was the highest among those of all the others. The water extract from Zizyphus jujuba Miller showed the highest antioxidative activity determined by TBARS method. Compared to the control, the inducing period associated with the oxidation degree was delayed up to 8 days in both the water extract from Chrysanthemum, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, and Paeonia lactiflora Pallas and the in ethanol extract from Chrysanthemum and Glycyrrhiz uralensis Fischer. These results support that water and ethanol extracts from 8 kinds of medicinal herbs contain antioxidative compounds.

  • Many plants, which collected from Korea, were applied to antitumor and cytotoxic screeing tests against sarcom 180 a ascitec in mice, V-79 KB and P388 cultured cells. The results are summarixed as follows:1) The total packed cell volum method has been used for the antineoplastic screening for from natural higher plants in Korea. By this method, we have found out that the root, leaf and stem of Tripterygiu, regelii Spragne & Taketa having strong antineoplastic activity and also Rumex Japonicus Houtt. Eragrositis ferru-ginea Beauv. and Patrinia scabio-saefolia Fischer showed significant activity to anticancer tumor while cynanchum wilfordii Hemsley, and Rosa polyantha Sieb. et Zacc. showed slight activity to antitumor. 2) Among the 13 tested plants, the root and stem of Tripterygium regelii Spragne & Taketa and Amethystanthus excisus Nakai showed strong antitumor activity by the V79 cytotoxic cell screening test. 3) Twelve plants, which are glowing in mountainous area of Korea tested to anticancer activity. From the results, Eragrositis ferru-ginea Beauv., Angelica gigas Nakai, Geranium sibiricum L., Patrinia scabio-saefolia Fisher, Cynanchum wilfordii Hemsley, and Rubia akane Nakai have been proved to be anti-cancer plants by using P388 cell cultured method. 4) Tripterygiu, resgelii Spragne & Taketa, Eragrositis ferru-ginea Beauv., Patrinia scabio-saefoli Fisher, Cynanchum wilfordii Hemsley and Rasa polyantha Sieb. et Zacc., var. genuina Thunb. showed strong anti-tumor activity both total packed cell volume method and Cytotoxicity method.

Current status on the development of molecular markers for differentiation of the origin of Angelica spp. (당귀(Angelica spp.)의 기원분석에 관한 분자생물학적 연구 현황 및 향후과제)

  • The dried root of Angelica species is used in traditional Chinese medicine in East Asia, particularly in Korea, China and Japan. Since the plant origin differs in these countries, they are often misused or adulterated in the commercial markets, resulting in distrust among the consumers. Enormous efforts have therefore been focused to distinguish the origin for the Angelica genus, by using morphological or cytogenetical analyses, and chemical markers based on biochemical analyses of secondary metabolites. DNA is considerably stable against different cultivation conditions, and to treatment and processing after harvesting of plants. Hence, several researches have been filed for the development of molecular markers, based on the single nucleotide polymorphisms in specific regions of DNA. However, there are several obstacles for application in the commercial markets, concerning the reproducibility, accuracy, sensitivity, and rapidity of these tests. In this review, we summarize the research achievements that help classify the origin of Angelica species, in particular, Angelica gigas Nakai. A. sinensis(oliv.) Diels, A. acutiloba Kitag., and A. acutiloba var. sugiyamae Hikino. Further researches are required for practical applications.

Chemical Weed Control in Direct-Seeded Angelica gigas (참부귀(富歸) 직파재배(直播栽培)에서 제초제(除草劑)에 의한 잡초방제(雜草防除))

  • This study was conducted to evaluate the effect of systematic herbicide application and polyethylene(PE) film mulching in direct seeded Angelica gigas Nakai. Freshly-collected-seed showed 86% of emergence rate and required 18 days of emergence duration. Cold treatment of seeds increased emergence rate by more than 90% and shortened 2 days of emergence duration. Paraquat) 1,1'-dimethyl-4,4'-bipyridylium dichloride was the most effective when applied just before emergence of A. gigas and provided 96% of emergence rate and 91% of weeding effect when applied 23 days after seeding. Soil applied herbicides used did not cause any phytotoxicity on emergence with application of 2-fold recommended rate and gave more than 90% of weeding effect. Black PE film mulching gave excellent emergence rate and weeding effect, and shortened emergence duration by 8 days.

  • To establish appropriate seed production system in Angelica gigas. Germination of seeds with different bolting years and the growth characters of plants originated from these seeds were in­vestigated. Seed weight from 1st year bolting plant was highest. In germinability according to seeds with different bolting years, seeds from 1st year bolting plant were higher than seeds from 2nd, 3rd years bolt­ing one. In growth characters of plants originated from seeds with different bolting years, as the bolting years became late, root growth increased and top growth decreased, therefore T/R ratio decreased.

Analysis of Index Component Content and Antioxidant Activity According to the Root Diameter of Angelica gigas Nakai (참당귀 뿌리 직경별 지표성분 함량 및 항산화 활성 분석)

  • Angelica gigas Nakai (AGN) is a traditional medicinal herb especially in Korea. It contains pyranocoumarins, which are major active components including decursin (D) and decursinol angelate (DA). This study was carried out to determine the change in active component content and antioxidant activity depending on the root diameter of AGN. Several processing steps are involved to use AGN roots as medicine. The dried AGN roots are divided into body (B), thick root (TkR), medium root (MR) and thin root (TnR) according to their diameter before cutting into medicine. The recovery rates of each root parts per 100 kg were measured as $32.3{\pm}2.5$, $9.0{\pm}1.0$, $39.3{\pm}2.1$ and $15.0{\pm}1.0%$, respectively and the mean diameters were measured as $51.95{\pm}4.55$, $7.05{\pm}0.89$, $2.88{\pm}0.49$ and $1.57{\pm}0.32mm$. Two index components, D and DA, were analyzed. The change of both D and DA content showed a similar tendency. Both D and DA content were increased as the root diameter decreased (higher in TnR). In addition, antioxidant activity was higher in B and TnR, and lower in TkR and MR than control. This study showed that the thinner the root diameter, the higher the D and DA content in AGN roots and that TnR has excellent antioxidant activity compared to other root parts, suggesting that the thinner root part of AGN could be used as a potential material.

  • Objectives: This analysis of prescription used for low back pain in the Yomun(腰門) chapter of ${\ll}$Donguibogam(東醫寶鑑); The Precious Mirror of Oriental Medicine${\gg}$ is designed to be helpful to practical use of clinics. Methods : Proscriptions used for low back pain in the Yomun(腰門) chapter of ${\ll}$Donguibogam(東醫寶鑑)${\gg}$ were classified and analyzed according to the frequency of proscriptions and the characteristics of each herbs in proscriptions(efficacy, used frequency, related organs etc.) Results and conclusions : After analysis, we obtained the following results : 1. The causes of low back pain are mainly eohyeol(瘀血), yangheo(陽虛), punghanseub(風寒濕). In care of low back pain, I suppose more efficiency that if Angelica gigas NAKAI(當歸) Cnidium officinale MAKINO(三芎) Prunus persica BATSCH(桃仁) is added when the cause is eohyeol(瘀血), or if Psoralea corylifolia L.(破古紙), Cinnamomum cassia PRESL(肉桂), Foeniculum vulare MILL(茴香), Eucommia ulmoides OLIV.(杜冲), Citrus unshiu MARKOVICH(陳皮) are added when the cause is yangheo(陽虛), or if Phellodendron amurense RUPR.(黃柏), Notopterygium incisum TING(羌活), Atractylodes Japonica KOIDZ.(蒼朮) are added when the cause is punghanseub(風寒濕).

Analysis of the Obesity-Related Research for Each Constituent Herb of Euiiin-tang (의이인탕 구성약물의 비만관련 연구 분석)

  • The purpose of this study is analyzed of exploratory research potential as anti-obesity agents of Euiiin-tang. Four Korean databases and 2 Korean Journals (Riss4U, KISS, OASIS, DBPIA, and Journal of Korean Rehabilitation Medicine, Journal of Korean Medicine of Obesity Research) were searched using search word 'individual herbs' and 'obesity', 'weight loss', 'fat', 'hypertension', 'hyperlipidemia', 'diabetes'. Clinical and Experimental Research published in the journal were analyzed, review research, studies of pharmacopunctures and studies of mixed herbal medicine were excluded. We collected 23 studies. Seven studies of Coicis Semen, 10 stdies of Ephedra Herba, 2 study of Angelica gigas Nakai, 3 studies Atractylodis Rhizoma Alba, 1 studies of Glycyrrhiza inflata Batal. Most studies were experiment researches which were composed of in vivo or in vitro, and clinical trial was 5 studies of Ephedra Herba. Main constituent herb, Coicis Semen, Ephedra Herba were thought to represent an anti-obesity effect. Through the result, we can assume to be likely effect of Euiiin-tang as obesity medicine.
